Background and purpose Atherosclerosis is the basis of cardiovascular disease. Ferroptosis is a form of programmed cell death characterized by lipid peroxidation, which contributes to atherogenesis. The plant extract PNS ( Panax notoginseng saponins ), containing the main active ingredients of Panax notoginseng , exhibits anti-atherogenic properties. Herein, we determined whether PNS and its major components could attenuate atherosclerosis by suppressing ferroptosis and revealed the underlying mechanism(s). Experimental approach The anti-atherogenic effects of PNS and their association with inhibition of ferroptosis was determined in apoE −/− mice. In vitro, the anti-ferroptotic effect and mechanism(s) of PNS components were demonstrated in the presence of ferroptosis inducers. Expression of ferroptosis markers and the ubiquitination of Keap1 were evaluated in USP2 −/− macrophages. Finally, the anti-atherogenic effect of USP2 knockout was determined by using USP2 −/− mice treated with high-fat diet (HFD) and AAV-PCSK9. Key results PNS inhibited ferroptosis and atherosclerosis in vivo. PNS suppressed ferroptosis and ferroptosis-aggravated foam cell formation and inflammation in vitro. Mechanistically, PNS and its components activated Nrf2 by antagonizing Keap1, which was attributed to the inhibition of USP2 expression. USP2 knockout antagonized ferroptosis and ferroptosis-aggravated foam cell formation and inflammation, thus mitigating atherosclerosis. USP2 knockout abolished inhibitory effects of PNS on foam cell formation and inflammation in vitro. Conclusion and implications PNS reduced USP2-mediated Keap1 de-ubiquitination and promoted Keap1 degradation, thereby activating Nrf2, improving iron metabolism and reducing lipid peroxidation, thus contributing to an anti-atherosclerotic outcome. Our study revealed the mechanism(s) underlying inhibition of ferroptosis and atherosclerosis by PNS. Graphical

At present, optical fiber surface plasmon resonance (SPR) biosensing technology faces two significant challenges, enhancing the detection performance and expanding the detected target types. To address this, this paper proposes a novel optical fiber SPR biosensor, modified with programmable polypeptide nanofibers (PNs) and polydopamine nanospheres covered gold nanoparticles (Au@PDA NPs). Peptide sequences have been applied in optical fiber SPR sensing due to their editable, self-assembling, and low-toxicity properties. In reported sensors, peptide sequences are engineered to self-assemble into peptide nanofibers, increasing the sensor’s specific surface area and providing customizable active sites. Au@PDA NPs further enhance the refractive index sensitivity and surface activity of the sensor. In this study, an SPR biosensor was employed to detect rabbit IgG. The results demonstrate a biological sensitivity of 0.0262 nm/(ng/mL) and a detection limit of 3.85 ng/mL. Compared to SPR sensors modified with single nanoparticles, the proposed sensor exhibits significant improvements in both sensitivity and detection limit. In addition to performance enhancement, this work also offers a systematic approach to improving the performance of sensors for different targets. By programming peptide sequences, various detectable substances can be detected. The method is cost-effective and highly efficient, which holds great significance for improving the commercial value of optical fiber SPR sensors.

Developing an easily synthesized and efficient adsorbent for separating Panax notoginseng saponins (PNS) from Sanqi extract remains a significant challenge. Here, a novel rosin-based polymer composite (RBM/DT) was synthesized using hydrogenated rosin glycol acrylate (HREGA), divinylbenzene (DVB), and N,N-dimethylaminoethyl methacrylate (DMAEMA), in combination with diatomite, a naturally occurring adsorbent. The physicochemical properties of RBM/DT were characterized by multiple analytical techniques, revealing a mesoporous structure, narrow particle size distribution, and good thermal stability. Following the application of RBM/DT for the purification of PNS, the content of PNS increased markedly from 49.32% to 88.33%. Quantum chemical calculations indicated that van der Waals forces, hydrogen bonding, and electrostatic interactions contributed to the process. Thermodynamic analysis confirmed that the adsorption was spontaneous, endothermic, and involved chemisorption. Collectively, these findings suggest that RBM/DT is an effective, thermally stable, and reusable biosorbent for PNS purification from Sanqi extract.
