Biodegradable scaffolds play an important role in tissue engineering, and appropriate degradation and resorption rates of these scaffolds are necessary to accommodate tissue growth. Synthetic polymers are frequently used because of their ease of production, good biocompatibility and controllable degradation rates. However, monitoring the degradation of these polymers in vivo by a noninvasive approach remains limited. In this study, we designed a composite scaffold by labeling poly(lactic-co-glycolic acid) (PLGA) with gold nanoclusters (Au NCs), which were used for tracking in vivo degradation through dual-modal fluorescence/computed tomography (CT) imaging. The diameter of the Au NCs was approximately 2.5 nm, and the emission peak was at a wavelength of 700 nm. After labeling PLGA with the Au NCs, the fluorescence intensity of the Au NC/PLGA composite scaffold reached 9.0 × 10 9 (p/s/cm 2 /sr)/(μW/cm 2 ), and the CT density of the scaffold increased to 200 HU. After the composite scaffold was implanted subcutaneously into nude mice, a continuous decrease in the fluorescence signal and CT value was observed. The mean fluorescence intensity was 8.3 × 10 9 , 3.17 × 10 9 , 2.26 × 10 9 , 2.11 × 10 9 , and 1.82 × 10 9 (p/s/cm 2 /sr)/(μW/cm 2 ) from the first week to the fifth week, respectively. The mean CT value changed from 222.6 to 185.9, 149.1, 112.5, and 55.2 (Hounsfield unit, HU) at the different timepoints. Compared with the change in the fluorescence intensity, the change in the CT value was similar to the change in the weight, and the Pearson correlation coefficient between the change in the CT value and weight was 0.8626. Furthermore, the structure and morphology of the scaffolds at different timepoints were analyzed by three-dimensional (3-D) reconstruction. This novel method of noninvasive dynamic monitoring of biodegradable polymers in vivo provides insight into choosing suitable biomaterials for tissue engineering and regenerative medicine.

Purpose Combination chemotherapy is gradually receiving more attention because of its potential synergistic effect and reduced drug doses in clinical application. However, how to precisely control drug release dose and time using vehicles remains a challenge. This work developed an efficient drug delivery system to combat breast cancer, which can enhance drug effects despite reducing its concentration. Methods Controlled-release poly-lactic-co-glycolic acid (PLGA) scaffolds were fabricated by E-jet 3D printing to deliver doxorubicin (DOX) and cisplatin (CDDP) simultaneously. Results This drug delivery system allowed the use of a reduced drug dosage resulting in a better effect on the human breast cancer cell apoptosis and inhibiting tumor growth, compared with the effect of each drug and the two drugs administrated without PLGA scaffolds. Our study suggested that DOX-CDDP-PLGA scaffolds could efficiently destroy MDA-MB-231 cells and restrain tumor growth. Conclusions The 3D printed PLGA scaffolds with their time-programmed drug release might be useful as a new multi-drug delivery vehicle in cancer therapy, which has a potential advantage in a long term tumor cure and prevention of tumor recurrence.

Biodegradable porous biomaterial scaffolds play a critical role in bone regeneration. In this study, the porous nano-hydroxyapatite/collagen/poly(lactic-co-glycolic acid)/graphene oxide (nHAC/PLGA/GO) composite scaffolds containing different amount of GO were fabricated by freeze-drying method. The results show that the synthesized scaffolds possess a three-dimensional porous structure. GO slightly improves the hydrophilicity of the scaffolds and reinforces their mechanical strength. Young’s modulus of the 1.5 wt% GO incorporated scaffold is greatly increased compared to the control sample. The in vitro experiments show that the nHAC/PLGA/GO (1.5 wt%) scaffolds significantly cell adhesion and proliferation of osteoblast cells (MC3T3-E1). This present study indicates that the nHAC/PLGA/GO scaffolds have excellent cytocompatibility and bone regeneration ability, thus it has high potential to be used as scaffolds in the field of bone tissue engineering.

Normally a useful tissue engineering scaffold should provide suitable micropores and fibrous structure to mimic the natural extracellular matrix (ECM), which can induce desired cellular activities and guide tissue regeneration. In this work, a microporous and micro/nanofibrous silk
fibroin-poly(lactic-co-glycolic acid) (SF-PLGA) scaffold prepared by thermal induced phase separation (TIPS) was described. Minitab statistical software was used to analyze the TIPS parameters effect on the pore size, porosity and fibre diameter. Results showed that the ECM-like architecture
could be achieved by TIPS with the mean porosity 81 ± 16% and the pore size among 0.0–66.7 μm. Besides, cell viability tests revealed that the cells could proliferate on the SF/PLGA scaffolds. These results reveal that the SF-PLGA scaffolds prepared by TIPS are suitable
for the future tissue engineering applications.

The repair of bone defects in patients with diabetes mellitus (DM) is a challenge in clinical treatment mainly because of inhibited osteogenesis caused by stem cell dysfunction. Our research found that high glucose induced the activation of GSK-3β, which played an important role in this process. Inhibited GSK-3β induced an acceptable osteogenic effect of lithium ions . However, whether lithium-containing biomaterials can exert a good bone-repair effect under the pathological state of DM is still unclear. Therefore, we synthesized a type of lithium-containing mesoporous bioactive glasses (Li-MBG), and explored their in vitro roles in regulating the proliferation, migration and osteogenic differentiation of BMSCs under the high-glucose microenvironment , and the underlying mechanism. Meanwhile, Li-MBG/PLGA composite scaffolds were prepared by 3D printing , followed by investigating their in vivo effects on repairing critical skull defects of DM mice. We found that Li-MBG reversed high glucose-induced suppression in the proliferation, migration and osteogenic differentiation of BMSCs by upregulating Itga3 and activating the β-catenin/Tcf7/Ccn4 signaling pathway. Li-MBG/PLGA composite scaffolds could effectively repair skull defects of DM mice by recruiting stem cells, presenting an excellent osteogenesis effect. Our findings provide references for clinical bone defect repair in DM patients.

Due to numerous merits, polyesters have been widely used for the preparation of porous scaffolds for biomedical applications. However, insufficient dimensional stability and weak cell interaction are two critical challenges to highly porous polyester scaffolds fulfilling their roles during applications. Here, we report the surface modification of PLGA scaffolds with air plasma for simultaneously tackling the dimensional shrinkage of PLGA scaffolds and improving scaffold–tissue integration. Highly porous PLGA scaffolds with interconnected pore structures and well orientated microtubules were prepared through lyophilization, and an air plasma process with varied duration was applied to the porous scaffolds. Water contact angle measurements indicated that moderate treatment (120 seconds) can significantly increase the wettability without distorting the morphology of and damaging the PLGA scaffolds. Further characterization (SEM, XPS, μ-CT, and theoretical computations) revealed that the plasma treatment enabled the generation of carboxyl groups on the surface of the PLGA scaffold, and the hydrophilic PLGA chain tends to extend under aqueous conditions in comparison to the hydrophobic pristine PLGA chain. Consequently, this could eliminate the hydrophobic-scaffold/water interface and weaken the driving force behind the distortion of the scaffolds in aqueous conditions, and finally inhibit overall shrinkage and maintain the microstructures of the porous scaffolds. Similar results were observed with porous PLA scaffolds, indicating the generality of air plasma treatment in inhibiting the shrinkage of polyester scaffolds. In the meantime, in vitro cell attachment experiments and in vivo subcutaneous implantation results consistently indicated that better maintaining the microstructure could significantly enhance the short-term cell attachment/proliferation on the scaffold and the long-term scaffold–tissue integration. These findings may inspire many creative and efficient strategies for the surface modification of polyester scaffolds to improve their performance for biomedical applications.

Pathogenic infection inevitably provokes chronic inflammation and stalls the normal orchestrated course of wound-healing cascades. However, current antibacterial therapies focus on the inherent bactericidal functions of biomaterials. To take full advantage of infection microenvironment (IME) peculiarities, an IME-activated nanocatalytic membrane consisting of electrospun poly (lactic-co-glycolic acid) (PLGA) scaffolds, MXene/Ag 2 S bio-heterojunctions (MX/AS bio-HJs), and lactate oxidase (LOx) for chronic cutaneous regeneration is devised and developed. In this “intelligent” system, PLGA membranes gradually degrade to lactate, and LOx consumes lactate to yield abundant hydrogen peroxide (H 2 O 2 ) in a microenvironment-responsive manner. In addition, MX/AS bio-HJs in membranes not only exert benign photothermal effects and generate reactive oxygen species upon NIR light, but also catalyze the produced H 2 O 2 to overwhelming hydroxyl radicals (·OH) through Fenton-like reactions, which all result in rapid synergistic sterilization. Furthermore, in vivo assays demonstrate that the nanocatalytic membranes reshape the stalled chronic wound into a regenerative wound by massacring bacteria, stopping bleeding, boosting epithelialization/collagen deposition of the wound beds, and promoting angiogenesis. As envisaged, the proposed tactic opens up a promising opportunity for arming membranes with IME-responsive nanocatalytic activity to remedy bacteria-invaded stalled full-thickness wounds.
