The colonization of Penicillium oxalicum SL2 on rice root surface increased Pb interception capacity of iron plaque and decreased Pb uptake by roots

Jianhao Tong,Hanxin Wu,Xiaohan Jiang,Jing Wang,Jingli Pang,Haonan Zhang,Ziming Xin,Jiyan Shi

The exploration of microbial resources to reduce Pb accumulation in rice attracted great attention. In this study, we found Penicillium oxalicum SL2, a Pb-tolerant strain with good capability of dissolving phosphorus and stabilizing Pb in soil, was able to colonize on the root surface of rice seedlings without additional carbon sources, and promoted the secretion of metabolites related to amino acid metabolism , organic acid metabolism, signal transduction and other pathways in rhizosphere exudates, in which the secretion of oxalate increased by 47.7 %. However, P. oxalicum SL2 increased Fe(II) proportion and Fe availability on the root surface, resulting in iron plaque content decrease. Moreover, by converting root surface Pb from Pb-Fe state to PbC 2 O 4 and Pb-P compounds, P. oxalicum SL2 increased Pb intercept capacity of iron plaque by 118.0 %. Furthermore, P. oxalicum SL2 regulated element distribution on the root surface, and reduced the relative content of Pb on the maturation zone of root tip , which was conducive to reducing Pb uptake by apoplastic pathway and the risk of Pb accumulation in root system . Our findings further revealed the interaction between P. oxalicum SL2 and rice root, providing a theoretical basis for the development and application of microbial agents in Pb-contaminated farmland.

The cellulose binding region in Trichoderma reesei cellobiohydrolase I has a higher capacity in improving crystalline cellulose degradation than that of Penicillium oxalicum

Jian Du,Xiu Zhang,Xuezhi Li,Jian Zhao,Guodong Liu,Baoyu Gao,Yinbo Qu

Commercial cellulase preparations for lignocellulose bioconversion are mainly produced by the fungus Trichoderma reesei . The maximum cellulose conversion of T. reesei cellulase mixture was 15%–20% higher than that of Penicillium oxalicum in the hydrolysis of corncob residue and Avicel. Nevertheless, both preparations hydrolyzed more than 92% of cellulose in NaOH-mercerized Avicel. When added to Avicel hydrolysis residue that was less reactive to P. oxalicum cellulases, cellobiohydrolase I (CBH I) from T. reesei resulted in a higher cellulose conversion than its homologous proteins from P. oxalicum and Aspergillus niger at the same protein loadings. Further domain exchange experiment attributed the high hydrolytic efficiency of T. reesei CBH I to its inter-domain linker and cellulose-binding domain. The results in part explained the superior performance of T. reesei cellulases on the degradation of native crystalline cellulose, and highlighted the important role of cellulose-binding region in determining the degree of hydrolysis by cellulases.

Mutation of a Conserved Alanine Residue in Transcription Factor AraR Leads to Hyperproduction of α-l-Arabinofuranosidases in Penicillium oxalicum

Liwei Gao,Shiying Li,Yanning Xu,Chengqiang Xia,Jiadi Xu,Jun Liu,Yuqi Qin,Xin Song,Guodong Liu,Yinbo Qu

α- l -Arabinofuranosidases are important in the degradation of plant polysaccharides and are used in several industrial processes. Although the use of filamentous fungi for the production of α- l -arabinofuranosidases is widely reported, there are few reports on strain engineering for enhanced production of these enzymes by fungi. In this study, the function of transcription factor AraR in l -arabinose release and catabolism by the fungus Penicillium oxalicum ( P. oxalicum ) is investigated. Also, a mutant of AraR, AraR A731V , is constructed to improve the production of α- l -arabinofuranosidases on the basis of the sequence homology between AraR and the xylanolytic gene activator XlnR. The AraR A731V -overexpressing strain can synthesize α- l -arabinofuranosidase in the absence of an inducer and shows a 54.1-fold increase in α- l -arabinofuranosidase production and a 7.4-fold increase in α-galactosidase production in the medium containing wheat bran. Determination of the transcript abundances of lignocellulolytic enzyme genes reveals significant upregulation of multiple α- l -arabinofuranosidase genes and downregulation of some cellulolytic and xylanolytic enzyme genes in the engineered strain relative to its parent. Taken together, the results suggest the conserved regulatory function of AraR in the family Trichocomaceae and provide a strategy for engineering fungal strains for enhanced α- l -arabinofuranosidase production. Graphical α- l -Arabinofuranosidases are important in plant cell-wall degradation and are used in many industries. In this study, it is shown that a point mutation of transcription factor AraR could increase α- l -arabinofuranosidase production by 54.1-fold in the fungus Penicillium oxalicum .

Optimization of an artificial cellulase cocktail for high-solids enzymatic hydrolysis of cellulosic materials with different pretreatment methods

Jian Du,Jingrui Liang,Xiuhua Gao,Guodong Liu,Yinbo Qu

Optimization of the composition of cellulase mixtures is an effective strategy to improve their hydrolytic efficiency and reduce protein demand during enzymatic degradation of lignocelluloses. In this study, the mixture design method was used to optimize the ratios of endoglucanase II (EG II), cellobiohydrolase I (CBH I) and β-glucosidase I (BG I) from Penicillium oxalicum in an artificial cellulase mixture for the hydrolysis of six different cellulosic materials. The optimal composition of enzyme mixture was distinctly different among not only cellulosic materials with different pretreatment methods but hydrolyses at different solids concentrations. CBH I was most critical for the hydrolysis of two acid-pretreated materials, probably due to its strong adsorption on lignin. A higher proportion of EG II was needed for the hydrolysis of ammonium sulfite pretreated wheat straw. The requirements of specific cellulase components were more pronounced at high solids concentrations, highlighting the importance of considering solids loading when optimizing cellulase cocktails.

Mechanistic investigation of sucrose monostearate-mediated antifungal effects against Penicillium oxalicum

Sun Yanru,Gao Jing,Jin Xianrui,Li Shuhan,Hou Yexuan,Fu Lu,Zhang Wenhui

Penicillium oxalicum is a major postharvest pathogen of kiwifruit, causing substantial economic losses. This study investigated the antifungal activity and mechanism of sucrose monostearate (SMS) against P. oxalicum . The minimum inhibitory concentration (MIC) of SMS was determined to be 1.25 g L − 1 , which significantly inhibited spore germination by 88.28% and reduced mycelial growth by 65.94% ( p  < 0.05). Mechanistically, SMS at 1.25 g L − 1 induced intracellular ROS accumulation by 1.7-fold, suppressed catalase (CAT) and peroxidase (POD) activities by 54% and 60%, and increased malondialdehyde (MDA) content by 2.5-fold, confirming severe membrane lipid peroxidation and content leakage. Scanning electron microscopy revealed marked hyphal deformation, and in vivo assays showed that SMS reduced kiwifruit lesion diameter by 83.7% compared to controls. These findings demonstrate that SMS, as a single-ester antifungal agent, disrupts fungal membrane integrity and exhibits strong potential for postharvest disease control.

Functional diversity and properties of multiple xylanases from Penicillium oxalicum GZ-2

Liao Hanpeng,Zheng Haiping,Li Shuixian,Wei Zhong,Mei Xinlan,Ma Hongyu,Shen Qirong,Xu Yangchun

A multiple xylanase system with high levels of xylanase activity produced from Penicillium oxalicum GZ-2 using agricultural waste as a substrate has been previously reported. However, the eco-physiological properties and origin of the multiplicity of xylanases remain unclear. In the present study, eight active bands were detected using zymography and all bands were identified as putative xylanases using MALDI-TOF-MS/MS. These putative xylanases are encoded by six different xylanase genes. To evaluate the functions and eco-physiological properties of xylanase genes, xyn10A , xyn11A , xyn10B and xyn11B were expressed in Pichia pastoris . The recombinant enzymes xyn10A and xyn10B belong to the glycoside hydrolase (GH) family 10 xylanases, while xyn11A and xyn11B belong to GH11 xylanases. Biochemical analysis of the recombinant proteins revealed that all enzymes exhibited xylanase activity against xylans but with different substrate specificities, properties and kinetic parameters. These results demonstrated that the production of multiple xylanases in P. oxalicum GZ-2 was attributed to the genetic redundancy of xylanases and the post-translational modifications, providing insight into a more diverse xylanase system for the efficient degradation of complex hemicelluloses.

Cloning, Expression, and Characterization of Endo-β-1,6-galactanase PoGal30 from Penicillium oxalicum

Zhang Xin,Wang Yibing,Liu Jiaqi,Wang Weiyang,Yan Xuecui,Zhou Yifa,Cui Jing,Yuan Ye

Because β-1,6-galactans are significant components in arabinogalactans from plant cell walls, identifying selective endo-β-1,6-galactanases is crucial to degrading these polysaccharides and to analyzing and modifying their structures. Here, we cloned and expressed in E. coli a novel endo-β-1,6-galactanase in the glycosidic hydrolase family 30 (GH30) from Penicillium oxalicum . Our recombinant PoGal30 hydrolase (1464 bp gene) that contains an N-terminal His-tag for purification by nickel affinity chromatography has a specific activity of 3.8 U/mg on the substrate de-arabinosylated gum Arabic (dGA) polysaccharide. The enzyme has 487 residues with a molecular mass of 60 kDa, an isoelectric point of 6, and functional pH and temperature optima of pH 2.5 to pH 5.0 and 40 °C, respectively. While the activity of PoGal30 is activated by Mg 2+ (5 or 50 mmol/L), it is completely inhibited by Cu 2+ and Fe 3+ (50 mmol/L) and partially inhibited by Hg 2+ , EDTA, and SDS (50 mmol/L). The enzyme demonstrates high specificity towards β-1,6-galactosidic linkages in dGA, but is inactive against aryl-glycosides and galactobioses with different linkages. Using PoGal30 is, therefore, an effective approach to analyzing the fine structure of polysaccharides and preparing bioactive oligosaccharides.

Screening of potential IL-tolerant cellulases and their efficient saccharification of IL-pretreated lignocelluloses

Yi-Xin Sun,Bing-Bing Shen,Hui-Ying Han,Yuan Lu,Bi-Xian Zhang,Yun-Fei Gao,Bao-Zhong Hu,Xiao-Mei Hu

Lignocellulosic biomass as one of the most abundant and renewable resources has great potential for biofuel production. The complete conversion of biomass to biofuel is achieved through the effective pretreatment process and the following enzyme saccharification. Ionic liquids (ILs) are considered as a green solvent for lignocellulose pretreatment. However, ILs exhibit an inhibitory effect on cellulase activity, leading to a subsequent decrease in the efficiency of saccharification. The screening of new potential IL-tolerant cellulases is important. In the current study, a fungal strain with a relatively high cellulase production was isolated and identified as Penicillium oxalicum HC6. The culture conditions were optimized using corn stover and peptone as the carbon source and nitrogen source at pH 4.0 and 30 °C with an inoculation size of 2% (v/v) for 8 days. It was found that P. oxalicum HC6 exhibited potential salt tolerance with the increase of the enzyme production at a salt concentration of 5.0% (w/v). In addition, high enzyme activities were obtained at pH 4.0–6.0 and 50–65 °C. The crude enzyme from P. oxalicum HC6 with good thermal stability was also stable in the presence of salt and ILs. Good yields of reducing sugar were obtained by the crude enzyme from P. oxalicum HC6 after the saccharification of corn stover that was pretreated by ILs. P. oxalicum HC6 with potentially salt-tolerant and IL-tolerant enzymes has great potential application in the enzymatic saccharification of lignocellulose.

Characterization of Novel Enzymes from Penicillium oxalicum and Their Role in the Degradation of RG-I Pectin

Yibing Wang,Qianqian Xuan,Tiantian Yuan,Baojin Wang,Zihan Zhang,Xiaoqian Xing,Xinyi Yang,Chen Meng,Kevin H. Mayo,Fan Li,Yifa Zhou,Ye Yuan

The degradation of complex plant polysaccharides, particularly Rhamnogalacturonan I (RG-I) pectin, is essential for unlocking the value of biomass. This study focuses on the enzymatic toolbox of Penicillium oxalicum for efficient RG-I breakdown. Here, we recombinantly expressed 15 novel pectinolytic enzymes and characterized their biochemical functions. The enzyme system consists of backbone-cleaving rhamnogalacturonan hydrolases and lyases, along with side-chain–degrading enzymes. Specific endo-β-galactanases (GH30, GH43, GH53) and an exo-β-galactosidase (GH35) collaboratively hydrolyze galactan side chains. Meanwhile, α-arabinanases (GH43) and exo-α-arabinofuranosidases (GH54, GH62) work synergistically to degrade arabinan side chains. Through coordinated action, these enzymes achieve complete degradation of complex RG-I structures. Our findings provide a comprehensive enzyme set for targeted polysaccharide deconstruction, offering valuable resources for biomass conversion and pectin valorization in industrial applications.
