Role of GTP-CHI links PAH and TH in melanin synthesis in silkworm, Bombyx mori

Ping Chen,Jiying Wang,Haiyin Li,Yan Li,Peng Chen,Tian Li,Xi Chen,Junjie Xiao,Liang Zhang

In insects, pigment patterns are formed by melanin, ommochromes, and pteridines. Here, the effects of pteridine synthesis on melanin formation were studied using 4th instar larvae of a wild-type silkworm strain, dazao ( Bombyx mori ), with normal color and markings. Results from injected larvae and in vitro integument culture indicated that decreased activity of guanosine triphosphate cyclohydrolase I (GTP-CH I, a rate-limiting enzyme for pteridine synthesis), lowers BH4 (6R- l -erythro-5,6,7,8-tetrahydrobiopterin, a production correlated with GTP-CH I activity) levels and eliminates markings and coloration. The conversion of phenylalanine and tyrosine to melanin was prevented when GTP-CH I was inhibited. When BH4 was added, phenylalanine was converted to tyrosine , and the tyrosine concentration increased. Tyrosine was then converted to melanin to create normal markings and coloration. Decreasing GTP-CH I activity did not affect L-DOPA (3,4- l -dihydroxyphenylalanine). GTP-CH I affected melanin synthesis by generating the BH4 used in two key reaction steps: (1) conversion of phenylalanine to tyrosine by PAH (phenylalanine hydroxylase) and (2) conversion of tyrosine to L-DOPA by TH (tyrosine hydroxylase). Expression profiles of BmGTPCH Ia , BmGTPCH Ib , BmTH , and BmPAH in the integument were consistent with the current findings.

Wet-adhesive and antibacterial PAH-TPP coacervates with self-mineralizing capability for cranial flap fixation

Weicheng Chen,Jiaxu Shi,Dachuan Liu,Kai Lu,Jianlong Fu,Jingxi Xu,Huan Wang,Zhiliang Guo,Li Dong,Di Li,Xin Li,Miodrag J. Lukic,Wei Xia,Song Chen,Bin Li

Repositioning and securing the cranial bone flap after craniotomy remain significant challenges in neurosurgery. Traditional fixation methods often suffer from weak mechanical strength, bioinertness, limited osteogenic capacity, and a lack of antibacterial properties, complicating clinical outcomes. Recent medical adhesives offer superior fixation but face significant limitations in cranial bone applications. In this study, we explored the application of PAH (Poly (allylamine) hydrochloride)-TPP (Tripolyphosphate) coacervate (PT) as a bone adhesive. The PT coacervate demonstrated excellent anti-swelling (anti-swelling ratio less than 1 %), self-healing, and injectable properties, as well as exceptional shape adaptability and cytocompatibility. Adhesion tests revealed its outstanding adhesion (99.06 ± 11.76 kPa for lap shear and 121.42 ± 16.73 kPa for end to end), long-term durability, and tunable adhesion strength. Furthermore, the coacervate demonstrated broad-spectrum antibacterial activity against both Gram-positive and Gram-negative bacteria (antibacterial rate more than 90 %), with mechanistic studies revealing promising strategies to address localized and systemic drug-resistant infections. Additionally, the coacervate's self-mineralizing properties significantly enhanced its osteogenic performance. In vivo studies confirmed its effective fixation, robust antibacterial activity, and improved osteogenesis, underscoring its suitability for cranial bone flap repositioning and fixation after craniotomy. In summary, this coacervate adhesive offers a promising therapeutic solution for addressing the challenges of cranial flap fixation in neurosurgery.

Characterization of PKCα-rutin interactions and its application as a treatment strategy for PAH by inhibiting ferroptosis

Haixia Che,Jie Yi,Xiaoting Zhao,Hong Yu,Xianyao Wang,Rui Zhang,Xin Li,Jia Fu,Qian Li

As a common plant-derived dietary flavonoid, rutin receives the widespread attention because of its good antioxidant bioactivities. Protein kinase Cα (PKCα) is a serine/threonine kinase that is involved in uncountable cellular processes, among which ferroptosis, a novel form of cell death, is triggered by lipid peroxidation and has been reported to associate with pulmonary arterial hypertension (PAH). But it is still not well appreciated how rutin inhibits ferroptosis in PAH and what function of PKCα plays in this process. In this study, we firstly observed whether rutin could prevent PAH by attenuating ferroptosis with PAH animal model and pulmonary artery smooth muscle cells (PASMCs) under hypoxia. Mitochondrial metabolomics and network pharmacology were employed to clarify the metabolic alteration and screen target proteins, results showed PKCα was a vital node in rutin regulating mitochondrial metabolism related to ferroptosis in PAH. Based on molecular docking and multispectral analysis, we found rutin could directly interact with PKCα through hydrogen bonds, which could induce static quenching, and then influence the secondary structure of PKCα. In conclusion, these findings mainly point to a novel mechanism that rutin protect PAH rat by modifying the structure, altering activity of PKCα, and then suppressing ferroptosis. This work was great helpful to reveal that the interaction behaviours between small molecules and bio-macromolecules are the critical point to develop the natural biological active ingredients and give an insight into the potential applications of flavonoid in health and disease.

Stimuli-responsive terpolymer mPEG-b-PDMAPMA-b-PAH mediated co-delivery of adriamycin and siRNA to enhance anticancer efficacy

Minghui Xu,Junmin Qian,Aili Suo,Weijun Xu,Rongrong Liu,Hongjie Wang

Combination of chemotherapy and small interfering RNA (siRNA)-based therapy has emerged as a promising approach for cancer treatment with the synergistic effect. In this study, a novel terpolymer methoxy poly(ethylene glycol)-block-poly(N,N-dimethylaminopropyl methacrylamide)-block-poly(acrylhydrazine) (mPEG-b-PDMAPMA-b-PAH) with a disulfide linkage between mPEG and PDMAPMA blocks was developed for the intracellular targeted co-delivery of adriamycin and P-gp siRNA. The terpolymer was synthesized by sequential reversible addition–fragmentation chain transfer (RAFT) polymerization of N,N-dimethylaminopropyl methacrylamide and N-tert-butoxycarbonyl-N′-acryl hydrazine (Boc-acrylhydrazine) in the presence of a PEGylated macro-RAFT agent, followed by Boc-deprotection. The terpolymer could chemically conjugate adriamycin via an acid-cleavable hydrazone bond and simultaneously condense the negatively charged siRNA through electrostatic interactions at an N/P ratio of 2. The resultant adriamycin-conjugated nanoparticles/siRNA complexes (ADR-NPs/siRNA complexes) showed a spherical morphology and had an average diameter of 186 nm. The release profiles of the two payloads from the ADR-NPs/siRNA complexes exhibited a pH/reduction dual-dependent sustained release behavior. The ADR-NPs/siRNA complexes could simultaneously deliver adriamycin and siRNA efficiently into MCF-7/ADR cells and significantly inhibit their growth in a synergistic fashion. All the results indicated that the terpolymer mPEG-b-PDMAPMA-b-PAH could serve as a potential vehicle for the combination of chemotherapy and gene therapy.

PAH-induced metabolic changes related to inflammation in childhood asthma

Wu Hao,Bao Yuling,Yan Tongtong,Huang Hui,Jiang Ping,Zhang Zhan,Li Lei,Wu Qian

Epidemiological studies have shown that PAHs may exert adverse effects on childhood asthma. However, the underlying molecular mechanism remains to be fully elucidated. This study aimed to investigate this process in view of metabolic pathways, especially one-carbon metabolism and tryptophan metabolism. Fifty asthmatic children and 50 control subjects were recruited for this study. Serum IgE and IL-17A levels were detected by ELISA. Serum PAH concentrations were measured by GC-MS. One-carbon-related metabolites and tryptophan metabolites were determined by UPLC-Orbitrap-MS. DNA methylation was analyzed by bisulfite sequencing PCR. ChIP assays were used to examine H3K4me3 enrichment on IL-17A gene. Multivariable linear regression was performed to evaluate the association between PAHs and childhood asthma mediated by intermediators. HE staining in lung tissue, IgE and IL-17A in BALF, metabolic profiles in urine, and Ahr , Il-17a , and Cyp1a1 gene expression were determined in PAH-exposed mice. Serum Fla level was associated with childhood asthma (OR = 1.380, 95% CI: 1.063–1.792), and had a great effect on one-carbon metabolites, especially SAH, SAM, and Ser, which exerted significant mediation effects on the relationship between the Fla concentration and asthma. Moreover, we did find significant mediation effects between serum Fla and asthma by LINE-1 DNA methylation and H3K4me3 levels in the IL-17A promoter region. The differential Trp metabolites, such as Trp, tryptamine, IA, IAA, indole, IAld, and IAAld, indicated that asthmatic children had increased indole-AhR pathway. Mediation analysis failed to show a mediator effect of Trp metabolites in the association between PAHs and childhood asthma. An animal study confirmed that PAH exposure increased methylation levels, and altered Trp metabolite-AhR-IL-17A axis, which may be influenced by gender. PAHs disturbed one-carbon metabolism to influence the methyl group refilling DNA methylation and histone methylation, and disturbed tryptophan metabolism to regulate Th17-cell differentiation, which may elevate serum IL-17A concentration in asthmatic children.

The naphthalene catabolic protein NahG plays a key role in hexavalent chromium reduction in Pseudomonas brassicacearum LZ-4

Huang Haiying,Tao Xuanyu,Jiang Yiming,Khan Aman,Wu Qi,Yu Xuan,Wu Dan,Chen Yong,Ling Zhenmin,Liu Pu,Li Xiangkai

Soil contamination by PAH and heavy metals is a growing problem. Here, we showed that a new isolate, Pseudomonas brassicacearum strain LZ-4, can simultaneously degrade 98% of 6 mM naphthalene and reduce 92.4% of 500 μM hexavalent chromium [Cr (VI)] within 68 h. A draft genome sequence of strain LZ-4 (6,219,082 bp) revealed all the genes in the naphthalene catabolic pathway and some known Cr (VI) reductases. Interestingly, genes encoding naphthalene pathway components were upregulated in the presence of Cr (VI), and Cr (VI) reduction was elevated in the presence of naphthalene. We cloned and expressed these naphthalene catabolic genes and tested for Cr (VI) reduction, and found that NahG reduced 79% of 100 μM Cr (VI) in 5 minutes. Additionally, an nahG deletion mutant lost 52% of its Cr (VI) reduction ability compared to that of the wild-type strain. As nahG encodes a salicylate hydroxylase with flavin adenine dinucleotide (FAD) as a cofactor for electron transfer, Cr (VI) could obtain electrons from NADH through NahG-associated FAD. To the best of our knowledge, this is the first report of a protein involved in a PAH-degradation pathway that can reduce heavy metals, which provides new insights into heavy metal-PAH contamination remediation.

Benzo(a)anthracene Targeting SLC1A5 to Synergistically Enhance PAH Mixture Toxicity

Yanwei Wang,Jiahui Zhao,Yipeng Xu,Jing Miao,Keyu Pan,Yihan Li,Yong Chen,Xuesong Liu,Ailin Zhao,Jingyu Qin,Tengfei Xu,Mingliang Fang

Human exposure to polycyclic aromatic hydrocarbons (PAHs) as mutagenic and carcinogenic pollutants in the environment often occurs in the form of mixtures. Although the mixture effects of PAHs have been previously recognized, the toxicological mechanisms to explain them still remain quite unclear. This study combined metabolomics and chemical proteomics methods to comprehensively understand the mixture effects of a PAH mixture including benzo(a)anthracene (BaA), benzo(b)fluoranthene (BbF), benzo(a)pyrene (BaP), and chrysene (CHR). Among them, BaA has shown a strong synergistic effect with other PAHs. Interestingly, BaA alone is not a potent oxidative stress inducer in liver cells but dose-dependently amplifies oxidative damage caused by the PAH mixture. Global metabolomics analysis results revealed damage to the antioxidant glutathione synthesis, which was caused by the glutamine depletion caused by BaA in the mixture. Subsequently, the label-free chemical proteomics and cellular thermal shift analysis (CETSA) demonstrated that the PAH mixture altered the thermal shift of glutamine transporter SLC1A5. Furthermore, Western blotting and the isothermal titration calorimetry (ITC) interaction measurements showed nanomolar KD values between BaA and SLC1A5. Overall, this study showed that BaA synergistically contributed to PAH mixture induced oxidative damage by targeting SLC1A5 to inhibit glutamate transport into cells, resulting in the inhibition of glutathione synthesis.

High phenanthrene degrading efficiency by different microbial compositions construction

Zhou Guoyan,Qiao Hongtao,Liu Yandong,Yu Xiongsheng,Niu Xiang

Microbial remediation has become the most promising technical means for the remediation of polycyclic aromatic hydrocarbons (PAHs) non-point source contaminated soil due to its low cost of treatment, complete degradation of pollutants, and in-situ remediation. In this study, in order to demonstrate the phenanthrene degrading microbial diversity, phenanthrene was chosen as the representative of PAHs and strains capable of degrading phenanthrene were isolated and screened from the sedimentation sludge and the bottom sludge of oil tank trucks, and high throughput sequencing was used to check the dominant strains with a good degrading effect on phenanthrene. Results showed even more than 50% of phenanthrene was degraded in all samples, the composition of PAH-degrading bacteria was diverse, and different environments constructed different functional microbial groups, which resulted in the microbial adapting to the diversity of the environment. Finally, a series of bacterial species with phenanthrene-degrading functions such as Achromobacter, Pseudomonas, Pseudochelatococcus, Bosea was enriched after nine transferring process. Overall, our study offers value information for the enrichment of functional degrading microbes of phenanthrene or other pollutants that more concern should be paid in not only the degradation rate, but also the diversity variation of microbial community composition.

Influencing factors of bioaugmentation treatment of PAH-contaminated soils in slurry bioreactors

Shiyu Bao,Likun Zhao,Yuanwen Liu,Lewei Yang,Peng Liu,Chaoqi Chen,Xuhui Mao

Polycyclic aromatic hydrocarbons (PAHs) are a group of contaminants with low bioavailability and biodegradability . As a result, removing PAHs from contaminated soils is often inefficient. In this study, soils spiked with fluoranthene and pyrene were inoculated with Aeromonas sp. BCP-3, a newly isolated strain from coke-plant sludge , and treated in slurry bioreactors . The removal of PAHs was significantly affected by aeration rate, slurry water-soil mass ratio, stirring rate, temperature, and the concentration of Tween-80. Such effects were attributed to changes in BCP-3 growth, PAHs solid-liquid mass transfer, and slurry particle size. Notably, under optimal conditions (i.e., aeration rate of 1.5 L/min, temperature of 30 °C, stirring rate of 200 rpm, water-soil mass ratio of 1.5:1, and Tween-80 concentration of 1.3 %), 96.3 % and 97.2 % of initially spiked fluoranthene and pyrene were removed within 12 days, respectively. A PAH-contaminated soil was collected from a local coking plant and treated under the same conditions, except for the Tween-80 concentration. For the real soil, the Tween-80 concentration was 4 % to ensure a significant proportion of PAHs released into the liquid phase. Twenty-one days after inoculation of BCP-3, the removal efficiencies of low molecular weight PAHs and high molecular weight PAHs were 82.2 % and 51.8 %, respectively. The removal efficiencies of PAHs were significantly higher than the slurry without BCP-3 or HgCl 2 -sterilized slurry. BCP-3 improved the bioavailability and biodegradability of PAHs, resulting in effective PAH removal in slurry bioreactors . Overall, successful bioaugmentation was achieved with the addition of Tween-80 and the optimization of operating conditions.

Biodegradation Potential of Bacillus sp. PAH-2 on PAHs for Oil-Contaminated Seawater

Xianghui Kong,Ranran Dong,Thomas King,Feifei Chen,Haoshuai Li

Microbial degradation is a useful tool for inhibiting or preventing polycyclic aromatic hydrocarbons (PAHs) widely distributed in marine environment after oil spill accidents. This study aimed to evaluate the potential and diversity of bacteria Bacillus sp. PAH-2 on Benzo (a) anthracene (BaA), Pyrene (Pyr), and Benzo (a) pyrene (BaP), their composite system, aromatic components system, and crude oil. The seven-day degradation rates against BaA, Pyr, and BaP were 20.6%, 12.83%, and 17.49%, respectively. Further degradation study of aromatic components demonstrated PAH-2 had a high degradation rate of substances with poor stability of molecular structure. In addition, the degradation of PAHs in crude oil suggested PAH-2 not only made good use of PAHs in such a more complex structure of pollutants but the saturated hydrocarbons in the crude oil also showed a good application potential.

Iron-microbe synergy in laccase deactivation and PAH bioremediation:Insights from four contrasting soils

Guanyu Chen,Zelin Hou,Rong Cai,Huayu Tao,Yongkang Wu,Hong Liang,Dawen Gao

The rapid deactivation of enzymes in soils—a major barrier to practical enzymatic remediation—remains mechanistically unresolved under realistic mineral-organic-microbial conditions. Here, we uncover the governing principles behind this bottleneck by investigating laccase inactivation during polycyclic aromatic hydrocarbon (PAH) biodegradation in four distinct soils (clay, red, black, loess). A critical paradox was observed: red soil achieved the highest benzo[ a ]pyrene removal rate (52.72% within 24 h) yet suffered the most rapid laccase activity loss (83% within 24 h). Multi-technique analysis (XPS, EPR, FTIR) linked this to a distinct conformational signature in red soil laccase—maximal α-helix content (21.4%), intensified T1 Cu signal (gII=2.3), and elevated -C O/-C-N bonds. A partial least squares structural equation model (PLS-SEM) revealed the core mechanism: iron-mediated enhancement of microbial fermentation activated T1 Cu sites, which subsequently governed overall laccase activity (path coefficient = 0.64, p  < 0.05). We propose a unified mechanism whereby soil iron drives a conformational shift in the T1 copper center (increased α-helix), optimizing it for initial substrate oxidation but simultaneously rendering it prone to catalytic deactivation. This establish a “mineral–microbe–enzyme” coupling framework, revealing that native soil iron catalytically tunes and subsequently inactivates the enzyme. This mechanistic understanding is critical for transitioning from empirical applications to the rational design of stabilized enzymatic remediation strategies.

Comparative performance of lignin-based slow-release biostimulants in remediating PAH-contaminated soils

Weixi Shi,Jinyu Hou,Zilu Qiu,Changmin Peng,Mengran Zhang,Changxun Dong,Wuxing Liu,Yongming Luo

PET-based lignin biostimulants removed up to 76.0% of soil PAHs. • SRB achieved 65.8% removal of recalcitrant HMW-PAHs. • Moderate nitrogen pulses enhanced PAH removal more than release stability. • PET treatments enriched Mycobacterium and increased bacterial diversity. • SRR increased positive bacteria-bacteria interactions to 39.2%. Polycyclic aromatic hydrocarbons (PAHs) pose a major challenge for soil remediation owing to their structural recalcitrance, and conventional bioremediation is often ineffective against high-molecular-weight PAHs (HMW-PAHs). In this study, two lignin-based slow-release biostimulant strategies were compared for the remediation of PAH-contaminated soil collected from a coking site over 120 days: physically encapsulated materials, including lignin-biochar (SRB) and lignin-resin (SRR) coated fertilizers, and a chemically synthesized aminated lignin material (CSK). All biostimulants were applied at an equivalent lignin input of 1% of soil dry weight. These biostimulants were designed to enhance PAH degradation by regulating nutrient availability and stimulating PAH-degrading microbial communities, while lignin may additionally contribute substrates associated with co-metabolic processes. The materials were characterized by SEM, FTIR, XPS, TGA/DSC, 1 H NMR, and elemental analysis, confirming their distinct structural features and successful modification. Nutrient-release assays showed that the physically encapsulated materials released 26.2% and 24.4% of their total nitrogen by day 5 for SRB and SRR, respectively, followed by continued release to 61.5% and 57.8% by day 25. In contrast, CSK exhibited the strongest slow-release capacity, with cumulative nitrogen release of 11.2% by day 5 and 46.1% by day 25, but maintained a consistently lower release amplitude. The physically encapsulated materials showed superior remediation performance, achieving ΣPAHs removal rates of 76.0% for SRB and 74.4% for SRR, compared with 66.0% for CSK. They were particularly effective against HMW-PAHs, with removal rates of 65.8% and 57.1%, respectively, compared with 47.7% for CSK. The physically encapsulated treatments maintained relatively high and sustained available nitrogen concentrations, with coefficients of variation of 0.16-0.17. Available nitrogen concentrations were strongly positively correlated with residual ΣPAH, MMW-PAH, and HMW-PAH concentrations ( r  = 0.8-0.9). Despite its greater material stability and slow-release capacity, CSK released only 11.2% of its total nitrogen by day 5 and 46.1% by day 25 and maintained relatively small temporal variation in soil available nitrogen (CV ≈ 0.04); this nutrient-supply pattern was associated with lower PAH degradation, particularly for HMW-PAHs. These results indicate that the magnitude and temporal pattern of nitrogen supply, rather than release stability alone, are important factors associated with biostimulation efficacy. Microbial analysis showed that the physically encapsulated materials increased bacterial α-diversity and enriched PAH-degrading genera such as Mycobacterium , which was significantly correlated with PAH removal. Co-occurrence network analysis further indicated that SRR promoted positive bacteria-bacteria interactions (39.2%) while maintaining low cross-domain antagonism (6.3%). Overall, under the tested conditions, the physically coated lignin-based slow-release formulations evaluated here provided an effective strategy for the targeted remediation of PAH-contaminated soils, particularly those containing substantial HMW-PAH fractions, with their performance supported by coordinated nutrient-release and microbial-community responses.
