Ammonia‑nitrogen has become a global issue with feed-dependent intensive culture modes, which may cause toxicity in aquatic animals by affecting immune regulation. Although numerous studies have investigated the acute and short-term toxic effect of ammonia‑nitrogen in fish, there is a lack of research on the long-term effects of exposure to low concentration of ammonia‑nitrogen. In this study, yellow catfish ( Pelteobagrus fulvidraco ) were exposed to water supplemented with ammonia (NH 4 Cl was used as the ammonia source) at concentrations of 0 (control), 8, 10, 12, 14, 16 and 18 mg/L for eight-weeks, and the effects on growth performance, histopathology, antioxidant and immune capacities of yellow catfish were investigated. The results showed that long-term ammonia exposure resulted in decreased weight gain rate (WGR) and serum antioxidant capacity, increased feed coefficient (FC), serum and tissue ammonia levels. They caused extensive histopathological changes and significantly impaired inflammatory and immune responses. On the other hand, ammonia exposure significantly affected the respiratory burst and the content of reactive oxygen species (ROS) of head kidney macrophages. Still, the above phenomenon can be reversed by inhibiting tumor necrosis factor- α ( TNFα ), indicating that TNFα may play a key role in this process. In conclusion, our findings indicate that extended exposure to ammonia nitrogen, even at low concentrations, has a detrimental effect on the well-being of aquatic organisms. This finding underscores the necessity of long-term environmental toxicological investigations of ammonia‑nitrogen.

Background Iron is an essential metal element for organisms, whose metabolism is regulated by many genes and also dietary iron sources. However, the characterization, distribution and the responses of iron metabolism-related genes to different iron sources were not clear in fish. Methods The full-length cDNA sequences of fifteen iron metabolism-relevant genes ( tf , tfr1 , hp , fpn1 , ho1 , ho2 , tfr2 , hjv , hepcidin , fth , ftl , ftm , irp1 , irp2 and hif2α .) were obtained via 3′ and 5′ RACE PCR from yellow catfish, a widely distributed freshwater teleost in China and other Asian countries. Their molecular characterizations were analyzed via the bioinformatic methods. Real-time quantitative PCR was used to explore their mRNA distribution in nine tissues. Their mRNA expression responses in four tissues (heart, brain, kidney and gill) were explored in yellow catfish fed diets with five iron sources, including ferrous sulfate (FeSO 4 ), ferrous bisglycinate (Fe-Gly), ferrous chloride (FeCl 2 ), ferric citrate (Fe-CA) and ferric oxide nanoparticles (Fe 2 O 3 NPs). Results Compared with mammals and other teleost, these members shared similar domains. Their mRNAs were expressed in nine tested tissues, but mRNA levels varied. Yellow catfish fed the diets containing Fe-Gly and Fe 2 O 3 NPs had higher iron contents in heart, brain, kidney and gill. Meantime, different dietary iron sources addition affected their mRNA expression differentially in brain, heart, kidney and gill. It should be pointed out that only three biological replicate tanks were used in the present feeding treatment , and more biological replicate tanks (more than five) should be emphasized in further researches. Conclusion Taken together, our study identified fifteen iron metabolism-relevant genes, explored their mRNA expression in nine tissues, and their mRNA expression in the responses to different dietary iron sources in four tissues, indicating their important regulatory function in iron metabolism and homeostasis.

This experiment was conducted to investigate whether dietary chenodeoxycholic acid (CDCA) could attenuate high-fat (HF) diet-induced growth retardation, lipid accumulation and bile acid (BA) metabolism disorder in the liver of yellow catfish Pelteobagrus fulvidraco. Yellow catfish (initial weight: 4·40 (sem 0·08) g) were fed four diets: the control (105·8 g/kg lipid), HF diet (HF group, 159·6 g/kg lipid), the control supplemented with 0·9 g/kg CDCA (CDCA group) and HF diet supplemented with 0·9 g/kg CDCA (HF + CDCA group). CDCA supplemented in the HF diet significantly improved growth performance and feed utilisation of yellow catfish (P < 0·05). CDCA alleviated HF-induced increment of hepatic lipid and cholesterol contents by down-regulating the expressions of lipogenesis-related genes and proteins and up-regulating the expressions of lipololysis-related genes and proteins. Compared with the control group, CDCA group significantly reduced cholesterol level (P < 0·05). CDCA significantly inhibited BA biosynthesis and changed BA profile by activating farnesoid X receptor (P < 0·05). The contents of CDCA, taurochenodeoxycholic acid and glycochenodeoxycholic acid were significantly increased with the supplementation of CDCA (P < 0·05). HF-induced elevation of cholic acid content was significantly attenuated by the supplementation of CDCA (P < 0·05). Supplementation of CDCA in the control and HF groups could improve the liver antioxidant capacity. This study proved that CDCA could improve growth retardation, lipid accumulation and BA metabolism disorder induced by HF diet, which provided new insight into understanding the physiological functions of BA in fish.

Simple SummaryYellow catfish (Pelteobagrus fulvidraco) is an economically important freshwater fish in Chinese aquaculture that is highly susceptible to hypoxia in culture water, which can lead to mass mortality. We first examined the expression levels of thesirt1gene across multiple tissues of yellow catfish and found that it was most abundantly expressed in the brain and intestine, then investigated its transcriptional response in the intestine during hypoxia. We found thatsirt1responds rapidly to hypoxic conditions and shows a dynamic transcriptional response associated with metabolic- and stress-related genes in fish. Whensirt1expression was experimentally suppressed, intestinal tissue damage was markedly aggravated. Conversely, treatment with resveratrol partially restored downstream gene expression and alleviated tissue injury. These findings advance our understanding of how fish respond to hypoxic stress at the transcriptional level and may provide a basis for further investigation into hypoxia adaptation in aquaculture.Hypoxia is a common stressor in aquaculture and severely compromises the physiological health of fish. The present study investigated the expression pattern and transcriptional response of thesirt1gene (sirtuin1) in yellow catfish (Pelteobagrus fulvidraco) under acute hypoxia stress. Sequence analysis of the yellow catfishsirt1gene indicated that the Sirt1 protein exhibits a high degree of sequence conservation among vertebrates. Tissue distribution analysis under normoxic conditions revealed thatsirt1mRNA was most abundantly expressed in the brain and intestine of yellow catfish. Results from the hypoxia stress trial (dissolved oxygen 1.1 ± 0.2 mg/L) demonstrated thatsirt1was the most hypoxia-responsive member of thesirtfamily, being significantly upregulated approximately 4.5-fold at 12 h. Subsequent functional validation experiments showed thatsirt1expression was significantly decreased at 6 h of acute hypoxia; together with the marked upregulation at 12 h observed in the time-course experiment, this suggests that the response ofsirt1to acute hypoxia is time-dependent. On this basis, a bidirectional intervention was performed using small interfering RNA (siRNA)-mediated knockdown ofsirt1and resveratrol-mediated activation to analyze the effects ofsirt1on downstream gene expression and intestinal histopathology. The results showed that inhibition ofsirt1led to compensatory upregulation ofpdk1, impairment ofampkαsignaling and a further decrease instat3expression, and aggravated hypoxia-induced intestinal histological damage, whereas resveratrol pretreatment partially restored the expression of the related genes and alleviated tissue injury. These findings indicate thatsirt1is a transcriptionally responsive gene involved in the response of yellow catfish to hypoxia stress, and provide a reference for further investigation into hypoxia adaptation in yellow catfish.

Simple SummaryMyostatin is a negative regulator of muscle growth and development, and mutational inactivation of this gene increases muscle mass. Knockout of themstnbgene inPelteobagrus fulvidracodid not increase muscle mass, although our previous study of themstnagene did. Instead, it reduced the body size and growth performance ofPelteobagrus fulvidraco. Through the present study, we found that the mutantPelteobagrus fulvidracohad a reduction in the number of vertebrae, the length, and the intervertebral distance inPelteobagrus fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantPelteobagrus fulvidraco. Histological comparison of the same sites in themstnmutant and wild groups ofPelteobagrus fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutPelteobagrus fulvidracothan in wild-typePelteobagrus fulvidraco.AbstractBased on obtainingmstnbgene knockout inPelteobagrus fulvidraco, a study on the effect of themstngene on skeletal morphology and growth was performed by comparing the number and length of the vertebrae of mutant and wild-type fish in a sibling group ofP. fulvidraco, combined with the differences in cells at the level of vertebral skeletal tissue. It was found thatmstnbgene knockdown resulted in a reduction in the number of vertebrae, the length, and the intervertebral distance inP. fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantP. fulvidraco. Further, histological comparison of the same sites in themstnmutant and wild groups ofP. fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutP. fulvidracothan in wild-typeP. fulvidraco. Our results demonstrated that when using genome editing technology to breed new lines, the effects of knockout need to be analyzed comprehensively and may have some unexpected effects due to insufficient study of the function of certain genes. Keywords:myostatin b;genome editing;Pelteobagrus fulvidraco;body length;skeleton;vertebral centrum

Nitrogen from ammonia is one of the most common pollutants toxics to aquatic species in aquatic environment. The intestinal mucosa is one of the key mucosal defenses of aquatic species, and the accumulation of ammonia nitrogen in water environment will cause irreversible damage to intestinal function. In this study, histology, immunohistochemistry, ultrastructural pathology, enzyme activity analysis and qRT-PCR were performed to reveal the toxic effect of ammonia nitrogen stress on the intestine of Pelteobagrus fulvidraco . According to histological findings, ammonia nitrogen stress caused structural damage to the intestine and reduced the number of mucous cells. Enzyme activity analysis revealed that the activity of bactericidal substances (Lysozyme, alkaline phosphatase, and ACP) had decreased. The ultrastructure revealed sparse and shortened microvilli as well as badly degraded tight junctions. Immunohistochemistry for ZO-1 demonstrated an impaired intestinal mucosal barrier. Furthermore, qRT-PCR revealed that tight junction related genes ( ZO-1, Occludin, Claudin-1 ) were downregulated, while the pore-forming protein Claudin-2 was upregulated. Furthermore, as ammonia nitrogen concentration grew, so did the positive signal of Zap-70 (T/NK cell) and the expression of inflammation-related genes ( TNF , IL-1β , IL-8, IL-10) . In light of the above findings, we conclude that ammonia nitrogen stress damages intestinal mucosal barrier of Pelteobagrus fulvidraco and induces intestinal inflammation.

Intestinal inflammation is a common challenge in intensive aquaculture, yet its pathogenesis remains unclear. While interleukin 22 (IL-22) is recognized as a critical regulator of cellular homeostasis during inflammation in higher vertebrates, its roles in fish are not well understood. This study established hypoxia-induced models in intestinal tissues and primary intestinal epithelial cells of yellow catfish to investigate the involvement of IL-22 in maintaining intestinal homeostasis. Results revealed that Pelteobagrus fulvidraco IL-22 ( Pf _ IL-22 ) was abundantly expressed in mucosal tissues, with the highest levels in the gill and intestine. Hypoxia induced pronounced intestinal injury, characterized by loosening of the lamina propria and extensive vacuolization, while activating hypoxia-inducible factor (HIF) signaling and markedly up-regulating IL-22 expression. IL-22 levels peaked at 24 h post-hypoxia, suggesting a role in early immune responses. Recombinant Pf _IL-22 also induced transcription of pro-inflammatory mediators, including IL-1β and tumor necrosis factor α (TNF-α), in primary intestinal epithelial cells, indicating a dual regulatory function in balancing protection and inflammation. Mechanistic analyses revealed that HIF-1α directly interacted with a hypoxia response element within the IL-22 promoter to drive transcription, as confirmed by dual-luciferase assays, electrophoretic mobility-shift assays, and HIF-1α knockdown. Silencing Pf_IL-22 significantly suppressed Th17 cell differentiation pathways, demonstrating its role in shaping downstream immune responses. These findings establish the HIF-1α/IL-22 axis as a key regulatory pathway modulating immune responses and alleviating intestinal inflammation, providing a basis for developing IL-22-targeted immunotherapies and selective breeding strategies in aquaculture.

This research aimed to investigate the effects of five dietary iron sources on growth performance, feed utilization, appetite regulation, antioxidant capacity, endoplasmic reticulum (ER) stress and inflammatory response in the intestine of yellow catfish Pelteobagrus fulvidraco . The experimental diets were formulated with five iron sources, including ferrous sulfate (FeSO 4 ), ferrous bisglycinate (Fe-Gly), ferrous chloride (FeCl 2 ), ferric citrate (Fe-CA) and ferric oxide nanoparticles (Fe 2 O 3 NPs). Yellow catfish (initial body weight: 2.50 ± 0.01 g/fish, mean ± SEM) were fed the experimental diets for 10 weeks. Fish fed the diets with Fe-Gly and Fe 2 O 3 NPs supplementation displayed better growth performance, iron bioavailability, food intake and higher expression of appetite-related genes. Compared to inorganic iron (FeSO 4 and FeCl 2 ) and Fe 2 O 3 NPs, organic iron (Fe-Gly and Fe-CA) exhibited improved intestinal histology, upregulated expression of tight junction-related genes and enhanced antioxidant capacity. Dietary Fe 2 O 3 NPs addition damaged the intestinal antioxidation capacity of yellow catfish by down-regulating the expression and activities of related antioxidant enzymes. Besides, fish fed the Fe 2 O 3 NPs diet also showed higher mRNA and protein levels of ER stress-and inflammation-related genes. Given the above findings, Fe-Gly seems to be a better iron additive for its benefits on growth performance and intestinal health to other iron sources.

Poultry by-products (PBPs) are increasingly used in aquacultural environments and have emerged as a potential source of veterinary antibiotics and pathogens; however, the ecological risks remain unclear. This study investigated tissue-specific accumulation of 34 antibiotics and microbial risks in yellow catfish (Pelteobagrus fulvidraco) following exposure to PBPs. Among the 11 tissues analyzed, the data suggest that over 93% of antibiotic residues may be derived from PBPs, with concentrations ranging from 1.21 to 354.31 ng g−1, with the highest concentrations observed in bile, kidney, and liver. Bioaccumulation of tilmicosin, enrofloxacin, and florfenicol occurred via both dietary intake and dermal exposure, with log bioaccumulation factor (BAF) values of up to 3.79 for bile. High-throughput 16S rRNA sequencing revealed the consistent occurrence of Acinetobacter and Mycobacterium across fish tissues, feed, and water, with 40% of dominant taxa identified as known or suspected cross-species potentially pathogenic genera. A significant reduction in skin microbiota diversity further indicates a possible risk of exposure-induced dysbiosis. These findings highlight PBPs as vectors of both antibiotic residues and microbial disturbance, underscoring the need for targeted control of PBP-derived risks to safeguard freshwater ecosystem health.

As an inevitable factor in aquaculture, ammonia plays a critical role in macrolide antibiotic resistance, leading to accumulating of antibiotic-resistant bacteria in fish skin mucus. In this study, four experimental groups were implemented to test the effects of ammonia alone or in combination with roxithromycin for 28 days on skin mucus microbial composition and the immune response of yellow catfish: CON (control), AN (50.00 mg L −1 total ammonia nitrogen, TA-N), ROX (100 μg L −1 roxithromycin), and HR (50.00 mg L −1 TA-N, 100 μg L −1 ROX). This study demonstrated that ammonia or roxithromycin exposure resulted in increased plasma ammonia content and decreased total antioxidant capacity. Compared with AN group, the combined exposure of ammonia and roxithromycin inhibited the skin mucus immune response. Microbial composition analysis showed that combined exposure of ammonia and roxithromycin had no significant effect on skin mucus α-diversity as compared with CON group. The abundance of Cetobacterium , Rhizobiales_Incertae_Sedis_uncultured and Acinetobacter was increased significantly with the combined effect of ammonia and roxithromycin, these bacteria may be potentially antibiotic-resistant. As compared with CON group, the combined exposure of ammonia and roxithromycin did not affect skin goblet cell counts. This study suggests that combined exposure to ammonia and ROX increases the risk of the emergence of antibiotic-resistant bacteria.

This study evaluated the effects of chronic exposure to copper nanoparticles (Cu-NPs) and waterborne copper (CuSO 4 ) on the reproductive system of yellow catfish ( Pelteobagrus fulvidraco ). Juvenile yellow catfish were exposed to 100 and 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO 4 for 42 days. The results showed clear reproductive defects in both female and male yellow catfish in the 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO 4 groups. Exposure to Cu-NPs or CuSO 4 inhibited folliculogenesis and vitellogenesis in the ovaries, and spermatogenesis in the testes , accompanied by elevation of the apoptotic signal. Ultrastructural observations also revealed damaged organelles of gonadal cells in both testes and ovaries. Most of the hypothalamic-pituitary-gonadal (HPG) axis genes examined and serum sex steroid hormones tended to be downregulated after Cu exposure. Metabolomic analysis suggested that gonadal estradiol level is sensitive to Cu-NPs or CuSO 4 . The heat map of gonadal metabolomics suggested a similar effect of 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO4 in both the ovaries and testes. Additionally, metabolomics data showed that the reproductive toxicity due to Cu-NPs and CuSO 4 may occur via different metabolic pathways. Cu-NPs tend to dysregulate the metabolic pathways of sphingolipid and linoleic acid metabolism in the ovary and the biosynthesis of amino acids and pantothenate and CoA in the testis. Overall, these findings revealed the toxicological effects of Cu-NPs and CuSO 4 on the HPG axis and gonadal metabolism in yellow catfish.

The present study investigated the growth performance, body composition, mineral content and intestinal health of yellow catfish juveniles fed different iron diets and explored the dietary Fe requirements for this fish species. Using iron oxide nanoparticles (Fe 2 O 3 NPs) as the Fe sources, we formulated six experimental diets with dietary Fe levels at 22.23, 28.28, 36.10, 44.53, 52.21, and 61.85 mg Fe/kg diet, respectively. Yellow catfish fed with 44.53 mg Fe/kg diet had the highest growth performance and feed utilization, while those fed 61.85 mg Fe/kg diet showed the lowest growth performance and feed utilization. Dietary Fe addition increased whole-body and intestinal Fe contents. Additionally, the addition of dietary Fe had an impact on whole-body crude protein, crude lipid, ash, calcium (Ca), magnesium (Mg), zinc (Zn), and manganese (Mn) contents, but did not whole-body moisture and copper (Cu) contents. Yellow catfish fed with 44.53 mg Fe/kg diet had an improvement in intestinal histology and an increase in expression of intestinal tight junction-related genes [ occludin and tight junction protein 1b ( zo-1b )] as well as antioxidant response genes [ superoxide dismutase 1 ( sod1 ), sod2 and catalase ( cat )], and had higher activities of antioxidant enzymes [CAT, total SOD (T-SOD) and total antioxidant capacity (T-AOC)]. However, malondialdehyde (MDA) content, and gene expression of glucose-regulated protein 78 ( grp78 ), eukaryotic translation initiation factor 2α ( eif2α ), activating transcription factor 4 ( atf4 ), inositol-requiring enzyme 1 ( ire1 ) (related to endoplasmic reticulum stress), and mRNA levels of tumor necrosis factor α ( tnfα ), interleukin 1β ( il1β ), il8 (related to inflammatory response) in the intestine were the lowest in 44.53 mg Fe/kg diet group. Conducting broken-line analysis of weight gain (WG) against dietary Fe levels, we revealed that the optimal Fe requirement was 45.28 mg Fe/kg diet, with Fe 2 O 3 NPs as Fe source.
