This study reports the structural characterization and in vitro antioxidant activity of a novel polysaccharide, PFH-3a from Fritillariae hupehensis bulbus (FHB) with a weight-average molecular weight (Mw) of about 92.34 KDa, which exhibits a unique glycosidic linkage pattern that has not been previously reported in FHB. Structure of PFH-3a was elucidated through NMR spin system assignment based on comprehensive analyses of 1D and 2D NMR spectroscopy, identification of sugar residue types by iterative matching against a self-constructed NMR database containing 1875 diverse sugar residues, monosaccharide composition analysis, 1D 1 H qNMR analysis, and glycosidic linkage determination. It revealed that PFH-3a comprises a backbone of β-D-Xyl p -( 1 →[3, 6 )-β-D-Gal p -( 1 →3, 6 )-β-D-Gal p -(1→] n , with two branches α-L-Ara f -(1→ and α-L-Ara f -(1→5)-α-L-Ara f -(1→ attached to position 3 of the main chain → 3 ,6)-β-D-Gal p -(1→. In vitro bioactivity assays demonstrated that PFH-3a exhibits dose-dependent antioxidant activity, indicating its potential as a natural antioxidant. However, further investigation is needed to explore the in vivo efficacy and the underlying mechanisms.

The combination of ultrasound and chemotherapy has been proposed as a promising strategy to achieve a better anticancer therapeutic efficacy. Here we present a facile strategy to construct novel bifunctional nanodroplets as smart vehicles for ultrasound and pH responsive delivery of anticancer agents. PFH is used as core and chitosan/alginate complexes are used as the stable shells of the nanodroplets. The effects of alginate/chitosan ratio, and the amount of surfactant as well as PFH on the size, size distribution, and encapsulation efficiency of nanodroplets are systematically investigated with the optimized formulation identified. The release of the encapsulated doxorubicin hydrochloride can be triggered by changing the pH value of the surrounding environment and the exposure to ultrasound. The nanodroplets also show strong ultrasound contrast via droplet-to-bubble transition as demonstrated by B-mode ultrasound imaging. The hemolytic activity and cytotoxicity are further studied, revealing the biocompatibility of the nanodroplets. The in vivo antitumor results demonstrate that the prepared droplets show excellent antitumor therapeutic efficacy and outstanding tumor-targeting ability. The proposed alginate/chitosan stabilized PFH nanodroplets represent an important advance in fabricating multifunctional therapeutic materials with great promises in the applications of combined antitumor therapies.

Purpose Hypoxia is considered to be obstructive to tumor treatment, but the reduced oxygen surroundings provide a suitable habitat for Bifidobacterium bifidum (BF) to colonize. The anaerobe BF selectively colonizes into tumors following systemic injection due to its preference for the hypoxia in the tumor cores. Therefore, BF may be a potential targeting agent which could be used effectively in tumor treatment. We aimed to determine whether a novel BF-mediated strategy, that was designed to deliver AP-PFH/PLGA NPs (aptamers CCFM641-5-functionalized Perfluorohexane (PFH) loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles) by aptamer-directed approach into solid tumor based on the tumor-targeting ability of BF, could improve efficiency of high intensity focused ultrasound (HIFU) treatment of breast cancer. Methods We synthesized AP-PFH/PLGA NPs using double emulsion method and carbodiimide method. Then, we evaluated targeting ability of AP-PFH/PLGA NPs to BF in vivo. Finally, we studied the efficacy of HIFU ablation based on BF plus AP-PFH/PLGA NPs (BF-mediated HIFU ablation) in tumor. Results The elaborately designed AP-PFH/PLGA NPs can target BF colonized in tumor to achieve high tumor accumulation, which can significantly enhance HIFU therapeutic efficiency. We also found that, compared with traditional chemotherapy, this therapy not only inhibits tumor growth, but also significantly prolongs the survival time of mice. More importantly, this treatment strategy has no obvious side effects. Conclusion We successfully established a novel therapy method, BF-mediated HIFU ablation, which provides an excellent platform for highly efficient and non-invasive therapy of tumor.

Thrombus is one of the culprits for global health problems. However, most current antithrombotic drugs are limited by restricted targeting ability and a high risk of systemic bleeding. A hybrid cell membrane-coated biomimetic nanosystem (PM/ [email protected] @P/R) was constructed in this paper to fulfil the targeted delivery of ginsenoside (Rg1) and perfluorohexane (PFH). Poly lactic-co-glycolic acid (PLGA) is used as carriers to coat Rg1 and PFH. Thanks to the camouflage of erythrocyte membrane (RM) and platelet membrane (PM), the nanosystem in question possesses remarkable features including immune escape and self-targeting. Therefore, a compact nano-core with [email protected] /R was formed, with a hybrid membrane covering the surface of the core, forming a “core-shell” structure. With its “core-shell” structure, this nanoparticle fancifully combines the advantages of both PFH (the low-intensity focused ultrasound (LIFU)-responsive phase-change thrombolysis) and Rg1(the antioxidant , anti-inflammatory and anticoagulant abilities). Meanwhile, PM/ [email protected] @P/R nanoparticles exhibits superior in-vitro performance in terms of ROS scavenging, anticoagulant activity and immune escape compared with those without cell membranes ( [email protected] /R). Furthermore, in the animal experiment in which the tail vein thrombosis model was established by injecting k-carrageenan, the combined treatment of LIFU and PM/ [email protected] @P/R showed a satisfactory antithrombotic efficiency (88.20 %) and a relatively higher biological safety level. This strategy provides new insights into the development of more effective and safer targeted biomimetic nanomedicines for antithrombotic treatments, possessing potential application in synergistic therapy field.

Laser-activated bioprobes with high photothermal conversion efficiency (IRPDA@PFH NDs) based on biocompatible IR-780 doped polydopamine perfluorocarbon nanodroplets (NDs) were developed. When protected by gelatin microspheres, their near-spherical morphologies can be easily observed with transmission electron microscope. Doping IR-780 (3 w/w % of added dopamine hydrochloride) can significantly enhance near-infrared (NIR) absorption and photothermal conversion efficiency to 57.7%. The enhanced NIR absorption and nonradiative relaxation are preferred to stronger photoacoustic (PA) signals and higher PA imaging definition; ultrasound (US) signals also increase more than 2.5 times because of easier phase change of NDs. These bioprobes had sensitive PA/US imaging capability with highly effective substitute utilizations, in which polydopamine was used either as a PA contrast or a photothermal agent. Perfluorocarbon can be used as an US contrast agent and temperature indicator. More importantly, the gray value increments of US increase with temperature in a general range from 35 to 55 °C. Especially, an approximate linear increasing of gray value in the optimized photothermal therapy (PTT) range from 35 °C to 50 °C could be used for the temperature monitoring and control of PTT. During PTT, the heated regions and the extent of photothermal heating can be visualized by US imaging. These findings indicate their great potential for biosensing and PTT monitoring.

Triple-negative breast cancer (TNBC) has inadequate treatment approaches and a poor prognosis. It is urgent to develop new treatment approaches for TNBC. The combination of photothermal therapy (PTT) and chemotherapy is a very effective potential therapy for TNBC. However, asynchronous accumulation, unclear efficacy, and toxic side effects hinder the further promotion of this method. Therefore, we designed and constructed a new type of nanocarriers, the cascade release near-infrared imaging (NIFI) & thermal-chemo combination nanoparticles (CNC NPs), that can release drugs through the cascade of ultrasound triggering and pH responding to achieve the synchronous tumor accumulation, monitoring and synergistic treatment of two functional molecules. The key material of CNC NPs is the polydopamine (PDA), which, through self-assembling, forms a rigid shell that contains doxorubicin (DOX) and NIF fluorescent dye IR780 on the surface of the perfluorohexane (PFH) microbubbles. The results show that CNC NPs have a hollow core-shell structure with an average particle size of 97.3 ± 27.2 nm and have exceptional colloidal stability and photothermal conversion efficiency. The NPs can effectively perform cascade drug release through ultrasound triggering and pH responding. CNC NPs have good in vivo biological safety and excellent fluorescence imaging, drug delivery, and therapeutic abilities in the TNBC models. These results provide an experimental basis for the development of new clinical treatment methods for TNBC.

Bacterial infection is a growing public health concern and causes a huge medical and financial burden. It is of significance to efficiently construct multifunctional platforms for bacterial point-of-care testing (POCT) and elimination. Herein, near-infrared (NIR) light-responded vancomycin-doped prussian blue nanoparticles (PB-VAN NPs ) with high efficient photothermal conversion was synthesized for binding, dual-mode portable detection, and elimination of bacteria. The PB-VAN NPs can bind to the surface of Gram-positive bacteria such as Staphylococcus aureus ( S. aureus ), forming complex of PB-VAN NPs / S. aureus . After being centrifugated, the suspension solution of PB-VAN NPs can stimulate perfluorohexane (PFH) to rapidly release oxygen (O 2 ) under NIR irradiation. Thus, the bacteria can be sensitively detected with portable pressure meter as signal reader, reporting a limit of detection (LOD) of 1.0 CFU mL -1 . On the other side, the sediment of PB-VAN NPs / S. aureus can be detected via thermal camera, reporting a LOD of 1.0 CFU mL -1 . Interestingly, the bacteria can be effectively inactivated with the local temperature elevation during temperature-based detection. The antibacterial efficiency reaches as high as 99.8%. The developed multifunctional nanoplatform not only provides a straightforward “mix-then-test” way for portable detection of bacteria with high sensitivity, also realizes high efficiency elimination of bacteria simultaneously. The developed strategy was further applied for promoting wound healing of bacteria-infected mice.

The research of short-chain fluorocarbon surfactants is crucial in effectively and environmentally extinguishing petrochemical fires. In this study, a short-chain fluorocarbon surfactant named 2 H ,2 H -perfluorooctanoic acid sodium salt (PFH-CA) was synthesized, and then characterized its chemical structure, thermal stability, surface activity, and foam property. Subsequently, PFH-CA was compounded with hydrocarbon surfactants including sodium dodecyl sulfate (SDS), dodecyl trimethyl ammonium chloride (DTAC), and N , N -dimethyldodecylamine- N -oxid (OB-2) at different molar ratios. The surface tension, interaction parameters, and foam property of the PFH-CA/hydrocarbon surfactant compounding systems were investigated. The analysis indicates that PFH-CA exhibits excellent surface activity and thermal stability concomitant with poor foam property. Introducing SDS, DTAC, and OB-2 enhances the foam performance and reduces the consumption of PFH-CA through synergistic interactions. Especially, PFH-CA/DTAC (1:4) system possesses the strongest interactions and the best performance, with critical micelle concentration ( CMC ) and surface tension at CMC ( γ CMC ) of 0.11 mmol/L and 20.90 mN/m, respectively. When concentration of PFH-CA exceeds 0.5 mmol/L, the foaming ability and foam stability stabilize at 30 cm and 90 %, respectively. The enhanced performance of PFH-CA/DTAC system is attributed to the electrostatic attraction between anionic and cationic surfactants, which facilitates the formation of micelles, subsequently leading to better surface activity and foam properties.
