Composting of 4-nonylphenol-contaminated river sediment with inocula of Phanerochaete chrysosporium

Danlian Huang,Xingmeng Qin,Piao Xu,Guangming Zeng,Zhiwei Peng,Rongzhong Wang,Jia Wan,Xiaomin Gong,Wenjing Xue

A composting study was performed to investigate the degradation of 4-nonylphenol (4-NP) in river sediment by inoculating Phanerochaete chrysosporium ( Pc ). Pc was inoculated into composting Reactor A, C and D, while Reactor B without inocula was used as control. The results showed that composting with Pc accelerated the degradation of 4-NP, increased the catalase and polyphenol oxidase enzyme activities in contaminated sediment. The dissipation half-life (t 1/2 ) of 4-NP in Reactor A, C and D with inocula of Pc were 2.079, 2.558, 2.424   days, while in Reactor B without inocula of Pc it was 3.239   days, respectively. Correlation analysis showed that the contents of 4-NP in sediment in Reactor A and D were negatively correlated with the actives of laccase, whereas no obvious correlation was observed in Reactor B and C. All these findings also indicated that Pc enhanced the maturity of compost, and the best composting C/N ratio was 25.46:1.

Biodegradation and detoxification of neonicotinoid insecticide thiamethoxam by white-rot fungus Phanerochaete chrysosporium

Anwei Chen,Wenjie Li,Xiaoxiao Zhang,Cui Shang,Si Luo,Ruoyu Cao,Doudou Jin

The extensive use of neonicotinoid pesticides in the past two decades caused serious impacts on many kinds of living beings. Therefore, it has been strongly suggested to detoxify and eliminate neonicotinoids’ residual levels in environment. Here, the degradation and detoxification of thiamethoxam (THX) by white-rot fungus Phanerochaete chrysosporium was conducted. Results shown that P. chrysosporium can tolerate THX and degraded 49% of THX after incubation for 15 days, and then 98% for 25 days at the initial concentration of 10 mg/L, which indicates the excellent degradation ability of this fungus to THX. Based on the by-products identified, THX underwent dechlorination , nitrate reduction , and C–N cleavage between the 2-chlorothiazole ring and oxadiazine. ( Z )-N-(3-methyl-1,3,5-oxadiazinan-4-ylidene)nitramide and 3-methyl-1,3,5-oxadiazinan-4-imine were identified as the main metabolites. The impacts of THX and its corresponding degradation intermediates on the growth of E. coil and Microcystis aeruginosa as well as the germination of rape and cabbage demonstrated that P . chrysosporium effectively degrades THX into metabolites and reduces its biotoxicity. The present work demonstrates that P . chrysosporium can be effectively used for degradation and detoxification of THX.

Biotransformation of cadmium-sulfamethazine combined pollutant in aqueous environments: Phanerochaete chrysosporium bring cautious optimism

Xueying Guo,Zhiwei Peng,Danlian Huang,Piao Xu,Guangming Zeng,Shuang Zhou,Xiaomin Gong,Min Cheng,Rui Deng,Huan Yi,Hao Luo,Xuelei Yan,Tao Li

Microorganism biotransformation of sulfamethazine (SMT) in aqueous environments is a major concern, especially considering their exposure to coexisting SMT and heavy metals. Phanerochaete chrysosporium ( P. chrysosporium ) is a more concerned Cadmium (Cd) and SMT hyper accumulation specie. This study, referring to metabolic mechanisms and application, was performed to investigate the single and combined effects of Cd-SMT, including toxicity, resistance, as well as the accumulation and biotransformation by P. chrysosporium . The results revealed that Cd-SMT co-contamination caused increasing active oxygen accumulation, and the number of antioxidant enzyme and non-enzymatic antioxidants were higher than that under the stress of their individual pollution. It was found that P. chrysosporium accumulated high levels of Cd with the increment of 6.98–23.96% induced by Cd-SMT co-contamination compared to under the stress of Cd individual pollution. What's more, the addition of Cd reduced the toxicity of SMT to P. chrysosporium . The decrease of malonaldehyde and the increase of protein also proved that P. chrysosporium held enormous potential to fit in the co-contaminated environment, and to remediate the co-contaminated water especially in the long-term treatment. These results undoubtedly contribute to the development of fungi-based technologies and the applications of P. chrysosporium in realistic environment rather than laboratory simulation environment.

Biodegradation, Biosorption of Phenanthrene and Its Trans-Membrane Transport by Massilia sp. WF1 and Phanerochaete chrysosporium

Gu Haiping,Lou Jun,Wang Haizhen,Yang Yu,Wu Laosheng,Wu Jianjun,Xu Jianming

Reducing phenanthrene (PHE) in the environment is critical to ecosystem and human health. Biodegradation, biosorption, and the trans-membrane transport mechanism of PHE by a novel strain, Massilia sp. WF1, and an extensively researched model fungus, Phanerochaete chrysosporium were investigated in aqueous solutions. Results showed that the PHE residual concentration decreased with incubation time and the data fitted well to a first-order kinetic equation, and the t1/2 of PHE degradation by WF1, spores, and mycelial pellets of P. chrysosporium were about 2 h, 87 days, and 87 days, respectively. The biosorbed PHE was higher in P. Chrysosporium than that in WF1, and it increased after microorganisms were inactivated and inhibited, especially in mycelial pellets. The detected intracellular auto-fluorescence of PHE by two-photon excitation microscopy also proved that PHE indeed entered into the cells. Based on regression, the intracellular (Kdin) and extracellular (Kdout) dissipation rate constants of PHE by WF1 were higher than those by spores and mycelial pellets. In addition, the transport rate constant of PHE from outside solution into cells (KinS/Vout) for WF1 were higher than the efflux rate constant of PHE from cells to outside solution (KoutS/Vin), while the opposite phenomena were observed for spores and mycelial pellets. The amount of PHE that transported from outside solution into cells was attributed to the rapid degradation and active PHE efflux in the cells of WF1 and P. Chrysosporium, respectively. Besides, the results under the inhibition treatments of 4°C, and the presence of sodium azide, colchicine, and cytochalasin B demonstrated that a passive trans-membrane transport mechanism was involved in PHE entering into the cells of WF1 and P. Chrysosporium.

Influence of cysteine and bovine serum albumin on silver nanoparticle stability, dissolution, and toxicity to Phanerochaete chrysosporium

Feng Yi,Guiqiu Chen,Guangming Zeng,Zhi Guo,Weiwei Liu,Zhenzhen Huang,Kai He,Liang Hu

The transport and environmental persistence of silver nanoparticles (AgNPs) after exposure to dissolved organic matter (DOM) are influenced by their stability and dissolution. In addition, their toxicity to microorganisms is influenced by the release of silver ions (Ag+). Here, we characterize the stability and dissolution of citrate-stabilized AgNPs, and their toxicity to a white-rot fungus, Phanerochaete chrysosporium (P. chrysosporium), following exposure to either cysteine (CYS) or bovine serum albumin (BSA). The results indicated that both CYS and BSA changed the diameter, zeta potential, and Ag+ dissolution of AgNPs. Bacterial viability and intracellular reactive oxygen species (ROS) levels were investigated to determine the toxicity of AgNPs to P. chrysosporium. In this study, CYS decreased the inhibitory effects of 100 μg L−1 Ag+ and 10 mg L−1 AgNPs in a concentration-dependent manner. At higher concentrations of CYS, the toxicity of Ag+ and AgNPs was reduced dramatically. However, cell viability decreased at higher BSA concentrations (≥50 mg L−1), suggesting a more complex reaction than simply binding to the released Ag+. Addition of 10 mg L−1 AgNPs significantly stimulated ROS production, but the addition of CYS and BSA decreased the ROS level in a concentration-dependent manner. In summary, our results provide useful information in understanding the fate, transformation, and toxicity of citrate-stabilized AgNPs in the natural environment.

Silver ion-enhanced particle-specific cytotoxicity of silver nanoparticles and effect on the production of extracellular secretions of Phanerochaete chrysosporium

Zhenzhen Huang,Piao Xu,Guiqiu Chen,Guangming Zeng,Anwei Chen,Zhongxian Song,Kai He,Lei Yuan,Hui Li,Liang Hu

This study investigated the influence of silver ions (Ag + ) on the cytotoxicity of silver nanoparticles (AgNPs) in Phanerochaete chrysosporium and noted the degree of extracellular secretions in response to the toxicant's stress. Oxalate production was elicited with moderate concentrations of 2,4-dichlorophenol (2,4-DCP) and AgNPs reaching a plateau at 10 mg/L and 10 μM, respectively. Increased oxalate accumulation was accompanied by higher activities of manganese peroxidase (MnP) and lignin peroxidase (LiP). However, the secretion of oxalate, MnP and LiP was significantly inhibited owing to Ag + incorporation into AgNP solution. Production of extracellular polymeric substances (EPS) significantly elevated with an increase in 2,4-DCP concentrations; however, after 24 h of exposure to 100 mg/L 2,4-DCP, an obvious decrease in EPS occurred, indicating that part of EPS could be consumed as carbon and energy sources to ameliorate biological tolerance to toxic stress. Furthermore, AgNP-induced “particle-specific” cytotoxicity was substantially enhanced with additional Ag + as evidenced by its significant negative impact on cellular growth, plasma membrane integrity, and morphological preservation compared with AgNPs at equal Ag concentration.

Physiological fluxes and antioxidative enzymes activities of immobilized Phanerochaete chrysosporium loaded with TiO2 nanoparticles after exposure to toxic pollutants in solution

Qiong Tan,Guiqiu Chen,Guangming Zeng,Anwei Chen,Song Guan,Zhongwu Li,Yanan Zuo,Zhenzhen Huang,Zhi Guo

Immobilized Phanerochaete chrysosporium loaded with TiO 2 nanoparticles (PTNs) are novel high-value bioremediation materials for adsorbing cadmium and for degrading 2,4-dichlorophenol (2,4-DCP). The real-time changes in H + and O 2 fluxes were measured using the noninvasive microtest technique (NMT). The H + influx increased after the addition of 2,4-DCP, and shifted to efflux following the addition of Cd 2+ . The O 2 flux decreased after the addition of both 2,4-DCP and Cd 2+ . A larger Cd 2+ flux was immediately observed after exposure to 0.5   mM Cd 2+ (−351.25   pmol   cm −2   s −1 ) than to 0.1   mM Cd 2+ (−107.47   pmol   cm −2   s −1 ). The removal of Cd 2+ by the PTNs increased more after treatment with the 0.5   mM exposure solution (27.6   mg   g −1 ) than with the 0.1   mM exposure solution (3.49   mg   g −1 ). The enzyme activities were analyzed to review the antioxidative defense system of PTNs in a solution containing various concentrations of Cd 2+ . The activities of the coenzyme nicotinamide adenine dinucleotide (NADH) oxidase as well as the enzyme catalase (CAT) plateaued at 6.5   U   g −1 FW and 9.7   U   g −1 FW, respectively, after exposure to 0.25   mM Cd 2+ . The activity of superoxide dismutase (SOD) increased gradually in solutions containing 0.1–0.6   mM Cd 2+ , and eventually reached a maximum (68.86   U   g −1 FW). These results illustrate how the antioxidative defense system and the physiological fluxes of PTNs respond to the stress caused by toxic pollutants .

Deciphering the biodegradation of thiamethoxam by Phanerochaete chrysosporium with natural siderite: Synergistic mechanisms, transcriptomics characterization, and molecular simulation

Shiye Zhu,Anwei Chen,Jiale Zhang,Si Luo,Jizhao Yang,Youzheng Chai,Jianhua Zeng,Ma Bai,Zhenghang Yang,Gen Lu

Fungi play vital roles in the fate of organic pollutants, particularly when interacting with minerals in aquatic and soil environments. Mechanisms by which fungi may mitigate pollutions in fungus-mineral interactions are still unclear. Inspired by biogeochemical cycling, we constructed a range of co-culture systems to investigate synergistic effects of the white-rot fungus Phanerochaete chrysosporium and the iron-bearing mineral siderite on thiamethoxam (THX) transformation, a common neonicotinoid pesticide. Co-culturing with siderite significantly enhanced THX transformation during the initial 10 days with a dose effect, achieving 86 % removal within 25 days. Fungi could affect siderite’s dissolution, transformation, and precipitation through their biological activities. These interactions triggered physiological adaptation and resilience in fungi. Siderite could enhance the activity of fungal ligninolytic enzymes and cytochrome P450, facilitating biotransformation. Genes expression related to growth, energy metabolism, and oxidative stress response upregulated, enhancing fungal resilience to THX. The primary THX degradation pathways included nitro-reduction, C-N cleavage, and de-chlorination. Molecular dynamics simulations provided insights into catalytic mechanisms of enzyme-THX interactions. Together, siderite could act as natural enhancers that endowed fungi to resist physical and chemical stresses in environments, providing insights into contaminants attenuation, fungal biomineralization, and the coevolution of the Earth's lithosphere and biosphere.

Transformation of Tetracycline by Manganese Peroxidase from Phanerochaete chrysosporium

Xuemei Sun,Yifei Leng,Duanji Wan,Fengyi Chang,Yu Huang,Zhu Li,Wen Xiong,Jun Wang

The negative impacts on the ecosystem of antibiotic residues in the environment have become a global concern. However, little is known about the transformation mechanism of antibiotics by manganese peroxidase (MnP) from microorganisms. This work investigated the transformation characteristics, the antibacterial activity of byproducts, and the degradation mechanism of tetracycline (TC) by purified MnP fromPhanerochaete chrysosporium. The results show that nitrogen-limited and high level of Mn2+medium could obtain favorable MnP activity and inhibit the expression of lignin peroxidase byPhanerochaete chrysosporium. The purified MnP could transform 80% tetracycline in 3 h, and the threshold of reaction activator (H2O2) was about 0.045 mmol L−1. After the 3rd cyclic run, the transformation rate was almost identical at the low initial concentration of TC (77.05–88.47%), while it decreased when the initial concentration was higher (49.36–60.00%). The antimicrobial potency of the TC transformation products by MnP decreased throughout reaction time. We identified seven possible degradation products and then proposed a potential TC transformation pathway, which included demethylation, oxidation of the dimethyl amino, decarbonylation, hydroxylation, and oxidative dehydrogenation. These findings provide a novel comprehension of the role of MnP on the fate of antibiotics in nature and may develop a potential technology for tetracycline removal. Keywords:tetracycline;manganese peroxidase;antibacterial potency;transformation products;transformation mechanism

Phanerochaete chrysosporium-driven quinone redox cycling promotes degradation of imidacloprid

Qingru Xie,Chao Xue,Anwei Chen,Cui Shang,Si Luo

This study systematically investigated the advanced oxidation processes driven by the white-rot fungus (WRF) Phanerochaete chrysosporium in the removal of the neonicotinoid pesticide imidacloprid (IMI). The quinone redox cycling driven by P. chrysosporium promotes the degradation of IMI in presence of Mn 2+ and Fe (III)-oxalate. Results showed that the degradation efficiency varied with the type of quinine mediator, and gallic acid (GA) was a more effective mediator than 1,4-benzoquinone (BQ). The removal rate of IMI was increased by 1.8-fold in the presence of 100 μM of Mn 2+ compared with that of the system without Mn 2+ . The addition of oxalic acid and EDTA promoted the degradation of IMI, and the effect of oxalic acid was more pronounced at the later stage (day 5 and day 6); the optimal Fe 3+ /oxalate ratio was 1:3. Time-course experiments revealed the maximum removal rates of IMI were 97.37%, 82.0% and 73.52% at the initial IMI concentration of 10, 20, and 30 mg/L, respectively, in the presence of 100 μM Mn 2+ , 100 μM Fe 3+ -300 μM of oxalate and 500 μM gallic acid. Under quinone redox cycling conditions, the good correlation between ·OH production rates and the enzymes activity was concluded, the quinines redox cycles promote enzyme activity, as well as the rate of ·OH production. Results from this study indicated the GA mediator-based system using P. chrysosporium offers a new method to advance the bio-oxidation process owing to its simple culture system, low-cost precursors, unique fungi, and degradation stability.

Extracellular enzyme mediated biotransformation of imidacloprid by white-rot fungus Phanerochaete chrysosporium: Mechanisms, pathways, and toxicity

Shiye Zhu,Anwei Chen,Youzheng Chai,Ruoyu Cao,Jianhua Zeng,Ma Bai,Liang Peng,Jihai Shao,Xiang Wang

As one of the widely used neonicotinoid insecticide, imidacloprid (IMI) poses serious threat on many kinds of non-target species. The acceleration of its removal from the contaminated environment is therefore an urgent task. Herein, the transformation of IMI by white-rot fungus Phanerochaete chrysosporium was examined, and the reaction mechanism underlying enzymes-mediated IMI biotransformation was elucidated. In addition, transformed products, pathways, and toxicity to Escherichia coli and Microcystis aeruginosa or the growth of rice seeds were investigated. P. chrysosporium could efficiently transform 93.5% of IMI for 6 days primarily through biotransformation, in which extracellular manganese peroxidase and lignin peroxidase substantially responded to transform IMI. IMI mainly underwent nitrate reduction , hydroxylation, and de-chlorination. A less neurologically toxic major metabolite of IMI, 6-chloronicotinic acid, was detected. Molecular dynamics simulation demonstrated that IMI stably bonded to active pocket of extracellular enzymes. This study reveals the key metabolic mechanism and fate of IMI in environment, and highlights the potential of fungi for IMI transformation.

Biodegradation of bisphenol A by alginate immobilized Phanerochaete chrysosporium beads: Continuous cyclic treatment and degradation pathway analysis

Jinqiong Wang,Yi Xie,Jie Hou,Xinyu Zhou,Jing Chen,Changhong Yao,Yongkui Zhang,Yonghong Li

The wide application of biodegradation technology makes it particularly significant to use immobilized microorganisms to achieve continuous degradation of pollutants. In this study, the performance of Ca-alginate (CA) immobilized Phanerochaete chrysosporium beads in a consecutive biocycle treatment of bisphenol A (BPA) effluent was researched. The effects of initial BPA concentration, biomass dosage, pH and curing time were investigated. Results showed that CA-immobilized P. chrysosporium can be successive cycle at least 9 times (12 h each), and each time the removal efficiency of BPA reached 100%. The degradation process of BPA in the treatment process was investigated. According to the analyses of glucose, ammonia nitrogen, total nitrogen, extracellular proteins and biomass, it was found that consumed carbon and nitrogen sources were used for the rapid growth of fungal hyphae and the synthesis of a series of proteins. These extracellular proteins can be utilized as a nitrogen source by P. chrysosporium , and played a key role in the removal of BPA. Biodegradation process followed the pseudo-first-order kinetics. Moreover, intermediates in the BPA degradation process were identified by gas chromatography-mass spectrometry (GC–MS) and liquid chromatography-mass spectrometry (LC-MS), and three possible degradation pathways were proposed. This strategy presented in the study provides a convenient and efficient biological method for the continuous treatment of BPA effluent.
