Chemical Composition, Antioxidant and Antimicrobial Activity of Pericarpium Citri Reticulatae Essential Oil

Bei Gao,Yulong Chen,Mingwei Zhang,Yujuan Xu,Siyi Pan

The chemical composition, antioxidant and antimicrobial activity ofPericarpium Citri Reticulatae(PCR) essential oil obtained using an improved Clevenger type apparatus were studied. Among the five differentPCRs examined the highest yield of essential oil was found in Chachi 2004 (harvested and stored in 2004) and the lowest in Chachi 2008 (harvested and stored in 2008). Fifty three different volatile compounds were determined, including terpenic hydrocarbons, alcohols, aldehydes, ketones and esters. D-limonene, one of terpenes, was the major constituent inPCR. The antioxidant capacity ofPCRessential oil varied considerably with the duration of storage time, and the oil from Chachi 1994 has the strongest ferric-reducing antioxidant power. In addition, the essential oil possessed varying degrees of antimicrobial activity against Gram-positive bacteria (Staphylococcus aureus,Bacillus subtilis,Bacillus cereus), exceptStreptococcus faecalis, while had no effect on Gram-negative bacteria (Escherichia coli,Enterobacter cloacae). Keywords:Pericarpium Citri Reticulatae;Chachi;chemical composition;antioxidant activity;antimicrobial activity

Ultra-small and biocompatible platinum nanoclusters with peroxidase-like activity for facile glucose detection in real samples

Yu Zhao,Jin Zhang,Danyang Xie,Haotian Sun,Shuqian Yu,Xiaolei Guo

The highly sensitive glucose detection based on the peroxidase-like properties of nanoclusters has been gained great interest. In this work, Pericarpium Citri Reticulatae polysaccharide (PCRP) stabilized platinum nanoclusters (Pt-PCRP NCs) were prepared by a green method in which potassium tetrachloroplatinate and PCRP were simply mixed without addition of other agents. Platinum nanoclusters (Pt NCs) had ultra-small size of 1.26 ± 0.34 nm. The hydrodynamic size of Pt-PCRP NCs was 29.7 nm, and zeta potential of which was -12.0 mV. Pt-PCRP NCs showed high biocompatibility toward HeLa cells and red blood cells. In addition, Pt-PCRP NCs catalyzed the decomposition of H2O2 to produce •OH, which further oxidized colorless 3,3′5,5′-tetramethylbenzidine (TMB) to blue oxidized 3,3′,5,5′-tetramethylbenzidine (oxTMB), exhibiting peroxidase-like property. The kinetics followed the Michaelis-Menten equation. More importantly, the colorimetric method for glucose detection using Pt-PCRP NCs had high selectivity and low detection limit for 0.38 μM. The established method based on Pt-PCRP NCs was used to precisely detect glucose detection in human serum, saliva, and sweat. Taken together, the prepared ultra-small and biocompatible Pt-PCRP NCs have good potential glucose applications in clinical diagnosis in the future.

N-doped carbon dots from pericarpium citri reticulatae for wide linear range sensing of hydroquinone

Peng Du,Zhengkun Chu,Jing Zhang,Jieyu Ma,Feng Cao,Jie Liu

Hydroquinone (H2Q) present in water is becoming a growing problem in both daily life and industry, with various harmful effects on human health. Therefore, the detection of H2Q is very important. The Carbon dot-based sensor is considered an effective method to detect H2Q. In this work, Pericarpium citri reticulatae carbon dots (P-CDs) were synthesized by a one-pot hydrothermal method for wide linear sensing of H2Q. An orthogonal test was used to expand the active site and fluorescence properties of P-CDs. Under optimal conditions, the P-CDs exhibit an extremely stable blue fluorescence and a quantum yield (QY) of up to 15.4%. In the range of 5–1000 μM, compared with the currently reported carbon dot-based H2Q sensors, the fluorescence intensity (FL) of P-CDs shows good linearity with the variation of H2Q concentration and a higher linear range. Moreover, the sensor demonstrates a low detection limit of 0.058 μM. The possible detection mechanism is based on the synergistic action of static quenching and IFE. This method has been successfully applied to the determination of H2Q in real samples, and the P-CDs-based sensor has the potential to detect H2Q in various complex scenes.

Metabolism and release of characteristic components and their enzymatic mechanisms in Pericarpium Citri Reticulatae co-fermentation

Fei Shen,Tingyu Wang,Renjie Zhang,Bin Zhong,Zhenqiang Wu

A co-fermentation strategy was explored to rapidly improve the characteristic components and quality of Pericarpium Citri Reticulatae (PCR) using Monascus anka and Saccharomyces cerevisiae , and the enzymatic mechanism was investigated. The results showed that the free flavonoid content of fermented PCR was 48.12% higher than that of unfermented PCR after 12 days of co-fermentation, resulting in stronger antioxidant activity. d -Limonene, γ-terpinene, proline (Pro), arginine (Arg), and serine (Ser) contributed the most to the flavors of citrus, herb, and sweet citrus based on odor and taste activity value analysis. Metabolomics and multivariate statistics showed that 55 components were differentially metabolized during co-fermentation, and ten metabolic pathways were closely related to metabolism. Furthermore, five hydrolases participated in the release and conversion of the active ingredients. This study provides an effective processing method for PCR and is conducive to the development of new PCR functional health foods.

Optimization of Ultrasound-Assisted Deep Eutectic Solvent Extraction, Characterization, and Bioactivities of Polysaccharide from Pericarpium Citri Reticulatae

Wen Yao,Chen Jiawen

The ultrasonic-assisted deep eutectic solvent method was used to extract the polysaccharides of Pericarpium Citri Reticulatae (PCRP), and the ultrasound-assisted DES extraction process was optimized by Box-Behnken response surface test using the extraction rate of the PCRP as an index; the in vitro activities of purified the PCRP(PCRPs-1) were investigated by determining the scavenging rate of DPPH• and ABTS• + as well as by enzyme inhibition assay. The monosaccharide composition was analyzed by HPLC. The best process conditions for response surface optimization were a material-liquid ratio of 1:37 g/mL, water content of 44%, time of 89 min, and power of 320 W. The polysaccharide extraction rate was measured to be 5.41%, which was well optimized when compared with that of the ordinary aqueous extraction method of 3.92%. By α-glucosidase and α-amylase inhibition activity test, it showed that the PCRPs-1 had hypoglycemic activity. The DPPH radical scavenging activity test and ABTS + scavenging activity test indicated that the PCRPs-1 had good biological activity. Analysis of the monosaccharide fractions showed that the PCRPs-1 consisted of mannose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, xylose, and arabinose, with molar ratios of 1:39.24:4.41:8.91:7.83:86.00:1.02:9.17. The activity studies showed that PCRPs-1 possessed certain hypoglycaemic and antioxidant activities.

Pulsed electric field-ultrasonic assisted extraction combined with macroporous resin for the preparation of flavonoids from Pericarpium Citri Reticulatae

Jinjing Zhan,Zijian Liang,Jian Li,Xinan Zeng,Guoliang Ou,Chuming Zhong

Pericarpium Citri Reticulatae (PCR) has long been perceived as a condiment and medicine in East Asian countries especially China. To establish a cost-effective method with the potential for industrial implementation to enhance extraction and purification of PCR flavonoids, four extraction methods (hot water extraction (HWE), ultrasound-assisted extraction (UAE), pulsed electric field-assisted extraction (PEF-AE), pulsed electric field combined with ultrasound-assisted extraction (PEF-UAE)) and five macroporous resins (AB-8, NKA-9, D-101, HPD-100, and HPD-950) were investigated and compared in this study. Results of total flavonoid content (TFC) and monomer flavonoid composition revealed that PEF-UAE was the most efficient extraction process. Response surface methodology (RSM) was employed to determine the optimum conditions of PEF-UAE (ultrasonic power 340 W, ultrasonic time 30 min, electric field strength 3 kV/cm, and 81 pulses). Under the optimum conditions, the PCR flavonoids extracted by PEF-UAE yielded the highest contents of total flavonoid, nobiletin, and tangeretin in all PCR flavonoid samples. Among the five macroporous resins, AB-8 was used to purify PCR flavonoids due to its higher adsorption and desorption capacities. A maximum purification efficiency was achieved under 6 h of adsorption time and 298 K of temperature, indicated by 2.47-fold, 2.78-fold, and 2.43-fold increases in the contents of hesperidin, nobiletin, and tangeretin, respectively. The purified PEF-UAE flavonoid extract also demonstrated the superior α-glucosidase inhibitory and radical scavenging activities. This study offered a prerequisite for the future utilization of PEF-UAE and AB-8 to produce PCR flavonoids for food and medicine applications. Novelty impact statement Pulsed electric field combined with ultrasound can significantly increase the extraction rate of flavonoids from Pericarpium Citri Reticulatae . The purification mechanism of AB-8 for Pericarpium Citri Reticulatae flavonoids has been explained. The Pericarpium Citri Reticulatae flavonoids extracted by pulsed electric field combined with ultrasound showed the best biological activities.
