Phytophthora infestans RXLR effector Pi23014 targets host RNA-binding protein NbRBP3a to suppress plant immunity

Wanyue Li,Zeming Liu,Yuli Huang,Jie Zheng,Yang Yang,Yimeng Cao,Liwen Ding,Yuling Meng,Weixing Shan

Phytophthora infestans is a destructive oomycete that causes the late blight of potato and tomato worldwide. It secretes numerous small proteins called effectors in order to manipulate host cell components and suppress plant immunity. Identifying the targets of these effectors is crucial for understanding P. infestans pathogenesis and host plant immunity. In this study, we show that the virulence RXLR effector Pi23014 of P. infestans targets the host nucleus and chloroplasts. By using a liquid chromatogrpahy-tandem mass spectrometry assay and co-immunoprecipitation assasys, we show that it interacts with NbRBP3a, a putative glycine-rich RNA-binding protein. We confirmed the co-localization of Pi23014 and NbRBP3a within the nucleus, by using bimolecular fluorescence complementation. Reverse transcription-quantitative PCR assays showed that the expression of NbRBP3a was induced in Nicotiana benthamiana during P. infestans infection and the expression of marker genes for multiple defence pathways were significantly down-regulated in NbRBP3 -silenced plants compared with GFP -silenced plants. Agrobacterium tumefaciens -mediated transient overexpression of NbRBP3a significantly enhanced plant resistance to P. infestans . Mutations in the N-terminus RNA recognition motif (RRM) of NbRBP3a abolished its interaction with Pi23014 and eliminated its capability to enhance plant resistance to leaf colonization by P. infestans . We further showed that silencing NbRBP3 reduced photosystem II activity, reduced host photosynthetic efficiency, attenuated Pi23014-mediated suppression of cell death triggered by P. infestans pathogen-associated molecular pattern elicitor INF1 , and suppressed plant immunity.

Biocontrol Mechanisms of Three Plant Essential Oils Against Phytophthora infestans Causing Potato Late Blight

Yongqiang Tian,Jianglai Wang,Qingqing Lan,Yang Liu,Jinfeng Zhang,Lu Liu,Xu Su,Rehmat Islam

Late blight, caused by the notorious pathogen Phytophthora infestans , poses a significant threat to potato ( Solanum tuberosum ) crops worldwide, impacting their quality as well as yield. Here, we aimed to investigate the potential use of cinnamaldehyde, carvacrol, and eugenol as control agents against P. infestans and to elucidate their underlying mechanisms of action. To determine the pathogen-inhibiting concentrations of these three plant essential oils (PEOs), a comprehensive evaluation of their effects using gradient dilution, mycelial growth rate, and spore germination methods was carried out. Cinnamaldehyde, carvacrol, and eugenol were capable of significantly inhibiting P. infestans by hindering its mycelial radial growth, zoospore release, and sporangium germination; the median effective inhibitory concentration of the three PEOs was 23.87, 8.66, and 89.65 μl/liter, respectively. Scanning electron microscopy revealed that PEOs caused the irreversible deformation of P. infestans , resulting in hyphal shrinkage, distortion, and breakage. Moreover, propidium iodide staining and extracellular conductivity measurements demonstrated that all three PEOs significantly impaired the integrity and permeability of the pathogen's cell membrane in a time- and dose-dependent manner. In vivo experiments confirmed the dose-dependent efficacy of PEOs in reducing the lesion diameter of potato late blight. Altogether, these findings provide valuable insight into the antifungal mechanisms of PEOs vis-à-vis late blight-causing P. infestans . By utilizing the inherent capabilities of these natural compounds, we could effectively limit the harmful impacts of late blight on potato crops, thereby enhancing agricultural practices and ensuring the resilience of global potato food production.

Inducible Sesquiterpene Synthase and a Phenylpropanoid Phytoalexin of Eggplants (Solanum melongena) Synergistically Promote Nonhost Resistance against Phytophthora infestans

Yantao Wang,Na Li,Caiyan Ma,Yanling Cai,Lei Pi,Sheng Yu,Zhiqing Wang,Suomeng Dong,Sanwen Huang,Shaoqun Zhou

Late blight caused by Phytophthora infestans is a devastating disease on potatoes (Solanum tuberosum) and tomatoes (Solanum lycopersicum). By contrast, eggplants (Solanum melongena) are a nonhost species of late blight, though the mechanisms underlying their resistance remain elusive. In this study, comparative transcriptomics analyses revealed that a sesquiterpene synthase-encoding SmTPS28 could enhance potato resistance against late blight when expressed as a transgene. Further targeted metabolomics analyses showed that syringaldehyde, a phenylpropanoid phytoalexin in eggplant leaves, could induce immune-related gene expression and protect potatoes from late blight when exogenously applied prior to pathogen exposure. Intriguingly, syringaldehyde exhibited a greater protective efficiency on SmTPS28-overexpressing potato plants than on their wild-type progenitors, suggesting functional synergism between these two components of nonhost resistance. These findings reveal novel perspectives of the nonhost resistance mechanisms of eggplants against late blight, and could facilitate the development of new tools and technologies for sustainable late blight management.

Nematophin, an Antimicrobial Dipeptide Compound From Xenorhabdus nematophila YL001 as a Potent Biopesticide for Rhizoctonia solani Control

Zhang Shujing,Liu Qi,Han Yunfei,Han Jinghua,Yan Zhiqiang,Wang Yonghong,Zhang Xing

This study was conducted to purify and identify metabolites of antimicrobial activity against phytopathogens from Xenorhabdus nematophila YL001. Three dipeptide compounds were purified from its cell-free cultural broth and identified as (±)-nematophin, cyclo (L-Pro-Gly), and N, N′-dimethyl-cyclo (L-Phe-L-Leu). Nematophin demonstrated a wider antifungal spectrum than the other two compounds. It also exhibited strong inhibitory effects on mycelial growth of Rhizoctonia solani and Phytophthora infestans with EC50 values of 40.00 and 51.25 μg/ml, respectively. Its (S)-configuration structure [(+)-nematophin] was also synthesized and exhibited higher antimicrobial activity than the enantiomeric mixture. The detached leaf assay revealed that nematophin possessed significant preventive and curative efficacy against R. solani on broad bean leaves showing corresponding control efficacies of 93.01 and 94.93% at 1,000 μg/ml, comparable to those of a chemical fungicide (carbendazim) at 500 μg/ml. Additionally, the pot experiments indicated that nematophin could effectively inhibit the disease extension on rice and broad bean plants caused by R. solani. Nematophin also exerted some adverse influences on the sclerotial development of R. solani by dramatically suppressing their formation and maturation at 40.00 μg/ml, as well as their germination at 15.00 μg/ml. Morphological and ultrastructural observations showed that the hyphae of R. solani became twisted, shriveled, and deformed at the growing points after exposure to nematophin at 40.00 μg/ml, and that the subcellular fractions also became abnormal concurrently, especially the mitochondrial structure. These results indicate that nematophin has great potential to be used as a bio-pesticide in agricultural production.

Mechanism of Chitosan Oligosaccharides in Inhibiting Phytophthora infestans and Synergistic Enhancement with Conventional Fungicides: A Strategy for the Management of Potato Late Blight

Zhenzhen Deng,Xunguang Luo,Xiaohui Ou,Chunyan Gao,Wanxin Hu,Changhong Li,Jinpeng Yu,Yuanyuan Zhang,Airong Huang,Shizhuo Liu,Shun Feng,Liang Jin,Maxime Bonnave,Francois Serneels,Xiumei Luo,Gengxin Xie,Pan Dong

Chitosan oligosaccharides (COSs) show broad-spectrum antimicrobial activity, yet their mechanism against Phytophthora infestans, the causal agent of potato late blight, remains unclear. This study evaluated the inhibitory effects, mechanisms, and field performance of COS against P. infestans. Results showed that COS significantly inhibited mycelial growth in three strains (T30–4, 3928A, 88069), with IC50 values of 0.10, 0.11, and 0.11 g/L, and at 0.5 g/L, it inhibited sporangial germination in T30–4. Transcriptomics revealed 727 differentially expressed genes (DEGs) enriched in fatty acid metabolism, membrane biogenesis, and transmembrane transport. Combined application with sublethal stress (high salt, high temperature, and high oxygen) or chemical fungicides (infinito and mancozeb) enhanced antioomycete efficacy. COS also induced systemic resistance in potatoes without phytotoxicity. Field trials confirmed that COS combined with infinito increased the yield by 31% and reduced disease severity by 35%. These findings support COS as a promising biocontrol agent for sustainable management of potato late blight.
