Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.

Hypoxia-Mimicking Microenvironment Scaffold for Enhanced Tendon Regeneration

Xiaoqing Cheng,Xing Yun,Yu Wei,Pengtao Shi,Ruichao He,Chen Yang,Lujian Liao,Min Wei,Qi Quan

Tendon, a connective tissue structure, serves the crucial role of transmitting force between muscles and bones. However, tendon injury repair continues to pose a significant challenge in clinical settings. In this study, we utilized single-cell RNA sequencing to delve into the cell populations and signaling pathways that are integral to tendon healing. Our findings suggest that hypoxia plays a pivotal role in activating macrophages, stimulating endothelial cell migration, and fostering fibroblast proliferation. Based on these insights, we have developed a PCL scaffold coated with DFOA, which effectively mimics a hypoxic environment to enhance tendon tissue regeneration. Furthermore, the PCL-DFOA scaffolds exhibit exceptional ability in promoting macrophage polarization and angiogenesis. This research offers a therapeutic strategy that harnesses the regenerative power of hypoxia to accelerate and optimize tendon healing processes.

Surface modification with hydrophilic and heparin-loaded coating for endothelialization and anticoagulation promotion of vascular scaffold

Yongchao Jiang,Haonan Wang,Xiaofeng Wang,Qian Li

Surface modifications are common approaches to promote endothelialization and resist thrombus, therefore obtaining long-term patency of vascular grafts. Herein, we developed a functional coating based on hyaluronic acid (HA), dopamine (DA), and heparin. In the present study, dopamine was firstly grafted to the HA molecules. The DA-grafted HA material was then applied to the surface of fibrous PCL scaffold via oxidation polymerization. Heparin was directly loaded during the coating, instead of grafting. The composite coating enhanced the surface hydrophilicity of PCL scaffold, as well as the mechanical properties. Notably, the coated scaffold could promote EC proliferation and angiogenesis behavior via the upregulation of CD31 gene expression regardless of heparin addition. It also showed more effective inhibition of platelet adhesion and blood clotting in vitro. These results lead us to the conclusion that this functional coating is great potential in treating cardiovascular diseases in terms of promoting endothelialization, reducing thrombus, and maintaining the long-term patency of vascular grafts.

Engineered Nanotopography on the Microfibers of 3D-Printed PCL Scaffolds to Modulate Cellular Responses and Establish an In Vitro Tumor Model

Linzhi Jing,Xiang Wang,Bin Leng,Ningping Zhan,Hang Liu,Shifei Wang,Yuyun Lu,Jie Sun,Dejian Huang

Scaffold-based three-dimensional (3D) cell culture systems have gained increased interest in cell biology, tissue engineering, and drug screening fields as a replacement of two-dimensional (2D) monolayer cell culture and as a way to provide biomimetic extracellular matrix environments. In this study, microscale fibrous scaffolds were fabricated via electrohydrodynamic printing, and nanoscale features were created on the fiber surface by simply leaching gliadin of poly(ε-caprolactone) (PCL)/gliadin composites in ethanol solution. The microstructure of the printed scaffolds could be precisely controlled by printing parameters, and the surface nanotopography of the printed fiber could be tuned by varying the PCL/gliadin ratios. By seeding mouse embryonic fibroblast (NIH/3T3) cells and human nonsmall cell lung cancer (A549) cells on the printed scaffolds, the cellular responses showed that the fiber nanotopography on printed scaffolds efficiently favored cell adhesion, migration, proliferation, and tissue formation. Quantitative analysis of the transcript expression levels of A549 cells seeded on nanoporous scaffolds further revealed the upregulation of integrin-β1, focal adhesion kinase, Ki-67, E-cadherin, and epithelial growth factor receptors over what was observed in the cells grown on the pure PCL scaffold. Furthermore, a significant difference was found in the relevant biomarker expression on the developed scaffolds compared with that in the monolayer culture, demonstrating the potential of cancer cell-seeded scaffolds as 3D in vitro tumor models for cancer research and drug screening.

Structurally sophisticated 3D-printed PCL-fibrin hydrogel meniscal scaffold promotes in situ regeneration in the rabbit knee meniscus

Hebin Ma,Bowen Xie,Hongguang Chen,Lifang Hao,Haigang Jia,Dengjie Yu,Yuanbo Zhou,Puzhen Song,Yajing Li,Jing Liu,Kaitao Yu,Yantao Zhao,Yadong Zhang

A meniscus injury is a common cartilage disease of the knee joint. Despite the availability of various methods for the treatment of meniscal injuries, the poor regenerative capacity of the meniscus often necessitates resection, leading to the accelerated progression of osteoarthritis. Advances in tissue engineering have introduced meniscal tissue engineering as a potential treatment option. In this study, we established the size of a standardized meniscal scaffold using knee Magnetic Resonance Imaging (MRI) data and created a precise Polycaprolactone (PCL) scaffold utilizing 3-Dimensional (3D) printing technology, which was then combined with Fibrin (Fib) hydrogel to form a PCL-Fib scaffold. The PCL scaffold offers superior biomechanical properties, while the Fib hydrogel creates a conducive microenvironment for cell growth, supporting chondrocyte proliferation and extracellular matrix (ECM) production. Physical and chemical characterization, biocompatibility testing, and in vivo animal experiments revealed the excellent biomechanical properties and biocompatibility of the scaffold, which enhanced in situ meniscal regeneration and reduced osteoarthritis progression. In conclusion, the integration of 3D printing technology and the Fib hydrogel provided a supportive microenvironment for chondrocyte proliferation and ECM secretion, facilitating the in situ regeneration and repair of the meniscal defect. This innovative approach presents a promising avenue for meniscal injury treatment and advances the clinical utilization of artificial meniscal grafts.
