Phytate enhances gut Parasutterella colonization to alleviate radiation injury

Yuan Li,Jiwei Qiu,Zhihong Liu,Huiwen Xiao,Bin Wang,Yanxi Dong,Yunong Xiao,Qi Wang,Jiali Dong,Ming Cui

Food as medicine shows promise for disease intervention or treatment. Here, we found phytate, an active ingredient of plant-based diets, exhibits properties in mitigating radiotherapy-related complications. Oral gavage of phytate restored hematogenic organ atrophy, elevated peripheral blood neutrophils and white blood cells, reduced inflammation, and improved gastrointestinal (GI) integrity in irradiated mice. Phytate intake modulated the gut microbiota, facilitating the colonization of symbiotic Parasutterella in GI tract, thus combating intestinal radiation toxicity. In vitro assays and untargeted metabolomics identified 3-phenyllactic acid (PLA) and N-acetyl-L-leucine (NL) as functional metabolites produced by Parasutterella . In vitro , ex vivo , and in vivo models showed that PLA induces M2-like polarization in macrophages, while NL reduced oxidative stress, both counteracting radiation toxicity and working synergistically. Our findings offer mechanistic insights into phytate for alleviating radiation-associated complications and suggest that Parasutterella and its metabolites might be employed as promising probiotics or postbiotics for cancer patients undergoing radiotherapy.

Effect of a high-collagen peptide diet on the gut microbiota and short-chain fatty acid metabolism

Fengfeng Mei,Zhouwei Duan,Muxue Chen,Jinfeng Lu,Meihui Zhao,Laihao Li,Xuanri Shen,Guanghua Xia,Shengjun Chen

In this study, a total of 24 male Sprague Dawley rats were randomly divided into 3 group (Collagen peptide of Salmon salar skin group, Ss- SCP ; Collagen peptide of Tilapia nilotica skin group, Tn -SCP and Model control group, MC) to investigate the impact of a high-collagen peptide diet on the gut microbiota and host health. After 16 days intervention, the body weights of the Ss -SCP and Tn -SCP intervention groups were significantly increased and the liver index was also remarkably higher than that of the MC group. The acetic acid and propionic acid levels in feces were both significantly increased in the diet high-collagen peptide groups and valerate acid level was lower than that in the MC group. With the intervention of a high-dose collagen peptide diet, the gut microbiota of the groups was shifted with increased abundance of Lactobacillus , Unidentified-Prevotellaceae , Allobaculum, and Parasutterella , whereas the Tn -SCP administration have caused low abundance of Anaerostipes, Blautia, and Fusicatenibacter . The relative abundance of Allobaculum as well as Parasutterella was positively correlated with propionic acid and acetic acid levels, respectively. In addition, Allobaculum abundance was negatively correlated with valerate acid level. The serum valerate acid content was potentially harmful to rat health and significantly increased in the groups intervened with collagen peptide. All together, these results showed that administration of diet high-collagen peptide shifts the gut microbiota in rats and induced a disturbance in short-chain fatty acid metabolism which is potentially harmful to health.

Celastrol ameliorates experimental autoimmune uveitis through STAT3 targeting and gut microenvironment reprofiling

Shengjin Xiang,Jinrun Chen,Mengyun Deng,Zixiang Wang,Xingyi Li,Dan Lin,Jianhong Zhou

Background Extensive research has revealed the favorable effects of celastrol (CEL) against various diseases, but the role of CEL in autoimmune uveitis remains unexplored. Methods We first assessed the prophylactical and therapeutical effects of CEL on autoimmune uveitis via rat experimental autoimmune uveitis model. After network pharmacology, functional enrichment and molecular docking analyses, we predicted the potential target of CEL and validated its effect on EAU by clinical and histopathological scores, Evans blue staining, immunofluorescence assay and western blotting. Then we evaluated the role of CEL in the gut environment by 16S rRNA sequencing and untargeted metabolomic analysis. Results We confirmed that CEL treatment suppressed the pathological TH17 response, inhibited the migration of inflammatory cells, and preserved the integrity of BRB via targeting STAT3-IL17 pathway. Furthermore, CEL was found to reduce the relative abundance of opportunistic pathogenic bacteria including Clostridium_sensu_stricto_1 , Parasutterella and GCA-900066575 , and enrich the relative abundance of beneficial Oscillospirales and Ruminococcus_torques_group in EAU rats by fecal 16S rRNA sequencing. Meanwhile, CEL treatment reshaped the gut metabolites in the EAU rats by increasing the relative concentrations of cholic acid, progesterone and guggulsterone, and decreasing the relative levels of isoproterenol, creatinine and phenylacetylglutamine. Conclusions CEL exerts its ameliorative effects on the experimental autoimmune uveitis through the dual mechanisms of targeting STAT3 and reprofiling the gut microenvironment.

The protective effect of small black soybean (Vigna Mungo L.) polysaccharide on acetic acid-induced gastric ulcer in SD rats and its impact on gut microbiota and metabolites

Zuanyuan Huang,Mengwei Hu,Xiuyi Peng,Rui Wang,Xiaoxiao Song,Junyi Yin

Small black soybean ( Vigna Mungo L.) is a kind of medicinal and edible plant with many potential health benefits for the human body. In this study, we investigated the potential anti-gastric ulcer effect of Vigna Mungo L. polysaccharides (VMP) using an acetic acid-induced gastric ulcer model in SD rats . The impact of VMP on the levels of antioxidant factors and growth factors in gastric tissue homogenate , serum metabolites, short-chain fatty acids (SCFAs) and gut microbiota in SD rats were analyzed. The results revealed that VMP exhibited a beneficial role in facilitating the healing process of gastric ulcers, specifically by reducing the area of gastric ulcers, increasing the levels of epidermal growth factor (EGF) and vascular endothelial growth factor (VEGF), and improving oxidative stress status. Additionally, VMP modulated 20 potential serum biomarkers associated with gastric ulcers, thereby exerting a significant impact on multiple crucial metabolic pathways. Notably, VMP notably increased the levels of SCFAs, including acetic acid, propionic acid , i-butyric acid, n-butyric acid, i-valeric acid, n-valeric acid, in the colonic contents of rats with gastric ulcers. Furthermore, VMP effectively ameliorated gut microbiota dysbiosis in gastric ulcer rats by reducing the relative abundance of Firmicutes , Lachnospiraceae_NK4A136_group , Quinella and Lactobacillus , while increasing the relative abundance of Bacteroidota , Bacteroides , Oscillospiraceae_UCG-005 , Muribaculaceae , Parasutterella and Ruminococcus . Correlation analysis showed that the relative abundance of Parasutterella and Lachnospiraceae_NK4A136_group were closely related to the changes of SCFAs and serum metabolites. These findings provide potential groundwork for the development of health products aimed at preventing and treating gastric ulcers.

Polysaccharides from Fu brick tea ameliorate obesity by modulating gut microbiota and gut microbiota-related short chain fatty acid and amino acid metabolism

Mingzhi Zhu,Jian Ouyang,Fang Zhou,Chenjie Zhao,Wan Zhu,Chunfang Liu,Peifang Huang,Jiafeng Li,Junwei Tang,Zhang Zhang,Jianan Huang,Miaomiao Wu,Kunbo Wang,Zhonghua Liu

Fu brick tea (FBT) is a traditional tea manufactured by solid-state fermentation of tea leaves ( Camellia sinensis ). Although anti-obesity effects have been reported for FBT, the associated role of FBT polysaccharides (PSs) and the underlying mechanisms remain unknown. In this study, we found that FBTPSs inhibited obesity, hyperlipidemia , and inflammation; improved intestinal barrier function; and alleviated gut microbiota dysbiosis in high-fat diet-fed rats. Akkermansia muciniphila , Bacteroides , Parasutterella, Desulfovibrio , and Blautia were the core microbes regulated by FBTPSs. FBTPSs regulated the production of gut microbiota-related metabolites, including short-chain fatty acids (SCFAs), branched-chain amino acids , and aromatic amino acids throughout the development of obesity, and regulated the SCFA-GPR signaling pathway . FBTPS-treated fecal microbiota transplant ameliorated obesity, alleviated gut microbiota dysbiosis, and improved gut microbiota-associated metabolites, suggesting that the anti-obesity effect of FBTPSs was gut microbiota-dependent. FBTPSs may serve as novel prebiotic agents for the treatment of obesity and dysbiosis of gut microbiota.
