Phoxim is a broad-spectrum organophosphorus pesticide widely used in the world; however, its neurotoxicity and potential risk pose a significant threat to human health and ecosystems. To address this issue, based on the same metal clusters and similar topologies, a MOF-on-MOF composite strategy, i.e., PCN222(Fe)@NU-1000, was developed for rapid evaluation of phoxim via ratiometric fluorescence and colorimetric dual-signal output. Herein, the inner core, PCN222(Fe), imparts peroxidase-like catalytic activity enabling colorimetric detection based on the 3,3′,5,5′-tetramethylbenzidine (TMB) and the solution decolourisation caused by acetylcholinesterase (AChE) products, while the outer shell, NU-1000, provides a strong fluorescence emission at 461 nm. This core-shell structure also facilitates fluorescence resonance energy transfer (FRET), whereby phoxim selectively quenches the fluorescence of NU-1000 without affecting PCN222(Fe), enabling ratiometric fluorescence sensing. This integrated dual-signal system-comprising ratiometric fluorescence and enzyme-mimetic colorimetry-ensures signal cross-verification and improved detection accuracy. The fluorescence mode exhibits a quantification range of 5.0 ng/mL to 4.0 μg/mL with a detection limit of 4.0 ng/mL, while the colorimetric mode shows a linear range from 2.5 ng/mL to 25.0 ng/mL and a detection limit of 1.76 ng/mL. Furthermore, the method accurately evaluates phoxim levels in fruits and vegetables such as apples and celery, achieving recoveries ranging from 95 % to 115 % and a relative standard deviation (RSD) of less than 9 %. This MOF-on-MOF dual-signal sensing strategy is designed to be simple, user-friendly, and highly reliable, providing significant value for the development of rapid and sensitive approaches in food and environmental science.

Here, an integrated cascade nanozyme with a formulation of Pt@PCN222-Mn is developed to eliminate excessive reactive oxygen species (ROS). This nanozyme mimics superoxide dismutase by incorporation of a Mn–[5,10,15,20-tetrakis(4-carboxyphenyl)porphyrinato]–based metal-organic framework compound capable of transforming oxygen radicals to hydrogen peroxide. The second mimicked functionality is that of catalase by incorporation of Pt nanoparticles, which catalyze hydrogen peroxide disproportionation to water and oxygen. Both in vitro and in vivo experimental measurements reveal the synergistic ROS-scavenging capacity of such an integrated cascade nanozyme. Two forms of inflammatory bowel disease (IBD; i.e., ulcerative colitis and Crohn’s disease) can be effectively relieved by treatment with the cascade nanozyme. This study not only provides a new method for constructing enzyme-like cascade systems but also illustrates their efficient therapeutic promise in the treatment of in vivo IBDs.

Copper is a common element in the environment and human body. Exposure to high concentrations of Cu2+ potentially causes health issues, such as Wilson and Alzheimer’s diseases. It is of great importance for the highly selective and sensitive detection of Cu2+. In this work, a porous metal–organic framework (MOF), PCN222, is employed for the selective and sensitive determination of Cu2+. The selectivity of PCN222 relies on the interaction between Cu2+ and the porphyrin core of PCN222. PCN222 stacked with porphyrin rings exhibits solvent adaptability, which overcomes the drawback of the natural porphyrinic enzyme (such as HRP). PCN222 is used for constructing a sensor which shows a linear range of Cu2+ concentration from 0.4 to 13 μmol L–1, and its limit of detection (LOD) is 50 nmol L–1. The response time is less than 3 s, which is faster than previous methods. This work opens up a new route to MOF applications in the detection of metal ions in complex environments.

Temporomandibular joint osteoarthritis (TMJ OA) is a degenerative joint disease with no complete cure at present. Notably, the inflammatory microenvironment in TMJ OA is modulated by oxidative stress, which impacts cartilage metabolism, chondrocyte apoptosis, inflammatory cytokine release, and extracellular matrix (ECM) synthesis. Thus, it is reasoned that reducing excess reactive oxygen species (ROS) in the chondrocyte microenvironment may be an effective therapeutic strategy for TMJ OA. Recently, cascade nanozymes, including Pt@PCN222-Mn, have been exploited to treat ROS-associated diseases. Nevertheless, cascade nanozymes are not employed for TMJ OA therapy. To fill this gap, it is explored whether the Pt@PCN222-Mn cascade nanozyme could be applied to the treatment of TMJ OA. The in vitro results demonstrate that the Pt@PCN222-Mn nanozyme can inhibit the production of inflammatory factors, the degradation of ECM, and the apoptosis of chondrocytes by inhibiting the ROS-nuclear factor kappa-B (NF- κ B_ and mitogen-activated protein kinase signaling pathways. The in vivo results further demonstrate that the Pt@PCN222-Mn nanozyme can delay the progression of TMJ OA in the rat unilateral anterior crossbite model. It is believed that insightful perspectives on the application of nanozymes in TMJ OA will be provided here.
