An in-situ gold growth self-catalytic signal amplification system enhanced hollow gold immunochromatography for ultrasensitive detection of p24 antigen in HIV infection

Chuanjiang Ran,Qingjie Zhang,Zhenglin Long,Yuhao Li,Xinyue He,Shengzhou Li,Chen Chen,Cong Cheng,Ying Xu,Wenli Zhang,Wei Guo,Yan Shen

In recent years, the global public health landscape has faced unprecedented challenges, notably with the COVID-19 outbreak and the wide spread of HIV. The rapid spread and serious harm of the virus has highlighted the critical need for fast, accurate, and accessible diagnostic tools. These requirements make lateral flow immunochromatographic assay (LFIA) become the most prominent point-of-care testing (POCT) technology at present. In order to find the balance between sensitivity, ease of operation, detection speed and cost of LFIA, hollow gold nanoparticles (HGNPs) with stronger optical signal, stability and simple synthesis process were used as immune labeling materials to construct LFIA platform (HGNPs-LFIA). HGNPs exhibited elevated catalytic activity in relation to the reaction between chloroauric acid (HAuCl 4 ) and hydroxylamine hydrochloride (NH 2 OH-HCl). This heightened activity consequently resulted in the deposition of new gold nanoparticles on the surface of the original HGNPs. At lower analyte levels, a deepening of the colors of the test line and the control line can be observed with naked eyes. Utilizing this special catalytic activity, an in-situ gold growth method was further used to amplify the detection signal of HGNPs-LFIA by self-catalytic, signal amplified HGNPs-LFIA (sa-HGNPs-LFIA) platform for ultrasensitive and rapid detection of p24 antigen in serum was constructed. The platform struck a good balance between high sensitivity and low false positives, and was capable of detecting p24 antigen as low as 1 pg/mL in serum within 30–40 min, which is 10 times more sensitive than the original HGNPs-LFIA and 20 times more sensitive than the commercially available Alere test strips, and further shortened the window of detection for acute HIV infection.

Rapid and sensitive detection of HIV-1 p24 antigen by immunomagnetic separation coupled with catalytic fluorescent immunoassay

Zhang Yun,Yang Hang,Yu Junping,Wei Hongping

In this study, a system of magnetic beads (MBs) coupled with catalytic fluorescent immunoassay for rapid and sensitive determination of HIV-1 capsid antigen p24 was developed. p24 was captured by antibody immobilized MBs, and the detection antibody was linked to horseradish peroxidase (HRP) through biotin–streptavidin recognition, catalyzing the oxidation of o -phenylenediamine (OPD) and hydrogen peroxide to produce a fluorescent product. This is the first reported utilization of the fluorescence of OPD oxidation product catalyzed by HRP for immunoassay. Optimization of conditions afforded a low detection limit of 0.5 pg/mL (3σ) for p24 with a linear range of 1.4–90.0 pg/mL. The assay exhibited good reproducibility with a relative standard deviation (RSD) of 4.4 %, 4.7 %, and 5.0 % for detecting 1.4 pg/mL, 22.5 pg/mL, and 45.0 pg/mL p24, respectively. The assay can be completed in less than 90 min. Moreover, the proposed method was successfully applied to detect p24 in spiked serum. This method overcomes the interference of MBs to the fluorescence signal and demonstrated higher sensitivity for detection of p24 than conventional ELISA kits. The system could be applied for detecting other antigens with high sensitivity, rapidity, specificity, and simple operation. A rapid and sensitive biosensing method coupling immunomagnetic separation and catalytic fluorescence for determination of HIV-1 p24 has been developed

An Ultrasensitive Electrochemical Immunosensor for HIV p24 Based on Fe3O4@SiO2 Nanomagnetic Probes and Nanogold Colloid-Labeled Enzyme–Antibody Copolymer as Signal Tag

Ning Gan,Xiaowen Du,Yuting Cao,Futao Hu,Tianhua Li,Qianli Jiang

An ultrasensitive portable electrochemical immunosensor for human immunodeficiency virus p24 (HIV p24) antigen detection has been developed, whereby the detection sensitivity was 1000 times higher than that of the ELISA method. Firstly, a novel HRP enzyme–antibody copolymer (EV-p24 Ab2) was synthesized through an EnVision regent (EV, a dextrin amine skeleton anchoring more than 100 molecules of HRP and 15 molecules of anti IgG), then incubated in the secondary antibody of p24. Secondly, the copolymer was immobilized on the gold nanocolloids (AuNPs) to fabricate a novel signal tag (AuNPs/EV-p24 Ab2). Subsequently, a sandwich-type immunoreaction would take place between the capture probe (silicon dioxide-coated magnetic Fe3O4nanoparticles (MNPs) labeled with the primary p24 antibody (MNPs-p24 Ab1)), p24 (different concentrations) and the signal tag [AuNPs/EV-p24 Ab2)] to form the immunocomplex. Finally, the immunocomplex was absorbed on the surface of screen printed carbon electrode (SPCE) by a magnet and immersed in the o-hydroxyl phenol (HQ) and H2O2. The large amounts of HRP on the signal tag can catalyze the oxidation of HQ by H2O2, which can induce an amplified reductive current. Moreover, the capture probe could improve the accumulation ability of p24 and facilitate its separation from the substrate through the magnet. Under optimal conditions, the proposed immunoassay exhibited good sensitivity to p24 within a certain concentration range from 0.001 to 10.00 ng/mL, with a detection limit of 0.5 pg/mL (S/N = 3). The proposed method can be used for real-time and early detection of HIV-infected people.

Effects on molecular interactions of hollow gold nanoparticles and antibody for sensitizing P24 antigen determination

Tao Wang,Chuanjiang Ran,Xinyue He,Shengzhou Li,Hongguang Xiang,Yan Shen,Jue Wang,Hongxia Wei

In recent years, with the rapid development of point-of-care testing, the application of lateral flow immunochromatography assay (LFIA) has become increasingly widespread. The key to the success of these detection technologies is the effective binding with diagnostic materials and detection antibody proteins. Although many researchers have tried to optimize antibody binding, a universally accepted strategy that can provide maximum performance has not been determined. In this study, the HIV infection P24 antigen was selected as the detection biomarker. Then the binding mechanism between hollow gold nanoparticles as diagnostic materials and detection antibodies was explored through dynamic light scattering, Fourier transform infrared spectroscopy, circular dichroism spectroscopy, and other methods. It was found that the binding efficiency is related to the change in protein secondary conformation during binding, hydrogen bonding, and van der Waals force maintain the binding mechanism between antibodies and nanoparticles. The main forces of particle complexation and the main binding site of the antibody were discussed and analyzed. Finally, an immunochromatographic system was constructed to evaluate the significant advantages of this platform compared to the common colloidal gold immunochromatographic system.

Fluorescence and visual immunoassay of HIV-1 p24 antigen in clinical samples via multiple selective recognitions of CdTe QDs

Tang Zhuoyun,Wei Zeliang,Huang Ke,Wei Yinhao,Li Dongdong,Yan Shixin,Huang Jin,Geng Jia,Tao Chuanmin,Chen Piaopiao,Ying Binwu

Human immunodeficiency virus (HIV) infection inflicts significant economic and social burdens on many countries worldwide. Given the substantial morbidity and mortality from HIV infection, there is an urgent need for accurate and early detection of the virus. In this study, immunofluorescence and visual techniques are described that detect the HIV-1 p24 antigen, which relied on selective recognition of Ag + /Ag nanoparticles (Ag NPs) and Cu 2+ /Cu + using cadmium telluride quantum dots (CdTe QDs). After the sandwich immunoreactions were accomplished, the alkaline phosphatase (ALP) hydrolyzed L-ascorbic acid 2-phosphate (AAP) to form ascorbic acid (AA) that further reduces Ag + and Cu 2+ to Ag NPs and Cu + , respectively. This method was highly sensitive and selective and could detect as low as 1 pg/mL of p24 antigen by naked eyes and had a good linearity in the concentration range 1–100 pg/mL. When using Ag + and Cu 2+ as media, the limit of detection (LOD) of the new method was 0.3 pg/mL and 0.2 pg/mL, respectively. Compared with clinical electrochemiluminescence immunoassay (ECLIA) results and clinical data, this method demonstrated good consistency for the quantification of HIV-1 p24 antigen in 34 clinical serum samples. In addition, this method could accurately distinguish HIV from other viruses and infections such as hepatitis B virus , systemic lupus erythematosus , hepatitis C virus , Epstein-Barr virus , cytomegalovirus , lipemia , and hemolysis . Therefore, our dual-mode analysis method may provide additional solutions to identify clinical HIV infection. Graphical abstract An immunofluorescence and visualization dual-mode strategy for the detection of p24 antigen was constructed based on immune recognition reaction and a phenomenon that cadmium telluride quantum dots (CdTe QDs) can selectively recognize Ag + /Ag nanoparticles (Ag NPs) and Cu 2+ /Cu + .
