Two newly-designed hydrocarbon surfactants, that is, poly(vinyl acetate)- block -poly(1-vinyl-2-pyrrolidone) (PVAc- b -PVP) and PVP- b -PVAc- b -PVP, were synthesized using reversible addition–fragmentation chain transfer polymerization and used to form CO 2 /water (C/W) emulsions with high internal phase volume and good stability against flocculation and coalescence up to 60 h. Their structures were precisely determined by nuclear magnetic resonance, gel permeation chromatography, thermal gravimetric analysis, and differential scanning calorimetry. Besides low temperature and high CO 2 pressure, the surfactant structures were the key factors affecting the formation and stability of high internal phase C/W emulsions, including the polymerization degrees of CO 2 -philic block (PVAc) and hydrophilic block (PVP), as well as the number of hydrophilic tail. The surface tension of the surfactant aqueous solution and the apparent viscosity of the C/W emulsions were also measured to characterize the surfactants efficiency and effectiveness. The surfactants with double hydrophilic tails showed stronger emulsifying ability than those with single hydrophilic tail. The great enhancement of the emulsions stability was due to decrease of the interface tension as well as increase of the steric hindrance in the water lamellae, preventing a frequent collision of CO 2 droplets and their fast coalescence. © 2018 Wiley Periodicals, Inc. We reported the preparation of the poly( N -vinyl pyrrolidone)-b-polyethylene glycol (PVP-b-PEG) double-hydrophilic block copolymer by atom transfer radical polymerization (ATRP). The PVP-b-PEG samples were characterized by nuclear magnetic resonance spectroscopy and Fourier translation infrared spectrum. By taking advantage of its exceptional binding affinity, the perhydroxycucurbit[6]uril ((HO) 12 CB[6]) was used to interact with PVP-b-PEG by hydrogen bond and constructed micelle-like core–shell aggregates with the physical cross-link PVP segments as the core and the extending PEG chains as the outer palisade. The size of aggregates was about 50–300 nm. The particle size of this type of assemblies could be controlled by the weight content of (HO) 12 CB[6]. The size distribution was broad at high concentrations. The broad size distribution at high (HO) 12 CB[6] concentration was due to the coexistence of isolated particles and clusters. These novel micelle-like core–shell aggregates could be controlled by pH value. At pH lower than 6 or pH larger than 13, hydrogen bond interactions were destroyed and the formed (HO) 12 CB[6]-induced PVP-b-PEG assemblies could be destructed.

Two new kinetic hydrate inhibitors (KHIs) named PVP-B and PVP-BP were synthesized successfully and compared with commercial KHIs such as PVP and Inhibex 55W. PVP-B and PVP-BP are both the ramifications of PVP. They were obtained by introducing a t-butyl group and both t-butyl and phenyl groups into PVP molecules, respectively. The hydrate inhibition performances of KHIs were assessed in a sapphire cell through two kinds of onset times of hydrate formation: TVO, the time when hydrate crystals are initially observed by naked eyes, and TPD, the time when rapid and continuous dropping of system pressure begins. TVO was used to evaluate an KHI’s inhibition performance to hydrate nucleation. The length of time period from TVO to TPD was used to evaluate a KHI’s inhibition performance to hydrate growth. The results demonstrate that both PVP-B and PVP-BP are superior to PVP and Inhibex 55W; PVP-BP has better inhibition performance than PVP-B because of the stronger steric hindrance effects of the phenyl group in PVP-BP molecules than those of the t-butyl group in PVP-B molecules. Additionally, we found TVO does not always increase with the increasing dosage of KHI while TPD does. The most suitable dosage of PVP-BP was determined to 0.5 wt % or so, at which the nucleation of hydrate is inhibited most perfectly. Finally, we demonstrated glycol could be used as synergist to improve the performance of PVP-BP remarkably. The most suitable dosage of glycol was determined to be 0.9 wt % or so. Our work presents not only two new KHIs but also important insights into the inhibition mechanism of KHIs.

Polymeric nanoparticles (NPs) are an important category of drug delivery systems, and their in vivo fate is closely associated with delivery efficacy. Analysis of the protein corona on the surface of NPs to understand the in vivo fate of different NPs has been shown to be reliable but complicated and time-consuming. In this work, we establish a simple approach for predicting the in vivo fate of polymeric NPs. We prepared a series of poly(ethylene glycol)-block-poly(d,l-lactide) (PEG-b-PLA) NPs with different protein binding behaviors by adjusting their PEG densities, which were determined by analyzing the serum protein adsorption. We further determined the protein binding affinity, denoted as the equilibrium association constant (KA), to correlate with in vivo fate of NPs. The in vivo fate, including blood clearance and Kupffer cell uptake, was studied, and the maximum concentration (Cmax), the area under the plasma concentration–time curve (AUC), and the mean residence time (MRT) were negatively linearly dependent, while Kupffer cell uptake was positively linearly dependent on KA. Subsequently, we verified the reliability of the approach for in vivo fate prediction using poly(methoxyethyl ethylene phosphate)-block-poly(d,l-lactide) (PEEP-b-PLA) and poly(vinylpyrrolidone)-block-poly(d,l-lactide) (PVP-b-PLA) NPs, and the linear relationship between the KA value and their PK parameters further suggests that the protein binding affinity of polymeric NPs can be a direct indicator of their pharmacokinetics.
