In situ embedding of glucose oxidase in amorphous ZIF-7 with high catalytic activity and stability and mechanism investigation
Glucose oxidase (GOx) has a great application potential in the determination of glucose concentration. However, its sensitivity to the environment and poor recyclability limited its broader application. Herein, with the assistance of DA-PEG-DA, a novel immobilized GOx based on amorphous Zn-MOFs (DA-PEG-DA/ [email protected] /PDA) was developed to impart excellent properties to the enzyme. SEM, TEM, XRD, and BET analyses confirmed that GOx was embedded in amorphous ZIF-7 with ∼5 wt% loading. Compared with free GOx, DA-PEG-DA/ [email protected] /PDA exhibited enhanced stability, excellent reusability, and promising potential for glucose detection. After 10 repetitions, the catalytic activity of DA-PEG-DA/ [email protected] /PDA can maintain 95.53 % ± 3.16 %. In understanding the in situ embedding of GOx in ZIF-7, the interaction of zinc ion and benzimidazole with GOx was studied by using molecular docking and multi-spectral methods. Results showed that zinc ions and benzimidazole had multiple binding sites on the enzyme, which induced the accelerated synthesis of ZIF-7 around the enzyme. During binding, the structure of the enzyme changes, but such changes hardly affect the activity of the enzyme. This study provides not only a preparation strategy of immobilized enzyme with high activity, high stability, and low enzyme leakage rate for glucose detection, but also a more comprehensive understanding of the formation of immobilized enzymes using the in situ embedding strategy.
A novel photonic crystal hydrogel inverse opal fluorescence enhancement platform for highly sensitive immunoassay of tumor markers
Cancer is a significant global health issue, necessitating early detection for effective diagnosis and treatment. We presented a novel method for detecting tumor markers using fluorescence-enhanced photonic crystal hydrogel inverse opal (PC-GELIO) membranes. We efficiently fabricated large-area, low-crack photonic crystal membranes using a homemade microfluidic chip. Composed of PEG-DA and PEG, the hydrogel replicated the photonic crystal structure, yielding photonic crystal hydrogel inverse opal membranes with a unique photonic band gap and inverse opal porous structure. These properties afford the PC-GELIO platform several advantages: Firstly, the high stability of the internal nanostructure in the large-area, low-crack photonic crystals film, prepared using the homemade microfluidic chip, is crucial for preserving the platform's exceptional fluorescence-enhanced sensing capabilities. Secondly, the peak wavelength emitted by fluorescent molecules aligns with the photonic band gap (PBG), significantly enhancing the fluorescence signal through the low-photon effect. Lastly, the high specific surface area provides numerous binding targets for biomolecule modification, and the hydrogel offers a uniform liquid environment conducive to biomolecule binding. We evaluated the fluorescence-enhanced sensing performance of the platform using alpha-fetoprotein (AFP). The limit of quantification (LOQ) for AFP was determined to be as low as 23 pg/mL, representing an order of magnitude improvement compared to non-enhanced platforms (0.29 ng/mL and 0.25 ng/mL). Consequently, this novel fluorescence-enhanced photonic crystal hydrogel inverse opal platform holds significant potential as a straightforward and efficient method for biomolecular detection.
High performance printed organic electrochromic devices based on an optimized UV curable solid-state electrolyte
Manufacturing cost is a major concern for electrochromic device (ECD) applications in smart windows for energy saving and low-carbon economy. Fully printing instead of a vacuum-based chemical vapor deposition (CVD) process is favored for large-scale fabrication of ECDs. To adapt to the screen printing process, a UV curable solid-state electrolyte based on lithium bis(trifluoromethane-sulfonyl) imide (LiTFSI) was specially formulated. It contains poly(ethylene glycol) diacrylate (PEG-DA), LiTFSI, water, and ethyl acetate. The optimized ECDs have achieved a 0.6 s bleaching time at 0.6 V and a 1.4 s coloring time at −0.5 V. The ECDs also exhibited excellent stability, which could endure 100 000 cycles of color switching while still maintaining 35% of transmittance change at a 550 nm wavelength. A demo ECD has been fabricated with a screen printed electrolyte, exhibiting stable switching between the clear state and patterned color state.
Pt Single-Atom Catalyst on Co3O4 for the Electrocatalytic Detection of Glucose
Developing an accurate, reliable, and stable glucose detection sensor poses a formidable challenge for the food industry. In this study, hydrothermal and wet chemical methods were employed to fabricate a Pt single-atom catalyst on Co3O4 (Co3O4/Pt SAs), where dispersed Pt atoms collaboratively catalyze the electrochemical reaction on the electrode surface. Subsequently, the integration of the microelectrode with polyethylene glycol diacrylate (PEG-DA) hydrogel and a dual-channel microfluidic chip facilitated continuous and rapid glucose detection. The addition of 5% PEG-DA hydrogel served to eliminate interference and expedite glucose detection, while the design of the dual-channel microfluidic chip generated microscale vortices, providing ample reaction opportunities for the target glucose while preventing interference from the mixing of previous and current reaction solutions. The research exhibited a favorable linear detection range from 1 to 800 μM, with a detection limit of 0.84 μM. Furthermore, validation of the device’s feasibility in real samples demonstrated a glucose recovery rate ranging from 94.9 to 104.4% in beverages.
