3D-printed vancomycin-eluting PGCL/MXene bifunctional scaffold for management of infected bone defects

Xipeng Chen,Yuanpei Cheng,Yongbo Li,Ze Tan,Han Wu

The clinical management of infected bone defects remains a significant challenge in orthopedic surgery. In this study, we developed a multifunctional composite scaffold by integrating MXene (Ti 3 C 2 ) nanosheets and vancomycin (Van) into a degradable poly(glycolide-co-caprolactone) (PGCL) polymer through precision 3D printing technology, with the aim of regenerating acutely infected bone defects. Systematic optimization established the optimal composition as 5 wt% Ti 3 C 2 and 5 wt% Van within the degradable PGCL polymer. Compared with PGCL scaffolds, a significantly thicker mineralized layer could be deposited on the PGCL@5 %Ti 3 C 2 /5 %Van surface in simulated body fluid (SPF). The in vitro results showed that the PGCL@5 %Ti 3 C 2 /5 %Van scaffold had antibacterial properties, cytocompatibility, osteoblast differentiation and extracellular mineralization. The in vivo results showed that the PGCL@5 %Van/5 %Ti 3 C 2 scaffold effectively inhibited infection caused by methicillin-resistant Staphylococcus aureus (MRSA) in bone defects and promoted bone repair. The potential mechanism of Ti 3 C 2 in anti-inflammatory and osteogenic induction was investigated through transcriptome analysis. These results suggest that Ti 3 C 2 is involved in the regulation of the PI3K‒Akt signalling pathway to increase osteogenic induction ability and regulate the inflammatory response by downregulating the NF-κB pathway through the deiodinase iodothyronine type II (DiO2). These findings indicate that the PGCL@5 %Ti 3 C 2 /5 %Van scaffold can enhance the regeneration of acutely infected bone defects.

Preparation and Properties of PGCL Fiber for Surgical Absorbable Suture

Pan Ying,Jiang Xianchen,Deng Yanlin,Hua Ming,Wu Pengfei,Yao Lirong,Pan Gangwei

Surgical sutures are among the earliest commercialized medical textile materials, facilitating tissue closure and accelerating wound healing through suturing. Absorbable medical sutures represent a significant advancement in this field. Poly(glycolide-co-ε-caprolactone) (PGCL) is a novel biodegradable polymer with excellent biocompatibility, and the development of PGCL medical fibers is of considerable importance. In this study, PGCL fibers were fabricated via melt spinning, and the effects of draw ratio and drawing temperature on their structure and properties of PGCL fibers were systematically investigated. The results demonstrated that the crystallization behavior and mechanical properties of PGCL fibers were highly dependent on the drawing parameters. Within an appropriate range of drawing temperature, higher drawing temperature and draw ratios contribute to increased crystallinity and enhanced mechanical performance. Specifically, when the as-spun fibers were drawn at 40 ℃ with a draw ratio of 4, the drawn fibers exhibited a crystallinity of 65.77% and a tensile strength of 1.917 cN/dtex, representing increases of 165.4% compared to the as-spun fibers. This work is expected to provide a preliminary guide for the industrial-scale production of the PGCL medical fibers.

Antibacterial and anti-inflammatory PGCL fibers via polydopamine-assisted surface modification for absorbable sutures

Mengting Jiang,Hanyu Bao,Ying Pan,Xingang Li,Amjad Farooq,Lirong Yao,Can Gao,Gangwei Pan

PDA-assisted dip-coating fabricated dual-functional PGCL sutures. • Optimized CS/IBU coating achieved ≥99.99% antibacterial activity. • Functionalized fibers showed rapid initial and sustained IBU release. • The coating preserved PGCL's hydrolytic degradation and mechanics. • Modified sutures demonstrated excellent L929 cytocompatibility. Traditional sutures often lack functional bioactivity and could act as substrates for bacterial colonization, leading to surgical site infections, postoperative inflammation, and impaired wound healing. In this study, to develop a multifunctional suture with synergistic antibacterial and anti-inflammatory properties, a dip-coating strategy was employed. Taking advantage of the universal adhesive properties of polydopamine (PDA), chitosan (CS) and ibuprofen (IBU) were immobilized onto poly(gly co lide-co-caprolactone) fibers. Systematic characterization revealed that PDA loading enhanced the surface activity of the fibers and significantly improved their adhesive capacity. At an optimized CS/IBU ratio (2 wt%:2 wt%) and dual-coating cycle, the fibers exhibited excellent mechanical performance and achieved antibacterial rates of ≥99.99% against Escherichia coli and Staphylococcus aureus . Notably, the sutures exhibited sustained drug release and excellent cytocompatibility. This research provides a promising strategy for the development of absorbable, multifunctional sutures with integrated antibacterial, anti-inflammatory, and controlled drug release capabilities. Application of PGCL/AP conductive composite scaffold combined with electrical stimulation in neural tissue engineering

Zhu Longchuan,Yan Hongye,Li Hongru,Pan Su,Yang Hanwen,Zhu Zhe,Qi Zhiping

Nerve regeneration and axonal growth can be significantly enhanced by electrical stimulation (ES) using conductive polymers. But in addition to ES, nerve cell function and how they react to substrate conductivity are also essential factors in nerve repair. As a result, ES, in conjunction with appropriate materials (such as tissue scaffolds), has been used to treat nerve damage and has demonstrated significant promise for peripheral nerve regeneration. In this work, electrospinning was used to create conductive composite scaffolds made of polyglycolic acid@polycaprolactone/aniline pentamer (PGCL/AP). We later verified that PC12 cells cultured on these scaffolds exhibited improved migration, proliferation, and vitality upon electrical stimulation. Furthermore, In the established 10-mm peripheral nerve injury model, the PAP + ES group achieved outcomes comparable to the autograft group in the assessment of functional and morphological recovery, encompassing SFI, myelination rate, and gastrocnemius muscle evaluation. Regulation of calcium delivery and pore structure via osteoinductive microspheres of HA/CS/PGCL for orbital bone regeneration

Zhou Xiaosong,Wang Zongliang,Wang Yu,Guo Min,Zhang Peibiao

Repairing orbital bone defects remains a complex challenge in craniofacial surgery. Existing bone repair materials are plagued by issues such as intricate preparation processes and inadequate consideration of the spatiotemporal characteristics of bone healing. This study aimed to enhance the osteoinductive activity of poly (glycolide-co-caprolactone) (PGCL)-based materials through bicomponent modification, thereby providing a basis for the design of clinical bone repair materials. Hydroxyapatite/calcium sulfate/PGCL (HA/CS/PGCL) composite microspheres were fabricated via electrostatically assisted spray evaporation (ESASE). Their physicochemical properties and in vitro/in vivo osteogenic efficacy were evaluated using degradation-mineralization assays, MC3T3-E1 cell differentiation assessments and a rabbit orbital bone defect model. Innovatively, CS mesocrystal pore structure regulation was introduced to synergize with HA, achieving multidimensional optimization of surface hydrophilicity, sequential calcium ion release and mechanical properties. Results showed that in vitro mineralization formed native-like hydroxyapatite structures within 14 days. On HA/CS/PGCL surfaces, the proliferation activity of MC3T3-E1 cells reached 1.8 times that of the PGCL group after 7 days, with a 1.6-fold increase in alkaline phosphatase activity. In vivo, the composite microspheres achieved a new bone volume ratio (BV/TV) of 78.72% at 10 weeks, resulting in complete closure of the bone defect. Their trabecular structural parameters (trabecular thickness, Tb.Th; trabecular number, Tb.N) outperformed those of the single-component groups. Histological staining confirmed their ability to induce ordered collagen arrangement and the formation of dense new bone. In conclusion, the HA/CS synergistic modification strategy achieved the sequential release of calcium ions during bone repair and spatial regulation of the microsphere surface pore structure. The superior performance of HA/CS/PGCL microspheres in repairing complex bone defects lays an experimental foundation for clinical translation. Further research into the interactions between ion release and cell signaling is warranted.

Synergistically Promoting Bone Regeneration by Icariin-Incorporated Porous Microcarriers and Decellularized Extracellular Matrix Derived From Bone Marrow Mesenchymal Stem Cells

Zhou Mengyang,Guo Min,Shi Xincui,Ma Jie,Wang Shutao,Wu Shuo,Yan Weiqun,Wu Feng,Zhang Peibiao

Multifunctionality has becoming essential for bone tissue engineering materials, such as drug release. In this study, icariin (ICA)-incorporated poly(glycolide-co-caprolactone) (PGCL) porous microcarriers were fabricated and then coated with decellularized extracellular matrix (dECM) which was derived from bone marrow mesenchymal stem cells (BMSC). The porous structure was generated due to the soluble gelatin within the microcarriers. The initial released ICA in microcarriers regulated osteogenic ECM production by BMSCs during ECM formation. The dECM could further synergistically enhance the migration and osteogenic differentiation of BMSCs together with ICA as indicated by the transwell migration assay, ALP and ARS staining, as well as gene and protein expression. Furthermore, in vivo results also showed that dECM and ICA exhibited excellent synergistic effects in repairing rat calvarial defects. These findings suggest that the porous microcarriers loaded with ICA and dECM coatings have great potential in the field of bone tissue engineering.
