Currently, the co-immobilization of multi-enzymes on a solid carrier and their applications in cascade catalytic reactions are primarily studied. In this study, a MOF/polymer porous membrane composed of DUT-5 incorporated polyvinylidene fluoride (PVDF) is constructed and employed in the co-immobilization of porcine pancreatic lipase (PPL) and glucose oxidase (GOx) for the first time. The loading amounts of DUT-5 nanoparticles were varied to optimize the subsequent immobilization efficiency for the two enzymes. The adsorption conditions, isotherms and kinetics for both PPL and GOx were investigated systematically, and the optimum adsorption conditions were determined and used in the co-immobilization of the two enzymes step by step. It is interesting that the maximum adsorption amounts for the two enzymes in the co-immobilization process are similar to the results obtained in the single enzyme immobilization, which indicates the better compatibility of PPL and GOx in the hybrid membranes. The catalytic activities of the co-immobilized enzymes were evaluated using a cascade oxidation reaction, in which H2O2 produced in the GOx-catalyzed oxidation of glucose worked as the oxidant in the subsequent oxidation of styrene. Further, the catalytic efficiency of the co-immobilized enzymes is compared to that of the mixed free PPL and GOx. The improved catalytic rates and better reusability of the co-immobilized enzymes highlight the broader applications of MOF-based membranes in the co-immobilization of two or more enzymes and the catalytic applicabality in the cascade organic reactions.

Graphical Lip synthing: A lipase-catalyzed, green, and efficient approach for the cross-aldol reaction promoted by water with high yield and good stereoselectivity is described. PPL, lipase from porcine pancreas, is first reported to catalyze direct asymmetric aldol reactions between aromatic aldehydes and cyclic ketones. More importantly, the catalytic activity of PPL was greatly promoted by a small quantity of water at 37 °C. A wide range of aromatic aldehydes reacted with cyclic ketones to provide the corresponding aldol products with high yields (up to 99 %) and moderate to good stereoselectivity (up to 90 % ee and 99:1 dr ).

PPL (lipase from porcine pancreas) was found to catalyze asymmetric cross aldol reactions of aromatic and heteroaromatic aldehydes with various ketones in ionic liquid ([BMIM][PF 6 ]) for the first time. Interestingly, PPL exhibited high catalytic activity and excellent stereoselectivity in this efficient and recyclable room temperature ionic liquid in the presence of moderate water, similar to the results obtained in organic solvents. High yields of up to 99%, excellent enantioselectivities of up to 90% ee, and good diastereoselectivities of up to >99:1 dr were achieved.

A nanocomposite (SBA-CIL-CS) which was combined mesoporous silica SBA-15 material with chitosan via ionic liquid as the bridging agent was successfully fabricated. The morphology and structure of the nanocomposite were characterized in detail with transmission electron microscope, fourier transform infrared spectroscopy, thermogravimetric analysis and nitrogen adsorption–desorption techniques. SBA-CIL-CS was investigated as an efficient support for immobilization of porcine pancreas lipase (PPL) and possessed high immobilization efficiency. The properties of immobilized enzyme (SBA-CIL-CS-PPL) such as activity, stability and reusability have been significantly improved, and a preferable pH and temperature tolerance were obtained as well. Results demonstrated the inorganic-organic nanocomposite could be used as an ideal support for enzyme immobilization.

Cutin and suberin in the rice seed coat serve as a lipid barrier that controls water entry into the seed. However, the molecular mechanisms underlying the permeability properties of seed coats remain largely unknown. In this study, we revealed that the rice EXTENSIN-like protein OsPEX1 influences seed germination by modulating water permeability in the seed coat. The germination rate of the gain-of-function pex1/pex1 mutant was lower than that of the wild-type seeds, although their seed vitality was comparable to that of the wild type. Morphological observations revealed that embryo growth in the pex1/pex1 mutant was inhibited. Further investigation indicated that water absorption in the seed coat around the micropyle was impaired during the early stage of germination. Integrated transcriptional and metabolic analyses suggested that the increased deposition of cutin and suberin may be associated with the overexpression of OsClo5 , a transcriptional repressor, in the mutant. It is proposed that abnormal deposition of cutin and suberin in the seed coat may be the major influencing factors contributing to the reduced impermeability of pex1/pex1 seeds. These results indicate that OsPEX1 plays a critical role in modulating water absorption in the seed coat during seed germination.

Soil salinity and drought are the two most common and frequently co-occurring abiotic stresses limiting cotton growth and productivity. However, physiological mechanisms of tolerance to such condition remain elusive. Greenhouse pot experiments were performed to study genotypic differences in response to single drought (4% soil moisture; D) and salinity (200 mM NaCl; S) stress and combined stresses (D + S) using two cotton genotypes Zhongmian 23 (salt-tolerant) and Zhongmian 41 (salt-sensitive). Our results showed that drought and salinity stresses, alone or in combination, caused significant reduction in plant growth, chlorophyll content and photosynthesis in the two cotton genotypes, with the largest impact visible under combined stress. Interestingly, Zhongmian 23 was more tolerant than Zhongmian 41 under the three stresses and displayed higher plant dry weight, photosynthesis and antioxidant enzymes activities such as superoxide dismutase (SOD), peroxidase (POD) catalase (CAT) and ascorbate peroxidase (APX) activities compared to control, while those parameters were significantly decreased in salt-stresses Zhongmian 41 compared to control. Moreover, Na + /K + -ATPase activity was more enhanced in Zhongmian 23 than in Zhongmian 41 under salinity stress. However, under single drought stress and D + S stress no significant differences were observed between the two genotypes. No significant differences were detected in Ca 2+ /Mg 2+ -ATPase activity in Zhongmian 41, while in Zhongmian 23 it was increased under salinity stress. Furthermore, Zhongmian 23 accumulated more soluble sugar, glycine-betaine and K + , but less Na + under the three stresses compared with Zhongmian 41. Obvious changes in leaf and root tips cell ultrastructure was observed in the two cotton genotypes. However, Zhongmian 23 was less affected than Zhongmian 41 especially under salinity stress. These results give a novel insight into the mechanisms of single and combined effects of drought and salinity stresses on cotton genotypes.

The co-occurrence of heavy metal contamination and biodegradable microplastic (polylactic acid, PLA) pollution poses increasing risks to terrestrial plant communities and soil functioning, yet species-specific responses to combined stress remain poorly understood. Cd and microplastics frequently co-occur in agricultural soils, where microplastics can alter cadmium mobility, bioavailability, and transport pathways, potentially modifying metal toxicity and plant stress responses compared with single-pollutant exposure. We investigated the responses of the invasive Bidens pilosa and the native Solanum nigrum grown in monoculture and mixed culture under combined cadmium (Cd) and biodegradable microplastic (PLA) stress by integrating plant growth, photosynthetic performance, oxidative physiology, and rhizosphere biochemical processes. Combined Cd–MP exposure markedly reduced plant growth, chlorophyll content (SPAD), photosystem II efficiency (Fv/Fm), nitrogen accumulation, biomass production, and rhizosphere enzyme activities associated with carbon, nitrogen, and phosphorus cycling. However, B. pilosa maintained greater physiological stability under stress, characterized by higher antioxidant enzyme activities (SOD, CAT, POD), lower reactive oxygen species (H 2 O 2 , O 2 ˙ − ) accumulation, and reduced lipid peroxidation (MDA), whereas S. nigrum exhibited stronger oxidative damage and functional impairment. Multivariate analyses further revealed that root antioxidant capacity was closely associated with rhizosphere microbial enzyme activity, suggesting a root-centered regulatory mechanism linking plant stress tolerance to soil functioning. Overall, the invasive species showed greater tolerance to combined contamination and maintained relatively higher rhizosphere functional activity than the native species, indicating that multi-pollutant stress may alter competitive interactions between invasive and native plants in contaminated environments.

Nitrogen plays a positive role in rice growth, whereas salt negatively affects it. However, rice varieties differ in nitrogen responses and whether nitrogen management can regulate the rice salt tolerance of all varieties remains largely unknown. We then investigated the effects of nitrogen levels on the responses of different rice varieties to salt stress through a comprehensive analysis including morphological, antioxidant physiological, and metabolomics-related indicators. A hydroponic experiment was conducted with three rice cultivars (Hanyou73, Meixiangzhan2, and Xiangyaxiangzhan) grown under two nitrogen treatments (HN: full nitrogen Kamura B nutrient solution and LN: 75% nitrogen reduction Kamura B nutrient solution) and two salt treatments (W: 0 mmol L −1 NaCl and S: 100 mmol L −1 NaCl). This resulted in four treatments: HNW, HNS, LNW, and LNS. Variety, salt, and variety x salt significantly affected the total fresh and dry weights of rice seedlings. Interestingly, compared with the HNS treatment, the LNS treatment increased the total dry weight of Hanyou73 by 32.88%, decreased the total dry weight of Xiangyaxiangzhan by 17.28%, whereas no significant change was detected in Meixiangzhan2. In addition, nitrogen and salt affected the photosynthetic pigment content, soluble protein content, hydrogen peroxide content, and antioxidant response. Moreover, metabolic analysis revealed that the metabolism of tryptophan, D-amino acid, and α-linolenic acid varied with nitrogen level across different salt treatments. This study demonstrated that nitrogen availability differentially regulates salt tolerance in rice varieties. For specific varieties, low nitrogen levels are more effective in reducing reactive oxygen species (ROS) and minimizing cellular damage in rice seedlings.

Nitrate serves not only as an essential nitrogen source but also as a signalling molecule to regulate plant growth and development. However, the mechanisms by which plants respond to nitrate to regulate root growth in rice remain largely unclear. In this study, we demonstrated that the overexpression of OsMADS27 significantly promoted root growth and nitrate absorption in response to nitrate supply, whereas RNA interference (RNAi) lines exhibited the opposite phenotype. OsMADS27 is predominantly expressed in the root, and the mutation of OsNAR2.1 impaired both the response rate and intensity of OsMADS27 under nitrate treatment. Notably, OsMADS27 is co-localised with OsNAR2.1, and the mutation of OsNAR2.1 affected the nuclear localisation of OsMADS27. Yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC) and co-immunoprecipitation (Co-IP) assays confirmed a direct protein interaction between OsMADS27 and OsNAR2.1. More importantly, chromatin immunoprecipitation followed by qPCR (ChIP-qPCR), dual-luciferase (Dual-LUC) and yeast one-hybrid (Y1H) assays demonstrated that OsMADS27 could directly regulate the expression of OsMADS57 to control root growth. Furthermore, OsMADS27 could also promote the expression of several nitrate transporter genes, including OsNRT1.2 , OsNRT2.1 , OsNRT2.3a and OsNRT2.4 , to regulate nitrate absorption in rice. Collectively, this study uncovers a novel pathway in which OsMADS27 promotes root growth in response to nitrate by directly regulating OsMADS57 expression in an OsNAR2.1-dependent manner. These findings will broaden our understanding and provide a novel regulatory network of OsMADS genes mediating root development through nitrate signalling in rice, which also has an important application value for molecular breeding to improve nitrogen use efficiency.

Ectomycorrhizal (EM) fungi play important roles in nutrient cycling and plant community establishment in forest ecosystems. Effects of EM formation on global alterations of the transcriptome and metabolome during plant-fungal interaction and the key metabolites involved in EM development are largely unknown. Here, dual RNA-Seq and untargeted metabolomic analyses were used to reveal stage-specific and core responses of Pinus yunnanensis and Lactarius deliciosus during mycorrhizal colonization. We found that L. deliciosus colonization in P. yunnanensis roots induced different transcriptional changes across three interaction stages, with a small core of genes consistently regulated at all stages. Concentrations of retinol (vitamin A) and retinoic acid increased while that of B group vitamins decreased during EM formation, which was coordinately regulated by these two plant-fungus partners, with L. deliciosus possibly playing a dominant role. Exogenous retinol altered the diameter and mantle thickness of P. yunnanensis - L. deliciosus EM tips and affected host plant growth and phosphorus acquisition . In the absence of L. deliciosus, exogenous retinol increased the root diameter and the number of root tips of P. yunnanensis . Furthermore, the concentration of auxin increased, but that of abscisic acid decreased during EM formation, and the genes involved in plant hormone signal transduction were gradually activated, and auxin and cytokinin signal transduction potentially played a positive role in this EM symbiosis. In conclusion, we propose that the interaction of P. yunnanensis and L. deliciosus alters vitamin metabolism, which may further affect plant hormone biosynthesis and signal transduction, modulating root morphology and EM traits.

The ancient bayberry demonstrates superior resistance to both biotic and abiotic stresses compared to cultivated bayberry, yet the underlying mechanisms remain largely unexplored. This study investigates whether long-term bayberry cultivation enhances stress resistance through modulation of tissue-specific microbes and metabolites. Employing microbiome amplicon sequencing alongside untargeted mass spectrometry analysis, we scrutinize the role of endosphere and rhizosphere microbial communities and metabolites in shaping the differential resistance observed between ancient and cultivated bayberry trees. Our findings highlight the presence of core microbiome and metabolites across various bayberry tissues, suggesting that the heightened resistance of ancient bayberry may stem from alterations in rhizosphere and endosphere microbial communities and secondary metabolites. Specifically, enrichment of Bacillus in roots and stems, Pseudomonas in leaves, and Mortierella in rhizosphere soil of ancient bayberry was noted. Furthermore, correlation analysis underscores the significance of enriched microbial species in enhancing ancient bayberry's resistance to stresses, with elevated levels of resistance-associated metabolites such as beta-myrcene, benzothiazole, L-glutamic acid, and gamma-aminobutyric acid identified through GC–MS metabolomics analysis. The beneficial role of these resistance-associated metabolites was further elucidated through assessment of their promotive and allelopathic effects, as well as their phytostatic and antioxidant functions in lettuce plants. Ultimately, our study delves into the intrinsic reasons behind the greater resistance of ancient bayberry to biotic and abiotic stresses by evaluating the impact of long-term planting on the microbial community and metabolites in the bayberry endosphere and rhizosphere, shedding light on the complex dynamics of host-microbial interactions.

Triclosan (TCS) is a broad-spectrum antimicrobial disinfectant widely used in pharmaceuticals and personal care products (PPCPs). Due to the extensive usage of PPCPs, TCS inevitably entered the environment and pose harmful effects on the ecosystem. Phytoremediation is an attractive approach to remove TCS from the environment. In this study, a gene encoding the anti-TCS nanobody was transformed into Arabidopsis thaliana ( A. thaliana ) to enhance the absorption capacity of TCS. Nanobodies are small antibody fragments (ca. 15 kDa) derived from the variable domain of camelid heavy-chain-only antibodies. We constructed two transgenic lines, the T-S-C line with nanobody expression throughout the plant and the T-S-P line with nanobody expression dominant in the roots, were constructed. The expression of nanobodies in A. thaliana alleviated the phytotoxicity of TCS. T-S-C and T-S-P exhibited significantly stronger tolerance to TCS toxicity than the wild type (WT), in either a solid medium system or a hydroponics system. Under the stress of TCS, the seedlings of both transgenic plants exhibited an increase in root length and fresh weight compared to those of WT. Moreover, in the presence of TCS, the activities of superoxide dismutase, peroxidase, catalase, and glutathione in transgenic plants were higher than those in WT. The concentration of TCS absorbed into the T-S-C and T-S-P plants from the solid medium increased by 50.0% and 24.1%, and from the hydroponics system increased by 55.6% and 38.0%, respectively, compared to those absorbed by WT. This study provides a proof of principle that transforming nanobodies into plants represents a novel technology to improve the efficiency of phytoremediation for environmental pollutants.
