Tetracycline, an animal antibiotic, may remain in milk to cause harm to human health. For economic reasons, the abuse of antibiotics is becoming more and more common. Therefore, the abuse of tetracycline has alarmed the dairy industry and many countries such as New Zealand, China, and the USA have proposed strict standards. Surface-enhanced Raman scattering (SERS) is an emerging detection method which has been applied in food detection with the advantages of no complex pretreatment, fast detection, and weak water environment interference. Considering the abuse of antibiotics in dairy industry, we used polydimethylsiloxane (PDMS) plasma cavity as SERS substrate to detect tetracycline in milk. We found that the enhancement ability of PDMS substrate is affected by addition of 4-amino-1-butanol and complex interplay in the milk--tetracycline system. The modified PDMS plasma cavity has high SERS sensitivity that allows us to achieve low detection limit of 0.28 μg/L. The correlation coefficient was 0.987. The detection of tetracycline in milk using PDMS substrate is quick (within 10 min) and it provides a possible method for in-site detection of tetracycline.

Superhydrophobic substrate is applied in matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) detection due to its confinement effect. The weak interaction of superhydrophobic surface with water/salts makes it potential in one-step enrichment and desalting of peptide in MALDI MS analysis. We fabricate a superhydrophobic substrate by spin-coating poly(dimethyl siloxane) (PDMS) on a candle soot layer. On this substrate, the peptide analytes can be confined and enriched in a small area due to the confinement effect and its strong hydrophobic interactions with PDMS. Meanwhile, the desalting can be easily realized by removing the residual solution after the absorption of analyst molecules due to the weak interaction between water/salt contaminants and the superhydrophobic surface. Using this substrate, angiotensin III (Ang III) in the presence of salt with high concentration (2 M or saturated) can be analyzed, and the peptide sequence coverage of 10 μg/mL myoglobin (MYO) and bovine serum albumin (BSA) digests is enhanced to 51% and 26%, which is 37% and 21% analyzed with the commercial ZipTipC 18 pipette tips. The LOD of bacitracin A (Bac A) in milk with this substrate is 100 pM and nearly 360 times lower than the LOD of standard testing method. This substrate has potential practical applications in proteomics research and actual sample analysis.

An aptamer-based sensitive method was developed here for detection of multiple foodborne pathogens in food matrix by surface-enhanced Raman scattering (SERS) technology. Polydimethylsiloxane (PDMS) film was first prepared and then coated with gold nanoparticles (AuNP) to act as an active substrate for the enhancement of Raman scattering. The as-prepared Au-PDMS film was functionalized with specific pathogen aptamers (Apt) to capture the targets. In addition, aptamers functionalized AuNP integrated with Raman reporters (4-Mercaptobenzoic acid (4-MBA)/Nile blue A (NBA)) were fabricated as pathogen-specific SERS probes. In this scheme, pathogens were first captured by Apt-Au-PDMS film and then bind with SERS probes to allow the formation of a sandwich assay to complete the sensor module for the detection of multiple pathogens. With Vibrio parahaemolyticus and Salmonella typhimurium as model targets, this protocol can selectively detect 18 cfu/mL and 27 cfu/mL, respectively. Furthermore, this platform can be successfully applied to detect pathogens in seafood samples with recoveries ranging from 82.9% to 95.1%.

Surface-enhanced Raman scattering (SERS) technology has been widely used in the field of environmental pollutant detection due to its fast and intuitive characteristics and low water interference. Herein, a low-cost and flexible SERS substrate was fabricated by transferring gold nanoparticles (NPs) onto polydimethylsiloxane (AuNPs@PDMS) membranes, which enabled highly sensitive detection of aromatic amine pollutants in wastewater after fire suppression. Finite time domain-difference calculations confirmed that the uniformly distributed NP gap significantly enhanced the localized electromagnetic field. The AuNPs@PDMS substrate performed satisfactorily in testing for rhodamine 6G (R6G) with a detection limit (LOD) of 5.5 × 10 −10 M. This substrate also demonstrated high reproducibility for detection of different concentrations of R6G and the relative standard deviation (RSD) was <7 %. These excellent SERS properties made AuNPs@PDMS a promising substrate in the detection of aniline and benzidine. Quantitative analysis of aniline and benzidine was achieved with LODs of 4.5 × 10 −9 and 7.8 × 10 −9 M, respectively. In addition, this substrate could be applied in rapid analysis of mixtures in wastewater after fire suppression using SERS without any complex pretreatments, displaying a great potential for rapid, high-sensitivity, and on-site detection of contaminants in fire-fighting sites.
