Tannic acid enhanced the removal of Phaeocystis globosa from seawater by Fe (II) activated persulfate

Mingda Che,Xianfeng Wang,Mei Cui,Renliang Huang,Wei Qi,Rongxin Su

The frequent outbreaks of Phaeocystis globosa ( P. globosa ) blooms in coastal areas have seriously threatened nuclear power safety. In this study, tannic acid (TA) was used to accelerate the removal of P. globosa from seawater by Fe (Ⅱ) activated persulfate (PS). In comparison with the Fe (Ⅱ)-PS system, the higher removal efficiency of P. globosa was obtained in the TA-Fe (Ⅱ)-PS system (91.2% vs 67.5%). The removal efficiency of fluorescence value, chlorophyll a, turbidity and total organic carbon (TOC) reached up to 90.1%, 91.2%, 61.8% and 51.6% with the addition of 10 mg L −1 of TA, 100 mg L −1 of Fe (Ⅱ) and 240 mg L −1 of PS in 420 min. The flocs images confirmed that TA played a dual role in regulating Fe (Ⅱ) concentration and activating PS, which facilitated the pre-oxidation of algae cells. Finally, the inactivated P. globosa settled to the bottom with the iron mineral flocs. In addition, positive environmental impacts were also validated due to the decrease in total organic carbon (TOC), total nitrogen (TN), total phosphorus (TP) and chemical oxygen demand (COD). The findings demonstrated that TA-Fe (Ⅱ)-PS system is a promising candidate for the removal of P. globosa from seawater.

Inhibitory effects of Ipomoea cairica extracts on the harmful algae Phaeocystis globosa

Dayong Liang,Hua Xiang,Jianrong Xia

Ipomoea cairica (L.) Sweet is an invasive plant that cause serious invasion and damage in South China. Phaeocystis globosa is a common harmful algal bloom species on the southeast coast of China. Both species cause great environmental disturbances and serious economic damage to the localregion. This study explored the potential inhibitory effects of I. cairica leaf extracts on P. globosa . The results showed that solitary cells growth was inhibited at extract concentrations higher than 0.25 % (v/v). Although the colony diameter did not change, and the colony number increased rapidly in the first 36 h, we found that cells in the colonies had been damaged using scanning electron microscope and SYTOX-Green staining at 48 h. In addition, the rapid light-response curve of cells treated with extracts decreased, along with down-regulation of photosynthesis-related genes ( psbA, psbD, and rbcL ), suggesting damage to the photosynthetic system. Finally, the activities of antioxidant enzymes including superoxide dismutase, peroxidase, and catalase increased with increasing treatment time, indicating that cells activate antioxidant enzyme defense systems to alleviate the production of reactive oxygen species (ROS). Increased ROS levels disrupt cell membranes, alter cellular ultrastructures, and ultimately lead to cell death. This study not only achieved the reuse of invasive plant resources, but also demonstrated that I. cairica leaf extract has potential value as an algaecide.

The mechanism of a new type of modified clay controlling Phaeocystis globosa growth

Qiu Lixia,Yu Zhiming,Cao Xihua,Ji Hena,Song Xiuxian

Phaeocystis globosa is a harmful algal bloom (HAB) species worldwide. Using modified clay (MC) to control HABs and to mitigate their adverse effects is currently a commonly used method in China. In this paper, the effects of oxidized composite modified clay (OXI-MC) on P. globosa were studied from different perspectives. The results show that the OXI-MC could effectively remove P. globosa and inhibit both the growth of residual algal cells and the formation of new colonies. The P. globosa algal biomass removal efficiencies after 3 h reached 90% at a dose of 0.1 g/L, and the number of colonies with different particle sizes was greatly reduced. Compared with those of the control, the superoxide dismutase (SOD) activity, catalase (CAT) activity, and malondialdehyde (MDA) content of the residual algae significantly increased, indicating that OXI-MC caused oxidative stress in the algal cells. In addition, we evaluated the effects of OXI-MC on the photosynthesis of residual microalgae and found that the maximal photochemical efficiency of photosystem II (PSII) under dark adaptation ( F v / F m ) and actual photochemical efficiency of PSII (ΦPSII) decreased, severely damaging the photosynthesis efficiency, implying that OXI-MC effected the photosynthesis system of P. globosa . The results of this study clarify that OXI-MC could remove the most of algal cells and break up the colonies of P. globosa by collision, flocculation, and releasing active substances, as well as inhibit effectively the growth and colony formation of residual P. globosa by causing oxidative stress, reducing photosynthesis activity, accelerating the degradation of polysaccharides, and inhibiting the formation of colonies.

Inhibition effect of natural flavonoids on red tide alga Phaeocystis globosa and its quantitative structure-activity relationship

Xiao Xi,Li Chao,Huang Haomin,Lee Ying Ping

Red tides that occur off coasts have become a worldwide phenomenon over the past decades. In order to mitigate the damage of the red tides on the aquatic ecosystems, it is crucial to develop a method for predicting algicidal activities that requires less labor and time, and most importantly, this method can quickly screen potential algicides to control red tides. In this study, we have investigated the algicidal activity of 19 natural flavonoids against a typical red tide alga, Phaeocystis globosa. Our results indicate that after 5 days of flavonoid exposure, the half inhibition concentrations (IC 50 ) ranged from 0.068 to 3.065 mg L −1 , which showed the strong algicidal activities of the flavonoids. Furthermore, quantitative structure activity relationship (QSAR) model has been carried out between negative scale logarithm (pIC 50 ) of the flavonoids and the corresponding molecular descriptors. The developed model was validated, both internally and externally, which displayed statistical robustness ( R 2 = 0.867, p = 0.0002, Q 2 LOO = 0.825, RMSEC = 0.182, Q 2 extF3 = 0.896, RMSEP = 0.161, CCC = 0.935). This indicates that the developed model was obtained successfully with satisfactory predictability and robustness for the future rapid screening of natural flavonoids with high inhibition activity on the red tide alga growth. Moreover, the main descriptors in the QSAR model were the molar refractivity, partition coefficient, lowest unoccupied molecular orbital, and highest occupied molecular orbital, illustrating that the molecular electro-chemical characteristics are significant in the algicidal actions of the flavonoids. Red tides frequently occur worldwide and have become a global environment problem. Flavonoids showed great potential in allelopathic control of the excessive growth of red tide algae. In this study, the algicidal activity of 19 natural flavonoids was investigated on a typical red tide organism Phaeocystis globosa . Futhermore, we applied the quantitative structure-activity relationship (QSAR) model to the experimental data. The model between molecular descriptors of flavonoids and their antialgal activity displays statistical robustness, and 4 of 45 selected molecular descriptors were obtained by regression of training set. The numbers in the figure represent the half inhibition concentration (IC 50 ) of flavonoids. Our results show that the algicidal activity of flavonoids is closely related to molar refraction, partition coefficient, lowest unoccupied molecular orbital, and highest occupied molecular orbital. The QSAR model can efficaciously predict the algicidal activity and provide insights into the inhibitory mechanisms of flavonoids.

Efficient removal of Phaeocystis globosa from seawater with the persulfate activation by arbutin-modified cellulose nanocrystals

Mingda Che,Cancan Shan,Wenjie Zhang,Yanyi Duan,Renliang Huang,Mei Cui,Wei Qi,Rongxin Su

Harmful algal blooms (HABs) from seawater have a severe threat to human health, aquaculture, and coastal nuclear power safety. Thus, it is highly desirable to explore environmentally friendly, efficient, and economic methods for controlling HABs. Herein, the arbutin-modified cellulose nanocrystals (AT-CNC) activated persulfate (PS), as a novel heterogeneous Fenton-like process, was proposed to remove Phaeocystis globosa ( P. globosa ) from seawater. The AT-CNC was synthesized via the surface modification of AT on CNC. The effects of AT dosage, CNC dosage, and PS dosage on the removal performance of P. globosa were investigated. With the addition of 530 mg/L AT-CNC (6 wt% AT/CNC of AT loading) and 120 mg/L PS, the removal percentage of chlorophyll a ( R c ), optical density at 680 nm ( R o ) and turbidity ( R t ) reached 97.7%, 91.9% and 85.2% at 24 h. According to electron paramagnetic resonance (EPR) spectra and radical quenching tests, the predominant free radicals inactivating P. globosa were hydroxyl radicals ( • OH). Additionally, the flocculation of the inactivated algae cells by AT-CNC was also critical for removing P. globosa. Moreover, a positive environmental impact was achieved in the AT–CNC–PS system due to the reduction of nitrogen, phosphorus and organic carbon contents. Based on the excellent removal performance for P. globosa , we believe that the AT-CNC activated persulfate is a promising option for HABs control.

Rapid monitoring and early warning of Phaeocystis globosa bloom based on an effective electrochemical biosensor

Tao Zhang,Chaoxin Zhang,Yingzhan Chen,Hongjie Liu,Shaopeng Wang,Yaling Liu,Yayi He,Man Zhang,Liwei Wang

Harmful algal blooms (HABs) have posed a significant threat to human society and the ecological environment. In particular, the outbreak of Phaeocystis globosa ( P. globosa ) bloom could affect coasts nuclear power safety. Unfortunately, current ecological monitoring tools fail to dynamically detect the densities of solitary cells from P. globosa in the pre-outbreak phases, thus affecting early interventions. In the study, an effective electrochemical DNA biosensor was developed to serve the rapid and effective detection of P. globosa DNA through a specific DNA probe strategy. Especially, its good specificity and lower limit of detection (LOD, 17 pg/μL or 1063 cells/L) met the monitoring requirement of solitary-cell population change of P. globosa before the cyst formation (threshold: 1.0 × 10 7 cells/L), which is the key step in the algal bloom outbreaks and influences the outbreak cycle and scale. Furthermore, the accuracy of this electrochemical biosensor for the quantitative detection of solitary-cell P. globosa was confirmed by using the classical microscopic examination techniques (r = 0.981, P < 0.001). Moreover, its applicability was also validated by actual sample testing (r = 0.996, P < 0.001). Therefore, the novel technology offers great potential to improve dynamic detection and early warning of P. globosa bloom.

A rapid removal of Phaeocystis globosa from seawater by peroxymonosulfate enhanced cellulose nanocrystals coagulation

Mingda Che,Cancan Shan,Renliang Huang,Mei Cui,Wei Qi,Jiří Jaromír Klemeš,Rongxin Su

Cellulose nanocrystals (CNC) are recognized as promising bio-based flocculants for controlling harmful algal blooms (HABs). Due to the charge shielding effect in seawater and the strong mobility of algae cells, CNC can’t effectively remove Phaeocystis globosa from seawater. To solve this problem, peroxymonosulfate (PMS) was used to enhance the coagulation of CNC for rapidly removal of P. globosa . The results showed that 91.7% of Chl-a, 95.2% of OD 680 , and 97.2% of turbidity of P. globosa were reduced within 3 h with the use of 200 mg L −1 of CNC and 20 mg L −1 of PMS. The removal of P. globosa was consisted of inactivation and flocculation. Notably, electron paramagnetic resonance (EPR) spectrums and quenching experiments revealed that the inactivation of P. globosa was dominated by PMS oxidation and 1 O 2 . Subsequently, CNC entrained inactivated algal cells to settle to the bottom to achieve efficient removal of P. globosa. The content of total organic carbon (TOC) and chemical oxygen demand (COD) decreased significantly, indicating that a low emission risk of algal cell effluent was produced in the CNC-PMS system. In view of the excellent performance on P. globosa removal, we believe that the CNC-PMS system has great potential for HABs treatments.

Inhibition of harmful algae Phaeocystis globosa and Prorocentrum donghaiense by extracts of coastal invasive plant Spartina alterniflora

Caicai Xu,Zhiwei Ge,Chao Li,Fanghao Wan,Xi Xiao

Harmful algal blooms (HABs) and the invasion of smooth cordgrass ( Spartina alterniflora ) have become important environmental problems in intertidal zones of China, which caused serious damage on the coastal ecological systems. By using S. alterniflora as algaecide, this helps to utilize this invasive plant, in addition, is expected to help to control the expansion of S. alterniflora . The potential of S. alterniflora in HABs mitigation was investigated on controlling Phaeocystis globosa (haptophyceae) and Prorocentrum donghaiense (dinophyceae). The growth of both HABs species was significantly inhibited at high concentrations, and P. globosa was more sensitive than P. donghaiense . Furthermore, the extracts of S. alterniflora reduced the effective quantum yield, photosynthetic efficiency, and relative maximal electron transport rate of both algal species at high concentrations, which implies a disruption on their photosynthetic system. Flavonoids, which were previously known as antialgal chemicals, were found to be abundant in the extracts of S. alterniflora by UPLC-MS detection. Our results revealed that the potential of S. alterniflora as a novel antialgal agent for controlling HABs, simultaneously, resource utilization possibility for the invasive plant S. alterniflora .

Programmed cell death process in freshwater Microcystis aeruginosa and marine Phaeocystis globosa induced by a plant derived allelochemical

Caicai Xu,Shumiao Yu,Jing Hu,Kokoette Effiong,Zhiwei Ge,Tao Tang,Xi Xiao

Harmful algal blooms (HAB) are a serious problem worldwide. Allelochemicals from natural plants were recently thought to be promising anti-algaecide in controlling harmful algae. However, the programmed cell death (PCD) process of algae under allelopathic pressure induced by 5,4′-dihydroxyflavone (5,4′-DHF) was poorly understood. In this study, two common and worldwide distributed microalgae, Microcystis aeruginosa and Phaeocystis globosa were selected as target algae, and the PCD processes induced by 5,4′-DHF were cross-compared between the two species. Both algae species were inhibited significantly by 5,4′-DHF with the relative sensitivity of 0.11. To uncover the PCD progress systematically, signals for PCD triggering, antioxidant enzyme activity, photosynthetic ability variation, caspase-like activities and typical indicators were investigated. In both species, typical indicators of PCD – phosphatidylserine externalization and chromatin condensation – were detected. The intracellular reactive oxygen species (ROS), nitric oxide (NO) and H 2 O 2 were the potential signal molecules to stimulate PCD, and caspase-like activities were activated with an elevation of cytochrome c indicating the initiation of PCD in both species. However, P. globosa responded to 5,4′-DHF immediately after 3 h with the elevation of ROS and not in M. aeruginosa . Antioxidant enzyme activities of superoxide dismutase (SOD) and catalase (CAT) in M. aeruginosa and P. globosa also showed different patterns on day 3. Specifically, SOD activity in M. aeruginosa increased significantly while it decreased significantly in P. globosa , CAT activity in M. aeruginosa decreased significantly while it increased significantly in P. globosa ( p < 0.05). Malondialdehyde (MDA) content in P. globosa increased significantly ( p < 0.001) while it showed no variation in M. aeruginosa . Overall, this study is one of the earliest studies to explore the inhibition and action mechanism of plant derived flavonoids on harmful algae from the perspective of PCD, and provide new insights into the antialgal mechanism of allelochemicals.
