A new composite, AgPW@PDA@Nisin, with shell-core structure was successfully synthesized by a polydopamine (PDA) surfaced conjugated nisin (an antibacterial 34 amino acid polycyclic peptide) as shell and polyoxometalates (Ag 3 PW 12 O 40 = AgPW) as core. The composite was characterized by the zeta potential , scanning electron microscopy (SEM), transmission electron microscope (TEM), X-ray diffraction analysis (XRD), fourier transform infrared (FT-IR). The AgPW@PDA@Nisin showed flower hierarchical structure and potential antibacterial activity against S. aureus ATCC29213. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of it were 4 and 32 μg/mL. AgPW@PDA@Nisin nanoflowers-induced bacterial death bears the characteristic of cell morphology, membrane integrity and permeability changing, nucleotide leakage. It indicated that the AgPW@PDA@Nisin interfere with the cell membrane, resulting in antibacterial activity against S. aureus. The cytotoxicity of the nanoflowers was low on HDF-a (human dermal fibroblasts) cells. A new class of hybrid inorganic-organic nanoflowers based on polyoxometalates and nisin with enhanced antibacterial properties can be developed for food preservation.

In this study, we successfully synthesized the red-fluorescence gold nanoclusters (AuNCs@nisin) while employing nisin as the template, and AuNCs@nisin showed the uniform particle size of around 3.5 ± 0.5 nm, and the corresponding excitation and emission wavelengths were 493 nm and 644 nm, respectively. Meanwhile, AuNCs@nisin was mainly composed by the elements of C, N, O, Au as well as S, and it was equipped with the functional groups of hydroxyl and amino. Significantly, the fluorescence of AuNCs@nisin showed the obvious response to thiourea based on the interaction between the metal core of AuNCs@nisin and thiourea, thus leading to the formation of Au-S bond and affecting the fluorescence intensity of AuNCs@nisin. Consequently, we achieved to establishing the detection of thiourea by AuNCs@nisin, and the corresponding linear range between the fluorescence intensity of AuNCs@nisin and the concentration of thiourea was 2 mmol/L-4 mol/L with a detection limit of 0.74 μmol/L. Consequently, the prepared AuNCs@nisin exhibited the potential of serving as a fluorescent probe for determining thiourea.

Freshness deterioration of salmon during cold storage poses significant safety concerns, highlighting the need for real-time monitoring strategies. Herein, a novel starch-pectin film incorporating Nisin-A and curcumin nano-emulsion (SP-NC film) was developed for real-time monitoring of salmon freshness. FTIR, XRD, and molecular dynamics simulations confirmed strong molecular compatibility and stable hydrogen bonding among film components. The SP-NC film exhibited high mechanical strength, with a tensile strength of 4.51 MPa and elongation at break of 43.58 %, as well as outstanding barrier properties reflected by low water vapor permeability (6.82 × 10 −10 g·mm −1 ·s −1 ·Pa −1 ) and oxygen permeability (6.10 × 10 −9 g·mm·mm −2 ·s −1 ). Under blue light, the SP-NC film reduced S. aureus and Salmonella survival rates to 1.85 % and 3.79 %, respectively. When applied to 4 °C refrigerated salmon, the SP-NC film extended shelf life from 4 to 8 days and provided visual spoilage indication. Overall, this study offers a valuable reference for the design of intelligent food packaging.

Nanocellulose has attracted extensive attention in the preparation of thermal insulation materials because of its high Young's modulus, high strength and low thermal expansion coefficient. In this study, nanocellulose microgels prepared by self-assembly of 2,2,6,6-Tetramethylpiperidoxyl oxidized nanocellulose (TOCNC) and Nisin were used as particle stabilizers, and acrylated epoxidized soybean oil (AESO) was used as oil phase to prepare Pickering emulsion, which was cured by heating to obtain biomass-based Pickering thermal insulation foam. Pickering foams prepared based on microgels with different Nisin contents have significant differences in thermal insulation performance. The results show that the addition of microgel particles can improve the thermal stability, increase the roughness of pore wall and reduce the thermal conductivity of AESO polymer foam. Among them, the Pickering foam prepared by microgel containing 0.06 wt% Nisin has the best thermal insulation performance and rougher pore wall surface, and its thermal conductivity is 0.040 W/(m·K), which is obviously lower than that of the foam prepared by TOCNC (0.083 W/(m·K)). Based on the structural design of nanocellulose microgel particles, this work innovatively realized the assembly of microgels at the oil-water interface and the regulation of thermal insulation performance through Pickering technology, and developed a biomass-based foam with improved thermal insulation effect, which provides a new idea for expanding the advanced application of nanocellulose in the field of thermal insulation materials.

The global waste rate of fresh meat products is approximately 20 – 30%, with the vast majority occurring in the consumer and retailer sectors. The use of antimicrobial preservative films can reduce fresh retail meat waste. In this study, we propose an environmentally friendly antimicrobial preservative film prepared using the residue of Pouteria campechiana pericarp as a substrate, sodium alginate as the film-forming agent, and nisin as the antimicrobial agent. This film was used to preserve duck breast meat. Nisin in the film slows the spoilage of duck breast meat by inhibiting and killing microorganisms, thereby extending its four-day shelf life. It is important to note that mouse-feeding experiments on the film have proven that it is safe and edible. This strategy provides ideas for the reuse of agricultural waste, and this edible antimicrobial film has great potential for the preservation of fresh meat.

Heat-resistant spore-forming bacteria are a primary cause of spoilage in ready-to-eat (RTE) foods, and effective control of spore contamination is critical for food preservation. Nisin, a GRAS antimicrobial peptide, exhibits potent activity against spore-forming bacteria, but its stage-specific effects during spore germination and outgrowth remain unclear. Heat-resistant Bacillus species were identified as the dominant spoilage bacteria in spoiled cooked beef, and B. cereus was selected to evaluate the inhibitory effects of nisin via phenotypic and transcriptomic assays. The minimum inhibitory concentration (MIC) of nisin against B. cereus spores was 400 µg/mL. Nisin slightly promoted spore germination, but significantly inhibited subsequent outgrowth of germinated spores. Time‑kill assays showed that 1 MIC nisin reduced viable counts to 3.09 log CFU/mL at 6 h compared with 8.05 log CFU/mL in controls; 2 MIC treatment for 2 h achieved a 0.65‑log reduction for intact spores but a 1.49‑log reduction for germinated spores. Nisin disrupted membrane permeability and integrity, triggered intracellular oxidative stress, leading to genomic DNA damage and reduced ATP synthesis. Transcriptomic analysis revealed 3049 differentially expressed genes, with disturbed energy metabolism and amino acid biosynthesis pathways. In cooked beef, nisin effectively inhibited B. cereus outgrowth and delayed product spoilage during storage. This study provides practical data support for nisin application to control spore-forming bacteria and extend the shelf life of RTE meat products.

Nisin is food preservative with good food safety and is widely used around the world. However, nisin degrades during food processing and storage, seriously affecting its antibacterial function. In order to improve the stability of nisin, nisin microcapsules were prepared by complex embedding nisin with pectin and chitosan. By selecting the mass ratio of pectin to nisin and chitosan, the microcapsules of nisin loading 200 mg with 81.15% encapsulation efficiency were prepared in aqueous solution of pH 4.0. The results of zeta potential of pectin, chitosan, nisin and microcapsules, infrared spectra and X-Ray diffraction spectra confirmed the embedding of nisin by pectin-chitosan. The adsorption isotherm of pectin and chitosan showed that chitosan and pectin mainly adsorbed by monolayer to form microcapsules. Compared with the same amount of free nisin, bactericidal efficiency decreased within 24 h, which showed there is a release process in microcapsules. However, after treated with 121 °C for 10 min, the sterilization activity loss of nisin was significantly lower than that of free nisin, which showed the thermal stability of nisin pectin microcapsules was greatly increased. The nisin microcapsules with good thermal stability and sustained-release effect can be better applied in future.

Hydrogels composed of biomaterials are promising drug delivery systems. In this study, we prepared an antibacterial hydrogel by incorporating the antimicrobial peptide nisin into bagasse cellulose nanofibrils, followed by mechanic shearing to obtain hydrogel microparticles as antimicrobial coatings for paper packaging. The concept of combining nisin with a hydrogel network helps to reduce the nisin depletion disadvantage in a nisin alone coating while achieving a prolonged antibacterial efficacy in application. First, antimicrobial hydrogels were constructed by electrostatic attraction between TEMPO-oxidized cellulose nanofibrils (TOCNF) and nisin. The rheological analysis confirmed that the hydrogel with a TOCNF/nisin ratio of 3:1 formed a good gel network accompanied by a high nisin loading efficiency. The obtained hydrogel exhibited controlled release of nisin in response to pH, with a slower rate at pH 7 than that of pH 5 and pH 9. Then, the obtained hydrogel has been processed to microparticles to facilitate its application as coating on paper packaging. Highly stable TOCNFs/nisin hydrogel microparticles were obtained under a water/hydrogel ratio of 4:1 and a mechanical shearing rate of 8000 rpm. The paper with hydrogel microparticles coating displayed an enhanced tensile strength by 14.67% compared to uncoated paper. As a packaging for cheese in storage at 4 °C for 7 days, the hydrogel microparticle coating paper can significantly reduce the growth of Listeria monocytogenes on the surface of cheese and show a sustained inhibition advantage over a nisin alone coating. The utilization of hydrogel microparticles as the coating slurry provides a versatile approach for paper treatment and endows the paper with a sustained release characteristic in actively preventing the bacterial proliferation on food surfaces during storage. The finding from this work is expected to broaden the application of nanocellulose and nisin into the field of food active packaging.

Bio-based nanomaterials with antimicrobial functions hold promise in replacing petroleum-based packaging for food preservation. A nanocellulose-based hybrid film with antimicrobial properties was developed from sugarcane bagasse and nisin. Cellulose nanofibrils (CNFs) were prepared from sugarcane bagasse pulp by mechanical grinding, and mixed with nisin to prepare CNFs/nisin nanohybrid films. The concentration of nisin has a remarkable influence on the mechanical, light transmission, gas barrier, and antimicrobial properties of these films. CNFs/nisin hybrid films with 1920 mg/L nisin exhibit good light transmission, relatively high tensile strength , low oxygen permeability, and low water vapor transmission rates. This hybrid film was used as a liner of low-density polyethylene plastic packaging for ready-to-eat ham; it completely inhibited Listeria monocytogenes during 7 days of storage at 4 °C. Such novel CNFs/nisin nanohybrid films are expected to expand the application of bagasse nanocellulose in active packaging for food preservation.

Recently, molybdenum disulfide functionalized with poly-ethylene glycol (PEGylated MoS 2 ) has been widely used as a new drug delivery vehicle in biomedical field. However, the weak antibacterial activity of PEGylated MoS 2 limits its application as an antibacterial agent. In this work, a novel silkworm-like conjugate of nisin loaded PEGylated MoS 2 ( [email protected] MoS 2 ) was developed for antibacterial application. The morphology and structure of PEGylated MoS 2 were strongly dependent on the Mo/S molar ratio of precursors during the solvothermal process. The silkworm-like skeleton was well kept after loading with nisin. A high level of reactive oxygen species (ROS) induced by the conjugate was an important cause of bacteria death. Due to the different structure of cell membranes, the sharp edges could more easily puncture into Escherichia coli ( E. coli ) as compared with Staphylococcus aureus ( S. aureus ) and produced more intracellular ROS, which improved the antibacterial activity of nisin against E. coli . As a result, [email protected] MoS 2 displayed the antibacterial activity against both gram-positive and gram-negative bacteria. Furthermore, the toxicity of the conjugate was very low. Therefore, the target conjugate of [email protected] MoS 2 may have great potential application as an antibacterial agent.

Nisin, the only antimicrobial peptide that has GRAS (generally considered as safe) status from both WHO and FDA, has the potential to become an important natural antibiotic as bacteria become more resistant to traditional antibiotics. Many studies have focused on how modification of nisin affects antimicrobial activity. In this study, nisin and chitosan were crosslinked using genipin to modify the N-terminal of nisin. The antimicrobial experiments were carried out on Staphylococcus aureus ATCC 6538. The results showed that nisin still retained antimicrobial activity after modification. When the crosslinking ratio of nisin to chitosan increased to 200: 1, MIC decreased from 48 μg/ml to 40 μg/ml. Less chitosan increased local number density and antimicrobial activity of nisin. When the local number density was high, adjacent peptides could cooperatively penetrate into the lipid membrane. The results of this study may help to better understand the antimicrobial mechanism and provide suggestions for the further modification of nisin.

To obtain a safe and biodegradable material for food packaging, this study developed a facile and green process of anchoring nisin onto oxidized cellulose through a simple Schiff base reaction for long-term antimicrobial active food packaging. To the best of our knowledge, this is the first report of such a packaging. The grafting reaction between the amino groups of nisin and the aldehyde groups of oxidized cellulose films was successfully established to obtained a nisin anchored cellulose films (NC) membrane; the structure was characterized with Fourier transform infrared spectroscopy , scanning electron microscopy, and thermogravimetric analysis . The NCs membrane had an outstanding transparent property of above 90% transmittance through 300 nm to 800 nm; while, the barrier properties against oxygen remain excellent. The thermal stability of the NCs membrane remained excellent and enabled an easier degradation than the original cellulose membrane. The NCs membrane displayed good antimicrobial activity against Alicyclobacillus acidoterrestris DSM 3922T in a long-term way, thus demonstrating good potential as food packaging material. The obtained results are promising and confirm the development of NCs membranes as green materials for food packaging.
