Based on a simple catechol-amine co-precipitation strategy and biomimetic adhesion characteristics, this study prepared a desensitized CL-20@PCPA core-shell energetic powder using the phenolic and amino compound containing low-cost polycatechol/polyamine (PCPA). The results indicate that the CL-20@PCPA composite powder exhibited an elevated characteristic drop height of impact sensitivity ( H 50 ) of 34.9 cm and a reduced friction explosion probability of 56% compared to pure CL-20. PCPA, containing catechol and amine groups, provided a stable platform for secondary modification to enhance the safety of the powder further. Notably, when the CL-20@PCPA@MXene powder was introduced into a propellant system, the resulting propellants displayed a high H 50 value of 20.3 cm while its heat of detonation remained unchanged. This study provides a simple and low-cost biomimetic interface modification strategy for preparing desensitized energetic materials.

Objective: To study the extraction process of agarwood active ingredients (AA) and investigate the safety and effectiveness of AA in the treatment of insomnia rats by nasal administration. Method: A β-cyclodextrin (β-CD) inclusion compound (a-β-CD) was prepared from agarwood essential oil (AEO), and the preparation process was optimized and characterized. The safety of AA in nasal mucosa was evaluated throughBufo gargarizansmaxillary mucosa and rat nasal mucosa models. Insomnia animal models were replicated by injectingp-chlorophenylalanine (PCPA), conducting behavioral tests, and detecting the expression levels of monoamine neurotransmitters (NE and 5-HT) and amino acids (GABA/Glu) in the rat hypothalamus. Results: The optimum inclusion process conditions of β-CD were as follows: the feeding ratio was 0.35:1.40 (g:g), the inclusion temperature was 45 °C, the inclusion time was 2 h, and the ICY% and IEO% were 53.78 ± 2.33% and 62.51 ± 3.21%, respectively. The inclusion ratio, temperature, and time are the three factors that have significant effects on the ICY% and IEO% of a-β-CD. AA presented little damage to the nasal mucosa. AA increased the sleep rate, shortened the sleep latency, and prolonged the sleep time of the rats. The behavioral test results showed that AA could ameliorate depression in insomnia rats to a certain extent. The effect on the expression of monoamine neurotransmitters and amino acids in the hypothalamus of rats showed that AA could significantly reduce NE levels and increase the 5-HT level and GABA/Glu ratio in the hypothalamus of insomnia rats. Conclusion: The preparation of a-β-CD from AEO can reduce its irritation, improve its stability, increase its curative effect, and facilitate its storage and transport. AA have certain therapeutic effects on insomnia. The mechanism of their effect on rat sleep may involve regulating the expression levels of monoamine neurotransmitters and amino acids in the hypothalamus. Keywords:agarwood;supercritical fluid extraction;β-cyclodextrin;nose-to-brain delivery;insomnia

Owing to excellent adhesion and easy process, dopamine has been widely used as an intermediate for adjusting the molecular structure of nanoparticles . However, the high cost of dopamine largely impedes its large-scale applications. Herein, alumina@poly(catechol-polyamine)@graphene oxide (denoted as Al 2 O 3 @PCPA@GO) multilayer core-shell hybrids were prepared via poly(catechol-polyamine) (PCPA) modification. The as-prepared hybrids were added into the carboxyl nitrile butadiene rubber (XNBR) latex to produce composites with high thermal conductivity because of the strong interaction between polar groups in Al 2 O 3 @PCPA@GO core-shell hybrids and C N groups in XNBR matrix. Finally, the maximum thermal conductivity (0.48 W/mK) was obtained with 30 vol% Al 2 O 3 @PCPA@GO/XNBR composite, which was about three times of pure XNBR matrix (0.16 W/mK). Moreover, 30 vol% Al 2 O 3 @PCPA@GO composite showed a high dielectric constant (16.84 at 100 Hz) and low dielectric loss tangent (0.45 at 100 Hz). Besides, the 30 vol% Al 2 O 3 @PCPA@GO/XNBR composite displayed a high tensile strength of 11.86 MPa, which is an advantage for practical applications. The developed method has a shorter reaction time and lower cost compared with poly(dopamine) coating, suggesting potential applications as thermally conductive materials for electronic equipment.

BACKGROUND Insomnia, a prevalent sleep disorder, detrimentally affects quality of life and is often challenging to manage with conventional treatments. This study delved into identifying and quantifying the main compounds by employing ultra-performance liquid chromatography–Q-Exactive-Orbitrap mass spectrometry, and further to evaluate the therapeutic potential of Rosa roxburghii folium (RRF) extract, with multiple pharmacological activities. Previous research had hinted at the efficacy of glycosides in influencing the γ -aminobutyric acid (GABAergic) system, which plays a pivotal role in sleep regulation. Utilizing a p -chlorophenylalanine-induced insomnia model in BALB/C mice, this investigation aimed to unravel the effects of various dosages of RRF extract on sleep quality and elucidated its mechanism of action. RESULTS A total of 66 compounds in the RRF extract were analyzed. Behavioral assessments demonstrated notable enhancements in sleep duration and latency. Biochemical analyses further corroborated these findings, revealing modulation in neurotransmitter levels indicative of a potential mechanism through the GABAergic and serotoninergic pathways. Additionally, histological evaluations suggested anti-inflammatory and antioxidant effects of the RRF extract. CONCLUSION The findings from this study underscored the therapeutic efficacy of RRF extract in combating insomnia, particularly highlighting its glycoside components’ role. The extract's significant improvement in sleep duration and latency, alongside its modulation of neurotransmitter levels, showcases its potential as a natural remedy for insomnia. Through its action on the GABAergic and serotoninergic pathways, as well as its anti-inflammatory and antioxidant effects, RRF extract emerges as a promising candidate for insomnia treatment, offering a holistic approach to sleep disorder management. © 2024 Society of Chemical Industry.

Delayed treatment of insomnia-related symptoms can harm physical health and increase the psychological burden. In addition to oral medications and some physical therapies, aromatherapy can help overcome some treatment-related side effects. Herein, parachlorophenylalanine (PCPA)-induced insomnia was established in Kunming (KM) mice, which were subjected to aromatherapy using five plants ( Jasminum sambac , Magnolia denudata , Rosa rugosa , Aloysia citriodora , and Abies balsamea ) essential oils (EOs). To determine the sleep-inducing effect of the five EOs, the rate of change in body weight, sleep latency, and total sleep time in mice were measured. Specific serum indices were used to evaluate the therapeutic effects of the tested drugs and PCPA modeling. Gas chromatography-mass spectrometry (GC-MS) was employed to identify active components in EOs. The five EOs contained multiple identical constituents and were rich in terpenoids, such as α-farnesene (28.42%), linalool (68.84%), and citronellol (23.78%). The EOs exhibiting varying effects on insomnia-induced weight loss. Nissl staining was used to examine and the number of neurons was elevated in the EO-treated groups when compared with the PCPA-induced group; however, the neuronal number was reduced in the hypothalamic tissues of the R. rugosa EO (RREO)-treated group. All EOs upregulated the expression of 5-HT1A and GABA A Rα1, as demonstrated by immunohistochemistry, western blotting, and reverse transcription-quantitative PCR results. In addition, EOs of A. citriodora and A. balsamea significantly upregulated the expression of 5HT 1A protein, whereas EOs of J. sambac and M. denudata exerted significantly different effects when compared with the model group, as determined by western blotting.

As a “green” radiation protection material with the potential to replace lead oxide, bismuth oxide has more research applications in the field of γ protection. However, due to their different surface properties, there are compatibility and interaction problems between metal oxides and rubber. To enhance the performance of bismuth oxide in neoprene, Bi 2 O 3 @PCPA-KH590 was prepared by coating bismuth oxide with polyphenol/polyamine and grafting KH590 on the surface of bismuth oxide. This allows for creating more active sites with the help of polyphenol/polyamine, enabling KH590 to be grafted onto bismuth oxide particles. The S functional group on the silane coupling agent is then cross-linked with the rubber substrate, enhancing the dispersibility of the bismuth oxide powder. This improvement not only improves the dispersibility of the bismuth oxide powder but also enhances the mechanical properties of the rubber composite material. Compared with pure CR, the mechanical properties of 60 wt% filler-modified powder composites can still be maintained at a high level, with the tensile strength reaching 12.83 Mpa. In sum, the proposed method appears feasible for preparing highly filled radiation protection rubber-based materials.

In order to obtain high dielectric properties nitrile rubber (NBR) dielectric elastomer composites, in this work, polypyrrole (PPy) was encapsulated on the surface of SrTiO 3 (STO) ceramic particles, and an insulating layer of poly (catechol/polyamine) (PCPA) was also covered on the outer surface of the STO-PPy core-shell particles (denoted as STO-PPy-PCPA particles), then the dielectric elastomer composites were prepared by adding STO and STO-PPy-PCPA to NBR, respectively. Due to the electrical conductivity of PPy, the dielectric constant of the STO-PPy-PCPA particles increases substantially. At the same time, the insulating layer of PCPA further prevents current leakage from the composite, which not only avoids premature electrical breakdown but also effectively reduces the dielectric loss. Compared with pure NBR, the dielectric constant of the prepared dielectric elastomer composites increased from 13 to 17.19 (20 phr STO-PPy-PCPA/NBR), and the actuated strain increased from 3.28 % to 13.86 % (5 phr STO-PPy-PCPA/NBR), which is 4.2 times that of pure NBR. This work successfully solves the current problems of requiring a large amount of ceramics to improve the dielectric constant and the tendency of conductive particles to agglomerate and premature breakdown. Graphical In this work, high performance NBR dielectric elastomer composites are obtained by synthesising the core-shell structure STO-PPy-PCPA and the actuated strain is enhanced from 3.28 % (pure NBR) to 13.86 (5 phr STO-PPy-PCPA/NBR).

With the increasing prevalence of insomnia-related diseases, the effective treatment of insomnia has become an important health research topic. Lavender (Lavandula angustifoliaMill.) essential oil (LEO) is a commonly used medicine for the treatment of insomnia and neurological disorders. However, neither the active components nor its sedative–hypnotic mechanism have been fully discovered. This study aimed to screen the main active terpenes and discover the possible mechanism of LEO through network pharmacology in the treatment of insomnia-related diseases, as well as to verify our hypothesis in insomnia mice. The results showed that, in LEO’s 15 potential active ingredients, beta-myrcene had strong sedative–hypnotic effects through the serotonergic synaptic pathway according to the network pharmacological prediction. Further, PCPA(DL-4-chlorophenylalanine)-induced insomnia mice were treated with beta-myrcene for one day or seven days. The quiet state of insomnia mice was increased effectively, and the hypnotic effect was enhanced by anaobarbital sodium by prolonging sleep duration, decreasing sleep latency, and increasing the rate of falling asleep. Beta-myrcene reduced the damage to hypothalamic neuron cells induced by PCPA and increased neurotransmitter levels of GABA, 5-HT, and Glu in the serum and hypothalamus of insomnia mice. Meanwhile, beta-myrcene exerted an improvement in insomnia by upregulating relevant genes and protein expression in the serotonergic synaptic pathway. These results support the merit of the sedative–hypnotic activity of LEO. Beta-myrcene, a terpene in LEO, may be the main source of its sedative–hypnotic properties. It may serve as a good potential compound in future clinical studies on coping with insomnia.

Cannabis sativa has been used for improving sleep for long history. Cannabidiol (CBD) has drown much attention as a non-addictive psychoactive component in Cannabis sativa extract. However, the effects of CBD on sleep architecture and it’s acting mechanism remains unclear. In the present study, we evaluated the sedative-hypnotic effect of cannabidiol (CBD), assessed the effects of CBD on sleep using a wireless physiological telemetry system. We further explored the therapeutic effects of CBD using 4-chloro-dl-phenylalanine (PCPA) induced insomnia model and changes in sleep latency, sleep duration and intestinal flora were evaluated. CBD shortened sleep latency and increases sleep duration in both normal and insomnia mice, and those effects were blocked by 5-HT 1A receptor antagonist WAY100635. We determined that CBD increases 5-HT 1A receptors expression and 5-HT content in the hypothalamus of PCPA-pretreated mice and affects tryptophan metabolism in the intestinal flora. These results showed that activation of 5-HT 1A receptors is one of the potential mechanisms underlying the sedative-hypnotic effect of CBD. This study validated the effects of CBD on sleep and evaluated its potential therapeutic effects on insomnia.

Insomnia remains a major public health challenge with limited safe treatment options. We demonstrate that mixed probiotics Lactobacillales ( Lactobacillus helveticus L551, Lacticaseibacillus paracasei L578, and Streptococcus thermophilus S709, group P) alleviates insomnia in PCPA-induced mice. P significantly reduced sleep latency and increased sleep duration, while ameliorating anxiety behaviors. The intervention attenuated insomnia-induced colonic structural damage and reactive oxygen species (ROS) accumulation, while restoring hippocampal CA1sp and hypothalamic Nissl body density, as well as normalizing brain glucose metabolism. Crucially, P intervention rebalanced neurotransmitter networks by enhancing serotonin (5-HT) synthesis while suppressing its catabolism, and reducing dopamine (DA) and norepinephrine (NE) levels. The P intervention significantly increased the abundance of Akkermansia and Acinetobacter compared to the insomnia model group (M). Notably, Acinetobacter exhibited significant positive correlations with N-acetylserotonin and gamma-aminobutyric acid (GABA) levels, while showing a significant negative correlation with 5-hydroxyindoleacetic acid (5-HIAA). These findings suggest its potential capacity to modulate tryptophan metabolism. In the M group, Staphylococcus was identified as the signature taxon, showing significant positive correlations with sleep latency and DA levels, and significant negative correlations with sleep duration and serotonin 5-HT levels. P intervention significantly reduced Staphylococcus abundance, concomitant with DA level normalization. In summary, Lactobacillales improve sleep quality and relieve anxiety in insomnia mice via modulating gut microbiota composition, thus regulating tryptophan metabolic pathways and sleep - related neurotransmitters.

Jujuboside A (JUA) is a saponin abundant in sour jujube seeds ( Ziziphus jujuba var. spinosa ). However, whether JUA improves insomnia and the consequent anxiety-like behaviors remains unclear. In this study, we evaluated the effects of JUA in para-chlorophenylalanine (PCPA)-induced insomnia in mice. JUA administration alleviated sleep disturbances and anxiety-like behaviors in PCPA-treated mice. PCPA reduced tight junction protein expressions, and altered gut microbiota composition, whereas JUA partially reversed these alterations. JUA increased the expression of tight junction proteins, especially claudin-1, providing structural and molecular evidence of preserved intestinal barrier-associated features. It also modulated the gut microbiota, as reflected by increased Bacteroidota and decreased Desulfobacterota . Along with these barrier-associated changes, JUA reduced systemic inflammation and oxidative stress. Furthermore, serum untargeted metabolomics analysis revealed that PCPA disrupted neuroactive metabolite profiles and amino acid metabolism-related pathways, whereas JUA partially reversed these metabolic alterations. Correlation analysis linked JUA-responsive metabolites with sleep parameters and anxiety-like behaviors. JUA corrected PCPA-induced central neurotransmitter imbalance and upregulated the expression of BDNF/TrkB. These findings collectively indicate that JUA has the ability to alleviate insomnia and anxiety-like behaviors induced by PCPA, supporting its potential as a bioactive compound from food sources in the management of sleep disorders and related emotional symptoms in diet.

Objectives:Insomnia is a highly prevalent sleep disorder involving complex neurochem-ical dysregulation; however, the spatial distribution of neurotransmitters and small-molecule metabolites across distinct brain microregions in the insomniac state re-mains poorly characterized. This study aimed to map region-specific metabolic perturba-tions in situ in a p-chlorophenylalanine (PCPA)-induced insomnia rat model using opti-mized matrix-assisted laser desorption/ionization mass spectrometry imaging (MAL-DI-MSI) integrated with targeted metabolomics validation.Methods:On-tissue chemical derivatization MALDI-MSI was optimized using α-cyano-4-hydroxycinnamic acid (CHCA) as the matrix on a Bruker tims TOF flex mass spectrometer. The rats received PCPA (400 mg/kg, intraperitoneal) for three days to establish the insomnia model. Metabolite identifi-cation was conducted using MetaboScape® software (2020b) and the Human Metabolome Database. Ultra-performance liquid chromatography–tandem mass spectrometry was em-ployed to quantify nine key neurotransmitters and metabolites across six brain microre-gions. Key synthetic enzyme expression was evaluated by immunofluorescence and West-ern blotting analyses.Results:TMP-TFB-derived brain slices clearly showed the distribu-tion of neurotransmitters in brain microregions. MALDI-MSI demonstrated that the spatial distribution and abundance of eight neurotransmitters were disturbed in brains of PCPA-induced insomniac rats. A total of 346 metabolites were characterized across six brain microregions (cerebellum, cortex, hippocampus, brainstem, hypothalamus, and stri-atum), with principal coordinate analysis revealing clear metabolic separation between control and insomniac rats. PCPA treatment markedly disrupted tryptophan and tyrosine metabolism, evidenced by decreased 5-HT, 5-HTP, and 5-HIAA, alongside region-specific alterations in DA, NE, HVA, and Ach. Additionally, GABA levels decreased in the hippo-campus, striatum, and hypothalamus, whereas glutamate increased throughout the brain. Targeted metabolomics validated the MSI findings, and Bland–Altman analysis confirmed good consistency between the two analytical platforms. PCPA further disturbed the meta-bolic enzymes MAOA, DDC, and TPH2 within the Trp-5-HTP-5-HT-5-HIAA metabolic pathway in the brainstem and TYH and DBA within the tyrosine-DA-NE metabolic pathway in the striatum. Conclusions:This study demonstrates that region-specific altera-tions in tryptophan and tyrosine metabolism pathways provide mechanistic insights into insomnia pathogenesis and potential therapeutic targets.
