PFASs May Risk Alzheimer’s Disease

Wang He-Ling,Fang Evandro F

We are now living in an aging society with increasingly older population, many of whom are suffering with or are at a high risk of developing dementia (1,2). Alzheimer’s disease (AD) is the world’s most common type of dementia, affecting about 50 million individuals worldwide currently, with that number set to rise to 82 million by 2023. There is a huge challenge in developing an anti-AD drug, and an enormous amount of effort has been devoted to this branch of research. It is encouraging that the 2 recently described anti-Aβ antibodies, lecanemab and donanemab, show clear benefits in slowing cognitive decline in patients with early-stage disease; however, rare but severe side effects (such as brain bleeding and swelling) still occur (10.1126/science.adj8110), indicating that there is likely a longway to go in developing a cure for AD (3,4).Although the hunt is on to develop an anti-AD drug to stop disease progression, it is also important and necessary to focus on prevention. It has been reported that lifestyle patterns and environmental toxins (natural and anthropogenic) increase the risk of dementia, with some estimates noting up to a 30% increase in AD risk. These factors are modifiable and reducing such risks could prevent or slow down the onset of dementia (5,6). Environmental toxins cover a broad spectrum of chemicals, including pesticides (organochlorine and organophosphate insecticides), toxic metals (such as aluminum), air pollutants, and especially industrial chemicals (6). Although we are exposed to trace amount of these contaminants during our daily lives, a lifelong accumulation could presumably reach to a toxic level, jeopardizing neuronal function and brain health.Per- and polyfluoroalkyl substances (PFASs) are a group of man-made chemicals that have been widely used in consumer products around the world since the 1950s. They have been very widely used and closely linked to our daily lives, being used, for example, on fabrics to provide stain resistance, and on cookware to keep food from sticking to it. While PFAS compounds facilitate our lives, they have brought environmental challenges with them, mainly due to their broad application, vast numbers (there are thousands of fluoroalkyl chemical compounds), and due to a chain of linked carbon and fluorine atoms that result in very slow breakdown over time, a long lifespan (7). Due to its widespread use, humans have been widely exposed to these chemicals, many of which can now be found in blood and cerebrospinal fluid (CSF) samples. In combination with data from laboratory animals, it is suggested these chemicals may cause a series of health issues, as summarized in Figure 1 (7,8).In this issue, Delcourt and colleagues (9) present a small human study showing patients with high CSF PFASs correlate with AD. Here, using the CSF samples from 8 patients hospitalized with suspected normal pressure hydrocephalus, they checked levels of 18 of the most common PFAS molecules. PFASs were detected in all 8 CSF samples, and a correlation was seen between high PFOS (perfluorooctane sulfonate) and cognitive impairment among the tested individuals. This discovery brings with it a sense of urgency in reducing exposure to PFASs in our daily lives and highlights the risks that it may bring in terms of our brain health. As this is a small-size proof-of-concept study, further large-size clinical validation and underlying molecular investigation are needed.E.F.F. is supported by Cure Alzheimer’s Fund (#282952), HELSE SØR-ØST (#2020001, #2021021, #2023093), the Research Council of Norway (#262175, #334361), Molecule AG/VITADAO (#282942), NordForsk Foundation (#119986), the National Natural Science Foundation of China (#81971327), Akershus University Hospital (#269901, #261973, #262960), the Civitan Norges Forskningsfond for Alzheimers sykdom (#281931), the Czech Republic-Norway KAPPA programme (with Martin Vyhnálek, #TO01000215), the Rosa sløyfe/Norwegian Cancer Society & Norwegian Breast Cancer Society (#207819), and Wellcome Leap (#104617).E.F.F. is the co-owner of Fang-S Consultation AS (Organization number 931 410 717); he has an MTA with LMITO Therapeutics Inc. Effect of coexisting Cd(Ⅱ) and As(V) on anionic PFASs sorption in soils: Models and mechanisms

Wei Zhang,Jianwei Li,Ruihua Huang,Xueqi Zhang,Yuting Wang,Dongmei Zhou,Qiming Xian

An in-depth understanding of the sorption behaviors of per- and polyfluoroalkyl substances (PFASs) in soil is essential to assess their environmental risks accurately. Due to chemical industry production and waste treatment, co-contamination soil of heavy metals (HMs)-PFASs has become a public concern worldwide. This study investigated soil sorption behaviors of PFASs including perfluorooctanoic acid (PFOA), perfluorooctanesulfonic acid (PFOS), and perfluorohexanesulfonic acid (PFHxS). A multiple linear regression (MLR) model was developed to predict the sorption of PFOS in soil. Validation results demonstrated that this model could effectively predict the distribution coefficients ( K d ) of PFOS based on soil organic carbon (OC), silt, clay, and free Fe/Al-oxide contents, exhibiting a strong predictive ability ( r 2  = 0.942, p  < 0.001). In six soils, HMs (Cd 2+ and As 5+ ) influence three anionic PFASs sorption primarily by altering the electrostatic and hydrophobic interactions between soil components and PFASs. The K d values of PFOS tend to rise with increasing Cd 2+ concentration but decline with increasing As 5+ concentration. In contrast, HMs have a relatively minor influence on the sorption of PFOA and PFHxS. Moreover, a nonlinear model was constructed for the first time to quantify the impact of HMs on PFASs sorption. The model achieves exceptional prediction accuracy when applied to both experimental data from this study and literature data. A comprehensive understanding of PFASs sorption behavior in soil under conditions of coexisting HMs is of great significance for formulating targeted degradation and mitigation strategies for co-contaminated sites.

Effects of per- and polyfluoroalkyl substances (PFASs) on hatching of Folsomia candida (Collembola) on soil

Liang Fang,Li Zhu,Wang Zinan,Ke Xin,Wu Longhua,Christie Peter,Luo Yongming

Egg hatching of the soil collembolan Folsomia candida and the effects of per- and polyfluoroalkyl substances were investigated. New and effective laboratory methods for egg hatching studies with soil collembolans were established. Per- and polyfluoroalkyl compounds (PFASs) have been used industrially worldwide and are persistent organic pollutants in many soils. Twenty eggs laid by synchronized adults of the collembolan Folsomia candida were added to each Petri dish containing compressed soil substrate mixed with perfluorooctanoic acid (PFOA), heptafluorobutyric acid (PFBA), or 6:2 chlorinated polyfluoroalkyl ether sulfonic acid (F-53B), and after 25 d of exposure the number hatched declined on average by 6.9%–49.7%, 10.3%–24.1%, and 3.4%–18.6%, respectively. PFASs delayed the peak of hatching by one day, and at different concentrations reduced the number of eggs hatched during the peak by 16.7%–30% and 23.3%–43.2% in PFOA and PFBA treatments, respectively. In the presence of F-53B the number of eggs hatched declined by 73.3% but the number of individuals increased by 29.3% at higher concentrations. The characteristics of egg hatching were stable and sensitive to PFASs, and may be suitable for use as indicators in the screening of contaminated soils for environmental risk assessment.

Occurrence and distribution of per- and polyfluoroalkyl substances (PFASs) in human livers with liver cancer

Ying Liu,Nan Lin,Cao Dai,Jianliang Xu,Yi Zhang,Mingxing Xu,Fei Wang,Yanjie Li,Da Chen

Per- and polyfluoroalkyl substances (PFASs) are anthropogenic compounds that are widely accumulated in human tissues, and the liver is considered a primary target organ for PFASs exposure. The occurrence and distribution of 21 PFASs in liver tissues with tumors (n = 55) and without tumors (n = 55) are investigated in this study. Eleven perfluorinated carboxylic acids (PFCAs) and five perfluorinated sulfonic acids (PFSAs) were detected at high frequencies (45.5%–100 %), while the detection frequencies of five perfluoroalkyl phosphate (PFPAs) were relatively lower (≤29.1 %). PFSAs and PFCAs accounted for up to 82.5%–92.7 % of the total PFASs. Although it was not found to be statistically significant, the concentrations of the total PFASs were slightly higher in the tumor liver samples (mean 64.3, range 5.70–303 ng/g) than those in the non-tumor liver samples (mean 62.7, range 4.08–240 ng/g).The perfluorooctanoic acid (PFOA), perfluorotridecanoic acid (PFTrDA), and perfluorobutanesulphonate (PFBS) showed significant differences (p < 0.05) between the tumor and non-tumor liver samples, and the different distribution levels of these three PFASs may have been a consequence of oxidative stress . The total concentrations of PFASs in the three age groups were in the decreasing order of middle-aged people (45–60) > old people (>60) > young people (<45). The PFASs in females were generally lower than in males, which may have been related to women's special excretion methods (such as childbirth and breastfeeding). The results should be valuable for further mechanistic studies regarding the toxic effects of PFASs in human livers.

Enhancing electrochemical reductive defluorination of PFASs using ZIF-67 modified cathode: Mechanistic insights and performance optimization

Xing Zhang,Han Wu,Yue Wang,Yafei Wang,Yangyang Deng,Yanping Zhu,Shengsen Zhang,Huub Rijnaarts,Harry Bruning,Junhao Qin,Qingqi Lin,Zhuobiao Ni,Rongliang Qiu

The effectiveness of electrochemical reductive defluorination is impeded by the low environmental concentrations of per- and polyfluoroalkyl substances (PFASs) and the robust nature of C − F bonds. In this work, we investigate the zeolitic imidazolate framework-67 (ZIF-67) as a promising catalyst for PFASs remediation. We show that ZIF-67 hold promise for simultaneous adsorption and reductive defluorination of 2-(trifluoromethyl) acrylic acid (TFMAA). Following a 48-hour treatment, the ZIF-67 modification significantly improves the adsorption of TFMAA onto the cathode surface, achieving removal and defluorination efficiencies of 99.66 % and 97.16 %, respectively (C 0 = 5 mg L −1 , applied voltage of −1.2 V vs. Ag/AgCl). Furthermore, the atomic H*, catalyzed by the Co−N 4 structure, boosts the reductive defluorination of TFMAA through indirect mechanisms, while the nitrogen loss due to carbonization further enhances the catalytic defluorination efficiency. This research presents a novel electrode material for PFASs defluorination and offers mechanistic insights into the defluorination process.

Efficient removal of per- and polyfluoroalkyl substances (PFASs) from stored rainwater by composite metal salt /polydimethyldiallylammonium chloride coagulants

Junhao Huang,Gangfu Song,Mingming Hao,Yitian He,Haotian Hao,Ruihua Li,Baoyou Shi,Xin Huang

ABSTRACT Stored rainwater, the primary source of drinking water in the villages and towns of the Loess Plateau in northwest China, has been found to contain per- and polyfluoroalkyl substances (PFASs) and lacks necessary treatment measures. Coagulation is a common water treatment process, and enhancing its efficacy in removing PFASs can significantly improve treatment efficiency, reduce costs, and minimize the environmental and health risks associated with perfluorinated compounds. This study investigated the removal efficiency of perfluorobutanoic acid (PFBA), perfluorobutanesulfonic acid (PFBS), perfluorooctanoic acid (PFOA), and perfluorooctanesulfonic acid (PFOS) using inorganic salt coagulants alone and in combination with polydimethyldiallylammonium chloride (PDMDAAC). The results indicated that the removal efficiencies of the four PFASs by polyferric chloride (PFCl) and polyaluminum chloride (PACl) increased with alkalinity. PDMDAAC significantly enhanced the coagulation removal efficiency of the four PFASs. The removal efficiency of the four PFASs was highest when the raw water pH was near 7. Within the molecular weight range of 0 to 500,000 for PDMDAAC, the removal efficiency of the four PFASs increased with increasing molecular weight. Charge neutralization is the primary coagulation mechanism for the removal of anionic PFASs. Therefore, this study provides guidance for selecting coagulants to remove PFASs from stored rainwater.

Insight into the uptake and translocation of per- and polyfluoroalkyl substances in hydroponically grown lettuce

Wang Wenfeng,Yuan Shu,Kwon Jung-Hwan

The prevalence of per- and polyfluoroalkyl substances (PFASs) in agricultural soils has raised concerns regarding the health risks associated with the consumption of PFAS-contaminated agricultural products. The present study investigated the uptake and translocation of nine PFASs in lettuce using a hydroponic setting. During the uptake experiments, long-chain PFASs (≥ C8) exhibited greater accumulations in lettuce roots, while short-chain PFASs (≤ C7) manifested preferential transport to the shoots. The average root concentration factors of PFASs were positively correlated with their log K ow values. A significantly negative relationship was found between the average translocation factors of PFASs and their molecular volume. Sorption of long-chain PFASs by lettuce roots was enhanced after heating the roots to increase the cell membrane permeability. The accumulation of perfluorododecanoic acid increased significantly in shoots of lettuce plants without roots as compared to whole lettuce plants. Results of the present study indicate that sorption to root surface tissues and efficiency in passing through the root Casparian strip are two important factors that affect the uptake and distribution of PFASs within plants.

Molecular mechanisms of per- and polyfluoroalkyl substances (PFASs) removal from sludge alkaline fermentation liquid by coagulation: Role of interactions between DOM and PFASs

Chengyu Gu,Sijia Ma,Yuan Lin,Yi Xue,Hongqiang Ren,Ke Xu

Per- and polyfluoroalkyl substances (PFASs), as highly recalcitrant pollutants, tend to accumulate in sludge, posing challenges for subsequent treatment and resource recovery. Sludge alkaline fermentation (SAF) produces sludge alkaline fermentation liquid (SAFL) with potential for circular-economy reuse, while simultaneously transferring PFASs from the solid phase into SAFL, thereby creating a trade-off between resource recovery and contamination. Coagulation is a critical step in SAFL separation, yet the fate of PFASs during this process remains poorly understood. This study evaluated the performance of polyaluminum chloride (PAC) and polyferric sulfate (PFS) in removing nine representative legacy and emerging PFASs from SAFL. High removal efficiencies were achieved across PFASs with varying chain lengths and headgroups, with long-chain PFASs (C ≥ 8) reaching up to 74% and emerging short-chain PFASs (C < 8) up to 55%. Spectroscopic analysis indicated that the hydrophobic interaction between PFASs and the Al/Fe based hydrolysates were the primary driving force for the direct removal of PFASs by coagulants. Dissolved organic matter (DOM) in SAFL played a facilitating role in PFASs removal, with proteins identified as the key contributors. DOM analysis revealed that additional coagulants facilitated the removal of protein-like substance and hydrophobic macromolecules that have strong affinities for PFASs. Molecular dynamics simulations suggested that Al/Fe-based hydrolysates increased electrostatic attraction and binding free energy, thereby increasing protein compactness and promoting co-precipitation removal of PFASs. These findings offer valuable insights for optimizing coagulation strategies in practical applications to effectively mitigate ecological risks in DOM-rich water contaminated with PFASs.

PFASs pose a threat to deep-sea cold seeps

Li Tang,Jing-Chun Feng,Rui Hou,Nuo Chen,Can-Rong Li,Si Zhang

Inputs of per- and polyfluoroalkyl substances (PFASs) threaten deep-sea cold seep ecosystems, yet their environmental behavior and risks remain poorly understood. We report a comprehensive investigation of 30 PFASs in seawater and sediment from South China Sea cold seeps. Emerging PFASs, notably 2,3,3,3-tetrafluoro-2-(1,1,2,2,3,3,3-heptafluoropropoxy) propanoic acid (hexafluoropropylene oxide dimer acid [HFPO-DA]) and sodium 1 H ,1 H ,2 H ,2 H -perfluoro-1-octanesulfonate (6:2 FTS), are widely detected. Seawater (0.75–3.52 ng/L) and sediment (0.45–2.55 ng/g dw) levels indicate moderate contamination, exceeding pristine open baselines but remaining below coastal impacts. Crucially, downward-decreasing sediment profiles are significantly disrupted by active methane seepage. Elevated fugacity fractions ( ff > 0.8) reveal a strong thermodynamic migration tendency driving secondary PFAS release into bottom waters via methane-induced “bubble irrigation.” ToxPi prioritization highlights pronounced matrix- and compound-specific vulnerabilities, with 6:2 FTS and perfluorobutanoic acid (PFBA) as primary hazard contributors. These findings identify cold seeps as overlooked hotspots for PFAS source-sink transformation, requiring integration into deep-sea chemical risk frameworks.

Complete defluorination of PFASs via photocatalytic reduction in water

Chong MeiChi,Zhou Qixin,Xu Jingyi,Wu Zhaohui,Zhu Enwei,Li Wenlu,Zhang Ling,Guo Yan,Zhu Yongfa

The environmental persistence of per- and polyfluoroalkyl substances (PFASs), driven by the exceptional stability of their C–F bonds, presents a formidable challenge for remediation. Herein, we report the 5,10,15,20-tetraphenyl (4-aminophenyl) porphyrin (TAPP) aggregates as visible-light-driven photocatalysts capable of achieving almost-100% defluorination of PFASs without chemical additives. Central to this process is the ultra-stable TAPP radical species (TAPP • ), which exhibits a lifetime exceeding 7 days under ambient conditions. Under visible-light irradiation, TAPP • generates reductive electrons with a potential of −2.68 V NHE , enabling injection into the C–F antibonding orbitals to initiate defluorination. The exceptional stability of TAPP • arises from intramolecular charge delocalization mediated by the synergistic overlap between the lone-pair electrons distribution of the amino groups and the highest occupied molecular orbital. This work develops a steady radical strategy that leverages charge-delocalization to engineer photocatalysts with highly reductive electron, offering an approach to address persistent environmental contaminants.

Efficient adsorption of short-chain perfluoroalkyl substances by pristine and Fe/Cu-loaded reed straw biochars

Na Liu,Yinhui Li,Minggu Zhang,Naiju Che,Xianliang Song,Yanli Liu,Chengliang Li

As the substitutes of legacy long-chain per-/polyfluoroalkyl substances (PFASs), short-chain PFASs have been widely detected in the environment. Compared to long-chain PFASs, short-chain PFASs have smaller molecules and are more hydrophilic. Therefore, they are more likely to experience long-distance transport and pose lasting environmental impacts. In this study, Fe-doped (R-Fe) and Cu-doped biochars (R-Cu) were prepared using reed straw biochar (R). The results showed that the PFBA and PFPeA sorption capacities of R-Fe were 25.81 and 43.59 mg g −1 , 1.65 and 1.55 times higher than those of R, respectively. The PFBA and PFPeA sorption capacities of R-Cu were 19.34 and 33.69 mg g −1 , 1.24 and 1.20 times higher than those of R, respectively. In addition, R, R-Fe, and R-Cu exhibited higher PFBA and PFPeA sorption capacities than the biochars previously reported. The excellent PFAS sorption performances of the biochars were attributed to the highly porous structure of R, which provided rich adsorption sites. Ion-pair sorption, pore filling, electrostatic interaction between the Fe/Cu and cationic groups on biochar and the anionic groups of PFASs, and hydrophobic interaction between the hydrophobic surface of biochar and the fluorinated tails of PFASs were the underlying sorption mechanisms. The biochars presented high removal rates (>86 %) of multiple PFASs (∑PFAS: 350 μg L −1 ) from synthetic wastewaters, including legacy and emerging PFASs of different chain lengths and with different functional groups. The biochars reported in this study are promising candidate adsorbents for treating waters contaminated with short-chain PFASs.

Superficially capped amino metal-organic framework for efficient solid-phase microextraction of perfluorinated alkyl substances

Sai Ouyang,Guifeng Liu,Sheng Peng,Jiating Zheng,Yu-Xin Ye,Juan Zheng,Yuanjun Tong,Yalan Hu,Ningbo Zhou,Xinying Gong,Jianqiao Xu,Gangfeng Ouyang

Perfluorinated alkyl substances (PFASs) were ubiquitously in the surface and groundwater. It is crucial and urgent to develop a rapid and ultrasensitive analysis method for the quantification of trace-level PFASs. Herein, a highly hydrophobic sorbent by capping phenylsilane groups on the surfaces of NH 2 -UiO-66(Zr) nanocrystals was used for efficient solid-phase microextraction (SPME) of PFASs in water samples. It was found that the superficially capped nanocrystals (NH 2 -UiO-66(Zr)-hp) exhibited both faster extraction kinetics and higher enrichment capacity than the non-capped nanocrystals. The extraction of eleven kinds of PFASs by NH 2 -UiO-66(Zr)-hp fiber reached equilibrium in 20 min. The enrichment factors of the NH 2 -UiO-66(Zr)-hp fiber ranged from 6.5 to 48, with a preference for long-chain PFASs over short-chain PFASs. It was proposed that superficial capping eliminated competitive moisture adsorption on the surfaces of the non-capped nanocrystals, thus facilitating the adsorption of PFASs through hydrophobic interaction. By using this new sorbent, the limits of detection of the SPME method as low as 0.035 to 0.616 ng·L −1 were achieved for the target PFASs. The recoveries of PFASs in the environmental water samples were 80.9%-120%. This study presents a new strategy for developing an efficient sorbent for PFASs by surface hydrophobic modification.
