Chronic refractory wounds are a common, costly and recurrent complication of diabetes. Platelet-rich plasma (PRP), a new therapy for chronic wounds, is limited by a long treatment period and unstable effects. FG-4592(Roxadustat) is a new prolyl-4-hydroxylation domain(PHD) inhibitor, which can stabilize hypoxia-inducible factor-1α(HIF-1α) effectively. It is a promising drug for wound repair and needs to be demonstrated via various appropriate application scenarios. This study investigated the effects of combining FG-4592 and PRP to promote diabetic wound healing. Diabetic rats were randomly assigned to five groups: nondiabetic control, diabetic untreated, diabetic + PRP, diabetic + FG-4592, and diabetic + PRP + FG-4592. Diabetes was induced with streptozotocin (STZ). A full-thickness skin defect was created, and FG-4592 (peritoneal injection) and PRP (periwound injection) were administered alone or together. Wound healing was assessed by histological analysis (Hematoxylin-Eosin (HE) and Masson staining) and protein expression of HIF-1α, VEGF(vascular endothelial growth factor), α-SMA(α-smooth muscle actin), CoL1α1(collagen type 1 alpha 1), and SDF-1(Stromal Cell-derived Factor 1) via Western blotting and qRT-PCR(Quantitative real time polymerase chain reaction). Immunohistochemistry(IHC) and immunofluorescence(IF) were also used to evaluate CD34 (Cluster of Differentiation 34), CD31(Cluster of Differentiation 31), VEGF(vascular endothelial growth factor), SDF-1(Stromal Cell-derived Factor 1), PCNA(Proliferating Cell Nuclear Antigen), and Integrin-β1 expression. The untreated diabetic group exhibited impaired wound healing, histological damage, and reduced expression of key proteins compared to the nondiabetic control group. Both PRP and FG-4592 alone improved wound healing, reduced damage, and upregulated protein expression. However, the combination of FG-4592 and PRP showed the most significant improvement. This study suggests that FG-4592 combined with PRP accelerates diabetic wound healing by enhancing endothelial progenitor cell recruitment, neovascularization, and cell proliferation and migration via upregulation of HIF-1α and its target genes. This combination therapy may provide a novel approach for treating chronic diabetic wounds.

Diabetic wounds remain a global challenge due to disordered wound healing led by inflammation, infection, oxidative stress, and delayed proliferation. Therefore, an ideal wound dressing for diabetic wounds not only needs tissue adhesiveness, injectability, and self-healing properties but also needs a full regulation of the microenvironment. In this work, adhesive wound dressings (HA-DA/PRP) with injectability were fabricated by combining platelet rich plasma (PRP) and dopamine-modified-hyaluronic acid (HA-DA). The engineered wound dressings exhibited tissue adhesiveness, rapid self-healing, and shape adaptability, thereby enhancing stability and adaptability to irregular wounds. The in vitro experiments demonstrated that HA-DA/PRP adhesives significantly promoted fibroblast proliferation and migration, attributed to the loaded PRP. The adhesives showed antibacterial properties against both gram-positive and negative bacteria. Moreover, in vitro experiments confirmed that HA-DA/PRP adhesives effectively mitigated oxidative stress and inflammation. Finally, HA-DA/PRP accelerated the healing of diabetic wounds by inhibiting bacterial growth, promoting granulation tissue regeneration, accelerating neovascularization, facilitating collagen deposition, and modulating inflammation through inducing M1 to M2 polarization, in an in vivo model of infected diabetic wounds. Overall, HA-DA/PRP adhesives with the ability to comprehensively regulate the microenvironment in diabetic wounds may provide a novel approach to expedite the diabetic wounds healing in clinic.

Chronic nonhealing wounds represent significant complications of diabetes, bearing a substantial burden and posing risks of disability or mortality. In diabetic wounds, continuous tissue fluid exudation, inflammatory cell migration, fibrosis, and bacterial biofilm formation create a “barrier”, which decreases the treating efficacy of therapeutics. To address these limitations, a recombinant human collagen type III microneedle patch (rhCol III-PRPM) loaded with platelet-rich plasma (PRP) was developed, in which methacrylated rhCol III (rhCol III-MA) loaded with PRP was utilized to form needle tips, while rhCol III-MA formed the base part of patch. RhCol III-PRPM featured adequate mechanical qualities, swelling capacity, and sustained in vitro release of growth factors from the activation of PRP for over 7 days. Leveraging the synergistic effects of rhCol III and PRP, rhCol III-PRPM patches facilitated cell proliferation, migration, and angiogenesis, and reduced oxidative stress. In animal experiment, this microneedle patch effectively promoted the healing of diabetic wounds during a 20-day treatment, partially owing to upregulating integrins and phosphorylated ERK protein levels. Diverging from other microneedle strategy, the rhCol III exhibited “dual functionality,” serving as both the microneedle patch matrix and therapeutic agent, promoting wound healing upon patch dissolution while delivering PRP. The combination of rhCol III and PRP in the form of microneedle patch offered a straightforward and efficacious way for effective diabetic wounds management, and held promise in bringing new possibility in clinical practice.

The occurrence of anastomotic leakage (AL) could lead to leakage of digestive fluid, which erodes surrounding organs, subsequently causing severe intra-abdominal infections, hemorrhaging, and even death. Preventing AL was crucial for significantly enhancing patient quality of life. Thus, we developed an injectable photosensitive hydrogel enriched with platelet-rich plasma (PRP) aimed at preventing AL. This study utilized the Schiff-base crosslinking reaction between photosensitive methacryloyl-substituted gelatin (GM) and oxidized dextran (OD), incorporating PRP to create a multifunctional, tri-crosslinked hydrogel (GM/OD@PRP) that effectively promotes AL healing. This GM/OD@PRP exhibited excellent mechanical properties, biocompatibility, and self-healing capabilities. The hydrogel was loaded with PRP, which was rich in various growth factors that stimulate fibroblast proliferation and angiogenesis, thereby increasing cell proliferation, vascular regeneration, and formation and ultimately promoting healing at the anastomotic site. Furthermore, the superior antimicrobial properties of GM/OD@PRP provide a relatively sterile environment at the healing site, reducing the possibility of abdominal infections. In a rat model of AL, the GM/OD@PRP notably enhanced anastomotic healing prevented the occurrence of fistulae, and demonstrated significant advantages in reducing abdominal adhesions. This GM/OD@PRP holds substantial potential for use in preventing AL and represents a promising new composite material for improving patient quality of life.

Osteoarthritis (OA) is a highly prevalent degenerative joint disease affecting over 500 million people globally, imposing substantial social and economic burdens. Current treatments, including platelet-rich plasma (PRP), are limited by rapid growth factor release, failing to achieve long-term efficacy. To address this, we developed an injectable PRP-loaded hydrogel via in situ assembly of metal-phenolic networks (MPNs), leveraging MPNs' multi-scale porous structure, adjustable mesh size, and dynamic drug interactions to enable "dynamic adhesion" and sustained growth factor release. In vitro studies with OA chondrocytes demonstrated stable PRP release for 14 days, while in vivo evaluations in OA rodent (rat) models confirmed therapeutic effects, including anti-inflammatory, antioxidant, and cartilage repair-promoting activities. This MPN-based hydrogel system validates a novel strategy for sustained PRP delivery, offering a convenient, safe, and effective approach to enhance long-term OA treatment outcomes with potential for clinical translation.

Sorption is prominent in low price, high efficiency, availability, and eco-friendliness. Organic porous materials have the characteristics of easy functionalization, diverse structure and stability, and show great potential in adsorption, energy storage, catalysis, and other fields. A mesoporous phenolic resin-type polymer (PRP) was successfully synthesized and modified by solid state reaction with maleic anhydride to prepare adsorbent (called as PRP-MAH) for sorption of Pb 2+ . The impact of reaction conditions (the pH value, reaction temperature, fresh concentration of solution, ionic strength and reaction time, etc.) was systematically studied. Characterization methods such as SEM, FTIR, and XPS indicated that the synthesized adsorbent PRP-MAH had regular morphology and good stability. The fitting of isothermal adsorption experiment data conforms to Langmuir sorption isotherm, and the sorption capacity reached 366.40 mg·g −1 at 308 K. The kinetic data were consistent with the quasi-second-order model, which indicated that the chemisorption might play the main role in the sorption process. Thermodynamic research manifested that the sorption of Pb 2+ by PRP-MAH was carried out by a spontaneous process at the study temperature. The studies show that PRP-MAH can remove Pb 2+ from water solution through ion exchange, electrostatic attraction, and surface complexation.

Amyloid-β oligomers (AβOs) are the most important toxic species in the brain of Alzheimer’s disease (AD) patient. AβOs, therefore, are considered reliable molecular biomarkers for the diagnosis of AD. Herein, we reported a simple and sensitive electrochemical method for the selective detection of AβOs using silver nanoparticles (AgNPs) as the redox reporters and PrP(95–110), an AβOs-specific binding peptide, as the receptor. Specifically, adamantine (Ad)-labeled PrP(95–110), denoted as Ad-PrP(95–110), induced the aggregation and color change of AgNPs and the follow-up formation of a network of Ad-PrP(95–110)-AgNPs. Then, Ad-PrP(95–110)-AgNPs were anchored onto a β-cyclodextrin (β-CD)-covered electrode surface through the host–guest interaction between Ad and β-CD, thus producing an amplified electrochemical signal through the solid-state Ag/AgCl reaction by the AgNPs. In the presence of AβOs, Ad-PrP(95–110) interacted specifically with the AβOs, thus losing the capability to bind AgNPs and to induce the formation of an AgNPs-based network on the electrode surface. Consequently, the electrochemical signal decreased with an increase in the concentration of AβOs in the range of 20 pM to 100 nM. The biosensor had a detection limit of 8 pM and showed no response to amyloid-β monomers (AβMs) and fibrils (AβFs). On the basis of the well-defined and amplified electrochemical signal of the AgNPs-based network architecture, these results should be valuable for the design of novel electrochemical biosensors by marrying specific receptors.

Blood vessel reconstruction is key for ischemic disease treatment by restoring microvascular perfusion and mitigating pathological tissue stiffening. During neovascularization, the mechanical and biochemical cues presented by the cellular-scale spatial heterogeneity of extracellular matrix (ECM) facilitate the endothelial cells (ECs) spreading and mechanotransduction, thereby driving angiogenesis. Therefore, developing ECM-mimetic proangiogenic biomaterials with compartmentalized spatial heterogeneity is highly desirable but challenging. Here, inspired by the spatially heterogeneous mechanical properties of natural tissues, we designed a series of microgel-annealed hydrogels (GMP) with compartmentalized stress heterogeneity. By integrating microfluidic-synthesized rigid microgels, the soft PRP-derived fibrin matrix was annealed with spatially mechanical domains that direct ECs-guided vascular morphogenesis through mechanotransduction, where rigid microdomains provide anchorage sites for cells adhesion, while soft interstitial matrix permits stalk cells migration and morphogenesis. In vitro, heterogeneous GMP hydrogel augmented ECs mechanotransduction via activating integrin β1-p-FAK-p-MLC signaling pathway and stabilizing VE-CAD/β-cat junctions, promoting 3D vasculogenesis and angiogenesis. In a rat myocardial infarction (MI) model, treatment with the heterogeneous hydrogel enhanced myocardial neovascularization, attenuated ventricular dilation and enhanced cardiac function. These findings not only provide valuable guidance for engineering proangiogenic biomaterials via ECM biomechanical mimicry, but also highlight the promise of spatially heterogeneous hydrogel with engineered mechanical cues essential for de novo blood vessel formation in the treatment of ischemic disease. Statement of significance This work develops a proangiogenic hydrogel that mimics the spatially heterogeneous mechanical properties of the native extracellular matrix (ECM). By annealing rigid microgels into a soft fibrin matrix, we create a biomaterial with compartmentalized stress domains that guide endothelial cell behavior. This engineered heterogeneity promotes 3D vascular network formation by enhancing key mechanotransduction pathways and stabilizing cell-cell junctions. Significantly, in a rat model of myocardial infarction, the hydrogel enhances neovascularization, attenuates tissue damage, and improves cardiac function. Our findings highlight that recapitulating ECM biomechanical heterogeneity is a transformative strategy for engineering biomaterials to drive functional blood vessel regeneration, offering a promising therapeutic approach for treating ischemic diseases.

Bacterial infection and excessive inflammation critically impede wound healing, necessitating the development of multifunctional bioactive materials. Herein, we report a biodegradable composite hydrogel (PPG) fabricated via projection-based 3D biophotocuring, comprising an interpenetrating double-network of gelatin methacryloyl (GelMA) and poly-L-lysine methacryloyl (PLMA), embedded with platelet-rich plasma (PRP). The resulting hydrogel exhibits potent antibacterial activity and robust inhibition of biofilm formation, alongside integrated anti-inflammatory and pro-angiogenic functions. In vitro, incorporating PRP promotes the migratory and proliferative activity of human skin fibroblasts and facilitated macrophage polarization toward the M2 phenotype. In vivo , PPG accelerates wound closure in bacteria-infected models by facilitating re-epithelialization, collagen remodeling, and neovascularization, as evidenced by sustained α-SMA and elevated CD31 expression. Mechanistically, PPG activates the PI3K/AKT/eNOS signaling axis, further supporting angiogenesis and extracellular matrix deposition. RNA-seq validation showed that PPG upregulated DEFB4 and SPRR2F, promoting wound healing and modulating inflammation. Collectively, this work presents a 3D-printed, bioactive hydrogel platform with synchronized antibacterial, immunomodulatory, and pro-regenerative properties, offering strong translational potential for treating infected wounds.

Uncontrolled hemorrhage following severe liver trauma
remains a
critical clinical challenge, as conventional hemostatic agents often
fail to simultaneously control diffuse bleeding and actively support
subsequent tissue repair. There is a pressing need for biomaterials
that integrate immediate hemostasis with pro-healing bioactivity.
Here, we report a tough composite sponge (SBC-AG@PRP) designed to
address this dual requirement. The scaffold is constructed by incorporating
gelatin into a sulfonated bacterial cellulose-alginate matrix, forming
a reinforced interpenetrating network that provides exceptional wet-state
toughness, rapid shape recovery, and high fluid uptake─properties
essential for conforming to dynamically moving liver wounds. This
mechanically optimized scaffold serves as a reservoir for platelet-rich
plasma (PRP), enabling the sustained release of growth factors over
7 days. In vitro , the released factors reprogram
macrophages toward a pro-reparative M2 phenotype and accelerate hepatocyte
migration. In a rat model of penetrating liver trauma, SBC-AG@PRP
achieves significantly faster hemostasis and reduced blood loss compared
to gauze and non-PRP controls. Beyond hemostasis, the controlled release
actively modulates the wound microenvironment by dampening inflammation,
promoting M2 macrophage polarization, and guiding organized collagen
deposition, ultimately facilitating functional liver healing. This
work establishes a synergistic strategy that couples mechanical design
with sustained biochemical signaling for the management of parenchymal
organ injuries.

Infected traumatic wounds remain a formidable clinical challenge because existing bioadhesives provide only tissue adhesion and lack concurrent antimicrobial and regenerative capabilities. Here, we developed an injectable bioadhesive (CγR) that integrates an “antibacterial–adhesive–regenerative” (AAR) triad by precisely tuning the ratio of carboxymethyl chitosan (CMCS), autologous platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA). CγR exhibits > 99.9% bactericidal efficacy against Escherichia coli and Staphylococcus aureus , delivers instantaneous wet-tissue adhesion (> 3 kPa), and sustains a gradient release of PDGF, TGF-β, and VEGF. In a rat model of infected full-thickness wounds, a single application achieved sterile sealing and complete epithelialization within 6 days, increased collagen deposition by 1.5-fold, and markedly drove macrophage polarization toward the prohealing M2 phenotype. By condensing the conventional “suture–antibiotic–dressing” regimen into a one-step procedure, CγR offers a biosafe and readily scalable platform for managing contaminated complex wounds and holds promise for clinical translation. Graphical The injectable bioadhesive CγR integrates carboxymethyl chitosan (CMCS), platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA) to provide antimicrobial, adhesive, and regenerative effects. CγR bioadhesive exhibits sustained release of growth factors, promotes rapid wound closure, and accelerates tissue regeneration in infected wound models.

Thrombin-triggered in situ anchoring enables stable underwater scaffold fixation for 48 h. • Bilayer biomimetic scaffold with strong water absorption captures and retains BMSCs. • CPTT@PRP synergistically upregulates CCL2 and CXCL8 to recruit BMSCs. • CPTT@PRP restores hyaline cartilage anisotropy. Utilising endogenous stem cells with bioactive biomimetic scaffolds to achieve in situ osteochondral regeneration represents a promising strategy in contemporary regenerative medicine. However, existing scaffold fixation methods (e.g., sutures, screws, or photocrosslinking) often cause secondary damage, involve complex procedures, or exhibit poor biocompatibility, and struggle to achieve efficient capture and long-term recruitment of stem cells from bone marrow blood. Herein, we propose a novel concept of “thrombin-triggered in situ anchoring” and integrate it into a type I collagen-based biomimetic bilayer osteochondral scaffold. Upon implantation, the thrombin loaded within the scaffold rapidly catalyzes the conversion of fibrinogen (released after microfracture) into a fibrin network, thereby achieving immediate and stable in situ fixation at the defect site. This adhesive property can be maintained underwater for up to 48 h, avoiding the drawbacks of conventional fixation methods. Concurrently, the scaffold exhibits a “sponge-like” strong water-absorption effect. After implantation into the microfractured defect, it rapidly absorbs bone marrow blood, maximizing the capture and retention of bone marrow mesenchymal stem cells (BMSCs). The sustained release of PRP further establishes a chemotactic gradient that actively guides the directional migration of BMSCs and modulates the microenvironment to induce their bidirectional differentiation. RNA sequencing results indicate that the scaffold synergistically upregulates CCL2 and CXCL8, promoting BMSC chemotaxis. In a rabbit critical-sized osteochondral defect model, CPTT@PRP achieved hyaline-like cartilage regeneration and subchondral bone repair within 12 weeks, improved collagen organization. This work applies the concept of enzymatic in situ anchoring to the design of osteochondral regeneration scaffolds, providing a novel strategy for microfracture-based osteochondral repair, and holds promise for extension to other fields of tissue engineering.
