Electrospinning and three-dimensional (3D) printing is widely used to fabricate bone tissue engineering scaffolds. However, cells are difficult to infiltrate into the nanoporous structure of traditional electrospun scaffolds by electrospinning, and also 3D printing techniques have the disadvantage of low print resolution. In order to solve these problems, we fabricated a 3D composite scaffold by infusing PCL/gelatin dispersed nanofibers into the meshes of PCL printing scaffold. The morphology of the composite scaffold was evaluated by a scanning electron microscope (SEM), which shows the micro-scale (100–300 μm) porous structure. The porosity of the composite scaffold is as high as 79.32 ± 8.32%. Mechanical testing results indicated that the compressive modulus of the 3D composite scaffold (30.50 ± 0.82 MPa) is significantly higher than that of the lyophilized electrospun scaffold (18.55 ± 0.56 MPa), which is attributed to the 3D printing scaffold. An in vitro study indicated that the 3D composite scaffolds have good biocompatibility. Through CCK-8 assay and fluoresce staining characterization, MC3T3-E1 cells exhibit a better proliferation and infiltration on the composite scaffold than on the PCL printing scaffold, which is due to the microporous structure of the electrospun scaffold. Altogether, these results confirm the good potential of an electrospun/3D printing composite scaffold applied to bone tissue repair.

The heart valve is a key structure for human blood circulation, and the development of artificial heart valves (AHVs) has become one of the research hotspots in the field of cardiovascular diseases. Compared to the vulnerability of biological valves to compression damage in transcatheter aortic valve replacement surgery (TAVR), polymer valves have shown superior performance in research. However, its structural differences from natural valves have limited its development. In this study, polycaprolactone gelatin (PCL-Gel) co-spinning directional nanofibers (FIB) were used to construct a three-layer structure of orientation layer-random layer-orientation layer imitating natural valves. Then, PCL-Gel/PAAm-co-PAA-Fe composite (COM-Fe) was prepared by iron ion crosslinking the oriented fiber membrane wrapped by polyacrylamide polyacrylic acid copolymer hydrogel (COM). The COM-Fe material has anisotropy similar to that of native valves and fully meets the thickness requirements for transcatheter surgery. In vitro simulated compression results showed that the COM-Fe material has no significant structural or strength loss after short-term curling compression. In vitro fluid dynamics results showed that the COM-Fe samples could fully achieve the parameters specified in ISO 5840-3:2021. In addition, COM-Fe materials showed excellent biocompatibility both in vitro and in vivo, and demonstrated anti-inflammation potential in a rat subcutaneous embedding model. It can be seen that biomimetic COM-Fe composite materials with good curling compression resistance and valve function have great potential for application in the direction of transcatheter AHVs.

ABSTRACT: Electrospun poly(ε-caprolactone) (PCL)/gelatin (GT) scaffolds were developed to provide controlled release of 7-ethyl-10-hydroxy camptothecin (SN-38). Acetic acid was introduced to improve the miscibility of PCL and GT to produce a homogeneous nanofiber membrane mixture. The effect of SN-38 content in binary mixtures on processability, fiber morphology, water sorption, swelling, and drug release was investigated. Electrospun PCL/GT blend nonwoven fibers showed fiber surface roughness, decreased PCL crystallinity, and increased swelling with increasing drug content of 1, 2, and 4 wt %. Additionally, increasing the SN-38 concentration reduced the degradation rate of the GT. Furthermore, we hypothesize the existence of a drug content saturation point in the monoaxial fiber to explain the different drug release patterns of PG2 compared with those of PG1 and PG4. The matrix also showed good biodegradation and anti-tumor function. Our results demonstrate that SN-38-loaded PCL/GT fibers can be obtained by electrospinning. The SN-38-loaded fibers merit further evaluation as a means to potentially prevent locoregional recurrence following surgical tumor resection.

The anticancer efficacy of chemodynamic therapy (CDT) is considerably restrained by the inadequate content of H 2 O 2 within tumor microenvironment (TME), unexpected oxidation of Fenton agents and low targeting efficiency of particulate therapeutic platforms. Herein for the first time, an implantable CDT platform was designed and synthesized by combining Fe 0 nanocrystal embedded mesoporous silica nanoparticles (FeMSN) and PCL-gelatin fibres (PG fibres), denoted as [email protected] fibres. FeMSN was fabricated via in situ reduction of ferrous ions within the mesopores of MSNs, and assembled at the surface of electrospun PG fibres by electrostatic interaction to form a fibrous mesh. The findings reflected that, in an acid condition, FeMSN nanoparticles can liberate from the composite fibres, and be effectively uptaken by 4T1 cancer cells. The FeMSN released may react with oxygen to generate H 2 O 2 in the acidic microenvironment, and subsequently transform H 2 O 2 to intracellular hydroxyl radicals, inducing enhanced cell-killing effect. By implanting the composite fibres locally at the tumor site of mice, promoted anticancer efficacy was observed with comparison of the intratumoral-injected FeMSN nanoparticles. This study suggests that [email protected] composite fibres can serve as a potential localized PDT platform for effective antitumor purposes, which may spark a series of follow-on investigations for localized cancer therapeutic technology.

Delayed endothelialization, the excessive proliferation of smooth muscle cells (SMCs), and persistent inflammation are the main reasons for the implantation failure of blood-contacting materials. To overcome this problem, an inflammation-responsive, core-shell structured microfiber scaffold is developed using polycaprolactone (PCL), selenocystamine-modified gelatin (Gel-Se), L-ascorbyl 6-palmitate (AP), and dexamethasone as the fiber shell, with poly ( l -lysine) (PLL) and heparin incorporated in the fiber core. Superhydrophilic microfiber scaffolds exhibit antifouling properties that inhibit protein adsorption and blood cell adhesion, thereby effectively mitigating the risk of acute thrombosis. The continuous release of heparin and sustained generation of nitric oxide (NO) through the catalytic decomposition of S-nitrosothiols by selenocystamine lead to a biomimetic endothelial function for the enhancement of blood compatibility. The inflammation-responsive compound AP can detoxify excess reactive oxygen species (ROS) while controlling the release of dexamethasone to reduce chronic inflammation. We demonstrate the ability of microfiber scaffolds to reduce thrombotic and inflammatory complications, inhibit SMC proliferation, and promote rapid endothelialization both in vitro and ex vivo. Hence, microfiber scaffolds are robust and promising for blood-contacting implants with enhanced antithrombogenicity and anti-inflammatory capabilities.

The treatment of severely damaged wounds is a serious challenge as these wounds have poor self-healing capacity, take longer to heal, and are prone to becoming chronic wounds. To solve this problem, PCL/gelatin electrospinning nanofiber membranes were combined with rGO-loaded chitosan non-woven fabrics to prepare biomimetic bilayer skin scaffolds. Among them, the PCL/gelatin nanofiber membrane was used to simulate the epidermis layer with a dense structure, and the chitosan non-woven fabric was used to simulate the dermis layer with a loose structure. When the concentration of the spinning solution was 7 %, and the average fiber diameter was 241 nm, HaCaTs had the best cell proliferation and adhesion, and the re-epithelialization process was fast. To enhance the induction effect of scaffolds on dermal cells, the electroactive substance rGO was used to give the scaffold electrical conductivity . Considering the biocompatibility, the concentration of GO dispersion was controlled at 1 mg/mL, and the obtained [email protected] 1rGO scaffolds had high conductivity, as well as the improved mitochondrial membrane potential of HFF-1 s which increased cell viability . So the induction of HFF-1 s migration was obvious. After cross-linking composite scaffolds with glutaraldehyde , the scaffolds still showed good cytocompatibility, at the same time, the peel strength was increased from 7.21 cN to 350 cN, and the structural stability of the nanofiber membrane layer in the liquid environment was also enhanced. Both the moisture absorption and moisture permeability of composite scaffolds were improved, which could maintain the moist environment of the wound to a certain extent. Altogether, results showed that electroactive biomimetic bilayer fibrous scaffolds have great application potential in promoting skin tissue regeneration.

Localized drug delivery systems (LDDSs), in the forms of fibers or hydrogel, have emerged as an alternative approach for effective cancer treatment, but suffer challenges in the limited efficacy originated from sole therapeutic functionality. Herein, a multifunctional LDDS, showing feasibility for minimally-invasive implantation, was designed and synthesized for on-site chemo-photothermal synergistic therapy. In this system, polydopamine (PDA) nanoparticles, loaded with doxorubicin (DOX), were assembled at the surface of electrospun PCL-gelatin (PG) fibers (PG@PDA-DOX). The composite PG@PDA-DOX nanofibers could effectively transform NIR light into heat and present excellent photostability. In addition, low pH and NIR irradiation enabled remarkably accelerated DOX release. The in vitro study of PG@PDA-DOX fibers showed effective anti-cancer effect with irradiation of 808 nm NIR by inducing cell apoptosis and suppressing cell proliferation. The in vivo study, by implanting PG@PDA-DOX nanofibers in the patient derived xenograft (PDX) model via minimally-invasive surgery, presented that the composite fibers can effectively inhabit tumor growth by the combined chemo-photothermal effect without clear systematic side-effects. This study has therefore demonstrated a minimally-invasive platform, in a fibrous mesh form, with both high therapeutic efficacy and considerable potential in clinical translation for liver cancer treatment.

Reduction of inflammation and thrombosis caused by implanted devices is critical for clinical success. To this end, the strategy based on programmable release of anti-inflammatory and anti-thrombotic agents from the widely-used polycaprolactone (PCL)/gelatin nanofiber scaffold is developed. The release of 2-O- d -Glucopyranosyl- l -ascorbic Acid (AA-2G) and heparin are controlled by reactive oxygen species (ROS)-responsive poly(ethylene glycol)-based β-thioether ester copolymer (PEGDA-EDT) and mesoporous silica nanoparticles (MSN) in the nanofiber, respectively. The in vitro assay demonstrate that the scaffolds are hemocompatible with the resistance of platelet adhesion; the control release of AA-2G prevents initial inflammation and oxidation of the blood cells, and the subsequent release of heparin entitles nanofibers with long-term anti-thrombotic capability. In addition, rapid endothelialization is obtained on the surface of nanofiber scaffolds for the further enhancement of the hemocompatibility. In vivo implant evaluation convinces that the nanofiber scaffolds possess high biocompatibility with the substantial resistance for inflammation and thrombosis. Hence, our work paves a new way to develop the anti-inflammatory and anti-thrombotic tissue-engineering substrates through programmable delivery of two or multiple drugs.
