Bio-decolorization of synthetic dyes by a novel endophytic fungus Penicillium janthinellum LM5 from blueberry pulp

Jufen Yan,Pei Wang,Longxin Wang,Qi Jin,Abdallah S. Ali,Yongsheng He,Ying Wang,Yuxuan Sun,Aoqiang Li,Waly Adwy,Rasha H. Ahmed,Xinya Han

Synthetic dyes from textile, paper, leather, and plastic processing, etc. are the majority of the colored effluents and have a harmful impact on human health and environment. Dye biosorption through endophytic fungi will be a challenge. A new fungus isolated from blueberry pulp was identified morphologically and genetically as Penicillium janthinellum LM5. It was utilized for the biosorption of synthetic dyes by forming mycelium particles with efficient decolorization properties in the dye solution. The results of decolorization experiments exhibited that dye type, pH, temperature and time were the important factors and the best decolorization condition on Congo red was pH 6.0, 30 ℃ and 48 h. Moreover, LM5 can achieve 90% decolorization rate even under the high salt concentrations of 200 mg/mL. Based on spectral analysis , the decolorization of Congo red was mainly attributed to the biosorption of mycelium and its pellet structure. Additionally, the fungus adsorbed Congo red can be as an excellent electrode material with a long-cycle performance and stable capacitance property, being a good way for recycling. In summary, this research employed a novel dye treatment method with simultaneous culture and decolorization through an endophytic fungus LM5, being a promising biomaterial in the application of dye wastewater treatment .

A proteomic-based investigation of potential copper-responsive biomarkers: Proteins, conceptual networks, and metabolic pathways featuring Penicillium janthinellum from a heavy metal-polluted ecological niche

Xin Feng,Jian Xu,Yu Liang,Guo-Li Chen,Xian-Wei Fan,You-Zhi Li

Graphical Our work not only provides the potential protein biomarkers indicating the Cu pollution, target metabolic steps for engineering the Cu-tolerant fungi during bioremediation, but also presents clues for insight into heavy metal tolerance mechanisms of other eukaryotes. Filamentous fungi-copper (Cu) interactions are very important in the formation of natural ecosystems and the bioremediation of heavy metal pollution. However, important issues at the proteome level remain unclear. We compared six proteomes from Cu-resistant wild-type (WT) Penicillium janthinellum strain GXCR and a Cu-sensitive mutant (EC-6) under 0, 0.5, and 3 mmol/L Cu treatments using iTRAQ. A total of 495 known proteins were identified, and the following conclusions were drawn from the results: Cu tolerance depends on ATP generation and supply, which is relevant to glycolysis pathway activity; oxidative phosphorylation, the TCA cycle, gluconeogenesis, fatty acid synthesis, and metabolism are also affected by Cu; high Cu sensitivity is primarily due to an ATP energy deficit; among ATP generation pathways, Cu-sensitive and Cu-insensitive metabolic steps exist; gluconeogenesis pathway is crucial to the survival of fungi in Cu-containing and sugar-scarce environments; fungi change their proteomes via two routes (from ATP, ATP-dependent RNA helicases (ADRHs), and ribosome biogenesis to proteasomes and from ATP, ADRHs to spliceosomes and/or stress-adapted RNA degradosomes) to cope with changes in Cu concentrations; and unique routes exist through which fungi respond to high environmental Cu. Further, a general diagram of Cu-responsive paths and a model theory of high Cu are proposed at the proteome level. Our work not only provides the potential protein biomarkers that indicate Cu pollution and targets metabolic steps for engineering Cu-tolerant fungi during bioremediation but also presents clues for further insight into the heavy metal tolerance mechanisms of other eukaryotes.

Anti-Diabetic Activity of a Novel Exopolysaccharide Produced by the Mangrove Endophytic Fungus Penicillium janthinellum N29

Zhuling Shao,Yingying Tian,Shan Liu,Xiao Chu,Wenjun Mao

Marine microorganisms often produce exopolysaccharides with novel structures and diverse biological activities due to their specific marine environment. The novel active exopolysaccharides from marine microorganisms have become an important research area in new drug discovery, and show enormous development prospects. In the present study, a homogeneous exopolysaccharide from the fermented broth of the mangrove endophytic fungusPenicillium janthinellumN29, designated as PJ1-1, was obtained. The results of chemical and spectroscopic analyses showed that PJ1-1 was a novel galactomannan with a molecular weight of about 10.24 kDa. The backbone of PJ1-1 was composed of →2)-α-d-Manp-(1→, →4)-α-d-Manp-(1→, →3)-β-d-Galf-(1→ and →2)-β-d-Galf-(1→ units with partial glycosylation at C-3 of →2)-β-d-Galf-(1→ unit. PJ1-1 had a strong hypoglycemic activity in vitro, evaluated using the assay of α-glucosidase inhibition. The anti-diabetic effect of PJ1-1 in vivo was further investigated using mice with type 2 diabetes mellitus induced by a high-fat diet and streptozotocin. The results indicated that PJ1-1 markedly reduced blood glucose level and improved glucose tolerance. Notably, PJ1-1 increased insulin sensitivity and ameliorated insulin resistance. Moreover, PJ1-1 significantly decreased the levels of serum total cholesterol, triglyceride and low-density lipoprotein cholesterol, enhanced the level of serum high-density lipoprotein cholesterol and alleviated dyslipidemia. These results revealed that PJ1-1 could be a potential source of anti-diabetic agent. Keywords:mangrove endophytic fungus;exopolysaccharide;structure;anti-diabetic activity

A novel biosorbent formed of marine-derived Penicillium janthinellum mycelial pellets for removing dyes from dye-containing wastewater

Ming-Xia Wang,Qi-Lei Zhang,Shan-Jing Yao

Mycelial pellets prepared using a marine-derived Penicillium janthinellum sp. strain (P1) were applied as biosorbents to remove dyes from aqueous solutions. Nine different dyes were studied and the effects of pH and salt concentration on the decolorization processes were investigated. The results showed that the mycelial pellet was effective in removing dyes from their water solutions. Moreover, it was found that pH was an important factor of the decolorization process and the best dye removal performance was achieved when pH   =   5.0. The adsorption process of the live mycelial pellets was not affected by high salt concentrations. In addition, the mycelial pellets retained 91.3% of the original decolorization activity after five reusing cycles without any desorption treatments. The adsorption isotherms indicated that the biosorption process agreed well with the Langmuir isotherm model with a maximum biosorption capacity of 344.83   mg   g(dry) − 1 and the kinetic adsorption results showed that the biosorption process followed a pseudo-second-order kinetic model.

Structural Characterization and Biological Evaluation of a Novel GlcNAc-Bearing Exopolysaccharide from the Mangrove Endophytic Fungus Penicillium janthinellum N29

Zhuling Shao,Luya Wang,Xin Wang

A novel, uniform acid heteropolysaccharide with a molecular weight of 13.14 kDa was isolated from the fermented broth of the mangrove endophytic fungusPenicillium janthinellumN29, designated as PJ3-1. A structural analysis showed that PJ3-1 is a straight-chain heteropolysaccharide containing GlcN, GlcNAc, Glc and GlcA in the ratio of 12.23:1.79:10.93:1.00. PJ3-1 markedly inhibits α-glucosidase activity and relieves insulin resistance in HepG2 cells. It alleviates lipid peroxidation and upregulates the activities of endogenous antioxidant enzymes. In addition, it also significantly scavenges hydroxyl radicals, ferrous ions, superoxide anions, DPPH radicals and ABTS radicals and enhances reducing power in a dose-dependent manner. Collectively, these findings suggest that PJ3-1 has the potential to serve as a natural antioxidant and hypoglycemic agent for functional food or pharmaceutical applications.
