Xin Wang,Dong Zhai,Xiaogang Yao,Yufeng Wang,Hongshi Ma,Xiaopeng Yu,Lin Du,Huixing Lin,Chengtie Wu

For therapy of tumor-induced bone defects, functional biomaterials with tumor therapy and bone regeneration are of great importance. In this study, 3D printing of calcium titanate (CaTiO 3 ) bioceramic scaffolds was prepared for the therapy of tumor-induced bone defects. The digital laser processing (DLP) based 3D printing technology was applied to prepared CaTiO 3 bioceramic scaffolds. It was found that the color of CaTiO 3 (CaTi) scaffolds was deeper with the sintering temperature rising. Interestingly, CaTi scaffolds with pink color were found with optimal compressive strength (13.44 ± 0.99 MPa) and satisfactory photothermal property when exposed to NIR laser. The temperature of pink CaTi scaffolds could be over 70 ℃ under NIR laser irradiation (1.32 W/cm 2 ). Attributing to such excellent photothermal performance, CaTi scaffolds had been used as a photothermal biomaterial for the effective therapy of the in vitro and in vivo bone tumor. Moreover, the prepared pink CaTi scaffolds possessed bone-forming in vitro and in vivo . Therefore, 3D printing of pink CaTi bioceramic scaffolds represents a promising strategy by using the bifunctional biomaterial for tumor-induced bone defect therapy.

An acidic polysaccharose (YL-D2N2) was isolated from crude polysaccharides of pink Auricularia cornea and characterized for its structural and antioxidant properties. YL-D2N2 consists of fucose, galactose, glucose, xylose, mannose and glucuronic acid in a molar ratio of 0.85: 1.50: 4.44: 27.52: 46.56: 19.13. It has a number-average molecular weight of about 52.811 kDa and a weight-average molecular weight of about 135.457 kDa. Structural characterization showed that YL-D2N2 consists of nine residues (Xyl p -(1→, Glc p A-(1→, →2)-Xyl p -(1→, →3)-Gal p -(1→, →3)-Man p -(1→, →4)-Glc p A-(1→, →2,3)-Man p -(1→, →3,4)-Glc p -(1→, →3,6)-Man p -(1→), with a backbone of →3)-β-D-Man p -(1→, →2,3)-α-D-Man p -(1→, →3,6)-α-D-Man p -(1 → and side chains containing β-D-Xyl p -(1 → and α-D-Glc p A-(1→. Notably, YL-D2N2 exhibits significant radical scavenging activity for superoxide anions, reaching 50.82 ± 0.64 % at a concentration of 3.2 mg/mL. Overall, YL-D2N2 exhibits a unique chemical structure and specialized applications for targeting superoxide anion radicals, providing valuable insights for further exploration of its structure-activity relationship.

LC-MS/MS-based metabolomic of pink Auricularia cornea grown on Lycium barbarum sawdust substrate

Lei Ye,Xuezhen Yang,Zhenzhu Huang,Bo Zhang,Qing Tian,Yu Huang,Wei Tan,Mingjun Feng,Xiaolin Li,Hongbin Xu

Driven by the need for sustainable agriculture, using Lycium barbarum (goji) sawdust as a substrate for mushroom cultivation increases productivity while reducing environmental impact. This study evaluates the effects of different proportions of goji sawdust on the growth, yield, and metabolic profiles of pink A. cornea . Six substrate formulations were tested, with 16 % goji sawdust supporting optimal yield and quality. LC-MS/MS-based metabolomics identified 1335 metabolites, mainly organic acids and derivatives, lipids and lipid-like molecules, organoheterocyclic compounds, and benzenoids. Biomarker analysis revealed twenty to thirty-five metabolites associated with substrate variations. KEGG enrichment highlighted key metabolic pathways, including ABC transporters, amino acid biosynthesis, D-amino acid metabolism, and aminoacyl-tRNA biosynthesis, contributing to substrate adaptation. While this study offers insights into the metabolic responses of pink A. cornea to goji sawdust, future research should incorporate absolute biomarkers quantification to further validate these results and optimize substrate compositions.

Impact of rutin on growth, development and oviposition of Bt-susceptible and Bt-resistant pink bollworm

Ling Wang,Dong Xu,Yi Yang,Weiguo Liu,Shengbo Cong,Wenjing Li,Nina Yang,Haichen Yin,Lirong Jin,Huaiheng Wu,Peng Wan

The evolution of pest resistance to insecticidal proteins from Bacillus thuringiensis (Bt) restricts the sustainable application of transgenic Bt crops, and exploring other ecofriendly insecticides to efficiently suppress Bt-resistant populations is thus urgently needed. The plant secondary metabolite rutin possesses pest resistance effects and plays an important role in plant resistance to pests, but the effect of rutin on pink bollworms is unclear. We investigated the effects of rutin on two types (Bt-susceptible and Bt-resistant) of pink bollworms ( Pectinophora gossypiella ). Rutin had significant inhibitory effects on the growth, development, and oviposition of both Bt-susceptible and Bt-resistant strains. For both strains, rutin ingestion by larvae did not significantly affect larval survival at 7 days after treatment or egg hatching rate but significantly decreased pupation rate, eclosion rate, neonate-to-adult survival, pupal weight and eggs per female and significantly prolonged larval developmental periods, and the effect increased with increasing rutin content. The response to rutin ingestion was basically consistent between Bt-resistant and Bt-susceptible strains, indicating that the Bt-resistant strain did not exhibit cross-resistance to rutin. These results provide a reference basis for green control and resistance management of pink bollworms and may be used as a component of comprehensive management for this pest.

Monotropein induces autophagy through activation of the NRF2/PINK axis, thereby alleviating sepsis-induced colonic injury

Jingchao Qiang,Rongrong Yang,Xueqing Li,Xuhui Xu,Mengyuan Zhou,Xiaomeng Ji,Yingzhi Lu,Zibo Dong

Sepsis is a systemic inflammatory disease that is caused by a dysregulated host response to infection and is a life-threatening organ dysfunction that affects many organs, which includes the colon. Mounting evidence suggests that sepsis-induced colonic damage is a major contributor to organ failure and cellular dysfunction. Monotropein (MON) is the major natural compound in the iris glycoside that is extracted from Morendae officinalis radix , which possesses the potent pharmacological activities of anti-inflammatory and antioxidant properties. This research evaluated whether MON is able to alleviate septic colonic injury in mice by cecal ligation and puncture. Colonic tissues were analyzed using histopathology, immunofluorescence, quantitative real-time polymerase chain reaction, and Western blot methods. It was initially discovered that MON reduced colonic damage in infected mice, in addition to inflammation, apoptosis, and oxidative stress in colonic tissues, while it activated autophagy, with the NRF2/keap1 and PINK1/Parkin pathways also being activated. Through the stimulation of NCM460 cells with lipopolysaccharides, an in vitro model of sepsis was created as a means of further elucidating the potential mechanisms of MON. In the in vitro model, it was found that MON could still activate the NRF2/keap1, PINK1/Parkin, and autophagy pathways. However, when MON was paired with the NRF2 inhibitor ML385, it counteracted MON-induced activation of PINK1/Parkin and autophagy, while also promoting inflammatory response and apoptosis in NCM460 cells. Therefore, the data implies that MON could play a therapeutic role through the activation of the NFR2/PINK pathway as a means of inducing autophagy to alleviate the oxidative stress in colonic tissues that is induced by sepsis, which will improve inflammation and apoptosis in colonic tissues.

LC-MS/MS Reveals Variation in Bioactive Flavonoid Profiles and MYB Regulatory Networks in Wild vs. Cultivated Strawberries (White, Pink, Red)

Mingzheng Duan,Ting Jiang,Xi Wang,Kangjian Song,Xu Xiao,Xiaoting Fu,Sijiu He,Jieming Feng,Muhammad Junaid Rao,Hengcui Guo

Strawberries ( Fragaria × ananassa ) are prized for bioactive flavonoids and anthocyanins, contributing to antioxidant capacity and health benefits. However, decades of breeding for yield and shelf-life have eroded their phytochemical diversity. To explore this trade-off, we compared wild strawberries ( Fragaria nilgerrensis , HM) with three cultivated varieties (pink-FY, red-RF, white-DX) using integrated LC-MS/MS metabolomics and transcriptomics. Wild HM strawberries contained twice the vitamin C (134.2 mg/100g), 2.5× more total flavonoids (138.7 mg/100g), and higher antioxidant activity (367 μg Trolox/g) than commercial varieties. In contrast, red-fruited RF showed abundant anthocyanins (pelargonidin-3-glucoside 6.6% abundance), while the wild HM genotype accumulated higher levels of flavonoids, with neohesperidin, lonicerin, hyperin and ternatumoside II collectively representing 20.9% of total flavonoids - concentrations 3-169 times greater than those observed in the three commercial varieties (FY, RF, DX). Transcriptomic analysis (RNA-seq, Illumina HiSeq 2500) revealed distinct MYB transcription factor associations: FxaYL_611g0111200 (MYB66) showed strong positive correlation with anthocyanin biosynthesis in RF (r=0.92, **p<0.005 ), while FxaYL_242g0472670 (MYB44-like) was significantly associated with HM's unique antioxidant flavonoid profile (r=0.96, ***p<0.0001 ). These regulatory relationships were corroborated by hierarchical clustering demonstrating varietal-specific expression patterns and principal component analysis (82.6% variance explained), collectively providing molecular targets for enhancing nutritional quality in strawberry breeding programs.

Lycopene attenuates di(2-ethylhexyl) phthalate-induced mitophagy in spleen by regulating the sirtuin3-mediated pathway

Xue-Yan Dai,Yi Zhao,Jing Ge,Shi-Yong Zhu,Mu-Zi Li,Milton Talukder,Jin-Long Li

Lycopene (Lyc) has been discussed as a potential effector in the prevention and therapy of various diseases. Di(2-ethylhexyl) phthalate (DEHP) is regarded as a universal environmental pollutant. To clarify the potential protective effect of Lyc on DEHP-induced splenic injury, 140 male mice were randomized into seven groups: control (distilled water), vehicle control (corn oil per day), Lyc (5 mg per kg BW per day), DEHP (500 or 1000 mg per kg BW per day), and DEHP combined Lyc group, respectively. All experimental animals were treated by oral gavage for 28 days. The results that showed DEHP exposure significantly up-regulated the mRNA and protein expression of the sirtuin family (except SIRT4-5), PGC-1α, OPA1, Drp1, MFN1/2, NRF1, TFAM, Parkin and PINK in DEHP-treated alone groups and the SOD2 and LC3-II protein expression were also in accordance with the above changes. These were accompanied with an increase of the number of inflammatory cells and rate of mitochondrial damage, and autophagosome formation in the spleen. Notably, Lyc supplementation facilitated all these changes to effectively return to the normal level, indicating that Lyc exerts protective effects against DEHP-induced splenic toxicity. Altogether, the protective effects of Lyc may be a strategy to ameliorate DEHP-induced spleen damage.

Pink Lotus Essential Oil and Alleviates on Free Fatty Acid Induced Steatosis in HepG2 Cells via PI3K/Akt and NF-κB Pathways

Runzhou Sun,Ruixin Xiao,Pengfei Lv,Feifei Guo,Yanling Gong,Meixing Yan

Pink lotus essential oil (PLEO) is the volatile components extracted from lotus flowers and there are few relevant research. The purpose of this study was to observe the effect of PLEO on NAFLD in vitro model and its possible mechanism. The ingredients of PLEO were determined by gas chromatography-mass spectrometry (GS-MS) and its lipid-lowering and hepatoprotective activities were investigated. HepG2 cells were treated with free fatty acid (FFA) to establish a cell model of NAFLD. Cell viability was evaluated by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method. Total cholesterol (TC), triglyceride (TG), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) were determined by Enzyme-Linked Immune Sorbent Assay (ELISA). Oil red O staining was performed to observe the lipid accumulation in the HepG2 cells. Lipid metabolism enzymes including fatty acid synthase (FAS), acetyl-coA carboxylase (ACC), stearoyl-CoA desaturase 1 (SCD-1), and carnitine palmitoyltransferase-1 (CPT-1), insulin signaling pathways including phosphatidylinositol 3 kinase (PI3K) and protein kinase B Akt, inflammatory signaling pathways such as nuclear factor kappa-B (NF-κB), were determined by Western blotting. There were 46 components determined in PLEO with many terpenoids compounds. PLEO decreased TC and TG contents in the FFA-treated HepG2 cells. Furthermore, PLEO inhibited TNF-α, IL-6 and IL-1β excretion, decreased NF-κB, FAS, ACC and SCD-1 while increased phosphorylation of NF-κB, PI3K, Akt, and CPT-1 expression. It is the first time to reveal that PLEO alleviates FFA-induced steatosis in HepG2 cells by regulating lipid metabolism, inhibiting inflammatory response, and improving insulin sensitivity.

Mutation of hop-1 and pink-1 attenuates vulnerability of neurotoxicity in C. elegans: the role of mitochondria-associated membrane proteins in Parkinsonism

Siyu Wu,Lili Lei,Yang Song,Mengting Liu,Shibo Lu,Dan Lou,Yonghong Shi,Zhibin Wang,Defu He

Mitochondrial dysfunction is considered as a critical mechanism in the pathogenesis of Parkinson's disease (PD). Increasing evidence supports the notion of mitochondria-associated membranes (MAMs) in mitochondrial dysfunction; yet little is known about the role of MAMs-related proteins in the pathogenesis of PD. Herein we exposed the nematode Caenorhabditis elegans to 0.5–10.0 μM rotenone (RO) or 0.2–1.6 mM paraquat (PQ) for 3 days. Our results showed that both RO and PQ induced similar Parkinsonism including motor deficits and dopaminergic degeneration. RO/PQ caused mitochondrial damages characterized by the increase of vacuole areas and autophagy vesicles, but the decrease of mitochondrial cristae. RO/PQ-impacted mitochondrial function was also demonstrated by the decrease of ATP level and mitochondrial membrane potential . Additionally, the attachment or surrounding of endoplasmic reticulum to the damaged mitochondria indicates ultrastructural alterations in MAMs. Using fluorescently labeled transgenic nematodes, we further found that the expression of tomm-7 and genes of Complex I, II and III was reduced, whereas the expression of pink-1 was increased in the exposed animals. To determine MAMs in toxicity toward PD, we investigated the mutants of hop-1 and pink-1 , encoding presenilin and PTEN-induced putative kinase 1 (PINK1) in mitochondria-associated membranes, respectively. Results demonstrated that the mutation of both hop-1 and pink-1 reduced the vulnerability of lethal, behavioral, and mitochondrial toxicity induced by RO/PQ. These findings suggest that presenilin and PINK1 play important roles in the RO/PQ-induced neurotoxicity through the mechanisms involved in mitochondria-associated membranes.

Low–temperature synthesis and chromatic properties of ferrum–zircon (Fe–ZrSiO4) pink coral ceramic pigment with the assistance of in–situ formed molten salt

Tao Wang,Xiang Zhang,Qiguo Hu,Xinhao Li,Tiangui Zhao,Lan Ma,Ting Sun,Jianmin Liu,Feng Jiang,Guo Feng

A novel low–temperature route has been developed to prepare Fe–ZrSiO 4 ceramic pigment with the assistance of in–situ formed molten salt. The effects of calcination temperature on the preparation and chromatic properties of the obtained pigments were investigated. When optimized calcination temperature is 900 °C, the as–prepared Fe–ZrSiO 4 ceramic pigment shows a pink coral with chromatic parameters of L* =56.64, a* =18.77 and b *=12.20. The obtained pigment shows a good dispersibility with average particle size of 2 μm, and can be susscessfully applied in high–temperature glazing coloration. The XRD and XPS results, as well as the changes in chromatic parameters after application in glaze, confirm that the doping of iron ions into ZrSiO 4 lattice is the main reason for the coloration of Fe–ZrSiO 4 ceramic pigment. The utilization of in–situ formed molten salt can provide a valuable reference for low–temperature synthesis of other ceramic materials.

Pink all-inorganic halide perovskite nanocrystals with adjustable characteristics: Fully reversible cation exchange, improving the stability of dopant emission and light-emitting diode application

Louwen Zhang,Ling Xu,Meng Zhu,Chen Li,Luying Li,Jun Su,Yihua Gao

In this work, we proposed a straightforward and effective strategy for preparing pink all-inorganic halide perovskite nanocrystal (NC) phosphors for light-emitting diode (LED) application. The pink perovskite NCs with varying Mn contents could be easily and steadily obtained via postsynthetic ion exchange at room temperature. The dependence of the size and optical properties of pink NCs on dopant content was systematically investigated. The reversible anion exchange processes of pink perovskite NCs were conducted with halide salts containing or excluding Pb. The contrasting results suggested that the irreversibility of Mn-emission was mainly affected by the fully reversible cation exchange between Pb 2+ and Mn 2+ . The pink NCs with better stability of dopant emission and high monodispersity were successfully developed by compounding with silica . A pink LED was further made by using the pink NCs/silica composites as pink phosphors on a 395 nm InGaN LED chip. This work provides a feasible strategy for the fabrication of pink LEDs, which may have great application prospects in the field of special lighting, like neon lights and plant growth lights.
