A DNA tetrahedral scaffolds-based electrochemical biosensor for simultaneous detection of AFB1 and OTA

Wenting Li,Yongqiang Shi,Xinai Zhang,Xuetao Hu,Xiaowei Huang,Nini Liang,Tingting Shen,Xiaobo Zou,Jiyong Shi

Herein, a bifunctional electrochemical biosensor based on the DNA tetrahedral scaffolds (TDNs) was proposed, OTA@TDNs and AFB1@TDNs were adopted for electrochemical signal output in response to OTA and AFB1 concentration, simultaneously. In order to increase the conductivity of the biosensor, highly porous gold (HPG) was loaded on electrode surface by pulse electrodeposition. Under optimal conditions, the P Fc displayed a linear range with AFB1 concentration between 0.05 ∼ 360 ng·mL −1 with the LOD of 3.5 pg·mL −1 . And the P MB selective and sensitive responses to OTA are achieved with a linear range of 0.05 ∼ 420 ng·mL −1 and a LOD of 2.4 pg·mL −1 . This biosensor has high sensitivity, selectivity and stability for OTA and AFB1 detection in peanut samples. The approach streamlines the experimental procedure, leading to significantly improve the detection efficiency of mycotoxins. Collectively, this method suggest a novel approach for the detection and monitoring of OTA and AFB1 in food sample.

Preparation of an OTA aptasensor based on a metal–organic framework

De-lei Li,Xi Zhang,Yuchan Ma,Yan Deng,Rong Hu,Yunhui Yang

A novel aptasensor for ultrasensitive and rapid detection of ochratoxin A (OTA) was developed using a metal–organic framework (MOF) as the signal unit. In this work, a MOF was utilized as the signal probe. The signal probe differs from traditional probes; the signal of the cadmium ions (Cd2+) from the MOF can be directly detected without acid dissolution and preconcentration, greatly simplifying the detection steps and reducing the detection time. Moreover, the MOF contains large amounts of Cd2+ ions, which could provide high electrochemical signals. Our report is the first time using an MOF as the electrochemical signal probe for an aptasensor application. A gold nanoparticle (Au NP)-doped molybdenum disulfide (MoS2) nanocomposite was employed as an immobilized matrix for the sensor because of its high electronic conductivity. Under the optimized experimental conditions, an ultrasensitive aptasensor for the detection of OTA exhibited a wide linear range of 0.05–100 ng mL−1 with a low detection limit (LOD) of 10 pg mL−1 OTA (at 3σ). With these merits, this stable, simple, low-cost, sensitive and selective electrochemical aptasensor shows promise for applications in environmental monitoring.

Synergistic Effects of DNA Structure for Ultrasensitive Detecting OTA in Grains

Yuan Wei,Lu Lixia,Lu Yichen,Xiong Xiong,Li Yi,Cui Xiaowen,Liu Yuanjian,Xiong Xiaohui

Ochratoxin A (OTA) is considered as one of the most hazardous mycotoxins commonly occurring in grains. Thus, the monitoring of OTA is extremely significant. Herein, a synergistic strategy to detect OTA in grains based on the combination of DNA tetrahedron-structured aptamer (DTA) and hybridization chain reaction (HCR) was proposed. Briefly, DTA was assembled on the gold electrode. In the presence of OTA, the trigger DNA on the top of the DTA was exposed to unfold the biotin-modified hairpin structure DNA to HCR. It could conjugate plentiful streptavidin-labeled horseradish peroxidase (SA-HRP) to form polyaniline (PANI), which was regarded as electrochemical active molecule to detect OTA. In this way, the synergistic effect of DTA and HCR not only can provide stable probe structure to improve the recognition rate of OTA, but also can supply a large number of templates to synthesize PANI under mild conditions. As a result, the biosensor has a good analytical performance for OTA in a linear range of 0.05 to 50 ng mL −1 , with a detection limit of 0.017 ng mL −1 . The accuracy of sensor is similar with high-performance liquid chromatography (HPLC) analysis.

A controllable Y-shaped DNA structure assisted aptasensor for simultaneous detection of AFB1 and OTA based on ARGET ATRP

Mingsan Miao,Liang Guo,Jinyan Xue,Yuzhen Jia,Zhenzhen Cui,Huai-Xia Yang

The development of a simple, rapid, and sensitive technology for the simultaneous detection of mycotoxins is of great significance in ensuring the safety of foods and drugs. Herein, a fluorescence aptasensor with high sensitivity and reproducibility for the simultaneous detection of aflatoxin B1 (AFB1) and ochratoxin A (OTA) was developed. In this sensing system, AFB1 and OTA aptamers were co-immobilized on the surface of magnetic beads (MBs) to form a Y-shaped structure through the principle of complementary base pairing, which were used as recognition probes to specifically capture the target. Activators generated by electron transfer for atom transfer radical polymerization (ARGET ATRP) was used as signal amplification strategy to improve the sensitivity. The initiator modified at the end of the antibody initiates the ARGET ATRP reaction. Different fluorescence signals were designed to achieve simultaneous detection of OTA and AFB1 with the limit of 18.62 and 1.60 fg/mL for AFB1 and OTA, respectively. In addition, experiments were conducted on three types of samples, and the recoveries of the two mycotoxins ranged from 87.30% to 109.50%, with relative standard deviations ranging from 0.50% to 4.92% under reproducible conditions. The results suggest that the developed aptasensor is sufficient to meet the different regulatory requirements of the two mycotoxins in food and drug safety and shows great potential.

In this paper, a cost effective immersion method based on the modification of polyurethane sponge has been developed in the application field of oil/water separation. Octadecanethio and polydopamine were used to grafted onto PU sponge which was roughened by etching method. The morphology, composition and surface properties of prepared Ota-PDA-PU sponges were characterized by various techniques. The characterization results of the modified sponge exhibited rough surface structure, excellent flexibility, and possessed super-hydrophobic characteristic with a high water contact angle (WAC) of 155 ± 1°. The as-prepared sponge displayed high absorption selectivity for different organic solvents and oils, the absorption capacity for sesame oil attained 40 times of its own weight. Meanwhile, the absorption selectivity has no decrease after 10 cycles of absorption-desorption process. More importantly, the as-prepared sponge could be used as a filter for oil-water separation.

Development of a Nafion-MWCNTs and in-situ generated Au nanopopcorns dual-amplification electrochemical aptasensor for ultrasensitive detection of OTA

Yujiao Hou,Nan Long,Qingbin Xu,Ying Li,Pengyue Song,Minghan Yang,Jiabo Wang,Lidong Zhou,Ping Sheng,Weijun Kong

Trace detection of ochratoxin A (OTA) in foods is essential to mitigate risks to human health. Herein, a label-free electrochemical (EC) aptasensor based on dual-signal amplification of Nafion dispersed multi-walled carbon nanotubes (Nafion-MWCNTs) and Au nanopopcorns was developed for ultrasensitive detection of OTA. Nafion solution prevented the leaching of MWCNTs, and the Nafion-MWCNTs modified screen-printed carbon electrode (SPCE) acted as the sensing substrate which facilitated the uniform distribution of the electrodeposited Au nanopopcorns. The in-situ generated Au nanopopcorns could not only load a large amount of aptamers for specific identification of OTA, but also promote the electron transfer of the sensing platform. The incorporation of Nafion-MWCNTs and Au nanopopcorns realized dual-amplification of the aptasensor due to the enhanced conductivity and the increased electroactive surface area of the electrode. The modified electrodes were characterized through scanning electron microscopy, X-ray photoelectron spectroscopy and EC evaluation. Under optimal conditions, the electrochemical impedance spectroscopy (EIS) was measured for the determination of OTA. The as-fabricated Au nanopopcorns/Nafion-MWCNTs impedimetric aptasensor displayed excellent sensitivity with a detection limit as low as 1 pg/mL and a wide linear range of 1 pg/mL-10 ng/mL for OTA. Practical application of the aptasensor in the spiked malt samples achieved satisfactory recoveries of 89.82–95.65 %, which was also successfully verified to detect OTA in eleven batches of actual malt samples collected from the local market. The creative aptasensor is simple, cost-effective, sensitive, and accurate, showing great promise for on-site monitoring of other trace contaminants in foods by simply replacing the aptamers.

Meilin Wei,Solairaj Dhanasekaran,Esa Abiso Godana,Qiya Yang,Yuan Sui,Xiaoyun Zhang,Guillaume Legrand Ngolong Ngea,Hongyin Zhang

Cryptococcus podzolicus Y3 is an antagonistic yeast that can control postharvest diseases of crops and mycotoxins , which is potentially valuable for ensuring food safety. Our previous study showed that C. podzolicus Y3 can efficiently degrade ochratoxin A (OTA), one of the harmful mycotoxins in the environment and food. However, the genomic information of C. podzolicus Y3 and its protein expression profile during OTA degradation are not available. In this study, whole-genome sequencing of C. podzolicus Y3 was analyzed. Protein expression during OTA degradation was investigated by the data-independent acquisition (DIA) technique. The genome size of C. podzolicus Y3 was about 29,181,249 bp length with 58.79% GC content and identified with 17 scaffolds, 9090 coding regions and 127 non-coding regions. The DIA identified 4315 proteins during OTA degradation, among which 101 proteins were significantly up-regulated and 116 were down-regulated. The differentially expressed proteins were mainly involved in reduced glutathione metabolism, phosphatidylinositol signaling system and mitogen-activated protein kinase signaling pathway, calcium ion homeostasis, endoplasmic reticulum protein processing and fatty acid metabolism. Our results provide the genome information of C. podzolicus Y3 for further functional researches. In addition, this study has revealed the toxicity and stress response mechanism of C. podzolicus Y3 during OTA degradation at the protein level.

Cascade amplifying aptasensor for positively correlated detecting OTA: Based on DNase I-assisted cyclic enzyme digestion and AgNPs@gel-enhanced fluorescence

Chenfei Han,Yu Xiao,Zhenjiang Liu,Daolin Du,Ming Li

To improve the performance of aptasensor, the coupling of novel material and strategy had been the hottest research with the promising prospects. In this study, a cascade amplifying aptasensor (CA-aptasensor), which had coupled the DNase I-assisted cyclic enzyme digestion and AgNPs@gel-enhanced fluorescence, had reflected the desired performance for positively correlated detecting OTA . For this purpose, the GO@PVP was used to adsorb and quench the free FAM-Apt, while the complex of OTA and FAM-Apt was digested by the DNase I. The released OTA allowed the free FAM-Apt to separate from the GO@PVP cyclically, thus the OTA level was amplified preliminarily. The AgNPs@gel could combine with the released FAM to form the metal enhanced fluorescence (MEF) effect, resulting in the cascade signal amplification. The OTA was detected at pg mL −1 level with an ultrasensitive LOD of 3.33 pg mL −1 and IC 50 of 13.6 pg mL −1 , which improved 90-fold and 728-fold than that of ELASA for same aptamer. The CA-aptasensor indicated high specificity, reliability and practicability. Meanwhile, the CA-aptasensor exhibited more highlights, including the efficient homogeneous system, low background signal and standardized aptamer recognizer. This study might provide a new approach for improving the performance of aptasensor, which could also guide the signaling cascade and positively correlated detection for other risk factors.

Rapid Visual Screening of OTA Based on Multicolor Electrochemiluminescence

Tao Qin,Tang Ning,OuYang Songtao,Jiang Yanjun,Luo Yun,Liu Yuanjian,Xiong Xiaohui

A multicolor electrochemiluminescence (ECL) apparatus based on a closed bipolar electrode (BPE) system was proposed for the visual testing of Ochratoxin A (OTA). Since the oxidation potentials of the accompanying luminophores ([Ru(bpy) 3 ] 2+ and [Ir(ppy) 3 ]) are different, a bidirectional color change from dark orange to yellow to forest green can be discovered at the anode of BPE by adjusting the current of the BPE. The presence of OTA was reflected by target-induced elimination of the Fc-aptamer from the cathode pole, leading to a reduction in the current of the BPE and a color change on the anode electrode. A low detection limit of 10 ng mL −1 of OTA could be identified with naked eyes. Combining sensitivity of electrochemistry with the intuitiveness of optical reading, the device holds great potential in the rapid food safety testing. Graphical Abstract

CRISPR/Cas12a-powered CLASA towards OTA ultrasensitive detection in cereal samples

Si Chen,Rong Fang,Yi Li,Fei Deng,Xing Liu,Danting Yang

Ochratoxin A (OTA) is classified as a Class 2B carcinogens and poses a significant risk bto the liver, kidney and other organs, potentially leading to carcinogenic and teratogenic effects. Accurate monitoring of OTA content in food is crucial for safeguarding human and animal health. Here, a novel approach called the C RISPR/Cas12a enzyme- l inked a ptamer- s orbent a ssay (CLASA) was developed, which integrates the high-performance trans -cleavage ability of CRISPR/Cas12a with the cost-effective and high-affinity properties of aptamer for ultra-sensitive OTA detection. In this strategy, we incubated aptamers on microwell plates, and used the fluorescence intensity changes caused by OTA and crRNA competitively bind OTA-specific aptamer to achieve the quantitative detection. Our sensitive CLASA method not only solves the problems of poor repeatability and relatively low sensitivity of fluorescence based-CRISPR/Cas12a reaction in solution, but also solves the defects of expensive antibody and false positive result in traditional ELISA method, achieving a limit of detection (LOD) as low as 171 fg·mL −1 with a broad linear range from 1 pg·mL −1 to 1000 pg·mL −1 . The presented CLASA approach was validated successfully by using commercial ELISA kit on rice and oat samples as well. Furthermore, by strategically modifying the sequences of aptamer and crRNA, our CLASA platform can be flexibly customized to detect a wide range of small molecular targets. This adaptability allows for the extension of our framework beyond nucleic acid substances, enabling the detection of non-nucleic acid compounds using the CRISPR-Cas12a system. As a result, our CLASA approach holds great promise as a versatile sensing kit with applications in various fields requiring sensitive and specific detection of diverse analytes.

Dual SERS-enhanced core-shell-satellite assemblies with Cu-doped ZIF-8 interlayer for OTA detection

Ying Yang,Wenqi Zhang,Lu Zhang,Ziyan Shi,Dexiang Wei,Weiwei Xia,Xiaoguang Xu

Ochratoxin A (OTA) is a highly toxic and bioaccumulative mycotoxin found in grains, threatening food safety. Surface-enhanced Raman spectroscopy (SERS) combined with immunochromatography enables rapid and sensitive detection of OTA. This study aimed to develop a novel core-shell-satellite SERS probe (Au@Ag@(Cu-ZIF-8)-AuNPs) for OTA detection. The structure comprised an Au@Ag nanoparticle core, coated with Cu-doped ZIF-8 and gold nanosatellites. This design generated dense electromagnetic “hot spots” through plasmonic coupling between the core and satellites. Simultaneously, Cu doping reduced the bandgap of ZIF-8, thus enhancing charge transfer efficiency for chemical signal amplification. The dual electromagnetic-chemical enhancement achieved a detection range of 0.1 fg/mL–1.0 μg/mL and an ultralow detection limit of 0.057 fg/mL, thereby surpassing the detection limits of conventional methods. This innovative probe, with its high sensitivity and wide dynamic range, offers a powerful platform for mycotoxin monitoring, hence advancing SERS-based rapid detection technologies crucial for food safety.

A fluorescent aptasensor for highly sensitive detection of OTA and AFB1 and constructing their logical operations

Qiu-Long Zhang,Xue Yang,Xuan-Ting Lin,Sha-Sha Chen,Zhu,Guang-Xiang Luan,Kun-Quan Chen,Zhi Zeng,Jian-Min Chen

In this study, a universal fluorescent aptasensor was developed for the sensitive detection of ochratoxin A (OTA) and aflatoxin B1 (AFB1). The design integrated a non-competitive transduction platform with highly efficient RNA-cleaving DNAzymes, forming the core of the sensing system. When OTA or AFB1 is present in the sample, the target mycotoxin specifically recognizes and binds to its corresponding aptamer. The strand displacement reaction is significantly hastened by this binding event, and in response, the DNAzyme is released as a consequence. The activated DNAzyme then triggers an amplified fluorescent signal, enabling quantitative detection of the target. The proposed aptasensor displayed sensitivity with a detection limit of 0.26 nM for AFB1 and 0.17 nM for OTA respectively. Beyond single-target detection, the platform also enabled simple and flexible implementation of logical operations between the two mycotoxins, along with the design of orthogonal experiments for multi-target analysis. Furthermore, specificity tests confirmed that other co-existing mycotoxins had negligible interference on the detection of AFB1 or OTA, verifying the sensor's high selectivity. To assess practical applicability, the aptasensor was utilized for the detection of AFB1 and OTA in actual samples, yielding recoveries of 93 %–101 % in milk and 101 %–107 % in corn flour, demonstrating the sensor's reliability in complex matrices. Collectively, this developed fluorescent aptasensor features portability, simplicity, cost-effectiveness, and broad adaptability, making it a promising alternative tool for detecting various mycotoxins in food safety monitoring and related fields.
