The role of Pediococcus pentosaceus in the gastrointestinne has received considerable attention in recent decades. This study aimed to investigate the effects of the short-term administration of P. pentosaceus on physiological characteristics, inflammation, and intestinal microecology in mice. In this study, 90 male C57BL/6J mice were divided into 15 groups, with 14 groups treated with a daily intragastric administration of different genotypes of P. pentosaceus. After three weeks of intragastric administration P. pentosaceus had a mild effect on mice. It could be seen that different P. pentosaceus strains had different effects on the gut microbiota and intestinal microecology. P. pentosaceus VCQYC5144M12 possessing an Enterolysin A operon may have been harmful, activating the expression of inflammatory factors, while P. pentosaceus DYNDL69M8 consisting of only a pediocin-like operon increased the abundance of beneficial bacteria and increased the content of acetic acid. The presence of various genotypes of bacteriocin may have been the explanation for variations among strains. This may provide theoretical support for further exploring the probiotic effect and patterns of P. pentosaceus.

Lactic acid bacteria (LAB) are used in fermented foods to overcome the shortcomings of traditional fermentation methods, such as long cycles and poor safety. However, the application of LAB in low-temperature foods is limited by their growth temperature. This study aimed to quantitatively compare the proteins of the YT2 strain acclimated at 37, 28, 20, 15, 8, and 4 °C (YT-4) and that grown at 37 °C (YT-37) using tandem mass tag (TMT) proteomics and bioinformatics analysis to elucidate the potential mechanism of cryotolerance of P. pentosaceus YT2. Altogether, 1416 quantitative proteins were identified, of which 747 were differentially abundant between the YT-4 and YT-37 strains. Functional enrichment analysis showed that the upregulated proteins were mainly involved in nucleotide transport, metabolism, translation, ribosomal structure and biogenesis, transcription, replication, recombination, and repair, while the downregulated proteins were involved in lipid, amino, and carbohydrate transport and metabolism. These results were validated for eight upregulated and five downregulated proteins using parallel reaction monitoring (PRM). Our study analyzed the potential mechanism underlying the improved cryotolerance of P. pentosaceus YT2 at 4 °C, which may enable the utilization of LAB in low-temperature applications.

Bamboo shoot boiled liquid (BSBL), a processing byproduct containing soluble proteins, peptides, amino acids, carbohydrates, and phenolics, is typically discarded, causing resource waste and environmental issues. This study analyzed metabolic changes in BSBL duringPediococcus pentosaceusB49 fermentation. The result of partial least squares discriminant analysis (PLS-DA) revealed significant metabolite profile differences across fermentation times (0 h, 24 h, 48 h, 72 h, 96 h). The most substantial alterations occurred within the first 24 h, followed by stabilization. Compared to unfermented BSBL, fermented samples exhibited significantly elevated signal intensities for 5,7-dimethoxyflavone, cinnamic acid, 3,4-dihydro-2H-1-benzopyran-2-one, 6,8-dimethyl-4-hydroxycoumarin, and 2-hydroxycinnamic acid (p< 0.05), showing upward trends over time. Conversely, (+)-gallocatechin intensity decreased gradually. Bitter peptides, such as alanylisoleucine, isoleucylisoleucine, leucylvaline, and phenylalanylisoleucine, in BSBL exhibited a significant reduction following fermentation withP. pentosaceusB49 (p< 0.05). KEGG enrichment indicated tyrosine metabolism (ko00350) and arginine/proline metabolism (ko00330) as the most impacted pathways. These findings elucidate metabolic regulation in BSBL fermentation, supporting development of functional fermented bamboo products.

C-NeuAc/OHEC hydrogels were prepared from carboxymethyl chitosan/N-acetylneuraminic acid (C-NeuAc) and oxidised hydroxyethyl cellulose (OHEC), and used as novel vehicles for the co-encapsulation of Pediococcus pentosaceus RQ-1 and isomaltose-oligosaccharide (IMO). The thermal stability, rheological behaviour, swelling kinetics, porosity, pH sensitivity, and degradability of the hydrogels were evaluated. The hydrogels were most stable and were suitable as vehicles for co-encapsulation of Pediococcus pentosaceus RQ-1 and IMO when C-NeuAc:OHEC = 5:4. In the presence of IMO, the encapsulation efficiency of the hydrogel was optimal and in the range of 91.02–94.63%. An in vitro assay revealed that the C-NeuAc/OHEC hydrogel provided good protection for Pediococcus pentosaceus RQ-1 against the harsh conditions of a simulated gastrointestinal tract. The survival rate of Pediococcus pentosaceus RQ-1 was only 77.0% in the presence of free IMO. By contrast, the survival rates of Pediococcus pentosaceus RQ-1 co-encapsulated with 0.5%, 1.0%, and 1.5% IMO were 84.4%, 90.8%, and 90.3%, respectively. Hence, IMO co-encapsulation significantly improved probiotic survival. Moreover, co-encapsulation with 1.0% IMO and 4 °C were the optimal storage condition for Pediococcus pentosaceus RQ-1. The present work showed that C-NeuAc/OHEC hydrogels are promising as probiotic co-encapsulation delivery vehicles.

In this study, carboxymethyl chitosan (CMCS) and N - acetylneuraminic acid (NeuAc) were used to develop C-NeuAc hydrogels to encapsulate Pediococcus pentosaceus QK-1. The mechanical properties, thermal stability, in vitro degradation, and pH sensitivity of the hydrogel were evaluated. The C-NeuAc concentration required for optimal hydrogel performance was 3% (w/v). Hydrogel swelling behaviour was effectively assessed by Fickian diffusion and Schott’s second-order kinetic models. The hydrogel demonstrated excellent biocompatibility and low in vitro cytotoxicity. An in vitro assay revealed that the viability of Pediococcus pentosaceus QK-1 in C-NeuAc had decreased by only 1.41 log (CFU/ mL) after exposure to simulated acidic gastric fluid. Moreover, the survival rate of the encapsulated and free Pediococcus pentosaceus QK-1 cells were 80.1% and virtually zero, respectively, after passage through the gastrointestinal tract. It was empirically determined that low temperature and freeze-drying were the ideal condition and method to ensure storage longevity of the hydrogel-encapsulated probiotic. Hence, C-NeuAc hydrogel is highly desirable as a food-grade probiotic delivery vehicle.

Pediococcus pentosaceus Li05, a strain of lactic acid bacteria isolated from the faeces of healthy volunteers, exhibited potential protective effects against various diseases. This study performed third-generation sequencing and detailed characterisation of its genome. The Li05 chromosome harboured conserved genes associated with acid resistance ( atp ), bile salt resistance ( bsh ), oxidative stress resistance ( hsl , dltA , and et al.), and adhesion ( nrd , gap , and et al.), whereas the plasmid did not contain antibiotic resistance or virulence genes. Following intervention with Li05 in loperamide-induced constipated mice, constipation symptoms improved. Meanwhile, alterations in gut microbiota, increased BSH activity in faeces, and modifications to the faecal bile acid profile were observed. Additionally, expression levels of TGR5 and TPH1 in the colon of the mice increased, leading to elevated 5-HT levels. When the TGR5 gene was knocked out or the TPH1 inhibitor LX1606 was administered to suppress 5-HT synthesis in constipated mice, the beneficial effects of Li05 on gastrointestinal motility and mucus secretion were reversed. Culturing intestinal organoids demonstrated that increased bile acids such as DCA, Iso-LCA, and EALCA could enhance 5-HT levels through the TGR5/TPH1 axis. Therefore, we concluded that Li05 regulated bile acid metabolism, subsequently increasing 5-HT levels through the TGR5/TPH1 axis, thus alleviating constipation. Graphical The mechanism of Li05 in alleviating constipation. Li05 intervention regulated gut microbiota and bile acid metabolism, increasing 5-HT levels through the TGR5/TPH1 activation to ameliorate constipation.

Improvement in the survival of the edible probiotic Pediococcus pentosaceus during the preparation and storage is important. In this study, the protective effects of isomaltose oligosaccharide in a protective agent (sodium glutamate, skimmed milk, and trehalose) on P. pentosaceus during vacuum freeze-drying and low-temperature storage were evaluated. Cell damage, and texture were evaluated using fluorescence analyses, and scanning electron microscopy, respectively, and a water activity assay was performed storage after 1 month of storage (−80, −20, 4, 25 °C). The viability of P. pentosaceus agents (24444, QK-1, MQ-1, and RQ-1) were evaluated for 9 months storage. The results showed that IMO increased 11.02% survival rate of P. pentosaceus QK-1 during the freeze-drying process, which was better than that of 24444, MQ-1, and RQ-1; the texture of the protective agent was thin, with a compact and granular cross-section. Decreased water activity (18.71–34.26%) and cell damage (8.32–10.53%), also enhanced survival rate (5.98–12.85%) were observed in the IMO-containing groups in storage. The survival rates of P. pentosaceus (QK-1, MQ-1, RQ-1) direct-injection agents (9 months) exceeded 90% under gastrointestinal satiety-mimicking conditions in vitro . Our data indicate that IMO has a good protective effect on the freeze-drying and storage processes of P. pentosaceus .

This study aimed to isolate and identify a fiber-degrading probiotic strain from traditional yak yogurt on the Qinghai–Tibet Plateau and evaluate its effects on rumen fermentation and microbial metabolism. Through carboxymethyl cellulose (CMC) screening, filter paper degradation, and straw degradation tests, a lactic acid bacterium —Pediococcus pentosaceus JASB0677—was obtained, with a straw degradation rate of 18.74%. P. pentosaceus JASB0677 exhibited tolerance to simulated gastric (pH 3.0, 1% pepsin, survival rate: 69.01%) and intestinal juices (pH 8.0, 1% trypsin, 63.18%) and showed bile salt resistance at 0.1%, 0.2%, and 0.3% concentrations (survival rates: 79.05%, 63.34%, and 52.32%, respectively). It also demonstrated strong antibacterial activity, with an inhibition zone diameter of 20.03 mm against Salmonella, and antioxidant activity, with 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radical scavenging rates of 46.52% and 39.17%, respectively. In vitro rumen fermentation revealed that P. pentosaceus JASB0677 significantly increased acetate and total volatile fatty acids (TVFAs) concentrations while reducing ammonia nitrogen levels ( p < 0.05). 16 S rRNA sequencing revealed that P. pentosaceus JASB0677 altered the rumen microbial composition by increasing the relative abundance of Proteobacteria , Actinobacteria , Enterobacter , Klebsiella , and Christensenellaceae_R-7_group . Metabolomic analysis revealed significant modulation of several metabolic pathways, especially those related to amino acid and lipid metabolism. Correlation analysis indicated that acetate and TVFAs concentrations were significantly positively correlated with the abundances of Proteobacteria , Actinobacteria , Enterobacter , and Klebsiella , as well as specific metabolites including mevalonic acid and L-leucine, while ammonia nitrogen showed the opposite trend ( p < 0.05). These results suggest that P. pentosaceus JASB0677 is a promising candidate functional feed additive for enhancing fiber utilization, maintaining microbial homeostasis, and improving rumen fermentation in ruminants.

Chronic obstructive pulmonary disease (COPD), a condition primarily linked to oxidative stress, poses significant health burdens worldwide. Recent evidence has shed light on the association between the dysbiosis of gut microbiota and COPD, and their metabolites have emerged as potential modulators of disease progression through the intricate gut-lung axis. Here, we demonstrate the efficacy of oral administration of the probiotic Pediococcus pentosaceus SMM914 (SMM914) in delaying the progression of COPD by attenuating pulmonary oxidative stress. Specially, SMM914 induces a notable shift in the gut microbiota toward a community structure characterized by an augmented abundance of probiotics producing short-chain fatty acids and antioxidant metabolisms. Concurrently, SMM914 synthesizes L-tryptophanamide, 5-hydroxy-L-tryptophan, and 3-sulfino-L-alanine, thereby enhancing the tryptophan-melatonin pathway and elevating 6-hydroxymelatonin and hypotaurine in the lung environment. This modulation amplifies the secretion of endogenous anti-inflammatory factors, diminishes macrophage polarization toward the M1 phenotype, and ultimately mitigates the oxidative stress in mice with COPD. The demonstrated efficacy of the probiotic intervention, specifically with SMM914, not only highlights the modulation of intestine microbiota but also emphasizes the consequential impact on the intricate interplay between the gastrointestinal system and respiratory health.

This study explored the effect of electrical stimulation (ES) and Pediococcus pentosaceus LL-07 ( P. pentosaceus LL-07) and Staphylococcus simulans QB7 ( S. simulans QB7) on the quality and microbial community of loin ham during the ripening. After the ES and starter culture treatments, the Aw and pH were decreased. Surface hydrophobicity, myogenic fiber fragmentation index (MFI), TCA-soluble peptide, amino nitrogen and free amino acids (FAAs) were also significantly higher than the control group (CK) ( P < 0.05). This increase was more significant in the E-S group (electrical stimulation followed by inoculation with P. pentosaceus LL-07 and S. simulans QB7) than the rest of the experimental group (E、S、S-E group). Furthermore, the CK and E-S groups were subjected to a bacterial community comparison experiment. The microbial diversity of these two groups was increased. Pediococcus spp. and Staphylococcus spp. became the dominant bacteria in E-S groups during the ripening. Correlation analyses show a strong correlation between protein hydrolysis, microorganisms and FAAs. In conclusion, the combination of ES and starter culture could promote protein hydrolysis, the accumulation of FAAS, and improves the bacterial community of loin ham.
