Patrinia scabiosaefolia , is used as wild vegetable in China for more than 2 000 years, with a variety of pharmacological activities, including anti-inflammatory, anti-tumor and hypoglycemic. Based on our ongoing research on chemical constituents and hypoglycemic activity of P. scabiosaefolia , 4 lignan compounds, (+)-isolariciresinol ( 1 ), 7 R ,7' R ,8 S ,8' S -(+)-neo-olivil-4- O - β - D -glucopyranoside ( 2 ), 4- O- methylcedrusin ( 3 ) and patrinian A ( 4 ), were isolated and identified. The hypoglycemic activity showed that compounds 2 and 3 could extremely significantly improve insulin resistance at 100 ( P < 0.001), 50 ( P < 0.001) and 25 µmol/L ( P < 0.01) in IR 3T3-L1 cells. While compound 4 only promoted glucose uptake by IR 3T3-L1 cells at 100 µmol/L ( P < 0.01). Western blotting experiments showed that compounds 2 and 4 up-regulated the protein expressions of p-IRS, PI-3K, p-AKT and glucose transporter 4 (GLUT4), and promoted the transcription of GLUT4 mRNA. Therefore, the mechanisms of compounds 2 and 4 were presumed to improve IR by activating PI-3K/AKT signaling pathway.

Introduction: Solar dermatitis, a condition triggered by excessive exposure to ultraviolet B (UVB) radiation, results in inflammatory skin damage marked by erythema, edema, and epidermal injury. Portulaca oleracea (PO) and Patrinia scabiosaefolia (PS) have been traditionally used in dermatological treatments, though their mechanistic pathways in UVB-induced skin injury are not fully understood. Methods: In this study, a mouse model of UVB-induced solar dermatitis was employed to evaluate the therapeutic potential of combined PO and PS (POPS) extracts. After UVB irradiation, POPS extracts were administered, and their bioactive compounds were identified through ultra-performance liquid chromatography-mass spectrometry (UPLC-MS). Network pharmacology, molecular docking, and pathway analysis were performed to identify key targets, focusing on the IL-17/CCL2 pathway and oxidative stress reduction. Results: Treatment with POPS extracts significantly diminished UVB-induced inflammation, erythema, and epidermal thickening in a dose-dependent manner. Network pharmacology and docking studies identified curvularin and olmesartan medoxomil as bioactive components with high affinity for IL-17 and IL-17RA targets, modulating the IL-17/CCL2 axis. In vivo experiments demonstrated that POPS extracts suppressed the expression of IL-17 and CCL2, reduced macrophage infiltration, and alleviated oxidative stress, effectively mitigating the symptoms of solar dermatitis. Conclusion: This study provides insight into the anti-inflammatory and protective properties of POPS extracts in solar dermatitis, highlighting their potential as a treatment through IL-17/CCL2 pathway modulation and oxidative stress reduction.

Histone lactylation, a recently identified post-translational modification, is closely linked to the pathogenesis and progression of malignant tumors, making it a promising therapeutic target for acute myeloid leukemia (AML). Increasing clinical and experimental evidence indicates that elevated serum lactate dehydrogenase (LDH) levels serve as both a diagnostic biomarker and an indicator of poor prognosis in AML, reflecting enhanced glycolytic activity and chemotherapy resistance. Patrinia scabiosaefolia Fisch (PS), a traditional medicinal herb, exhibits broad pharmacological activities, including heat-clearing, detoxifying, antibacterial, antiviral, and antitumor effects. However, the molecular mechanisms underlying its antileukemic activity, particularly in AML, remain insufficiently defined. To elucidate how DEPS exerts therapeutic effects in AML through the HIF-1α–histone lactylation axis, we systematically established the association between DEPS-mediated inhibition of histone lactylation and cellular hypoxia. Lactylation pan-antibody incubation confirmed preliminarily that DEPS significantly inhibits histone lactylation modification. Lactate restoration experiments further demonstrated that DEPS markedly suppresses lactate-induced activation of HIF-1α signaling, reducing downstream metabolic proteins and VEGFA expression. Under hypoxic conditions, AML cells displayed increased proliferation and histone lactylation, both of which were attenuated by DEPS treatment. Sodium L-lactate enhanced the expression of HIF-1α and its downstream effectors (HK2, PDK1, PKM2, GLUT1, LDHA, and VEGFA), whereas subsequent DEPS exposure significantly reversed these changes. Ultimately, DEPS inhibits H3/H4 lysine lactylation, induces G2/M cell-cycle arrest, and promotes apoptosis in THP-1 and HL-60 cells, including doxorubicin-resistant HL-60 cells. Collectively, our findings reveal previously uncharacterized antileukemic mechanisms of DEPS involving suppression of the HIF-1α signaling pathway and histone lactylation. These results highlight the importance of epigenetic regulation in AML and support the therapeutic potential of DEPS in AML treatment and reversal of chemoresistance. Graphical
