Sodium p -perfluorous nonenoxybenzene sulfonate (OBS)—one of the main alternatives to perfluorooctane sulfonate—has been increasingly detected in both aquatic environments and human bodies. Therefore, the pathogenic risks of OBS exposure warrant attention, especially its central nervous system toxicity mechanism under long-term exposure. In this study, the effects and mechanisms of OBS on the zebrafish brain at 40 days post exposure were examined. The results demonstrated that at 3.2 μg/L, OBS had no significant effect on the zebrafish brain, but 32 μg/L OBS caused depression or poor social behavior in zebrafish and reduced both their memory and survival ability. These changes were accompanied by histological damage and cell apoptosis. Furthermore, OBS caused the accumulation of excessive reactive oxygen species in the fish brain, leading to oxidative stress and subsequently cell apoptosis. Moreover, an imbalance of both inflammatory factors (IL-6, IL-1β, IL-10, TNF-α, and NF-κB) and neurotransmitters (GABA and Glu) led to neuroinflammation . Additionally, 32 μg/L OBS induced decreases in mitochondrial membrane potential and Na + -K + -ATPase activity, leading to both mitochondrial structural damage and the emergence of mitochondrial autophagosomes , partly explaining the neurotoxicity of OBS. These results help to analyze the target sites and molecular mechanisms of OBS neurotoxicity and provide a basis for the scientific evaluation of its health risks to humans.

Concerns regarding the possible health and environmental hazards of sodium p -perfluorous nonenoxybenzene sulfonate (OBS), a new substitute for perfluorooctane sulfonate, have been raised by its detection in a range of environmental matrices, animals, and people. Despite its widespread presence, research on the toxic effects and toxicokinetics of OBS remains limited. Therefore, in this study, we aimed to provide the first detailed investigation into how tissue distribution, bioaccumulation, and OBS exposure cause oxidative damage in adult zebrafish. Zebrafish were exposed to OBS at doses of 0, 10, and 100 μg/L for 28 days, following a 28-day depuration phase. Our findings indicate that OBS accumulated rapidly in zebrafish, with log kinetic bioconcentration factor (logBCF k ) values ranging from 2.48 to 2.70 and slow elimination rates ( k e = 0.0490–0.0643 d −1 ). The blood, liver, and muscle were responsible for over 50 % of the total body burden of OBS, with tissue concentrations decreasing in the following order: blood > liver > intestine > gills > gonad > brain > muscle, suggesting a tissue-specific accumulation pattern. Molecular docking analysis showed that OBS exhibited binding energies of −2.71 and −3.82 kcal/mol with organic anion transporter 1 and human serum albumin, respectively. Additionally, chronic exposure to OBS resulted in significant liver damage in zebrafish, including ballooning, cytoplasmic vacuolisation, and oxidative stress. This study provides valuable experimental data on the toxicokinetics and in vivo toxicology of OBS in zebrafish, offering important insights for environmental risk assessments.

Perfluorooctane sulfonate (PFOS) is a representative persistent organic pollutant that exerts toxic effects on aquatic organisms. As an alternative to PFOS, sodiump-perfluorous nonenoxybenzene sulfonate (OBS) has been frequently detected in aquatic environments and human tissues in recent years. However, its toxic effects on aquatic organisms and potential health risks to humans remain unclear. Zebrafish embryos are transparent and amenable to in vivo manipulation and observation. Therefore, in the present study, we investigated its developmental toxicity in zebrafish embryos, with PFOS as the positive control. We exposed zebrafish embryos to different concentrations of OBS (15, 20, and 25 mg/L) and PFOS (15 mg/L) for 2–168 h post fertilization (hpf) and then examined physiological and gene expression changes. At 24 hpf, spontaneous twitches in the 25 mg/L OBS group decreased to (5 ± 0.34)/min. By 48 hpf, the 20 mg/L OBS group’s hatching rate was (47.78 ± 2.22)%, significantly lower than the control. At 72 hpf, heart rates in both the PFOS and OBS groups were elevated, at 82 ± 0.6, 84.5 ± 0.5, 89.4 ± 0.3, and 93.7 ± 0.4, respectively. Similarly to PFOS, OBS induced developmental toxicity in zebrafish embryos. In addition, both OBS and PFOS exposure downregulated the expression level of anti-apoptotic Bcl-2 in zebrafish embryos, with a notable 0.53-fold decrease observed in the 25 mg/L OBS group. Conversely, they upregulated the expression levels of pro-apoptotic Bax, Caspase-3, and Caspase-9, with Caspase-3 expression increasing 1.14-, 1.5-, and 1.7-fold in the 15 mg/L PFOS, 20 mg/L OBS, and 25 mg/L OBS groups, respectively. These OBS- and PFOS-induced changes in gene expression increased apoptosis, suggesting that OBS can induce developmental toxicity in zebrafish embryos, and that its effect is comparable to that of PFOS. Therefore, considering its aquatic toxicity, measures aimed at limiting or remediating OBS pollution in the environment are necessary.

Fluorine-containing wastewater seriously threatens human health due to the wide application of sodium p -perfluorous nonenoxybenzenesulfonate (OBS). Herein, a novel Fe-Co bimetallic biochar (Fe-Co@BC) with abundant pore structure, functional groups and quickly electron transfer capability was prepared using hydrothermal method. Meanwhile, Fe-Co@BC-Ⅰ exhibited a superior catalytic capacity for degradation of OBS in PMS system, and the removal efficiency of OBS reached to 94.1 % within 60 min. After three cycles, the removal efficiency of OBS still up to 80 % and the Fe-Co had almost no leaching (Fe: 0.006 mg/L; Co: 0.023 mg/L). Further mechanistic insights were verified that high catalytic capacity and stability ascribed to the synergistic effects of Fe and Co, where Fe-Co mutual interaction promoted the formation of Fe 2+ and Co 2+ and accelerated electron transfer of catalyst. In addition, the quenching experiments and electron paramagnetic resonance (EPR) proved that the degradation of OBS can be mainly ascribed to indirect oxidation of SO 4 −• , O 2 −• and •OH, and the possible degradation pathway of OBS was proposed in the Fe-Co@BC/PMS system. This study provides an insight into bimetallic prepared and activation PMS degradation fluorinated wastewater.

Sodium p -perfluorous nonenoxybenzene sulfonate (OBS), a novel alternative to perfluorooctanesulfonate (PFOS), is widely used in industry as a surfactant, firefighting foam and photographic material. The occurrence of OBS in the aquatic environment has been recently reported, but little information is available on its accumulation and toxic effects in aquatic organisms. In this study, zebrafish larvae (3 d post-fertilization) were subjected to OBS (10, 100 μg/L) and PFOS (10 μg/L) for a period of 48 h, followed by a 24 h of depuration period. The bioconcentration and depuration kinetics, oxidative stress and possible molecular mechanisms of OBS and PFOS were investigated in zebrafish larvae. Our results showed that the uptake and depuration of both OBS and PFOS fitted well with a first-order kinetic model. The uptake rate constant of OBS was similar to that of PFOS, but the depuration rate constant was much higher than PFOS with a half-life of 69.7–85 h for OBS and 222.2 h for PFOS. The calculated BCFs of OBS and PFOS were 238.0–242.5 and 644.2, respectively. In our acute toxicity assay, the enhanced expression of Nrf2 protein accompanied by the upregulation of CAT and SOD protein expressions indicated OBS and PFOS induced oxidative stress in zebrafish larvae, and the Nrf2-ARE signaling pathway was involved in this process. Collectively, OBS has a lower bioconcentration potential than PFOS, but its toxic effect on oxidative stress was comparable to PFOS in zebrafish larvae.

Sodium p-perfluorous nonenoxybenzenesulfonate (OBS) has been proposed as a substitute for perfluorooctanesulfonic acid (PFOS), yet it has garnered increasing attention due to its environmental persistence and potential toxicity. Despite these concerns, the neurotoxic mechanisms of OBS remain unclear. This study investigates and compares the neurotoxic effects and mechanisms of OBS and PFOS inCaenorhabditis elegans. L4-stage worms were exposed to OBS (0.1–100 μM) or PFOS (100 μM) for 24 h. Neurobehavioral analysis showed that OBS exposure induced concentration-dependent neurobehavioral deficits, with 100 μM OBS significantly reducing pharyngeal pumping rate (29.8%), head swing frequency (23.4%), and body bending frequency (46.6%), surpassing the effects of PFOS. Both compounds decreased the fluorescence intensity of dopaminergic, glutamatergic, and γ-aminobutyric acid neurons and downregulated neurotransmitter-associated genes. They also increased ROS generation and inhibited antioxidant gene expression. Molecular docking revealed that OBS had a stronger binding affinity to p38 MAPK key protein (PMK-1) than PFOS. OBS and PFOS upregulatedpmk-1andskn-1, modulating oxidative stress and neuronal function.pmk-1mutation differentially affected OBS-induced neurobehavioral changes and gene expression alterations. Our findings indicate that OBS exhibits stronger neurotoxicity than PFOS inCaenorhabditis elegans, mediated through the PMK-1 pathway. These results highlight the need for further investigation into the safety of OBS as a PFOS alternative.

As emerging alternatives to perfluorooctane sulfonate (PFOS), 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (6:2 Cl-PFESA) and sodium p -perfluorous nonenox-benzenesulfonate (OBS) were frequently detected in the four freshwater fish species collected from Poyang Lake. Median concentrations of 6:2 Cl-PFESA and OBS in fish tissues were 0.046–6.0 and 0.46–5.1 ng/g wet weight, respectively. The highest concentrations of 6:2 Cl-PFESA was found in fish livers, whereas OBS was found in the pancreas, brain, gonads, and skin. The tissue distribution pattern of 6:2 Cl-PFESA is similar to that of PFOS. The tissue/liver ratios of OBS were higher than those of PFOS, suggesting that OBS has a greater tendency to transfer from the liver to other tissues. The logarithmic bioaccumulation factors (log BAFs) of 6:2 Cl-PFESA in three carnivorous fish species were greater than 3.7, whereas those of OBS were less than 3.7, indicating that 6:2 Cl-PFESA had a strong bioaccumulation potential. Notably, sex- and tissue-specific bioaccumulation of OBS has also been observed in catfish. Most tissues (except the gonads) exhibited higher OBS concentrations in males than in females. However, no differences were found for 6:2 Cl-PFESA and PFOS. Maternal transfer efficiency of OBS was higher than that of 6:2 Cl-PFESA and PFOS in catfish ( p < 0.05), indicating that OBS presents a higher risk of exposure to males and offspring through maternal offloading.

Selectivity is the key for adsorption or recovery of per- and polyfluoroalkyl substances (PFAS) from aqueous environments. In this study, the fluorinated metal–organic frameworks (MOFs) with different fluorine contents were prepared and investigated to evaluate the selective adsorption of an emerging PFAS, sodium p‑perfluorous nonenoxybenzenesulfonate (OBS). Adsorption of OBS increased with the increasing fluorine content of MOFs, and the best fluorinated MOFs showed an excellent selective adsorption capacity. The influence of various co-existing hydrocarbon substances on OBS adsorption was slight. The adsorption amount of OBS was 15-fold to that of the hydrocarbon surfactants (n-caprylic acid). The selectivity was attributed to the fluorinated structures of MOFs whose oil contact angle was up to 101°, and thus the oleophobicity of fluorinated structures made MOFs repel hydrocarbon compounds. Molecular dynamics simulations and density functional theory calculations indicated that the fluorinated structures were able to capture OBS molecules from the long distances (roughly 1–100 nm) via hydrophobic interaction, and then formed a F–F non-covalent interaction which was highly probable to be driven by a dispersion force to further attract OBS fluorinated tails within shorter distances (less than 3.5 Å). The combination of oleophobic and fluorophilic effects contribute to the exclusive selectivity of fluorinated MOFs for OBS molecule.

Sodium p -perfluorous nonenoxybenzene sulfonate (OBS), an economical alternative to perfluorooctane sulfonate (PFOS) in multiple industrial fields, is widely detected in the environment. The toxicity of OBS has received increasing attention. Pituitary cells are components of the endocrine system and act as vital regulators of homeostatic endocrine balance. However, the effects of OBS on pituitary cells remain unknown. The present study explores the effects of OBS (0.5, 5, and 50 μM) on GH3 rat pituitary cells after treatment for 24, 48, and 72 h. We found that OBS significantly inhibited cell proliferation in GH3 cells with remarkable senescent phenotypes, including enhanced SA-β-gal activity and expression of senescence-associated secretory phenotype (SASP)-related genes, cell cycle arrest, and upregulation of the senescence-related proteins γ-H2A.X and Bcl-2. OBS caused significant cell cycle arrest of GH3 cells at the G1-phase and concomitantly downregulated the expression of some key proteins for the G1/S transition, including cyclin D1 and cyclin E1. Consistently, the phosphorylation of retinoblastoma (RB), which plays a central role in regulating the cell cycle, was prominently reduced after OBS exposure. Furthermore, OBS notably activated the p53-p21 signalling pathway in GH3 cells, as evidenced by increased p53 and p21 expressions, enhanced p53 phosphorylation, and augmented p53 nuclear import. To our knowledge, this study is the first to reveal that OBS triggers senescence in pituitary cells via the p53-p21-RB signalling pathway. Our study demonstrates a novel toxic effect of OBS in vitro, and provides new perspectives for understanding the potential toxicity of OBS.

Per- and polyfluoroalkyl substances (PFASs) are toxic to microorganisms , thereby affecting microbial communities in sludge and soil, but how to repair the toxicity of microorganisms remains unclear. In this study, rutin, an antioxidant, was added into a culture medium with an aerobic denitrification bacteria, Pseudomonas stutzeri , under the exposure of sodium perfluorononyloxy-benzenesulfonate (OBS) to evaluate the repair mechanisms of rutin to the toxicity of OBS to the bacteria. The results showed that rutin could repair the damage of OBS to cell structures, and reduce the death rates of the bacteria under OBS exposure. The dosage of rutin reduced the effect on the inhibition of denitrification ability of P. stutzeri under OBS exposure. Compared with the bacteria exposed to single OBS, the dosage of rutin resulted in that the death rates recovered from 96.2% to 66.4%, the growth inhibition rate decreased from 46.5% to 15.8%, the total nitrogen removal rate recovered from 66.9% to 100%, and the NO 2 − content recovered from 34.5 mg/L to 0.22 mg/L. The expressions of key denitrification genes ( napA , nirS , norB , nosZ ) were recovered after adding rutin under OBS exposure. Rutin changed the positive rate of reactive oxygen species , the relative superoxide dismutase and catalase activities in the bacteria which exposed to OBS. The mechanism by which rutin repaired the toxicity of OBS to P. stutzeri related to inhibiting the activities of antioxidant and denitrification enzymes rather than affecting the expressions of genes involved in these enzymes. This study sheds light on the repair method of micro-organics and reveals the repair mechanisms under PFASs exposure.

In this study, the internal relationships among oil bodies (OBs), the protein–phospholipid interactions in aqueous phase, oil–water interface behavior, and the stability of reconstituted OBs were analyzed from the bulk phase, interface, and macro perspectives, and the stability mechanism of OBs was discussed. OB proteins and phospholipids were combined through hydrophobic and electrostatic interactions, resulting in the stretching of protein conformation. OB proteins and phospholipids act synergistically to increase interface pressure and the rate of increase in interface pressure with relatively stable elastic behavior, which is beneficial to the formation and stability of interfacial films. When OBs were reconstituted by an OB protein–phospholipid complex system, phospholipids bound to OB proteins through hydrophobic and electrostatic interactions. OB proteins and phospholipids uniformly covered the oil droplet surface of reconstituted OBs to form a stable interfacial film, which maintained the stability of OBs. The addition of phospholipids significantly reduced the particle size of OBs prepared by OB proteins in a dose-dependent manner, and particle size decreased with the increase in phospholipid content (p< 0.05). Phospholipids increased the net surface charge, enhanced electrostatic repulsion, and improved the physicochemical stability of reconstituted OBs. The stability mechanism elucidated in this study provides a theoretical basis for the demulsification of peanut OBs.Keywords:stability mechanism;peanut OBs;protein;phospholipid;interaction

Per/polyfluoroalkyl
substances (PFASs) have been confirmed to have
negative health effects in animal models. As a novel alternative to
legacy PFASs, sodium ρ-perfluorous nonenoxybenzenesulfonate
(OBS) raises growing concerns over environmental exposure and biological
toxicity. Herein, colitis models were constructed for exploring the
toxicity of OBS in intestinal homeostasis during the recovery period.
The results verified the gut toxicity of OBS, which increased serum
interleukin-1β/6/17α/22 (IL-1β, IL-6, IL-17α,
IL-22) levels, decreased colonic mucus secretion, and changed gut
microbiota. During colitis recovery, no statistically significant
difference was observed in colonic mucus secretion and related gene
expression in dextran sulfate sodium-OBS (DSS-OBS) compared with the
DSS; compared with the CON group, the unchanged mucus secretion and
gene expression in DSS-OBS ghad still a statistically significant
difference. It indicated that the intestinal barrier disruption did
not worsen after the OBS exposure, but the recovery might be delayed.
Moreover, OBS exposure significantly changed gut microbiota during
the recovery period, suggesting that disrupted gut microbiota might
be associated with the delay of OBS-mediated inhibition of intestinal
barrier repair. Additionally, network toxicology analysis identified
the action targets of OBS (including Nuclear Receptor Subfamily 1
Group I Member 2 (NR1I2), Solute Carrier Family 22 Member 5 (SLC22A5),
Glutathione S-Transferase Pi 1 (GSTP1), etc.), which mediate gut microbiota
disorder and intestinal barrier dysfunction, and serve as potential
biomarkers for OBS-induced intestinal-related diseases. Collectively,
OBS exhibits definitive gut toxicity. Although short-term OBS exposure
did not exacerbate the progression of colitis, it delayed the recovery
of intestinal function. Among, multiple target sites, coupled with
the gut microbiota, commonly mediate the gut toxicity of OBS. It suggested
that the potential health risks of OBS cannot be overlooked, and the
relevant departments should strengthen the management and develop
next-generation alternatives.
