Physalis alkekengi L. is one of the most popular berries in the world, and its calyx is rich in nutrients. Still, the study of soluble dietary fiber (SDF) from Physalis alkekengi L. calyx (PC) has not been reported. This study aimed to extract Physalis alkekengi L. calyx soluble dietary fiber (PCSDF), and the optimal extraction process was investigated using response surface methodology. After ultrasonic pretreatment, the yield of PCSDF reached 8.74 ± 0.17% under the conditions of liquid-solid ratio of 30 mL/g, thermostable α-amylase concentration of 2000 U/mL, dispase concentration of 20,000 U/mL, amyloglucosidase concentration of 40,000 U/mL. For the first time, PCSDF was characterized by UV spectroscopy, infrared spectroscopy, scanning electron microscopy and thermogravimetric analysis, and the composition of the monosaccharides, namely mannose, glucuronic acid, rhamnose, galacturonic acid, glucose, galactose, xylose, arabinose and fucose, was identified by HPLC. For antioxidant activity, the half inhibitory concentration (IC 50 ) against DPPH, ABTS + , ·OH, and ·O 2 − radicals were 3.67, 2.96, 5.45, and 2.90 mg/mL, respectively, and there was a specific Fe 3+ reducing capacity. In terms of hypoglycemic feature, the IC 50 values for the inhibitory ability of α-glucosidase and α-amylase were 1.20 and 9.21 mg/mL, respectively. For lipid-lowering potential, the IC 50 value for pancreatic lipase inhibitory ability for lipid-lowering was 16.95 mg/mL. This paper fills the research gap of PCSDF and provides a basis for further evaluation of PCSDF using in vivo studies. Graphical Abstract

In the present study, Physalis alkekengi L. calyx total flavonoids (PCTF) were extracted using the ultrasound-assisted ethanol method and separated and purified using macroporous resin AB-8. Physalis alkekengi L. calyx total flavonoid-chitosan (PCTF-CS) composite films containing 0.05 %, 0.10 % and 0.15 % PCTF were prepared using the purified PCTF laminated with chitosan (CS) and compared with single CS films, respectively, to investigate their mechanical properties, barrier properties, optical properties, microstructure, crystallography, thermal stability, water contact angle, particle size and zeta potential, antioxidant property, antimicrobial property, and preservation effect on chilled beef. The PCTF-CS films with PCTF additions had darker colors and higher mechanical and barrier properties than the CS films. In addition, the addition of PCTF improved the antioxidant and antimicrobial properties of the CS films. It enhances the freshness retention of fresh beef, and effectively inhibits the rise of weight loss, pH, total bacteria, total volatile basic nitrogen, and thiobarbituric acid reactive substances in beef, prolonging the shelf life of beef. These results indicate that the addition of PCTF can provide CS films with superior functional properties and bioactivities and that PCTF-CS composite films are a potential and promising packaging material for food preservation.

The design of electrode materials for sensitive electrochemical sensors is important for the rapid detection of ascorbic acid. Using Physalis alkekengi L. husks (PaLH) as the carbon precursor, PaLH-derived carbon materials (PCM) were prepared by high temperature carbonization, and hierarchical porous carbon materials (KPCM) were prepared by KOH activation. The carbon materials were characterized by scanning electron microscopy, X-ray diffraction, and isothermal nitrogen adsorption and desorption. KPCM had a larger specific surface area (1345.45 m 2 g −1 ) and more micro/mesopores than PCM without KOH activation. Two types of carbon materials, PCM and KPCM, were used as electrode modifiers, and the electrochemical properties of KPCM/GCE and PCM/GCE (GCE = glassy carbon electrode) were investigated. KPCM/GCE had a higher electron-transfer rate, higher electrochemically active area, smaller charge-transfer resistance, wider potential window, and higher catalytic activity for ascorbic acid detection. An ascorbic acid electrochemical sensing platform constructed with KPCM/GCE showed high sensitivity (334.03 μA mM −1 cm −2 and 134.52 μA mM −1 cm −2 ) and a low detection limit (0.92 μM) ( S/N = 3) in the linear range of 0.005–1.6 mM. This method was applied to the analysis and determination of ascorbic acid in chewable vitamin C tablets.

Ethnopharmacological relevance Physalis alkekengi L. ( P. alkekengi ) is widely used in traditional medicine and functional foods for its various medicinal properties. However, its toxicological profile has not been comprehensively assessed. Aim of the study This study aimed to characterize the developmental toxicity and cardiotoxicity of P. alkekengi using a Danio rerio ( D. rerio ) embryo model and to identify key toxic constituents to support safety optimization. Materials and methods D. rerio embryos were exposed to the fruit and calyx extracts of P. alkekengi as well as membrane-fractionated preparations to assess toxic phenotypes. Chemical profiling was performed using ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry (UPLC-Q-Exactive Orbitrap-MS), and mechanistic insights were explored using apoptosis-related assays and network toxicology analysis. Results The fruit extract of P. alkekengi exhibited stronger developmental toxicity and cardiotoxicity effects compared to the calyx extract. The 5–10 kDa non-polysaccharide fraction of the fruit was identified as the primary toxic source, with Glycidyl stearate (GS) being the key cardiotoxic component in this fraction. Removal of the most toxic 5–10 kDa fraction during the extraction process significantly reduced the toxicity of the fruit extract. Conclusion The toxicity of P. alkekengi fruit extract is concentrated in the non-polysaccharide 5–10 kDa fraction, with GS being the main contributor to the toxicity in this fraction. Membrane-based separation technology to remove this fraction can enhance the safety of the extract. This study provides a framework for the safety evaluation and optimization of traditional medicinal plants.
