The Influence of Triphenyltin Exposure on the Osmoregulatory Capacity of Marine Medaka (Oryzias melastigma) at Different Salinities

Teng-Zhou Li,Cheng-Zhuang Chen,Shao-Ying Xing,Ling Liu,Ping Li,Zhi-Hua Li

Triphenyltin (TPT) is an organotin pollutant widely found in the aquatic environment. It has endocrine-disrupting and osmotic pressure toxicity. In this study, the physiological and biochemical effects of TPT and various salinities were investigated in different tissues (gut, gill, and brain) of marine medaka. The exposure experiments were conducted for 42 days in different salinities (0, 15, and 30 ppt) without TPT exposure and in different salinity groups with TPT exposure concentrations of 100 ng/L, respectively. The results showed that the Na+-K+-ATPase (NKA) and Ca2+ATPase activity had significant tissue-specific differences, with the highest activity observed in the gills, indicating their major contribution to osmoregulation. Changes in salinity also resulted in significant alterations in the ion concentrations of the gut and gill tissues in the 0-C and 15-C groups. While the changes in Na+and Cl−were relatively stable, the presence of TPT disrupted the regulation of Ca2+and K+. In conclusion, substantial variations were observed in the osmoregulatory capacity of marine medaka tissues. Environmental concentrations of TPT had little effect on osmotic enzyme activity but interfered with the regulation of Ca2+and K+concentrations in the tissues. This study provides valuable insights into the osmotic toxicity of TPT in aquatic environments with different salinities.

The effect of tris (1,3-dichloro-2-propyl) phosphate on the early embryonic heart development of Oryzias melastigma

Sujing Wang,Na Zheng,Qirui An,Xiaoqian Li,Yining Ji,Yunyang Li,Changcheng Chen,Zhifei Xiu

The flame retardant tri (1, 3-dichloro-2-propyl) phosphate (TDCIPP) is widely present in environmental media and organisms. People have paid much attention to the growth and developmental toxicity of TDCIPP, but there is little information about its cardiotoxicity and potential mechanisms. In this study, marine medaka ( Oryzias melastigma ) embryos were exposed to TDCIPP solutions (0, 0.05, 0.5, 5, and 50 μg/L) for 21 days to investigate the adverse effects of TDCIPP on cardiac development. The results showed that TDCIPP exposure altered the heart rate at different stages of embryonic development. In addition, 50 μg/L TDCIPP resulted in increased sinus venosus (SV)–bulbus arteriosus (BA) distance, pericardial cysts, and cardiac linearization in newly hatched fish. During embryonic development, the expression level of key genes regulating cardiac development is disturbed. The early stage of cardiac development is the sensitive window period for the toxic effects of TDCIPP. Oxidative stress was observed in newly hatched juveniles, but no significant lipid peroxidation damage was observed. In addition, vitellogenin (VTG) levels in juvenile fish were significantly reduced. Our results show that TDCIPP exposure induces cardiotoxicity in marine medaka embryos, which is induced in the early stages and promotes heart defects by amplifying inflammatory responses at a later stage.

Ultrasensitive label-free electrochemical immunosensors for detecting marine medaka (Oryzias melastigma) vitellogenin based on novel Cu2O–BSA nanoparticles and anti-lipovitellin monoclonal antibody

Zhenzhong Zhang,Shaoguo Ru,Jun Wang,Dong Wang,Lingchao Zhao,Hayan Teng,Zhikuang Dai,Wenzhe Zhang,Ailing Yang

Vitellogenin (Vtg) is a biomarker for environmental estrogens, and its sensitive detection method is vital for the detection of weak estrogenic activity. This study aimed to develop sensitive electrochemical immunosensors for marine medaka ( Oryzias melastigma ) Vtg based on novel nanomaterials. Firstly, Cu 2 O–BSA nanoparticles (NPs) with a diameter of 120−150 nm was synthesized, and monoclonal antibody against lipovitellin (anti-Lv mAb) was prepared. Then two simple and efficient label-free immunosensors for the detection of Vtg were established using Cu 2 O–BSA NPs and gold nanoparticle (Au NPs) as signal amplification elements and anti-Lv mAb as the detection probe. These two immunosensors have good sensitivity, selectivity, reproducibility for Vtg quantification, and are easy to use. Moreover, the BSA/Anti-Lv mAb/Au/Cu 2 O–BSA/GCE immunosensor exhibited a linear range from 0.128 to 5 × 10 4 pg/mL, with the lowest detection limit (LOD) of 0.09 pg/mL, which is considerably lower than those of the previously reported Vtg detection techniques. Finally, the performance of the immunosensor was evaluated by quantifying Vtg concentrations in plasma of male marine medaka exposed three estrogenic bisphenols. The results demonstrated that the BSA/Anti-Lv mAb/Au/Cu 2 O–BSA/GCE immunosensor was an easily-operated, ultrasensitive and accurate detection method for weak estrogenic activity.

Conversion of Retinoids along the Marine Food Chain Contributes to Adverse Impacts on the Spine, Liver, and Intestinal Health of the Marine Medaka (Oryzias melastigma)

Haisu Liu,Lei Xie,Yang Xiao,Ruiwei Ran,Yuhang Fang,Baoling Yang,Liying Tan,Juanchan Xu,Songhui Lu,Yuelei Dong,Lei Cui

Marine microalgae serve as an aquaculture bait. To enhance algal cell growth and breeding profits, high-intensity light conditions are standard for cultivating bait microalgae, potentially altering microalgal metabolite production. This research revealed that Thalassiosira pseudonana, when subjected to high-intensity light conditions, accumulated significant quantities of retinal (RAL) that transferred through the food chain and transformed into all-trans retinoic acid (atRA) in marine medaka. The study further explored the toxic effects on individual fish and specific tissues, as well as the mechanisms behind this toxicity. The accumulation of atRA in the liver, intestine, and spinal column resulted in structural damage and tissue inflammation, as well as oxidative stress. It also down-regulated the gene transcription levels of key pathways involved in immune function and growth. Furthermore, it disrupted the homeostasis of the intestinal microbial communities. The implications for wildlife and human health, which are influenced by the regulation of microalgal metabolite accumulation and their transfer via the food chain, require further investigation and could hold broader significance.

Early-life exposure to semicarbazide induced neurodevelopmental toxicity associated with neurotransmitter in Oryzias melastigma

Qiaoyue Xu,Willy Baeyens,Chunqing Liu,Davie Cappoen,Myriam Hsu,Yue Gao,Deqi Xiong

Semicarbazide (SEM) is a hydrazine-derived compound that enters the environment from various sources and exhibits complex toxicological mechanisms. There is growing concern about pollutant-induced toxicity during early development, as well as the long-term effects. In this study, Oryzias melatigma embryos were exposed to 10, 50, and 100 μg/L SEM for 14 days then reared in clean water for an additional 90 days until adulthood. Transcriptomic analysis showed that differentially expressed genes were primarily enriched in neurotransmitters and lipid metabolism-related pathways. Specifically, several neurotransmitter system-related gene expressions were perturbed, accompanied by reduced gamma-aminobutyric acid (GABA) and dopamine levels, as well as decreased glutamate decarboxylase (GAD) and acetylcholinesterase (AChE) activity. Altered gene expressions associated with lipid-related pathways suggest a possible link with the lipid metabolism and a potential role of SEM-induced neurodevelopmental effects. Notably, SEM residues were still detected in the liver and brain of adult in the 100 μg/L group even after the recovery periods. Abnormal neurotransmitter levels were also observed in adult fish, accompanied by reduced exploratory activity and anxiety-like behavioral changes. Altogether, these findings suggest that SEM-induced developmental neurotoxicity may be associated with disruptions in neurotransmitter homeostasis and metabolic disturbance, and early-life SEM exposure may lead to long-term neurodevelopmental effects even after a recovery period. This study provides new evidence for the neurotoxic effects of SEM exposure in aquatic organisms and highlights the importance of considering pollutant exposure during early development in ecological risk assessments.

Secondary PVC microplastics are more toxic than primary PVC microplastics to Oryzias melastigma embryos

Bin Xia,Qi Sui,Yushan Du,Liang Wang,Jing Jing,Lin Zhu,Xinguo Zhao,Xuemei Sun,Andy M. Booth,Bijuan Chen,Keming Qu,Baoshan Xing

Irregular-shaped and partially degraded secondary microplastics (SMP) account for the majority of MPs in marine environments, yet little is known about their effects on marine organisms. In this study, we investigated the embryotoxicity of polyvinyl chloride SMP and primary microplastics (PMP) to the marine medaka Oryzias melastigma . This study aimed to determine the physical impacts of MPs and, for the first time, elucidate the underlying mechanisms of physical toxicity. SMP shortened hatching time and induced higher teratogenic effects on larvae relative to PMP, indicating a higher toxicity from SMP. Physical damage from SMP to the chorion surface appears to be the main toxicity mechanism, caused by their irregular shape and reduced aggregation relative to PMP. In contrast, real-time changes in oxygen demonstrated that hypoxia caused by greater PMP adsorption to the chorion surface contributes to the toxicological responses of this material relative to SMP. Modulation of genes involved in hypoxia-response, cardiac development and hatching confirmed the toxicity mechanisms of PMP and SMP. The chemical contribution to observed toxicity was negligible, confirming impacts derived from physical toxicity. Our findings highlight the negative effects of environmentally relevant SMP on the marine ecosystems .

Deciphering the TLR-mediated immunotoxic adverse outcome pathway of pentachlorophenol in marine medaka (Oryzias melastigma)

Yu Zhang,Jizhuang Chu,Shixuan Cui,Qiang Kong,Lihua Cui,Yumiao Zhou,Shudi Yao,Youru Zhang,Huanxin Zhang

Pentachlorophenol (PCP) is a persistent organic pollutant commonly detected in marine environments, and recent studies have confirmed its adverse effects on the immune systems of aquatic organisms. However, the potential molecular mechanisms of action and subsequent downstream damage caused by PCP remain unclear. This study employed transcriptomics, metabolomics, biochemical and histopathological analyses, and molecular dynamics simulation to explore the immunotoxic effects and associated downstream damage induced by PCP in marine medaka ( Oryzias melastigma ). The results indicated that PCP at environmentally relevant concentrations (1 and 10 μg/L) activates the Toll-like receptor (TLR) signaling pathway, significantly upregulating the expression levels of pro-inflammatory cytokines and triggering inflammatory responses. Using the adverse outcome pathway (AOP) analysis framework, the binding of PCP to TLR5 was identified as the molecular initiating event. This event triggers a series of key events, activation of the TLR signaling pathway, an enhanced inflammatory response, increased oxidative stress levels, disrupted amino acid metabolism, and liver injury, ultimately leading to impaired immune system function (adverse outcome). This study establishes the mechanistic linkage between TLR5 and metabolic dysfunction in PCP immunotoxicity, providing novel toxicological insights and theoretical support for the ecological risk assessment of PCP and related marine pollutants.

Semicarbazide conferred developmental toxicity in Oryzias melastigma embryos by oxidative stress and energy metabolism disorder

Qiaoyue Xu,Chunqing Liu,Si Chen,Xishan Li,Deqi Xiong

The residual antibiotics in aquatic animals present potential risk to ecological environment and food safety. Here, we uncover the biochemical reaction and molecular mechanisms in marine medaka embryos exposed to semicarbazide (SEM) at environmental relevant concentrations (10, 50 and 100 μg/L) for 14 days then followed by 7 days clean water recovery period. The decreased levels total superoxide dismutase (T-SOD) and catalase (CAT), and increased levels in malondialdehyde (MDA) content suggesting SEM induced severe oxidative stress and excessive reactive oxygen species (ROS). These effects further contributed to morphological changes and cardiac dysfunction. Meanwhile, oxidative stress activated genes associated with inflammatory responses ( tlr1, cyp1a1, ahr, il-1β ), implying potential suppression of innate immune functions. Furthermore, elevated activities of pyruvate kinase (PK), hexokinase (HK), and lactate dehydrogenase (LDH), along with decreased succinate dehydrogenase (SDH) activity, indicated SEM activate glycolysis pathway and disrupt Tricarboxylic Acid cycle (TCA) cycle leading to mitochondrial function and energy metabolism imbalance. Notably, these adverse effects cannot be completely eliminated after the short recovery period, implying a potential long-term toxicity of SEM. This study provides new insights into the toxicity effect SEM on the early developmental of marine organisms, its underlying regulatory mechanisms and implications for SEM risk management.

Ecotoxicological insights into UV-P-induced immunotoxicity and bacterial infection risk in marine medaka (Oryzias melastigma)

Li Wu,Mingshan Song,Lifang Wen,Sisi Wei,Zhenqi Xin,Pengzhi Qi,Xiaojun Yan,Yongjiu Xu,Baoying Guo,Weifeng Wang

Benzotriazole ultraviolet stabilizers (BUVs) are widespread and bioaccumulative aquatic contaminants. However, their immunotoxicity and underlying molecular mechanisms remain largely unexplored. In this study, we measured the concentrations of ten BUVs in the East China Sea and selected UV-P for further study based on its highest detected concentration. Consequently, marine medaka ( Oryzias melastigma ) were exposed to an environmentally relevant concentration (3.5 μg/L) and two elevated concentrations (35 and 350 μg/L) of UV-P for 28 days. Immunological parameters, histopathological alterations, and transcriptomic profiles were examined at the end of exposure. Additionally, bacterial challenge tests using Pseudomonas fluorescens were performed to assess host resistance. The results revealed an initial increase in the levels of alkaline phosphatase, immunoglobulin M, C-reactive protein, and complement component 3, followed by a significant decrease with increasing UV-P concentration. Histopathological damage was observed in immune tissues, including the liver, intestine, kidney, and gills. Transcriptomic analysis indicated significant enrichment of differentially expressed genes in multiple immune-related pathways following UV-P exposure. The key pathways affected mainly included cytochrome P450-mediated metabolism and the complement and coagulation cascades. The 24-h post-infection assessment revealed dose-dependent increases in bacterial loads and mortality rates in UV-P-exposed fish. Collectively, these findings demonstrate that UV-P induces immunotoxicity and increases the risk of bacterial infection in marine medaka. This study highlights the potential ecological threats posed by BUVs and offers valuable insights into the immunotoxic mechanisms of these contaminants.

Temperature-dependent toxicity and mechanisms of florfenicol on the embryonic development of marine medaka (Oryzias melastigma)

Xingying Guo,Xu Gong,Jinhui Wang,Tan-Duc Nguyen,Syed Shabi Ul Hassan Kazmi,Jiezhang Mo,Feng Hua,Wenhua Liu,Zhen Wang

The extensive use of antibiotics and their persistence in the environment have seriously threatened marine ecosystems in recent years. The frequent occurrence of extreme weather due to climate change has also increased the uncertainty of effective toxicity identification and risk assessment of the chemicals of concern. This study aimed to investigate the toxic effects and potential mechanisms of florfenicol (0.5, 10, 100, 500 and 1000 μg/L) on the embryonic development of marine medaka ( Oryzias melastigma ) through 21-d experiment under three temperature exposure scenarios (20, 25, and 30 °C). Considering embryo development, the highest level of florfenicol at 1000 μg/L decreased the hatching success at 25 °C, whereas the total inhibition of hatching was observed at 20 °C regardless of florfenicol concentrations of concern. The ATP content was inhibited by the highest florfenicol dose at 20 °C, while stimulated ATP content at 20 °C and 30 °C, compared to 25 °C. Fluctuation of temperatures, especially at 20 ℃, induced oxidative stresses, including increased MDA contents and decreased CYP450 and HSP90 contents. For inflammatory response-related genes ( nf-κb , nlrx1 , pycard , caspase 1 , and il-1β ), an increase of florfenicol dose led to gene upregulation at 25 °C. Conversely, gene upregulation was also observed for all treatments at 30 °C, while predominantly downregulation was observed for all treatments at 20 °C. For genes related to DNA damage and apoptosis ( pi3k , akt , foxo1 , tp53 , bcl-2 , bax , apaf-1 , caspase 9 , and 3 ), alterations in gene expression were evident for all florfenicol treatments at both 20 °C and 30 °C. Therefore, this study suggests that the combined effects of florfenicol and temperature may disrupt the normal function of mitochondria, impacting ATP production, thereby inducing oxidative stress, inflammatory response, DNA damage, and apoptosis, ultimately resulting in decreased hatching rates and increased mortality. Furthermore, the key findings of this study reveal the complex interactions between florfenicol and temperature and emphasize the need to consider temperature when identifying toxicity and mechanisms, as well as the ecological risk assessment of florfenicol in marine environments.

Multigenerational and persistent toxicity of environmentally relevant concentrations of Benzo[a]pyrene in Marine Medaka (Oryzias melastigma)

Dong Sun,Xuer Zhang,Shuyuan Chen,Qian Liu,Yuna Huang,Hui Zhao,Meina Duan

Benzo[ a ]pyrene (BaP) is a well-known environmental pollutant with extensively documented toxicological hazard. While transgenerational BaP effects have been documented, the reversibility of these effects following exposure cessation at different generational stages remains unexplored. This study employed a novel exposure methodology on marine medaka ( Oryzias melastigma ) with two key regimens: including continuous multi-generational BaP exposure (G1F1-G3F3) and post-exposure recovery (non-BaP, G1F2-G1F3 and G2F3). Integrated analyses were conducted across phenotypic, developmental, reproductive, and molecular biological endpoints. The results showed that: (1) BaP significantly inhibited growth, development, and reproduction of marine medaka, increasing offspring deformity rate and mortality rate. Most notably, 100 % offspring mortality occurred in both G2F4 and G3F4 generations, as well as the post-exposure recovery in G2F4; (2) 200 μg·L −1 BaP damaged male gonads, decreasing sperm counts and increasing testicular non-cellular areas, and the non-cellular areas found in G2F3 and G3F3 were exacerbated; (3) molecular analyses demonstrated systematic disruption of the hypothalamic-pituitary-gonadal (HPG) axis, with significant downregulation of key genes of steroidogenic enzymes ( cyp17a , 17βhsd , cyp19a ) and reproductive hormone receptors ( erα , arα ). These findings demonstrate that environmentally relevant concentrations of BaP induce irreversible transgenerational reproductive impairment that persists beyond exposure termination. This underscores the critical need to reassess environmental quality criteria for BaP, while also emphasizing the importance of comprehensive ecological risk assessment and the necessity of incorporating trans/multigenerational testing into standard ecotoxicological frameworks.

Mechanisms of action and adaptive responses to diclofenac and meloxicam during the early life stages of Oryzias melastigma

Chuan Liang,Yang Chen,Yunzhe Ling,Peirun Li,Jiahao Liu,Xuantian Li,Yanhua Xu,Zhiying Liu

Non-steroidal anti-inflammatory drugs (NSAIDs), though designed to target specific molecular pathways, pose significant environmental risks to non-target organisms, particularly marine fish. This study investigated the toxicity mechanisms and adaptive responses to diclofenac (DCF) and meloxicam (MEX) during the early life stages (ELS) of Oryzias melastigma at environmentally relevant concentrations over a 31-day period. Mechanistic investigations of sub-lethal effects were conducted using Enzyme-Linked Immunosorbent Assay (ELISA), RNA sequencing (RNA-Seq) and quantitative PCR (qPCR). The results revealed that cyclooxygenase (COX) inhibition disrupted the renin-angiotensin system, leading to an accumulation of angiotensin II and cardiovascular developmental defects. Additionally, downregulation of the pla2 gene reduced substrates essential for COX enzyme activity, exacerbating the effects. Although NSAIDs are known to affect the digestive system, no significant effects on developmental factors were observed. RNA-Seq and qPCR analyses revealed an adaptive upregulation of key genes, including ace2 and cyp7a1 , involved in cardiovascular and metabolic regulation. Furthermore, 16S rRNA sequencing identified shifts in the microbial community, particularly in g_Rubritalea and g_Sphingomonas genera. Both the upregulated genes and the altered microbial taxa likely played a role in mitigating toxic effects and promoting homeostasis. Moreover, molecular docking suggested that MEX exhibited stronger sub-lethal effects than DCF, likely due to its higher binding affinity to COX. These findings provide valuable insights into NSAID toxicity mechanisms in marine fish, highlighting the importance of adaptive responses in countering environmental stress and underscoring the long-term ecological risks of chronic NSAID exposure.
