The aim of the current study was to compare the antitumor efficiency of Phellinus igniarius polysaccharides (PIP) from six different origins and preliminarily investigate its potential mechanisms. PIP was extracted using the microwave extraction method. The corresponding in vivo antitumor efficacy was assessed in Kunming mice bearing H22 tumors and Gansu PIP (GPIP) was identified to have a significantly higher antitumor efficacy compared with the control group (P<0.05), while no significant difference was observed following treatment with PIP from different origins (P>0.05). The spleen index of the GPIP group significantly increased compared with the saline and CTX groups (P<0.01). The in vitro MTT assay of GPIP on HepG2 cells indicated that GPIP had no direct cytotoxicity. The serum immune cytokines of interleukin‑2, interleukin‑12 and interferon‑γ were assessed using the ELISA method. The concentration of all three serum cytokines significantly increased compared with saline and CTX groups (P<0.01) indicating that activating the immune system may be a potential antitumor mechanism. These results demonstrated that GPIP has great potential as a natural antitumor agent with immunomodulatory activity.

Gut microbiota dysbiosis has been reported as a risk factor in the development of type 2 diabetes mellitus (T2DM). Polysaccharides fromPhellinus igniarius(P. igniarius) possess various properties that help manage metabolic diseases; however, their underlying mechanism of action remains unclear. Therefore, in this study, we aimed to evaluate the effect ofP. igniariuspolysaccharides (SH-P) on improving hyperglycemia in mice with T2DM and clarified its association with the modulation of gut microbiota and their metabolites using 16S rDNA sequencing and liquid chromatography–mass spectrometry. Fecal microbiota transplantation (FMT) was used to verify the therapeutic effects of microbial remodeling. SH-P supplementation alleviated hyperglycemia symptoms in T2DM mice, ameliorated gut dysbiosis, and significantly increased the abundance ofLactobacillusin the gut. Pathway enrichment analysis indicated that SH-P treatment altered metabolic pathways associated with the occurrence and development of diabetes. Spearman’s correlation analysis revealed that changes in the dominant bacterial genera were significantly correlated with metabolite levels closely associated with hyperglycemia. Additionally, FMT significantly improved insulin sensitivity and antioxidative capacity and reduced inflammation and tissue injuries, indicating improved glucose homeostasis. These results indicate that the ameliorative effects of SH-P on hyperglycemia are associated with the modulation of gut microbiota composition and its metabolites. Keywords:Phellinus igniarius;polysaccharide;hypoglycemia;gut microbiota;metabolites;fetal microbiota transplantation

Phellinus igniarius is a well-known edible and medicinal fungus consumed in Asia for centuries. In the present study, a polysaccharide named PIP-1 was obtained from Phellinus igniarius mycelia, its physicochemical properties were determined, its detailed structures were elucidated by analysis of its depolymerized product, and its antioxidant and antitumor activities were evaluated. Results showed that PIP-1 had a high molecular weight, a high intrinsic viscosity and a linear repeating backbone composed of glucopyranose (Glc p ), galactopyranose, and mannopyranose joined by α-(1 → 4), α-(1 → 3), and α-(1 → 6) linkages, and single α‑terminal‑D‑Glc p as side chains 6- O -linked to the main chain according to the GC-MS and NMR results, and exhibited triple helical structure. Pharmacological results revealed that PIP-1 could effectively scavenge hydroxyl radicals, partly scavenge DPPH radials and chelate ferrous metal ions. Furthermore, PIP-1 showed potent inhibitory effects on growth of HT-29 and MCF-7 cells. These findings suggested that high-molecular-weight PIP-1 containing triple helical structure and only α-type glycosidic bond holds promise as novel functional foods and agents to promote human health.

A novel Cu nanoparticles/reduced graphene oxide–chitosan (CuNPs/r-GO–chitosan) composite film modified glassy carbon electrode (GCE) was fabricated by dispersing CuNPs uniformly on a stable r-GO–chitosan thin film through electrodeposition process. The modified electrode was characterized by cyclic voltammetry , scanning electron microscopy (SEM) and electrochemical impedance spectroscopy (EIS), and exhibited efficiently electrocatalytic oxidation toward monosaccharides with high stability. The good electrocatalytic activity of this modified electrode might be attributed to the synergistic effect of r-GO and CuNPs, and the stability might be attributed to the r-GO and chitosan thin matrix film. When the CuNPs/r-GO–chitosan/GCE was used as an electrochemical sensor in high performance anion exchange chromatography-direct current amperometric detection (HPAEC-DC) flowing system for the determination of monosaccharides under constant working potential of +0.55 V, the detection limits ( S / N =3) ranged from 0.006 to 0.02 mg L −1 for the analyzed sugars, and the dynamic linear ranges spanned from 0.02 to 500 mg L −1 . The proposed method has been applied for the determination of monosaccharide composition of crude polysaccharides from phellinus igniarius real samples, and the results were satisfactory.

In many clinical studies, prebiotics have been used as adjuvant therapy for inflammatory bowel disease (IBD). Phellinus igniarius polysaccharide (PIP) possesses great anti-inflammatory and prebiotic activities. Herein, we developed an orally deliverable PIP-loaded chitosan-modified PLGA nanomedicine (CS-PIPP) to investigate its anti-inflammatory effect in vitro and in vivo. Dextran sodium sulfate (DSS)-induced colitis model was established to evaluate the preventive effect of CS-PIPP on IBD. This study characterized that CS-PIPP had a size of 288.7 ± 5.49 nm, positive zeta potential, and showed good stability over four weeks. The in-vitro study suggested that CS-PIPP had enhanced phagocytosis by macrophages, which could further significantly inhibit M1-like macrophages phenotype and regulate lipopolysaccharide (LPS)-induced inflammatory cytokines. The in-vivo study revealed that CS-PIPP prominently prevented intestinal inflammatory damage and protected the integrity of the intestinal barrier. Moreover, CS-PIPP increased the contents of short-chain fatty acids (SCFAs) and positively regulated the gut microbiota. Specifically, CS-PIPP reduced enteropathogenic microorganisms while increasing the beneficial microbiota, including Lactobacillus and Akkermansia , which revealed the potential of CS-PIPP as prebiotics. Generally, CS-PIPP promoted the anti-inflammatory effect of PIP, so it could be regarded as a novel and potent nanoformulation to treat IBD.

Phellinus igniarius is an important medicinal and edible fungus with diverse biological activities. This study aimed to investigate the effects of aqueous extract from P. igniarius (API) on the treatment of hyperuricemia (HUA) and related kidney damage. The chemical constituents of API were determined. The therapeutic effects of API on HUA and renal injury were assessed in adenine/potassium oxonate (PO)-treated mice. The constituent analysis of API revealed a predominance of polysaccharides (33.4 %), followed by total flavonoids (9.1 %), and total triterpenoids (3.5 %). Compared to control, the adenine/PO treatment greatly elevated serum uric acid (UA) levels but this elevation was attenuated by API. In the liver, the expression and activity of xanthine oxidase (XOD) were increased by HUA which were diminished by API. Furthermore, API was found to enhance the expression of UA transporter ABCG2 in the kidney and intestine of HUA mice, suggesting elevating UA excretion. Additionally, API ameliorated HUA-induced renal injury, as indicated by reduced serum BUN/creatinine levels, decreased glomerular and tubular damage, and lowered fibrotic levels. Network pharmacology analysis predicted that P. igniarius may regulate mitochondrial function to improve HUA-related renal injury. This prediction was then substantialized by the API-induced upregulation of NAD+/NADH ratio, ATP level, SOD2 activity, and expression of SOD2/PCG-1α/PPARγ in the kidney of HUA mice. Our results demonstrate that API may effectively ameliorate HUA by reducing UA production in the liver and enhancing UA excretion in the kidney and intestine, and it might be a potential therapy to HUA-related renal injury.

Bioactivity screening revealed that the EtOAc extract from the culture broth of Phellinus igniarius SY489 exhibited remarkable α -glucosidase inhibitory activity, with an IC 50 value of 1.92 μg/mL. Activity- and ultraviolet (UV) profile-guided isolation led to the discovery of four anti-diabetic styrylpyrones ( 1 – 4 ), including two novel compounds, phelignidins A ( 1 ) and B ( 2 ). Compounds 1 and 2 represent a rare structural type of styrylpyrone dimer, in which one of the pyrone moieties exists in an open-ring state. The absolute configurations of the new compounds 1 and 2 , as well as the previously unresolved compound 3 , were established. Compounds 1 – 4 were effective in α -glucosidase inhibition, anti-glycation, and antioxidant assays, surpassing or being comparable to the positive control drugs, with minimal cytotoxicity. Furthermore, studies on α -glucosidase inhibition mechanisms suggested that these compounds interact with α -glucosidase at a single binding site, causing secondary structure unfolding and exerting inhibitory activity via a mixed-type mechanism. These results provide an important basis for developing novel, low-toxicity, multi-target anti-diabetic drugs from edible and medicinal fungi.

Background: Phellinus igniarius (P. igniarius) is an important medicinal and edible fungus in China and other Southeast Asian countries and has diverse biological activities. This study was performed to comparatively investigate the therapeutic effects of wild and cultivated P. igniarius on hyperuricaemia and gouty arthritis in rat models. Methods: UPLC-ESI-qTOF-MS was used to identify the chemical constituents of polyphenols from wild P. igniarius (WPP) and cultivated P. igniarius (CPP). Furthermore, WPP and CPP were evaluated in an improved hyperuricaemia rat model induced by yeast extract, adenine and potassium oxonate, which was used to examine xanthine oxidase (XO) activity inhibition and anti-hyperuricemia activity. WPP and CPP therapies for acute gouty arthritis were also investigated in a monosodium urate (MSU)-induced ankle swelling model. UHPLC-QE-MS was used to explore the underlying metabolic mechanisms of P. igniarius in the treatment of gout. Results: The main active components of WPP and CPP included protocatechuic aldehyde, hispidin, davallialactone, phelligridimer A, hypholomine B and inoscavin A as identified by UPLC-ESI-qTOF-MS. Wild P. igniarius and cultivated P. igniarius showed similar activities in reducing uric acid levels through inhibiting XO activity and down-regulating the levels of UA, Cr and UN, and they had anti-inflammatory activities through down-regulating the secretions of ICAM-1, IL-1β and IL-6 in the hyperuricaemia rat model. The pathological progression of kidney damage was also reversed. The polyphenols from wild and cultivated P. igniarius also showed significant anti-inflammatory activity by suppressing the expression of ICAM-1, IL-1β and IL-6 and by reducing the ankle joint swelling degree in an MSU-induced acute gouty arthritis rat model. The results of metabolic pathway enrichment indicated that the anti-hyperuricemia effect of WPP was mainly related to the metabolic pathways of valine, leucine and isoleucine biosynthesis and histidine metabolism. Additionally, the anti-hyperuricemia effect of CPP was mainly related to nicotinate and nicotinamide metabolism and beta-alanine metabolism. Conclusions: Wild P. igniarius and cultivated P. igniarius both significantly affected the treatment of hyperuricaemia and acute gouty arthritis models in vivo and therefore may be used as potential active agents for the treatment of hyperuricaemia and acute gouty arthritis.

Phellinus igniariusis a medicinal fungus rich in terpenoids with potential biological activities. In this study, theP. igniariusterpenoid-enriched extract (PITT) was optimized using single-factor experiments combined with response surface methodology based on a Box–Behnken design. The optimal extraction conditions were determined as extraction temperature 64 °C, extraction time 21 min, ethanol concentration 78%, and liquid-to-solid ratio 20 mL/g, yielding a total terpenoid content of 7.078 mg/g. The chemical profile of PITT was characterized by liquid chromatography–tandem mass spectrometry (LC–MS/MS), revealing the presence of multiple putative terpenoid compounds, including betulin, curdione, and several sesquiterpenoids. The antioxidant activity of PITT was evaluated using DPPH, ABTS, and hydroxyl radical scavenging assays, showing dose-dependent effects. Furthermore, an H2O2-induced oxidative stress model in RAW 264.7 cells was employed to assess cellular antioxidant activity. Pretreatment with PITT significantly reduced intracellular reactive oxygen species (ROS) levels, enhanced the activities of endogenous antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase), and decreased malondialdehyde (MDA) levels. These results indicate that PITT exhibits notable antioxidant activity and may serve as a potential natural antioxidant source for further development. However, further studies are required to elucidate the underlying mechanisms.

Introduction Although Phellinus igniarius has been shown to treat hyperuricemia (HUA) and gout, a self-inflammatory disease caused by purine metabolism disorders, the underlying mechanisms remain unclear. Polysaccharides are among the main components of P. igniarius with anti-inflammatory and immunoregulatory properties. Methods Here, we examined the therapeutic effects of polysaccharide SH-P-1-1 isolated from P. igniarius on HUA and gout, and explored the underlying mechanism, focusing on gut microbiota and metabolite regulation. Results SH-P-1-1 significantly decreased uric acid and creatinine levels, reduced xanthine oxidase and adenosine deaminase activities, alleviated kidney damage, and reduced urate deposition in joints in model rats. It increased the abundance of Blautia and Muribaculaceae but reduced that of the Lachnospiraceae_NK4A136_group , Lactobacillus , and Turicibacter . SH-P-1-1 also significantly modulated the metabolic profile and was closely related to some metabolic pathways, such as tryptophan metabolism, relevant to HUA and gout. The beneficial activities of SH-P-1-1 correlated with gut microbiota composition and differential metabolites, including short-chain fatty acids. Discussion Overall, this study demonstrates the potential of SH-P-1-1 as a natural supplement for preventing and managing HUA and gout.

To achieve the conversion of waste Phellinus igniarius substrate into functional foods, we employed enzymatic hydrolysis-centrifugal fractionation method to extract highly uniform spherical cellulose nanocrystals (CNCs, 14–28 nm) from the waste Phellinus igniarius substrate. These CNCs were used to construct an emulsion delivery system for carotenoids and polyphenols. We utilized biphasic fluorescence staining and Cryo-SEM to visually confirm the adsorption of CNCs at the interface. Furthermore, interfacial shear rheology was applied to perform time-, frequency-, and strain-dependent analyses. These methods provided direct, dynamic, and quantitative evidence that the CNCs rapidly adsorb at the oil-water interface and form a solid-like film dominated by elastic behavior (G′ > G″). This conclusively demonstrates that the emulsification mechanism follows a Pickering-type stabilization. Stabilized emulsions exhibit excellent storage and environmental stability, effectively delaying the degradation of bioactive compounds while preserving their antioxidant activity. In vitro simulated digestion experiments demonstrated that the Pickering mechanism of the CNC-stabilized emulsion offers the advantage of retarding lipid digestion. The bioaccessibility of the four encapsulated bioactive compounds, namely β-carotene, lutein, zeaxanthin, and dihydroquercetin, reached 46.19 ± 1.20 %, 44.66 ± 0.56 %, 40.72 ± 1.17 %, and 38.97 ± 1.16 %, respectively. This study successfully established a bioactive compound delivery system, enabling the conversion of waste Phellinus igniarius substrate into functional food.
