Electrospinning Trilayer Dressing of PLA/ZnO with Controlled Released Profiles for Visible pH Monitoring and Effective Exudate Management

Jun Zhang,Chao Hou,He Yu,Md All Amin Newton,Binjie Xin,Tingxiao Li

To address persistent challenges in wound care, including exudate management, infection prevention, and healing monitoring, this study developed a multifunctional fiber dressing using electrospinning technology. The system integrates a patterned hydrophobic polylactic acid/zinc oxide (PLA/ZnO) inner layer providing antimicrobial protection and exudate drainage; a polyacrylonitrile/polydopamine (PAN/PDA) intermediate water transport layer enabling unidirectional exudate transfer; and an outer PAN-based functional membrane incorporating sodium polyacrylate (SPA), phenol red (PSP), and tetracycline hydrochloride (TCH) for controlled drug release and visual wound status monitoring. The comprehensive evaluation revealed exceptional performance: rapid exudate removal within 3.35 s, 96 h sustained antibacterial drug release (68.6% cumulative release), pH-responsive chromatic transition (yellow-to-red at pH 5–9), and significant antimicrobial efficacy against Escherichia coli and Staphylococcus aureus (inhibition zone diameters of 10 and 18 mm, respectively). The dressing exhibited a balance of mechanical strength (6.1 MPa tensile strength) and breathability (13.4 mm/s air permeability rate). By integrating three core functions─efficient exudate management, antimicrobial regulation, and intelligent healing monitoring─this multifunctional architecture demonstrates clinical applicability in postoperative care and chronic wound therapy.

Preparation and characterization of unidirectional water-transported bilayer PLA/ZnO-PAN/SPA nanofibrous membrane for wound healing

Chao Hou,Md All Amin Newton,Binjie Xin,Tingxiao Li

Wound infection caused by excessive exudate leads to overhydration of wounds and prevents wound healing. Thus, there is a need for a wound dressing to be developed that can unidirectionally export excess biological fluid while inhibiting bacterial infection. Herein, a nanofibrous bilayer membrane with a unidirectional water transport function was fabricated by electrospinning as a novel wound dressing, which can export excess biological fluid efficiently and further inhibit bacterial infection. The inner layer of the bilayer nanofiber membrane is polylactic acid (PLA)-zinc oxide (ZnO) nanofibers, which are hydrophobic and antibacterial. The outer layer is polyacrylonitrile (PAN)-sodium polyacrylate (SPA) nanofibers, which are hydrophilic and liquid-absorbent. The bilayer nanofibrous membrane's morphology, mechanics, thermal stability, wettability , water absorption, gas permeability, and unidirectional water transport ability were characterized and evaluated. The study showed that the nanofibrous bilayer membrane exhibited excellent physicochemical properties and biological performance, and its mechanical properties can reach 4.6 MPa. In addition, when the thickness of the hydrophobic layer is 3 µm and the thickness of the hydrophilic layer is 44 µm, the double-layer fiber membrane can effectively export the excess biological fluid in 5.09 s and prevent the wound from wetting again. The nanofibrous bilayer membrane with excellent water absorption, air permeability , thermal stability, and mechanical strength provides a feasible solution for chronic wound healing.

Additive manufacturing of antibacterial PLA-ZnO nanocomposites: Benefits, limitations and open challenges

Wei Juene Chong,Shirley Shen,Yuncang Li,Adrian Trinchi,Dejana Pejak,Ilias (Louis) Kyratzis,Antonella Sola,Cuie Wen

Polymeric biomaterials such as polylactic acid (PLA) play a prominent role in the advancement of biomedical additive manufacturing (AM). PLA offers indeed a very advantageous combination of thermo-mechanical properties and functional attributes, as it is biobased, biodegradable, biocompatible and easy to print. However, PLA can be damaged by common sterilization methods and is sensitive to most chemical disinfectants, and this may impair its widespread usage. One of the most promising ways to overcome this shortcoming is to provide PLA with embedded antibacterial activity by the addition of appropriate fillers such as zinc oxide (ZnO) nanoparticles. After a detailed introduction to the basic properties of PLA and ZnO nanoparticles , the present review analyzes the main variables that govern the antibacterial activity of PLA-ZnO nanocomposites. Current applications and related manufacturing processes are also presented to showcase the importance of having embedded antibacterial functions in demanding applications such as food packaging and wound dressing. Emphasis is then placed on the emerging literature of the AM of PLA-ZnO nanocomposites, with a focus on fused filament fabrication (also known as fused deposition modeling). Existing gaps and hurdles related to the development and 3D printing of such composites is critically discussed. It is envisioned that a deeper understanding of the processability, thermo-mechanical behavior, biocompatibility and antibacterial efficacy of additively manufactured PLA-ZnO nanocomposites will foster their adoption in the biomedical field and, ultimately, in all circumstances where it is crucial to limit infection transmission.

Preparation, characterization and antibacterial evaluation of PLA/ZnO nanofiber membranes loaded with thymol by coaxial electrospinning

Dan Huang,SuQiu Si,Zhuoming Chen,Binjie Xin

In this study, a novel nanofiber material possessing antibacterial and antioxidant properties was fabricated through coaxial electrospinning, utilizing polylactic acid (PLA), thymol (THY), and nano zinc oxide (ZnO) as the raw materials. THY and ZnO was used to endows the membranes with good antibacterial and antioxidant properties. The surface morphology of the PLA/Z@T nanofiber membrane was characterized using scanning electron microscopy (SEM). Additionally, the successful formation of the core-shell structure was verified through transmission electron microscopy (TEM), fluorescence microscopy, and Fourier transform infrared spectroscopy (FTIR). Additionally, both ZnO and THY exhibit strong antibacterial properties, and their synergistic effect significantly enhances the antibacterial activity of the composite nanofiber membranes against Staphylococcus aureus . The drug sustained release percentages within 24 h for PLA@T and PLA/Z@T were 15.91 % and 14.82 %, respectively. The addition of zinc oxide does not significantly affect the release efficiency of thymol. After continuous release for 288 h, these percentages increased to 73.35 % and 72.94 %, respectively. By assessing antioxidant activity through DPPH and ABTS assays, it was found that PLA/Z@T exhibited higher activity compared to the control group. Subsequently, the in vitro antibacterial performance was investigated, revealing an antibacterial rate higher than that of the control group, the antibacterial rates against Staphylococcus and Escherichia coli reached 99.75 % and 98.49 % respectively. In conclusion, the results demonstrate that PLA/Z@T nanofiber membranes possess excellent antibacterial efficiency, sustained antibacterial performance, and antioxidant properties, offering a novel strategy for antibacterial materials.

Preparation and characterization of unidirectional water conducting double-layer PVA/CS/ZnO-PLA/ZnO nanofiber membranes for wound healing

Zhiyuan Chen,Binjie Xin

During the wound healing process, the surrounding tissue fluid plays a crucial role. However, excessive exudate can lead to wound infection and over-hydration, thereby delaying the healing process. Therefore, developing a novel wound dressing that can unidirectionally remove excess biological fluids and effectively inhibit bacterial infection is particularly important. This study successfully utilized electrospinning technology to prepare a nanofiber dual-layer membrane with unidirectional water transport functionality. Its inner layer consists of polylactic acid (PLA) and zinc oxide (ZnO) nanofibers, providing excellent hydrophobic and antibacterial properties. The outer layer is composed of polyvinyl alcohol (PVA), chitosan (CS), and nanoscale zinc oxide (ZnO) nanofibers, offering outstanding hydrophilicity and liquid absorption capability. Experimental results demonstrate that this nanofiber dual-layer membrane exhibits outstanding physical, chemical, and biological properties, with a mechanical strength of up to 6.4 Mpa and breathability 17.47 mm/s and antibacterial property is outstanding. In practical applications, this dressing can effectively expel excess biological fluids within just 5 s, preventing wound over-hydration and providing an innovative and feasible solution for the effective treatment of chronic wounds.

Biodegradable bilayer active packaging with unidirectional release: PLA/ZnO/MgO outer layer and gelatin electrospun mat incorporating eugenol-thymol for cucumber preservation

Yunge Zhang,Mi Li,Yuanchang Cao,Jian Ju,Qingmin Chen,Yanli Ma,Cuicui Li

Novel biodegradable bilayer film with unidirectional release. • PLA/ZnO/MgO outer layer improves barrier and UV-blocking. • Inner gelatin/EUG-THY layer ensures long-term antimicrobial activity. Fresh cucumbers are highly perishable due to moisture loss, microbial infection, and oxidative deterioration. In this study, a biodegradable bilayer active packaging film with unidirectional release was developed for cucumber preservation. The outer PLA layer was reinforced with 2 wt% of a 1:1 ZnO/MgO nanoparticle mixture, exhibiting enhanced hydrophobicity, mechanical strength (tensile strength 62.75 MPa), reduced water vapor permeability (88.7 g/m 2 ·24 h), and excellent UV-blocking. The inner layer was prepared by electrospinning gelatin loaded with 0.2–0.4 wt% eugenol (EUG) and thymol (THY). Unidirectional release evaluation showed that >92% of EUG and THY were released toward the food simulant via a non-Fickian diffusion mechanism ( n  = 0.54–0.57). The films exhibited concentration-dependent antimicrobial and antioxidant activities. During 21 days of storage at 4 °C, cucumbers packaged with the EUG/THY-loaded bilayer films showed significantly lower weight loss, reduced color deterioration, and enhanced antioxidant enzyme activities. The 0.4% EUG/THY formulation extended cucumber shelf life to 18–20 days. This bilayer design offers a promising strategy for eco-friendly active packaging with unidirectional release for fresh produce preservation. Investigation of physico-mechanical, thermal, morphological, and antibacterial effects of bio-based epoxidized soybean oil plasticizer on PLA-ZnO nanocomposites as flexible food packaging

Mina Ghoroghi,Sara Estaji,Mohammad Iman Tayouri,Reza Jahanmardi,Marcos A.L. Nobre,Hossein Ali Khonakdar

Polylactic acid (PLA) has attracted considerable attention because of its excellent properties compared to petroleum-based materials. However, improving its toughness, crystallization, and functionalities is challenging. Using a laboratory internal batch mixer, we produced unique blended nanocomposites comprising polylactic acid (PLA) as the host matrix, epoxidized soybean oil (ESO) as the plasticizer, and zinc oxide nanoparticles (ZnO NPs) as the reinforcements. The SEM-EDS test showed that neat PLA had a smooth surface, but the PLA-ZnO nanocomposite had rough micrographs and a uniform dispersion state of the nanoparticles. It was also found that adding ESO to the blend nanocomposites created a matrix droplet structure, and the size of the oil domains decreased as the ZnO content increased. All results derived from FTIR, XRD, and DSC tests were consistent with the morphological features in which ZnO NPs acted as the blend’s compatibilizer and heterogeneous nucleating agent. Our team observed that adding ESO decreased the PLA-ZnO nanocomposites’ glass transition temperature (T g ) by an average of 2 °C and increased the crystallization rate. It was also confirmed that the presence of ESO improved the flexibility and reduced the strength of the PLA. The tensile strength was further enhanced by incorporating ZnO into the blend. Adding 5 wt% of ZnO NPs increased the tensile strength of the PLA containing 10 wt% ESO by approximately 3.5 MPa. Furthermore, the tensile modulus was predicted using theoretical models; for example, the Paul model fitted with experimental findings. The addition of ESO increased the overall surface free energy of the nanocomposites. On the other hand, increasing ZnO content resulted in high contact angles and antibacterial properties of the blended nanocomposites. The favorable characteristics of the resulting films emphasized the potential use of these nanocomposite films as a promising choice for food packaging materials.
