A major citrus postharvest pathogen,Penicilliumitalicum(P. italicum), causes substantial economic losses in citrus. In this study, a citral nanoemulsion containing polymethoxylated flavonoids (PMFs), the antimicrobial compounds from citrus, was prepared. The antifungal activity and potential antifungal mechanisms of the nanoemulsion againstP. italicumwere evaluated. The results showed that the growth ofP. italicumwas effectively inhibited by the nanoemulsion, with a minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of 62.5 and 250 mg L−1, respectively. The nanoemulsion significantly inhibited spore germination and mycelial growth, and it altered the morphology ofP. italicum. In addition, the permeability of the cell membrane increased with increasing nanoemulsion concentrations, as evidenced by a rapid rise in extracellular electric conductivity and stronger red fluorescence from mycelia (propidium iodide staining). Compared with the control, the nanoemulsion treatment induced a decrease in total lipid and ergosterol contents inP. italicumcells by 64.61% and 60.58%, respectively, demonstrating that membrane integrity had been disrupted. The results indicated that the PMFs-loaded nanoemulsion exerted antifungal activity againstP. italicumby disrupting cell membrane integrity and permeability; such a nanoemulsion may be used as a potential fungicide substitute for preservation in citrus fruits. Keywords:antifungal mechanism;citrus blue mold;membrane integrity;membrane permeability;P. italicum

Citrus is susceptible to Penicillium digitatum ( P. digitatum ) infection in post-harvest storage, resulting in enormous economic losses. This study aimed to investigate the antifungal activity and potential mechanism of the combination of Polymethoxylated flavones (PMFs) and citral (two natural antifungal components derived from citrus) against P. digitatum in vitro and citrus fruit. The results show that PMFs can enhance the antifungal activity of citral nanoemulsion, and PMFs-loaded citral nanoemulsion (PCT) has significant antifungal activity in a concentration-dependent manner. PCT can evidently inhibit spore germination and mycelial growth in vitro , and effectively control the growth of green mold on postharvest citrus fruit. Furthermore, PCT treatment resulted in the alteration of mycelia morphology, accumulation of reactive oxygen species, and membrane lipid peroxidation. These changes can disrupt the normal structure and function of the cell membrane, as evidenced by the reduction of total lipid and ergosterol content in the mycelia and the stronger red fluorescence of the cells emitted after PI staining. Based on the above results, we infer that PCT has a strong inhibitory effect on P. digitatum , and its potential mechanism is related to the destruction of the cell membrane. Therefore, PCT can be considered as a botanical fungicide for citrus preservation.

Polymethoxylated flavones (PMFs) are a class of flavonoid compounds predominantly present in citrus and are renowned for their extensive biological activity (antiobesity, antioxidant and cardiovascular protection). Recent advancements have elucidated the biosynthetic pathway of PMFs, although the transcriptional regulatory mechanisms involved are not yet fully understood. In this study, we used PMF-rich Bingtangcheng ( Citrus sinensis Osbeck) fruit (BTC) and treated it with exogenous methyl salicylate (MeSA). Our study demonstrates that MeSA treatment dynamically regulates the biosynthesis of PMF monomers in citrus peels during low-temperature storage (10 °C), with 0.2 mmol · L -1 MeSA combined with 14-day storage effectively enhancing PMF accumulation. The transcriptomic analyses demonstrated that MeSA treatment activated the expression of essential genes associated with the PMF biosynthetic pathway, including FNSII , F3'H , and COMT . Additionally, MeSA also activated the biosynthesis and signaling pathways of other phytohormones. A transcription factor, CitERF92 , from the ERF family B3 group, was significantly induced by MeSA and strongly correlated with the expression of CitFNSII-1 . Dual-luciferase assays and electrophoretic mobility shift assays demonstrated that CitERF92 could bind to and significantly activate the promoter activity of CitFNSII-1 . Subsequent transient overexpression experiments confirmed that CitERF92 markedly increased the content of PMF monomers in the BTC peel. These results reveal the CitERF92-CitFNSII-1 module as a novel regulator of PMF biosynthesis, enhancing the understanding of the regulatory network that controls PMF production in citrus.

Introduction Citri Reticulatae Pericarpium Viride (CRPV, Qing Pi in Chinese) has been widely used in traditional Chinese medicine. Polymethoxylated flavonoids (PMFs), which are a special group of flavonoids with strong antitumor activity, are broadly distributed in citrus peels. However, systematic investigation of antitumor PMFs in CRPV has received little attention to date. Objectives An MCF-7 cell biospecific extraction method integrated with neutral loss/diagnostic ion filtering-based HPLC-QTOF-MS/MS strategy was developed for rapid and specific profiling of antitumor PMFs and systematic identification of PMFs in CRPV. Methodology By incubating MCF-7 cells with CRPV extract, potential antitumor PMFs specifically bound to cells and were isolated. Then, by systematic investigation of fragmentation pathways, neutral loss and diagnostic ion filtering strategies were proposed to comprehensively and accurately identify PMFs. Results Sixteen antitumor PMFs were unambiguously or tentatively identified. Among them, minor compound 15 (5-hydroxy-6,7,8,3′,4′-pentamethoxyflavone with a free hydroxyl group at C-5) exhibited excellent antitumor activity, with an IC 50 value of 2.81 ± 0.76 μg/mL, which is lower than that of 5-fluorouracil (IC 50 , 4.92 ± 0.83 μg/mL). Nobiletin ( 12 ) and tangeretin ( 16 ), two major PMFs, presented moderate antitumor activities with IC 50 values of 13.06 ± 1.85 and 17.07 ± 1.18 μg/mL, respectively, and their contents were sensitively and precisely determined. Conclusions To the best of our knowledge, this is the first report on the systematic investigation of antitumor PMFs in CRPV. The study will lay a foundation for the quality control and clinical application of CRPV.

Previous studies have shown that polymethoxylated flavonoids-loaded citral emulsion (PCT) can inhibit the growth and reproduction of Penicillium in citrus; however, PCT is difficult to apply to fruit preservation due to its high fluidity and volatility. Therefore, in this study, we combined PCT with chitosan (CS) to investigate the effect of a composite coating on citrus preservation. The results showed that compared to the control group, the CS-PCT group could effectively reduce the decay rate and maintain moisture availability, color difference, and hardness. Moreover, the contents of nonenzymatic antioxidants and volatile substances with antimicrobial activity were better preserved. In addition, the activities of related antioxidant enzymes were greater in the treatment group, and the expression of the corresponding enzyme-encoding genes was upregulated. Consequently, CS-PCT treatment could effectively maintain fruit quality and improve the resistance of citrus fruits during storage; moreover, it can be considered a nontoxic and efficient citrus preservative.

Background The efficient and timely determination of polymethoxylated flavones (PMFs, primarily nobiletin and tangeretin) and flavanone glycosides (primarily hesperidin) in Citri Reticulatae Pericarpium (CRP) is of paramount importance for the production of CRP and the evaluation of its efficacy. Conventional analytical methods including chromatography-based approaches commonly provide high sensitivity and selectivity, but require bulky equipment and complicated procedures performed by professional technicians and are thus inconvenient in practical applications. Therefore, there is a clear need for portable and miniaturized sensing platforms that can rapidly and simultaneously detect PMFs and hesperidin in CRP product. Results A state-of-the-art three-dimensional porous graphene electrode was first fabricated by direct laser scribing of a poly(ether-ether-ketone) (PEEK) film for electrocatalysis of nobiletin , tangeretin and hesperidin . Kinetic analysis was conducted to investigate the reaction mechanisms of these three flavonoids at such prepared PEEK-laser induced graphene (PEEK-LIG) electrodes. Since the as-prepared PEEK-LIG electrodes exhibited high electrocatalytic efficiency towards these three flavonoids , a portable electrochemical sensing platform assembled with a smartphone, a miniatured electrochemical workstation, and an integrated PEEK-LIG microchip was developed. Consequently, the developed portable electrochemical sensing platforms exhibited great sensitivity and low detection limits for both PMFs and hesperidin. More importantly, tests conducted on real CRP extract samples demonstrated that the developed portable electrochemical sensing platform exhibited high validity, high reliability, as well as excellent reproducibility. Significance This is the inaugural report on the portable and simultaneous determination of PMFs and hesperidin in the pericarp of Citrus Reticulata , which may be utilized for differentiating CRP products. Furthermore, the portable and powerful electrochemical sensing platforms developed could also potentially be applied for a wide range of analytes, thanks to their simple and rapid fabrication and determination processes.

HBP-A is the main active component of a traditional Chinese medicine Huaizhen Yanggan Capsule, for the remarkable treatment of knee osteoarthritis (KOA). This study aimed to elucidate the ameliorative effect of HBP-A on meniscus hypertrophy and mineralisation in KOA and the molecular mechanism of its action. An Hartley guinea pig model of KOA that underwent anterior cruciate ligament transection (ACLT) and a model of rat primary meniscus fibrochondrocytes (PMFs) were used to investigate the ameliorative effect of HBP-A on meniscal hypertrophy and calcification and its signal transduction mechanism of action. The results show that Guinea pig's meniscus width, as well as the area of meniscus calcification and meniscus and articular cartilage injury score, were significantly reduced in the HBP-A intervention group compared to the ACLT group. The expression levels of mtrix metalloproteinase 13 (MMP13), runt-related transcription factor 2 (Runx2), Indian hedgehog (Ihh), alkaline phosphatase (ALP), and ankylosis homologue (ANKH) at the protein and gene level significantly decreased in the HBP-A intervention group compared to the ACLT group. In vitro study, apoptosis, hypertrophy, and calcification of rat PMFs after 10% stretch force were significantly improved with HBP-A intervention. Western blot and RT-qPCR showed that hypertrophy, calcification, and p38 MAPK signalling pathway-related markers of PMFs were incredibly depressed in the HBP-A intervention group compared to the 10% stretch force group. In conclusion, HBP-A can slow down meniscus hypertrophy and mineralisation induced by abnormal mechanical loading, and its mechanism of action may be through the p38-MAPK signalling pathway.
