PHMG-KH560 as antifungal and reinforcement media for paper materials

Minghao Jia,Xiaoxia Huang,Xiaogang Zhang,Gang Hu

Improving the strength decrease caused by fungal metabolites is still an arduous task in the field of paper materials protection. A set of damaged Kangxi Dictionary from Qing Dynasty was chosen as samples in this work. The main goal of this work is to develop high-efficiency and eco-friendly media with the characteristics of antifungal and reinforcement, to preserve paper materials and continue the historical and scientific research value of cultural heritage. PHMG as an efficient antifungal was used in this work. For better behavior, the epoxy group of KH560 can be induced by the amino group in PHMG by ring opening reaction to form PHMG-KH560 colloidal complex. Spraying PHMG-KH560 on paper could establish Si-O-C bonding and increased the hydrogen bond between polymeric material and paper fibers, which simultaneously inhibited fungi and improved mechanical strength of the original paper, even after the artificial aging. PHMG-KH560 had certain antifungal activity against five fungi, such as Aspergillus versicolor, and Aspergillus Niger , and had better inhibitory effect on Cladosporium herbarum, Trichoderma viride , and Penicillium expansum . The ancient writing ink on the paper sprayed with PHMG-KH560 did not affect its visual beauty, which also indicated that the complex reagent was expected to be applied to the antifungal and reinforcement of paper materials.

Construction of a stable and superhydrophilic PHMG/TA/Fe(III) composite coating-modified membrane for for oil–water separation

Qianyu Wei,Qiuhu Lin,Zezhen Zhang,Lili Wu,Chaocan Zhang

Polyvinylidene fluoride (PVDF)/polyhexamethylene guanidine (PHMG)/tannic acid (TA) coatings have some degree of hydrophilicity but lack stability. To improve the hydrophilicity and stability of antimicrobial PVDF/PHMG/TA membranes, Fe(III) is introduced into the composite system. As a weakly acidic hydrophilic nanoion, Fe(III) can enhance the hydrophilicity and resistance to contamination of the membranes. Furthermore, the stability can be enhanced because a three-dimensional cross-linked coating structure is formed by the complexation of Fe(III), TA and PHMG. This work investigates the effects of different tannic acid concentrations and pH conditions on the hydrophilicity, anti-pollution and stability of the modified coatings. The surface morphology of the membranes was analyzed by field-emission scanning electron microscopy. Additionally, the hydrophilicity and the antipollution performance were evaluated based on the water contact angle, submerged oil contact angle, emulsion flux and flux recovery rate. The stability was evaluated by the changes in flux recovery rate and water contact angle before and after emulsion scouring and ultrasonic treatment. The results show that the modified membranes have favorable hydrophilicity and stability after the addition of Fe(III). The water contact angle can be reduced to 19°, the pure water flux can reach 13,850 L·m −2 ·h −1 , and the emulsion flux can reach 2650 L·m −2 ·h −1 . Moreover, 99.68% flux recovery was achieved after 7 days of deionized water flushing, and the water contact angle remained almost unchanged.

Preparation and Antifouling Property of Polyurethane Film Modified by PHMG and HA Using Layer-by-Layer Assembly

Huihui Yuan,Chenli Xue,Jiaqian Zhu,Zhaogang Yang,Minbo Lan

To reduce the possibility of bacterial infection and implant-related complications, surface modification on polyurethane (PU) film is an ideal solution to endow hydrophobic PU with antibacterial and antifouling properties. In this work, a variety of polyhexamethylene guanidine/ hyaluronic acid (PHMG/HA) multilayer films were self-assembled layer-by-layer on PU films using polyanions, carboxyl-activated HA, and polycations PHMG by controlling the concentration of these polyelectrolytes as well as the number of layers self-assembled. Attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) spectra, water contact angle (WCA), and A Atomic force microscope (AFM) of PU and modified PU films were studied. Protein adsorption and bacterial adhesion as well as the cytotoxicity against L929 of the film on selected PU-(PHMG/HA)5/5-5 were estimated. The results showed that PU-(PHMG/HA)5/5-5 had the best hydrophilicity among all the prepared films, possessing the lowest level of protein adsorption. Meanwhile, this film showed efficient broad-spectrum antibacterial performance as well as significant resistance of bacterial adhesion of more than a 99.9% drop for the selected bacteria. Moreover, almost no influence on cell viability of L929 enhanced the biocompatibility of film. Therefore, the modified PU films with admirable protein absorption resistance, antimicrobial performance, and biocompatibility would have promising applications in biomedical aspect. Keywords:modified PU film;protein adsorption;antibacterial;hyaluronic acid (HA);polyhexamethylene guanidine (PHMG)

Preparation and Antibacterial Properties of Polyelectrolyte Complexed Nanoparticles Aggregated from PHMG and Sodium Caffeate

Mingyuan Tang,Xiaoxian Hao,Yuanyuan Kang,Xiaofeng He,Haichao Zhao

In this study, we synthesized polyelectrolyte complexed nanoparticles using an ion exchange reaction between poly(hexamethylene guanidine hydrochloride) and sodium caffeate. The morphology of the obtained antiparticle was observed by scanning electron microscopy, and FT-IR and XPS were employed for the structural characterization. The antimicrobial properties of E. coli and S. aureus were characterized through minimum inhibitory concentration (MIC), growth curve analysis, plate colony counting method, and crystal violet method. Notably, the sample showed a 100% bactericidal rate against E. coli at 0.095 μg/mL and against S. aureus at 0.375 μg/mL within 1 h, demonstrating excellent antimicrobial performance against E. coli and S. aureus. The CA-PHMG-containing acrylic resin coatings exhibited exceptional antimicrobial and antiadhesive properties when examined under an inverted fluorescence microscope, particularly at a 4% weight concentration of the antibacterial agent. This study holds vast potential for development in the field of antimicrobial coatings.

Optimizing the Bacteriostatic and Cytocompatibility Properties of Poly(hexamethylene guanidine) Hydrochloride (PHMG) via the Guanidine/Alkane Ratio

Yu Rao,Jinghong Wang,Huanhuan Wang,Hong Wang,Rong Gu,Jie Shen,Qing Hao,John L. Brash,Hong Chen

The emergence of “superbugs” is not only problematic and potentially lethal for infected subjects but also poses serious challenges for the healthcare system. Although existing antibacterial agents have been effective in some cases, the side effects and biocompatibility generally present difficulties. The development of new antibacterial agents is therefore urgently required. In this work, we have adapted a strategy for the improvement of poly(hexamethylene guanidine) hydrochloride (PHMG), a common antibacterial agent. This involves copolymerization of separate monomer units in varying ratios to find the optimum ratio of the hydrocarbon to guanidine units for antibacterial activity. A series of these copolymers, designated as PGB, was synthesized. By varying the guanidine/hydrophobic ratio and the copolymer molecular weight, a structure-optimized PGB was identified that showed broad-spectrum antibacterial activity and excellent biocompatibility in solution. In an antibacterial assay, the copolymer with the optimum composition (hydrophobic unit content 25%) inhibited >99% Staphylococcus aureus and was compatible with mammalian cells. A polyurethane emulsion containing this PGB component formed transparent, flexible films (PGB-PU films) on a wide range of substrate surfaces, including soft polymers and metals. The PGB-PU films showed excellent bacteriostatic efficiency against nosocomial drug-resistant bacteria, such as Pseudomonas aeruginosa and methicillin-resistant S. aureus (MRSA). It is concluded that our PGB polymers can be used as bacteriostatic agents generally and in particular for the design of antibacterial surfaces in medical devices.

A robust biaxially stretching strategy for fabrication of biodegradable and antimicrobial PBAT-based composite films

Yanjun Sun,Haihao He,Xuehua Jia,Meiqiong Chen,Ning Chen,Yinghong Chen

In this study, the biaxially stretching and reactive blending strategies were combined to fabricate the high-performance antimicrobial biodegradable polybutylene adipate-terephthalate (PBAT)/polylactic acid (PLA) based composite film with polyhexamethylene guanidine hydrochloride (PHMG) being as the efficient antimicrobial agent. Various measurements, including scanning electron microscopy (SEM), Fourier transform infrared (FTIR) analysis, 2D-small angle X-ray scattering (2D-SAXS), gas barrier test, mechanical property test, antimicrobial activity test, etc., were employed to carefully characterize the obtained films. The experimental results showed that the biaxially stretching could induce the orientation of polymer molecular chains and formation of submicron-thick layered thin-lamella structures of PLA dispersed phases in PBAT matrix in the presence of initiator dicumyl peroxide (DCP) and PHMG, thus significantly enhancing the crystallinity and mechanical properties of the obtained film, as well as improving the gas barrier and hydrophilicity-oleophobicity characteristics. At a stretching ratio of 3.5 × 3.5, the tensile strength of the prepared film could reach 61.9 MPa, and meanwhile the oxygen and water vapor permeability rates were reduced to 3.1 × 10 −14 and 5.1 × 10 −14  g/(cm s Pa), respectively. In addition, the incorporation of PHMG endowed the film with highly efficient antibacterial properties, e.g. a film with 1.0–1.5 wt% PHMG content could achieve a 100 % inhibition rate against Escherichia coli (E. coli) and a 99.9 % inhibition rate against Staphylococcus aureus (S. aureus). The further lychee preservation experiments demonstrated that the PHMG incorporating PBAT/PLA composite film remarkably extended the preservation time of fruit (40 % longer). The prepared antimicrobial PBAT/PLA/PHMG film exhibited the high safety and economic benefits, thus showing the promising application prospects in the field of food packaging.

Development of antifouling antibacterial polylactic acid (PLA) -based packaging and application for chicken meat preservation

Xiaoying Zhao,Tianyu Chen,Jiaxin Liu,Xinning Wang,Yunxuan Weng

Microbial contamination is the leading cause of food spoilage and food-borne disease. Here, we developed a multifunctional surface based on polylactic acid (PLA) bioplastic with antifouling and antibacterial properties via a facile dual-coating approach. The surface was designed with hierarchical micro/nano-scale roughness and low surface energy. Bactericidal agent polyhexamethylene guanidine hydrochloride (PHMG) was incorporated to endow the film with bactericidal activity. The film had good superhydrophobic, antifouling and antibacterial performance, with a water contact angle of 154.3°, antibacterial efficiency against E. coli and S. aureus of 99.9 % and 99.6 %, respectively, and biofilm inhibition against E. coli and S. aureus of 63.5 % and 68.9 %, respectively. Synergistic effects of antibacterial adhesion and contact killing of bacteria contributed to the significant antibacterial performance of the film. The biobased biodegradable film was highly effective in preventing microbial growth when applied as antibacterial food packaging for poultry product, extending the shelf life of fresh chicken breast up to eight days.

Guanidine-modified polysaccharide conditioning layer designed for regulating bacterial attachment behaviors

Jianxin Wen,Xiaomei Liu,Zhuoyue Han,Zhijuan Wang,Hidetoshi Saitoh,Hua Li

Biofouling has been persisting as a global problem due to the difficulties in finding efficient and environmentally friendly antifouling coatings for long-term applications. Initial attachment of bacteria on material surface and subsequent formation of biofilm are the predominate phenomena accounting for subsequent occurrence of biofouling. Among the various factors influencing the bacterial attachment, conditioning layer formed by organic macromolecules usually plays the key role in mediating bacterial attachment through altering physicochemical properties of substrate surface. In this study, a guanidine-modified polysaccharide conditioning layer with the capability of tuning the bacterial attachment is constructed and characterized. Dextran, a polysaccharide widespread in bacteria extracellular polymeric substances (EPS), is oxidized by sodium periodate, and cationic polymer polyhexamethylene guanidine hydrochloride (PHMG) is anchored to oxidized dextran (ODEX) by Schiff base reaction. AFM characterization reveals morphological changes of the polysaccharide conditioning layer from tangled chain to island conformation after the PHMG modification. The guanidine-based dextran conditioning layer promotes attachment of both P. aeruginosa and S. aureus and disrupted bacterial cytomembranes are seen for the attached bacteria due to electrostatic interaction of the electropositive guanidine group with the electronegative bacteria. The guanidine-based dextran conditioning layer shows a low survival ratio of 22 %-34 % and 1 %-4 % for P. aeruginosa and S. aureus respectively after incubation in the bacterial suspension for 72 hours. The results would give insight into further exploring the potential applications of the newly designed polysaccharides conditioning layer for combating occurrence of biofouling.

Ros-responsive hydrogel with size-dependent sequential release effects for anti-bacterial and anti-inflammation in diabetic wound healing

Zhipeng Sun,Yilin Ding,Zetao Wang,Huitong Luo,Qi Feng,Xiaodong Cao

Bacterial infection and the resulting inflammatory cascade are key factors hindering diabetic ulcers healing. The essential prerequisites for promoting diabetic ulcer healing are to sequentially achieve rapid sterilization, prevent reinfection, and modulate the inflammatory response. Inspired by the size-dependent sequential toxin release behavior of “Malaysian exploding ant”, we develop a ROS-responsive “exploding hydrogel” (E-gel) with size-dependent sequential release effects, which sequentially performs photothermal sterilization, persistent antibacterial activity, and immune regulation. Specially, polyhexamethylene guanidine (PHMG, P) and mesoporous polydopamine nanoparticles loaded with α-lipoic acid (α-LA@MPDA NPs, LD) were encapsulated into sulfhydryl-modified hyaluronic acid (HA-SH) through disulfide bonds to form a PLD/E-gel. The high temperatures generated by the photothermal effects of MPDA can kill over 90 % of bacteria within 10 min. During the swelling period of PLD/E-gel, PHMG diffuses out and effectively prevents the wound from re-infection. Along with the degradation of PLD/E-gel, submicron-sized α-LA@MPDA NPs are released from the cross-linked network to scavenge excessive ROS and modulate the inflammatory cascade. Animal experiments shows that PLD/E-gel accelerates the healing of infected diabetic wounds in SD rats by rapidly killing bacteria, inhibiting bacterial recolonization, relieving wound inflammation, and facilitating collagen deposition. To sum up, PLD/E-gel has a great application potential for diabetic wound healing.
