Wearable electrochemical sensors for glucose analysis hold significant potential for non-invasive monitoring of sweat biomarkers. However, the complex and expensive preparation process has limited the application of these sensors. In this study, we proposed an electrochemical glucose biosensor based on poly (2-hydroxyethyl methacrylate) (PHEMA) hydrogel initiated via iron ion and phenol hydroxyl groups. Copper nanoparticles are then incorporated into the hydrogel, enhancing the conductivity and electrocatalytic performance of the hydrogel electrode. The composite hydrogel, characterized by its rich metal coordination and hydrogen bonds, demonstrated remarkable cyclic adhesion properties, stretchability, and self-healing capability. Subsequently, this hydrogel is modified onto a screen-printed electrode (SPE) to create an in vitro skin patch for glucose detection in human sweat. The biosensor exhibits a wide linear range for glucose concentration of 0–0.2 mM and 0.2–4 mM, with low detection limit of 9.99 μM. The sensor exhibits good stability, selectivity and flexibility, and demonstrates a strong correlation with glucose measurements obtained from real sweat using a glucose assay kit. This sweat sensor holds great development potential, providing broad prospects for future non-invasive detection applications.

Ocular bacterial infections are typically associated with elevated levels of reactive oxygen species (ROS). Nevertheless, for treating ocular bacterial keratitis (BK), broad-spectrum antibiotics in eye drops or ointments are unable to inhibit ROS and encounter swift clearance and reduced bioavailability. This work developed antibiotic levofloxacin (LEV)-loaded hollow ceria nanoparticles (hCe NPs, ROS scavengers), which were embedded into poly-hydroxyethyl methacrylate (pHEMA) hydrogels to prepare dual-functional contact lenses (LEV@hCe-pHEMA), enabling extended ocular drug delivery and enhanced bioavailability. The integration of LEV@hCe NPs within pHEMA contact lenses preserved good optical transmittance (> 90.0 %), fortified UV-blocking capacities (200–400 nm), achieved controllable LEV release (84.2 % within 120 h), and enhanced ROS scavenging-antioxidative potential (78.4 % within 60 min). In vitro cytotoxicity evaluations revealed low cytotoxicity (cell viability >95.0 %) of the hCe-pHEMA contact lenses and affirmed their good biocompatibility. Notably, LEV@hCe-pHEMA exhibited significant antibacterial efficacy against S. aureus ATCC29213 (89.8 %) and E. coli ATCC25922 (94.2 %), demonstrating their therapeutic potential. In vivo safety evaluations in rabbit models showed no ocular irritation or pathological changes during a 7-day wearing period, confirming the good biocompatibility of the hCe-pHEMA lenses. LEV@hCe-pHEMA contact lenses could be utilized to treat rabbit BK model induced by S. aureus . It could near-completely remove the keratitis (within 7 days), reducing corneal edema and further recovering corneal transparency. The results suggested that LEV@hCe-pHEMA contact lenses could be employed as promising dual-functional smart ocular drug delivery systems for non-invasive ocular therapy.

Non-invasive, continuous monitoring of sweat biomarkers using flexible sensors enables real-time tracking of indicators such as glucose, electrolytes, and lactate, facilitating early disease detection and improved health management. In this paper, a flexible screen-printed electrode (labeled as Cu@PHEMA/SPE) based on a Cu 2+ doped poly(2-hydroxyethyl methacrylate) (PHEMA) hydrogel was prepared by a simple free radical polymerization method, which is used for glucose and ascorbic acid detection in sweat. This hydrogel exhibits excellent mechanical properties (184 kPa and 346 %), self-healing capabilities and recycled self-adhesive performance. Cyclic voltammetry (CV) and chronoamperometry techniques were employed to investigate the electrochemical properties of the hydrogel electrode in an alkaline solution and the flexible electrode in a neutral solution, with respect to glucose and ascorbic acid. The flexible electrode demonstrated a wide linear range, high sensitivity, and a low detection limit. Specifically, glucose detection ranged from 0 to 2 mM, exhibiting sensitivities of 122.24 and 24 μA/mM/cm 2 , with a detection limit of 4.5 μM. Moreover, it can detect ascorbic acid in the range of 0–220 μM, with sensitivities of 799.85 and 201.94 μA/mM/cm 2 , and a detection limit of 0.96 μM. Furthermore, the electrode displayed good selectivity for interfering substance such as uric acid (UA), dopamine (DA), lactic acid (LA), sodium chloride (NaCl), and potassium chloride (KCl), and it maintained performance despite external deformation. Notably, real sweat samples analysis demonstrated strong concordance with the results obtained using commercially available kits. These results indicate that this flexible electrode has great potential for application in wearable devices.

Plant-based insect repellents are environmentally-friendly and play a practical and economic role in preventing the transmission of diseases to humans. Preformed 2-hydroxyethyl methacrylate-co-ethylene glycol dimethacrylate (pHEMA) hydrogels were functionalized with pedant triazinyl-β-cyclodextrin (T-β-CD) with the aim of improving the ability to host and release plant-based repellents. The structural and thermal properties were investigated by FTIR and thermogravimetric analysis, respectively. The most potent mosquito repellents, geraniol, methyl salicylate and trans-cinnamaldehyde, were screened from 35 plant-based compounds through Y-tube olfactometer assay. Pendant T-β-CDs did not affect the swelling of the hydrogels and exhibited low cytotoxicity towards mouse embryo fibroblasts, and no skin irritation or allergy was observed on human skin treated with modified hydrogels loaded with repellents. The physicochemical properties of the repellent and its affinity to form complexes determined the role played by the CDs in the loading and release. Methyl salicylate possessed an optimal stability constant KS (232.19 M−1) and thus it got a longer protection duration with higher repellent activity. Increasing the crosslink density resulted in decreases in the degree of swelling, the amount of T-β-CDs bonded to hydrogels, and the duration of complete repellency. Additionally, pHEMA hydrogels with pendant T-β-CDs showed good recyclability. The new hydrogel formulation loaded with plant-based repellents may provide a suitable, eco-friendly and safe approach to achieve protection against mosquitoes.
