Synthesis of carbon quantum dots with iron and nitrogen from Passiflora edulis and their peroxidase-mimicking activity for colorimetric determination of uric acid

Liang Chunfeng,Lan Yuwei,Sun Zihua,Zhou Liya,Li Yanzhou,Liang Xinyuan,Qin Xingzhen

Carbon quantum dots co-doped with iron and nitrogen (Fe@NCDs) were synthesized by using Passiflora edulis Sims ( P. edulis ) as a precursor. The Fe@NCDs exhibit outstanding peroxidase-mimetic activity owing to successful doping with iron resulting in a behavior similar to that of iron porphyrins. In the presence of H 2 O 2 , the Fe@NCDs catalyze the oxidation of the peroxidase substrate 3,3′,5,5′-tetramethylbenzidine (TMB) with a color change from colorless to blue. The blue oxidation product has a characteristic absorption peaking at 652 nm. A colorimetric assay was worked out for uric acid (UA) that measures the hydrogen peroxide produced during oxidation of UA by uricase. Response is linear in the 2–150 μM UA concentration range, and the limit of detection is 0.64 μM. The method was applied to the determination of UA in (spiked) urine, and recoveries ranged from 92.0 to 103.4%. Schematic representation of the fabrication of iron and nitrogen co-doped carbon dots (Fe@NCDs) using Passiflora edulis Sims as carbon-based materials. First, uric acid (UA) was oxidized to generate H 2 O 2 by uricase. Then, the Fe@NCDs catalyzed the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to form blue-colored oxidized TMB (oxTMB) in the presence of H 2 O 2 . UA can be quantified based on the theory.

Degradation by Vc-H2O2, characterization and antioxidant activity of polysaccharides from Passiflora edulis peel

Xia Li,Guozhu Zhang,Jing Li,Tiemin Jiang,Huiying Chen,Peijun Li,Yuan Guan

In this paper, water extracted polysaccharides from Passiflora edulis peel (WPEP) was degraded by ascorbic acid-hydrogen peroxide (Vc-H 2 O 2 ). The degradation conditions were optimized by response surface methodology (RSM). The 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate of degraded WPEP (DWPEP) was 45.38% under optimal conditions (Vc-H 2 O 2 concentration 9.3 mM, temperature 52°C, time 1.4 hr). And the molecular weight (Mw) of DWPEP (38 kDa) was 32.20% (WPEP was 118 kDa). WPEP and DWPEP were mainly composed of galacturonic acid. Their backbone structures, functional groups and β-glycoside linkage patterns are similar, however, still with some differences. Antioxidant tests showed that WPEP and DWPEP had strong DPPH radical scavenging ability (93.03% and 81.70%), moderate hydroxyl radical scavenging ability (53.99% and 26.59%), and low reducing power (0.698 and 0.52). In conclusion, these differences in structures and properties may be an important factor affecting the antioxidant activity of polysaccharides. Novelty impact statement In this paper, the Vc-H 2 O 2 system was used to degrade the polysaccharides extracted from Passiflora edulis peel. The molecular weight, intrinsic viscosity, and rigid conformation changed obviously. The degraded polysaccharides are valuable to expand their application and to improve the practical efficacy.

Physicochemical characterization and the hypoglycemia effects of polysaccharide isolated from Passiflora edulis Sims peel

Yuan Guan,Hefei Sun,Huiying Chen,Peijun Li,Yang Shan,Xia Li

One polysaccharide, designated as WPEP-A, was isolated from Passiflora edulis Sims peel and its hypoglycemic effects on diabetic db/db mice were evaluated. Physicochemical characterization showed that WPEP-A was composed of galactose, glucose, xylose, rhamnose, galacturonic acid and glucuronic acid with a molecular weight of 9.51 × 104 Da. We observed an inhibition in weight gain and blood glucose levels. Glucose tolerance and insulin tolerance improved after the administration of WPEP-A. In addition, our data showed increased antioxidant enzyme activities. Furthermore, the levels of serum insulin and triglyceride decreased with the recovery of liver damage. Meanwhile, positive changes in short chain fatty acid content were observed, and the mRNA levels of glucagon-like peptide 1 receptor, glucagon and prohormone convertase 3 were up-regulated in the intestinal tract. In summary, our results showed that WPEP-A had hypoglycemic activity and improved intestinal function in diabetic mice, which may contribute to the attenuation of the hypoglycemia effects.

In vitro binding capacities, physicochemical properties and structural characteristics of polysaccharides fractionated from Passiflora edulis peel

Yongjin Sun,Ke Yang,Xin Zhang,Lifen Li,Huixiang Zhang,Li Zhou,Jun Liang,Xia Li

This study aimed to determine structural characteristics, physical properties and binding effects of polysaccharide and its fractions from Passiflora edulis peel on lipid , cholesterol and bile acids . The crude polysaccharide (WPEP) was extracted using hot water extraction method, fractionated using DEAE-52 cellulose column, and purified by Sepharose CL-6B column to obtain four polysaccharide fractions (WPEP-F1, WPEP-F2, WPEP-F3, and WPEP-F4). The physicochemical properties and structural characteristics were analyzed using several methods. The chemical-binding effects were also determined. Among all polysaccharide samples, WPEP-F2 had the highest total sugar content (77.63%). The viscosity of all polysaccharide samples was increased. Among the four polysaccharide fractions, WPEP-F2 showed the best emulsifying activity. WPEP-F1 was mainly composed of glucose, galactose , mannose , and arabinose , whereas WPEP-F2, WPEP-F3, and WPEP-F4 were principally composed of galacturonic acid. SEM analysis showed that the four fractions had different microstructures. Among the polysaccharide components, WPEP-F2 had the highest lipid-binding effect (9.3 g/g). It also had the best binding effect on taurocholic acid , deoxycholic acid , glycocholic acid , and cholesterol (50.87%, 69.12%, 30.22%, and 78.68%, respectively), but WPEP-F3 had the best binding effect on cholic acid (53.99%). The lipid, cholesterol, and bile acid-binding activities of WPEP-F2 could be attributed to its high viscosity, porous appearance structure, and sugar content in addition to its triple helix structure. The findings lay a theoretical foundation for developing Passiflora edulis peel polysaccharide-related foods and promoting application of the peel polysaccharide in functional food industries.

Effects of Drought Stress Induced by Hypertonic Polyethylene Glycol (PEG-6000) on Passiflora edulis Sims Physiological Properties

Ying Qi,Lingling Ma,Muhammad Imran Ghani,Qiang Peng,Ruidong Fan,Xiaojing Hu,Xiaoyulong Chen

Passion fruit is known to be sensitive to drought, and in order to study the physiological and biochemical changes that occur in passion fruit seedlings under drought stress, a hypertonic polyethylene glycol (PEG) solution (5%, 10%, 15%, and 20%) was used to simulate drought stress in passion fruit seedlings. We explored the physiological changes in passion fruit seedlings under drought stress induced by PEG to elucidate their response to drought stress and provide a theoretical basis for drought-resistant cultivation of passion fruit seedlings. The results show that drought stress induced by PEG had a significant effect on the growth and physiological indices of passion fruit. Drought stress significantly decreased fresh weight, chlorophyll content, and root vitality. Conversely, the contents of soluble protein (SP), proline (Pro), and malondialdehyde (MDA) increased gradually with the increasing PEG concentration and prolonged stress duration. After nine days, the SP, Pro and MDA contents were higher in passion fruit leaves and roots under 20% PEG treatments compared with the control. Additionally, with the increase in drought time, the activities of antioxidant enzymes such as peroxidase (POD), superoxide dismutase (SOD) and catalase (CAT) showed an increasing trend and then a decreasing trend, and they reached the highest value at the sixth day of drought stress. After rehydration, SP, Pro and MDA contents in the leaves and roots of passion fruit seedlings was reduced. Among all the stress treatments, 20% PEG had the most significant effect on passion fruit seedlings. Therefore, our study demonstrated sensitive concentrations of PEG to simulate drought stress on passion fruit and revealed the physiological adaptability of passion fruit to drought stress. Keywords:Passiflora edulisSims;drought stress;hypertonic polyethylene glycol;physiological characteristics

Healing Potential of Propolis Extract–Passiflora edulis Seed Oil Emulgel Against Excisional Wound: Biochemical, Histopathological, and Cytokines Level Evidence

Puneet Gupta,Ashish Singh,Neelam Singh,Faraat Ali,Ayushi Tyagi,Sadish K. Shanmugam

Propolis is rich in natural bioactive compounds, and considering its importance for many skin therapies, emulgel was prepared. This study examines how a propolis extract (PE) and Passiflora edulis seed (PS) oil emulgel affect rat deep skin wound healing. Based on preset criteria of maximum drug content and optimum drug permeation through the stratum corneum along with drug retention in the skin layers, an optimized emulgel formula based on Box-Behnken factorial design was prepared and used for subsequent in vitro and in vivo evaluations. In vivo wound-healing activities of emulgel and control treatments were investigated in a rat model. The optimized emulgel formula exhibited superior healing activity compared with plain PE suspension-treated rats on day 14 of wounding. Histopathological investigations of hematoxylin and eosin and Masson's Trichrome-stained skin sections supported this effect. Emulgel promotes cutaneous wound healing through a variety of mechanisms, including anti-inflammatory through modulation of cytokines tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 production, and promotion of collagen fiber formation, all of which contribute to tissue remodeling. Furthermore, when compared with propolis suspension, emulgel showed significant antioxidant and anti-inflammatory effects. Emulgel significantly increased the skin's hydroxyproline level, antioxidant potential, wound contraction, increased penetration, and localized propolis deposition across the skin. Incorporation of PS oil into the emulgel accelerates the tissue regeneration process. The findings suggest that 5% propolis emulgel could be used as an alternative to treat wounds.

Structural and rheological characterization of pectin from passion fruit (Passiflora edulis f. flavicarpa) peel extracted by high-speed shearing

Yupeng Lin,Fengping An,Hong He,Fang Geng,Hongbo Song,Qun Huang

In this study, the structural and rheological properties of pectin extracted by high-speed shearing (HSS–P) at 50 °C ultrapure water and conventional heating methods (CE50-P and CE90-P) from passion fruit ( Passiflora edulis f. flavicarpa ) peel were evaluated and compared. The HSS-P has loose, honeycomb-like surface morphology, lower molecular weight and higher polydispersity index compared to CE50-P and CE90-P. Monosaccharide composition indicated that HSS-P had more side chains, which were caused by its highly branched molecular structure, short main chains and a high molar ratio of RG-I in the molecular configuration. FTIR spectra showed that high-speed shearing led to a decrease in the linearity of the main chain of the pectin molecule. NMR spectra indicated that the molecular structure of HSS-P was a linear chain of homogalacturonan (HG) inserted into rhamnogalacturonan I (RG-I) possibly with arabinans and galactans as the principal side chains. Furthermore, the effects of pectin concentration, sucrose , CaCl 2 and pH on the rheological properties of different pectins were investigated. All pectin exhibited pseudoplastic fluid behavior at different environmental conditions. The HSS-P samples showed the greatest increase in the apparent viscosity and viscoelasticity in the presence of sucrose (60%–100%) and CaCl 2 (0.1–0.2 g/100 mL), pH changes and increased pectin concentration. The HSS-P was easily combined with the co-solutes and its viscoelasticity showed stronger stability under temperature changes. In conclusion, the high-speed shearing results in a unique molecular structure that contributed to the excellent rheological properties of pectin. Thus providing theoretical guidance for further application of high-speed shearing in extracting pectin.

Valorization of Passiflora edulis Peel for the Fabrication of Recyclable Nanosilver/Ceria-Cellulose Aerogels for Efficient Catalytic Reduction of Methyl Orange

Ho Thanh Gia-Thien,Nguyen Phung Anh,Tran-Pham Phuong Khanh,Phan Hong Phuong,Pham-Nguyen Ai Vi,Nguyen Thi Thu Thuy,Do Ba Long,Nguyen Duc Quy,Le Thanh Quang,Pham Thi Thuy Phuong,Van Nguyen Thi Thuy,Nguyen Tri

Developing fully sustainable and recyclable catalysts from agricultural waste remains a major challenge for advanced wastewater treatment. Herein, we report an integrated, zero-waste strategy to fabricate a hierarchically structured Ag/ceria-cellulose porous catalyst derived exclusively from purple Passiflora edulis peel waste. Phytochemical-rich peel extracts were employed as green reducing and stabilizing agents for the in situ synthesis of Ag nanoparticles, while the residual cellulose-rich biomass was converted into a three-dimensional porous aerogel via microwave-assisted extraction and freeze-drying. Incorporation of ceria nanorods endowed the aerogel with abundant oxygen vacancies and strong metal–support interactions, effectively suppressing Ag nanoparticle agglomeration and promoting interfacial electron transfer. The optimized Ag/ceria-cellulose aerogel exhibited uniformly dispersed Ag nanoparticles with an average size of 7.84 nm. Under mild conditions (0.5 wt% Ag loading, catalyst dosage of 0.5 g L −1 , and MO/NaBH 4 molar ratio of 1/200), over 95% of methyl orange was reduced within 30 min, following pseudo-first-order kinetics with an apparent rate constant of 0.14 min −1 . Owing to its monolithic and mechanically robust architecture, the aerogel catalyst could be readily recovered by simple filtration and retained more than 75% of its initial activity after five consecutive cycles. The superior catalytic performance and durability originate from the synergistic interplay between Ag nanoparticles, oxygen vacancy-rich ceria nanorods, and the interconnected cellulose aerogel matrix, which collectively enhance active-site accessibility, charge transfer efficiency, and structural stability. This work demonstrates a scalable and circular pathway for transforming passion fruit peel waste into high-value, reusable catalysts for wastewater remediation.

Flash extraction optimization of low-temperature soluble pectin from passion fruit peel (Passiflora edulis f. flavicarpa) and its soft gelation properties

Yupeng Lin,Hong He,Qun Huang,Fengping An,Hongbo Song

Low-temperature soluble and soft gelation (LT-SG) pectin belonging to special gum is an important food additive. Flash extraction optimization of pectin from passion fruit ( Passiflora edulis f. flavicarpa ) peel at low temperature (50   °C) by response surface methodology and LT-SG properties was investigated in the present study. Under the optimized conditions of a rotation speed of 5000   r/min, extraction time of 9   min, and the liquid–solid ratio of 42:1   mL/g, the pectin yield was 7.52%. The DE value of 57.02% indicated it was high methoxyl pectin, which easily dissolved at a low temperature (70   °C). Gel texture analysis revealed that the soft gelation properties were depended on the sucrose concentration and pH in the pectin solution. Both adding sucrose and decreasing pH refined gel network structure, as results, the gel texture was strengthened.

Eco-friendly simultaneous extraction of pectins and phenolics from passion fruit (Passiflora edulis Sims) peel: Process optimization, physicochemical properties, and antioxidant activity

Dongxue Huo,Jincheng Dai,Siyu Yuan,Xiaoqing Cheng,Yonggui Pan,Lu Wang,Ruimin Wang

The objective of this study was to simultaneously extract passion fruit ( Passiflora edulis ) peel pectins and phenolics using deep eutectic solvents, to evaluate their physicochemical properties and antioxidant activity. By taking L-proline: citric acid (Pro-CA) as the optimal solvent, the effect of extraction parameters on the yields of extracted passion fruit peel pectins (PFPP) and total phenolic content (TPC) was explored by response surfaces methodology (RSM). A maximum pectin yield (22.63%) and the highest TPC (9.68 mg GAE/g DW) were attained under 90 °C, extraction solvent pH = 2, extraction time of 120 min and L/S ratio of 20 mL/g. In addition, Pro-CA-extracted pectins (Pro-CA-PFPP) and HCl-extracted pectins (HCl-PFPP) were subjected to high performance gel permeation chromatography (HPGPC), Fourier transform infrared spectroscopy (FT-IR), thermogram analysis (TG/DTG) and rheological measurements. Results verified that the Mw and thermal stability of Pro-CA-PFPP were higher than those of HCl-PFPP. The PFPP solutions featured a non-Newtonian behavior, and compared with commercially pectin solution, PFPP solution exhibited a stronger antioxidant activity. Additionally, passion fruit peel extract (PFPE) exhibited stronger antioxidant effects than PFPP. The results of ultra-performance liquid chromatography hybrid triple quadrupole-linear ion trap mass spectrometry (UPLC-Qtrap-MS) and high performance liquid chromatography (HPLC) analysis showed that (−)-epigallocatechin, gallic acid, epicatechin, kaempferol-3- O -rutin and myricetin were the main phenolic compounds in PFPE and PFPP. Our results suggest that Pro-CA can be considered as an eco-friendly solvent for high-efficient extraction of high-value compounds from agricultural by-products.

Preparation and characterization of zein–caseinate–pectin complex nanoparticles for encapsulation of curcumin: pectin extracted by high-speed shearing from passion fruit (Passiflora edulis f. flavicarpa) peel

Xiefei Li,Yupeng Lin,Yumeng Huang,Xin Li,Fengping An,Hongbo Song,Qun Huang

BACKGROUND Pectin extracted by high-speed shearing from passion fruit peel (HSSP) is a potentially excellent wall material for encapsulating curcumin, which has multiple advantages over pectin prepared by heated water extraction. HSSP was used to fabricate complex nanoparticles of zein–sodium caseinate–pectin for encapsulation of curcumin in this study. The influence of heating on the physicochemical properties of the composite nanoparticles was also investigated, as well as the effect of composite nanoparticles on the encapsulation efficiency, antioxidant activity and release characteristics of curcumin. RESULTS The nanoparticles were formed through electrostatic interactions, hydrogen bonds and hydrophobic interactions between the proteins and HSSP. A temperature of 50 °C was more favorable for generating compact and small-sized nanoparticles, which could effectively improve the encapsulation efficiency and functional properties. Moreover, compared to other pectin used in the study, the nanoparticles prepared with HSSP showed the best functionality with a particle size of 234.28 ± 0.85 nm, encapsulation rate of 90.22 ± 0.54%, free radical scavenging rate of 78.97% and strongest protective capacity in simulated gastric fluid and intestinal release effect. CONCLUSION Zein–sodium caseinate–HSSP is effective for encapsulating and delivering hydrophobic bioactive substances such as curcumin, which has potential applications in the functional food and pharmaceutical industries. © 2024 Society of Chemical Industry.

Effects of thermal drying methods on characteristic volatile organic compounds of passion fruit (Passiflora edulis Sims) peel: A multi-analytical approach combining GC–MS, GC-IMS, E-nose, and multivariate statistical analysis

Lingshuai Meng,Meirun Zhu,Xu Song,Junjie Yi,Guangcan Tao,Siyao Wu,Tingting Zheng,Sen Cao,Su Xu

This study systematically evaluated thermal drying methods effects on characteristic volatile organic compounds of passion fruit peel using E -nose, GC–MS, GC-IMS combined with multivariate statistical analysis. Specifically, GC–MS exhibited higher sensitivity for hydrocarbons and other compounds, whereas GC-IMS was esters, aldehydes, and alcohols. Multiple Factor Analysis confirmed that the combination of these two methods was shown to more comprehensively evaluate the volatile flavor compounds and aroma characteristics of passion fruit peel. Overall, HPD exhibited the best retention of characteristic volatile organic compounds (e.g., Methyl benzoate-M, Nonanal, Decanal), generated new compounds (e.g., (E)-2-octenal, Benzaldehyde, 2-methylbutanal) and enriched the aroma characteristics (e.g., apple, chocolate, fruity). Therefore, HPD was recommended as the optimal pretreatment method for obtaining the best aroma quality of passion fruit peel. Besides, methyl benzoate could serve as a key marker for evaluating the aroma quality of passion fruit peel dried by VMD.
