A novel multimodal aptasensor for Patulin detection in fruit products based on high-performance RuMOF@hydrogel and versatile pericarp-derived carbonized polymer dots

Liangrui Lv,Qin Tao,Yijing Kuang,Tingting Zhang,Xiaoying Wang

Patulin (PAT) is a widespread fruit toxin. Trace-level PAT exposure can cause serious harm to human health. Herein, a multimodal PAT aptasensor was designed based on Ru(bpy) 3 2+ -based metal organic framework composited hydrogel (RuMOF@hydrogel) and versatile banana peel-derived carbonized polymer dots (BPPDs). RuMOF@hydrogel modified magnetic-electrode exhibited excellent anodic and cathodic electrochemiluminescence (ECL) emission and stability. Meanwhile, the BPPDs could enhance anodic ECL of RuMOF@hydrogel, and also show excellent fluorescence (FL) and photothermal (PT) properties. With the aid of PAT-triggered hybridization chain reaction and magnetic separation, ECL, FL, and PT responses could be recorded concurrently. The detection limit can reach as low as 0.25 fg mL −1 . The ratiometric ECL quantitation ensured the sensitivity and accuracy of this assay. And visual FL and portable PT modes contributed to the utility. Furthermore, this aptasensor demonstrated better performances than HPLC in fruit products and the protocol can be extended to determine various contaminants in foods.

Label-free fluorescence aptasensor for the detection of patulin using target-induced DNA gates and TCPP/BDC-NH2 mixed ligands functionalized Zr-MOF systems

Xiaohai Yan,Gengan Du,Hong Chen,Qiannan Zhao,Qi Guo,Jianlong Wang,Zhouli Wang,Wei Song,Qinglin Sheng,Yane Luo,Yahong Yuan,Tianli Yue

Patulin (PAT) is an unsaturated lactone mycotoxin primarily produced by Penicillium expansum and Aspergillus clavatus . Given the potential health risks and economic losses associated with PAT, the rapid detection of PAT using fluorescent aptasensors is of significant importance in evaluating food safety. However, it easily increases the cost and complexity caused by signal labeling. We combined TCPP/BDC-NH 2 mixed ligands functionalized Zr metal–organic frameworks (Zr-MOF mix ) and terminated three-stranded DNA gates (ttsDNA gates) to fabricate a label-free fluorescent aptasensor for PAT detection. The Zr-MOF mix system was synthesized via a one-pot strategy and could be used to address the problem of pore size limitation and increase the loading amounts of dyes. TtsDNA gate was integrated into the Zr-MOF mix system to control the release of dyes, exhibiting a high signal-to-background ratio. The single-stranded aptamer region in ttsDNA gate situated away from the surface of the Zr-MOF mix , resulting in a natural release of dyes in the absence of PAT. While binding to PAT resulted in target-induced conformational changes that helped form the hairpin structure of the aptamer. This structure hindered the release of dyes from the pores of Zr-MOF mix , thus reducing the fluorescence signals intensity. The stimuli-responsive DNA-gated material provides a platform for PAT analysis under conditions of a low limit of detection (0.871 pg/mL). Furthermore, the excellent specificity and anti-interference of the fluorescent aptasensor make the system suitable for the analysis of apple juice samples. This label-free strategy is cheaper and simper compared with labeled detection, especially for the development of multi-target-detection.

Competitive binding strategy for reliable signal-off surface enhanced Raman scattering sensing in protecting apples from patulin without external interference

Yang Zhang,Xinchen Wu,Limei Yin,Pierre Picchetti,Tianxi Yang,Ruiyun Zhou,Chuping Zhao,Shanshan Xue,Zhepeng Zhang,Xiaobo Zou,Zhiming Guo

Patulin is a toxin that can be found in damaged or moldy fruits, especially apples. Very high detection sensitivity can be achieved using the SERS (Surface Enhanced Raman Scattering) technique, which provides information about molecular vibrations. However, unexpected desorption of target toxins from the surface of the SERS substrate and interference from other substances pose a major challenge. Here we investigate a competitive binding strategy for reliable SERS analysis of PAT. Core-Raman molecule-shell (CMS) structures and programmable, specific DNA interactions are utilized to regulate Raman signal intensity without affecting the embedded Raman molecules. Specfically, aptamer-functionalized CMS structures are combined with metal-organic scaffold nanoparticles that possess complementary DNA sequences. In the presence of PAT, binding of the aptamer to the toxin leads to degradation of the hybrid nano assembly, which can be measured by a change in the SERS signal. This SERS strategy enables turn-off sensing in apple juice over a wide range (10 pg/mL to 1 μg/mL), with a detection limit as low as 0.02 ng/mL. Analysis of real samples gives results consistent with high-performance liquid chromatography (HPLC) test. The excellent selectivity and sensitivity of the SERS sensor demonstrate its potential for a wide range of SERS sensor applications.

Operando Photoelectrochemical Surface-Enhanced Raman Spectroscopy: Interfacial Mechanistic Insights and Simultaneous Detection of Patulin

Shuda Liu,Na Dong,Shuyun Meng,Yuye Li,Yuqing Li,Siyuan Wang,Dong Liu,Tianyan You

Comprehending the biosensing mechanism of the biosensor interface is crucial for sensor development, yet accurately reflecting interfacial interactions within actual detection environments remains an unsolved challenge. An operando photoelectrochemical surface-enhanced Raman spectroscopy (PEC-SERS) biosensing platform was developed, capable of simultaneously capturing photocurrent and SERS signals, allowing operando characterization of the interfacial biosensing behavior. Porphyrin-based MOFs (Zr-MOF) served as bifunctional nanotags, providing a photocurrent and stable Raman signal output under 532 nm laser irradiation. Aptamer was used to bridge the Zr-MOF and the silver-encased gold nanodumbbells (AuNDs@AgNPs). The simultaneous in situ acquisition of target-induced PEC and SERS signal responses facilitated the correlation of electron transfer information from the photocurrent with the distance information from the SERS signal. It revealed the biosensing mechanism in which target-induced aptamer conformational bending drove the Zr-MOF to approach the electrode. However, the increase in charge transfer observed through conventional electrochemical methods contradicts the conclusions drawn from the operando PEC-SERS analysis. Comprehensive analysis indicated that redox probes introduced during the non-in-situ measurement process became adsorbed within the MOF pores, potentially affecting the judgment of the biosensing mechanism. In addition, the operando PEC-SERS biosensor simultaneously obtained two independent signals, providing self-verification to improve the accuracy and reliability of patulin detection. The linear ranges were 1 pg mL–1–10 ng mL–1 for the PEC method and 1 pg mL–1–100 ng mL–1 for the SERS method, respectively. This work provides a powerful tool for determining the interface characteristics of biosensors.

An ultrasensitive ratiometric aptasensor based on the dual-potential electrochemiluminescence of Ru(bpy)32+ in a novel ternary system for detection of Patulin in fruit products

Liangrui Lv,QingQing Chen,Chunyang Jing,Xiaoying Wang

To sensitively monitor trace-level of toxic patulin (PAT), an ultrasensitive PAT ratiometric aptasensor based on the dual-potential electrochemiluminescence (ECL) of Ru(bpy) 3 2+ was first proposed. Noteworthily, Ru(bpy) 3 2+ -doped trimetallic nanocube ( [email protected] ) innovatively integrated the luminophore and cathode coreaction accelerator (CCA), which could generate strong cathodic ECL in the existence of low concentration of K 2 S 2 O 8 . Simultaneously, anthocyanin-derived carbon quantum dots (anth-CQDs) prepared from purple potato skins was first served as a green anodic coreactant. And SiO 2 -coated anth-CQDs ( [email protected] 2 ) exhibited excellent performance for enhancing anodic ECL of [email protected] Based on this, a novel ternary ECL system was established. In the presence of PAT, the ECL intensity ratio of anode to cathode ( I ECL-A / I ECL-C ) was significantly increased, and a low detection limit of 0.05 pg mL −1 was obtained. Moreover, when proposed method and high performance liquid chromatography (HPLC) were simultaneously applied to series of fruit products, the obtained results were completely consistent, reflecting its practicability.
