Pectins, a complex class of polysaccharides, are prominently represented by HG-chains, which are both abundant and extensively studied. Bacteroides species exhibit a remarkable ability to metabolize plant- and animal-derived polysaccharides, including the degradation of HG-type pectins through polysaccharide utilization locus (PUL). However, the effects of structurally diverse HG-type pectins on PUL expression, metabolite production, and metabolic pathways of Bacteroides remain unclear. Addressing this, this study chose HG-type pectins with different molecular weights from citrus (CP) and pomelo (PP), identified Bacteroides species capable of utilizing these pectins, studied the resulting metabolites through non-targeted metabolomics coupled with short-chain fatty acids analysis, and examined the activation of PUL and metabolic pathways by transcriptomic studies. The results showed that Bacteroides thetaiotaomicron A4 and Bacteroides caccae K9 could utilize HG-type pectins with different molecular weights. The production of propionic acid by Bacteroides thetaiotaomicron A4 was significantly affected by the molecular weight of pectins. Utilizing CP, B. thetaiotaomicron A4 enriched metabolites such as carbohydrates, amino acids, peptides, amines, and significantly enhanced pathways such as sphingolipid metabolism and drug metabolism (other enzymes) through PUL75, PUL63, and PUL55. In contrast, when using PP, B. thetaiotaomicron A4 enriched similar metabolites and further upregulated pathways related to sphingolipid metabolism and pyrimidine metabolism. The molecular weight of HG-type pectins differentially affected the expression of carbohydrate-active enzymes and metabolic pathways, resulting in different metabolite profiles. This study aims to contribute to the understanding of structure-activity relationship between pectins and gut microbiota and to inform precision nutrition strategies.

At present, the situation of air pollution is still serious, and research on air filtration is still crucial. For the nanofiber air filtration membrane, the diameter, porosity, tensile strength, and hydrophilicity of the nanofiber will affect the filtration performance and stability. In this paper, based on the far-field electrospinning process and the performance effect mechanism of the stacked structure fiber membrane, nanofiber membrane was prepared by selecting the environmental protection, degradable and pollution-free natural polysaccharide biopolymer pullulan, and polyvinylidene fluoride polymer with strong hydrophobicity and high impact strength. By combining two kinds of fiber membranes with different fiber diameter and porosity, a three-layer composite nanofiber membrane with better hydrophobicity, higher tensile strength, smaller fiber diameter, and better filtration performance was prepared. Performance characterization showed that this three-layer composite nanofiber membrane had excellent air permeability and filtration efficiency, and the filtration efficiency of particles above PM 2.5 reached 99.9%. This study also provides important reference values for the preparation of high-efficiency composite nanofiber filtration membrane. Keywords:electrospinning;biopolymer;composite structure;air filtration;nanofiber membrane

Water-in-water (W/W) Pickering emulsions have aligned with the increasing consumer demands for health foods due to their entirely aqueous formulation with oil-free. In this work, Pullulan (PUL) and Hydroxypropyl methylcellulose (HPMC) were used to construct polysaccharide-based W/W Pickering emulsion system. PUL and HPMC concentrations played crucial roles in droplet sizes and phase behavior of PUL/HPMC Pickering emulsions. Cellulose nanocrystals (CNCs) and cellulose nanofibrils (CNFs) were used as stabilizers to prepare PUL/HPMC Pickering emulsions and the stabilization mechanisms were investigated. PUL/HPMC Pickering emulsions stabilized by CNCs with a rigid rod-like structure and strong interfacial adsorption capacity exhibited smaller and more uniform droplet morphology and higher storage stability, whereas the flexible fibrillar CNFs tended to form entangled networks in the continuous phase and bridge droplets, resulting in droplet deformation and coalescence. Furthermore, environmental stability of PUL/HPMC Pickering emulsions revealed that increasing CNCs concentration could significantly enhance their centrifugal and thermal stability. This work would provide a novel insight into the stabilization behavior of nanocellulose with different morphologies in W/W Pickering emulsions and broaden potential applications of W/W Pickering emulsions in functional food fields.

In this study, pullulan-in-hydroxypropyl methylcellulose (PUL/HPMC) water-in-water Pickering emulsions stabilized by cellulose nanocrystals (CNCs) were prepared for the encapsulation and cultivation of L. plantarum . Results demonstrated that CNCs were mainly adsorbed at the PUL-HPMC interface, thereby effectively inhibiting the aggregation and coalescence of emulsion droplets. L. plantarum was encapsulated in emulsions and primarily distributed in the PUL phase. It was found that the optimal encapsulation concentration of L. plantarum was 9.67 log CFU/g, and the optimal addition of MRS broth was 1.0 wt%. Finally, the viable counts of L. plantarum increased from 4.44 to 7.42 log CFU/g during 30 h of cultivation. Furthermore, an initial pH 5.5 afforded the optimal cultivation performance for L. plantarum , with viable counts increasing from 4.00 to 7.06 log CFU/g during 24 h of cultivation. This study provides a novel microculture platform for efficient L. plantarum cultivation and fully aqueous emulsion systems with food-grade application potential for probiotics-based functional foods.
