Screening of multifunctional PGPB from the Qinghai-Tibet Plateau and their effects on tomato growth and quality

Yi Guoyun,Wang Rong,Li Juan,Geng Guigong,Cheng Liang

The identification of indigenous plant growth-promoting bacteria (PGPB) from the Qinghai-Tibet Plateau is vital for developing region-specific microbial inoculants to improve crop productivity in high-altitude agricultural ecosystems. The solubilizing strains were isolated from faba bean leaves and roots collected in alpine farmlands of Qinghai using selective media. Following this, multifunctional PGPB with plant growth-promoting traits and biocontrol activities were identified. Selected PGPB strains were introduced into the rhizosphere of tomato plants to assess their impact on growth promotion. Results revealed that strain H18 displayed nitrogen fixation, phosphorus and potassium solubilization, IAA production and ACC deaminase activity. Additionally, it also exhibited antagonistic effects against eight fungal plant pathogens. Physiological and molecular analyses identified strain H18 belongs as Bacillus atrophaeus. This strain significantly improved tomato seed germination and enhanced growth during seedling and flowering stages, including parameters such as plant height, crown width, and stem diameter. Furthermore, during the fruiting stage, it increased yield and improved fruit quality. In conclusion, strain H18 is a multifunctional PGPB with excellent biocontrol and growth-promoting effects.

Enhanced Cd extraction of oilseed rape (Brassica napus) by plant growth-promoting bacteria isolated from Cd hyperaccumulator Sedum alfredii Hance

Fengshan Pan,Qian Meng,Sha Luo,Jing Shen,Bao Chen,Kiran Yasmin Khan,Jan Japenga,Xiaoxiao Ma,Xiaoe Yang,Ying Feng

Four plant growth-promoting bacteria (PGPB) were used as study materials, among them two heavy metal-tolerant rhizosphere strains SrN1 (Arthrobacter sp.) and SrN9 (Bacillus altitudinis) were isolated from rhizosphere soil, while two endophytic strains SaN1 (Bacillus megaterium) and SaMR12 (Sphingomonas) were identified from roots of the cadmium (Cd)/zinc (Zn) hyperaccumulator Sedum alfredii Hance. A pot experiment was carried out to investigate the effects of these PGPB on plant growth and Cd accumulation of oilseed rape (Brassica napus) plants grown on aged Cd-spiked soil. The results showed that the four PGPB significantly boosted oilseed rape shoot biomass production, improved soil and plant analyzer development (SPAD) value, enhanced Cd uptake of plant and Cd translocation to the leaves. By fluorescent in situ hybridization (FISH) and green fluorescent protein (GFP), we demonstrated the studied S. alfredii endophytic bacterium SaMR12 were able to colonize successfully in the B. napus roots. However, all four PGPB could increase seed Cd accumulation. Due to its potential to enhance Cd uptake by the plant and to restrict Cd accumulation in the seeds, SaMR12 was selected as the most promising microbial partner of B. napus when setting up a plant–microbe fortified remediation system.

Phytoremediation effect of Scirpus triqueter inoculated plant-growth-promoting bacteria (PGPB) on different fractions of pyrene and Ni in co-contaminated soils

Xiao Chen,Xiaoyan Liu,Xinying Zhang,Liya Cao,Xiaoxin Hu

At present, few reveal the mechanism of inoculation plants with PGPB to remediate PAH-metal co-contaminated soil by analyzing the chemical speciations of contaminants. This study investigated the influence of inoculation plants with PGPB on different fractions of pyrene and Ni in rhizospheric and non-rhizospheric soil. The results demonstrated that the addition of PGPB brought the extensive increase of FDA activities in pyrene-Ni co-contaminated soil. PGPB increased the resistance of plants in nickel and pyrene-Ni contaminated soil, but decreased the plant biomass in single pyrene contaminated soil. The addition of PGPB efficiently decreased bioaccessible fractions of pyrene and increased the bioavailability of Ni in both rhizospheric and non-rhizospheric soil. Although inoculation plants with PGPB significantly increased the accumulation of Ni in single Ni and pyrene-Ni co-contaminated soil, the poor bioavailability of Ni in rhizospheric soil still restricted the phytoremediation of the heavy metal. The presence of pyrene hindered the inoculated plant from accumulating Ni to some extent. On the contrary, the presence of Ni significantly promoted the degradation of pyrene in both rhizospheric and non-rhizospheric soil after inoculation plants with PGPB.

Evaluation of the Effect of Plant Growth Promoting Endophytic Bacteria from Pinellia ternata Using an Efficient Organic Silica Hybrid Monolithic Column

Zhang Panpan,Yuan Bo,Xu Mengke,Cao Xiaoying,Xue Lingwei,Jiang Jihong

This study aims to investigate the plant growth promoting bacteria (PGPB) in the medicinal plant Pinellia ternata (P. ternata), and evaluate the effect of PGPB on the growth and alkaloid content of P. ternata. Bacteria were isolated from P. ternate, and 16S rRNA gene sequencing of partial isolates was carried out. 23 kinds of representative bacteria were classified into 7 genera: Acinetobacter, Bacillus, Enterobacter, Pseudomonas, Kosakonia, Rhizobium and Stenotrophomonas. The results of the plant growth promotion screening showed that 23 strains could produce different levels of Indole-3-acetic acid (IAA), 1-aminocyclopropane-1-carboxylicacid deaminase (ACC), biofilm and certain enzymes. Potential PGPB were selected to evaluate their abilities to promote the growth of wheat. The results indicated that strain PtB01 (Rhizobium nepotum) and PtB24 (Bacillus methylotrophicus) significantly promoted the growth of the wheat seedlings. These two strains and their co-culture group were inoculated on P. ternata aseptic seedlings. It was found that these strains increased shoot length by 194%, root length by 307%, leaf area by 232%, fresh weight by 231%, dry weight by 165%, and the number of fibrous roots by 200%. We also designed a novel organic silica hybrid monolithic column through silicon dioxide modification and tested the influence of two strains on the alkaloid in P. ternata aseptic seedlings. The result showed that the content of alkaloid was increased by 140% after inoculation PGPB in P. ternata. PGPB from the medicinal plant P. ternata promoted the growth of the host plant and influenced the alkaloid content of P. ternata.

The potential of Co-cultivation of plant growth promoting bacteria (PGPB) and Chlorella sp. LZ-29 to enhance the biomass and protein content of Chlorella sp. LZ-29

Zhilai Lu,Zhe Luo,Min Liao,Xiaomei Xie,Mingdi Fan,Yue Xiao,Yunqiang Zhu

Two plant growth promoting bacteria (PGPB) were isolated. • PGPB could significantly enhance the biomass and protein content of Chlorella. • PGPB could enhancing the nitrogen assimilation and cell division of Chlorella. • PGPB could inhibit the flow of carbon metabolism to fat. The co-cultivation of microalgae with bacteria that stimulate microalgae growth and metabolic changes is a promising strategy for improving microalgae yield and quality. In this study, two plant growth promoting bacteria (PGPB), Bacillus safensis BAI-6 (BAI-6) and Bacillus altitudinis FEN-9 (FEN-9), were isolated from a long-term open cultivation system of Chlorella sp. LZ-29. Both strains were confirmed to produce the growth-promoting phytohormone indole-3-acetic acid (IAA) and vitamin B12. The changes in biomass, protein, fat, polysaccharide, pigment and other active components of Chlorella sp. LZ-29 under co-cultivation with these strains were investigated. Compared with the axenic Chlorella culture, co-cultivation with BAI-6 or FEN-9 alone increased biomass by 46.57% and 58.75%, respectively, while the dual-strain co-culture achieved the highest biomass increase of 66.46%. Protein content increased by 7% with BAI-6, 2.56% with FEN-9, and 13.37% in the dual-strain co-culture. Conversely, fat content decreased by 3.05%, 3.67%, and 3.48%, respectively, while polysaccharide content also declined in all co-culture treatments ( p  < 0.05). The synergistic effect of BAI-6 and FEN-9 in dual co-culture resulted in significantly enhanced biomass and protein production ( p  < 0.01), likely due to increased secretion of IAA and vitamin B12, which promoted nitrogen assimilation and cell division, while inhibiting carbon flux towards lipid synthesis. These findings demonstrate that the dual-strain co-culture system has significant potential for enhancing both the yield and protein content of Chlorella sp. LZ-29.
