Mosses play a crucial role in environmental protection, ecological preservation, and horticulture. While the effects of nanomaterials on angiosperms have been widely studied, their impact on bryophytes remains underexplored. In this study, we investigated the effects of mesoporous silica nanoparticles (MSNs) and virus-like mesoporous silica nanoparticles (VMSNs) on the model moss species Physcomitrium patens ( P. patens ). Our results revealed that MSNs, with an average size of approximately 123 nm, are nontoxic to P. patens and enhance its salt tolerance. The expression of key genes involved in stress responses were significantly induced in MSN-treated plants under salt stress, including peroxidase ( POX ), L-ascorbate oxidase ( L-AO ), alternative oxidase ( AOX ), and calcium-dependent protein kinase ( CPK ). MSN treatment reduced the accumulation of H 2 O 2 and O 2 ·− , increased Ca 2+ signaling, and modulated reactive oxygen species (ROS) homeostasis, collectively improving moss tolerance to salt stress. MSNs were observed on the cell surface, in intercellular space, and within the cytosol and vesicles. They were transported bidirectionally between rhizoids and apical leaves. This study provides novel insights into the distribution, transport, and functional mechanisms of MSNs in mosses, offering a valuable foundation for the application of nanomaterials in plant stress biology and ecological management of bryophytes.

Major physiological and developmental innovations are key to land-plant adaptation and evolution, but their genetic basis remains to be fully understood. Here, we show that lipocalins in the moss Physcomitrium patens regulate various physiological and developmental processes. Phylogenetics analyses reveal three major groups of plant lipocalins, including a newly identified group that is restricted to seedless plants. We demonstrate that the temperature-induced lipocalin gene in P. patens ( PpTIL ) is functionally conserved with flowering plant homologs in response to abiotic stresses. PpTIL not only regulates protonemal development and the transition from two-dimensional to three-dimensional growth but also affects many other processes, such as lipid transport and metabolism, auxin biosynthesis and transport, and chlorophyll catabolism. Further, PpTIL also operates antagonistically with SAFEGUARD1 in protecting chloroplast grana and envelopes from singlet oxygen stress. These findings provide major insights into the role of lipocalins in land-plant evolution.

Cytochrome P450 monooxygenase 98 (CYP98) is a critical rate-limiting enzyme of the phenylpropanoid pathway. One of the end-product of the phenylpropanoid pathway is a lignin monomer, although the occurrence of lignin in bryophytes is controversial. Here we investigated the functions of PpCYP98 in Physcomitrium patens by transcriptome and metabolome analyses. We identified 5266 differentially expressed genes (DEGs) and 68 differentially abundant secondary metabolites between wild-type and ΔPpCYP98 gametophores . Of the identified metabolites, 23 phenolic acids were identified, with only one showing upregulation. Among the phenolic acids, 4-coumaroyl tartaric acid and chlorogenic acid showed significant decreases. Declines were also observed in coniferylaldehyde and coniferin , precursor substances and downstream products of the lignin monomer coniferyl alcohol , respectively. Thus, the pre-lignin synthesis pathway already exists in bryophytes, and PpCYP98 plays vital roles in this pathway. Besides, most flavonoids show significant reductions, including eriodyctiol, dihydroquecetin, and dihydromyricetin, whereas naringenin chalone and dihydrokaempferol were increased after PpCYP98 knockout. Therefore, the synthesis of flavonoids shares the core pathway with phenylpropanoids and mainly starts from caffeoyl-CoA, that is the compound of divergence between the two pathways in moss. PpCYP98 showed systemic effects on metabolisms, including carbohydrate, fatty acid , and hormonal signaling transductions, suggesting that PpCYP9 8 might indirectly regulate carbon influx allocation. Our results demonstrated roles of PpCYP98 were essential for the development of the early landing plant.
