Phytophthora sojae is an oomycete pathogen that causes significant agricultural losses worldwide, mainly the soybean damping-off and root/stem rot. To counter this, copper oxide nanoparticles (CuO NPs)/reduced graphene oxide (rGO) composites (CuO-rGO) have been proposed as a potential solution in nano-agriculture. In this study, we aimed to investigate the inhibitory mechanisms of CuO-rGO, CuO NPs, and CuCl2 on P. sojae through phenotypic and transcriptomic analysis. Even a low concentration of 10 mg Cu2+ L−1, CuO-rGO, CuO NPs, and CuCl2 exhibited remarkable inhibitory effects on the pathogen. This was due to the capacity of these three forms of Cu to induce oxidative stress, which caused disruptions in the mycelial cell membrane, intracellular structure, carbon metabolism, organic acid metabolic processes, and amino acid metabolism. Among them, CuO-rGO showed a better slow-release effect and long-lasting antimicrobial properties, making it more suitable for practical agricultural use. Furthermore, the transcriptomic analysis revealed distinct patterns of antioxidant enzyme-related genes among treatments. Compared to the other two treatments, the CuO-rGO treatment showed higher expression of differential expression genes (DEGs) associated with superoxide dismutase (SOD) and peroxiredoxin (PRDX). The CuO NPs treatment had the most significant expression of DEGs related to catalase (CAT), while the DEGs linked to glutathione peroxidase (GPX) were significantly expressed in the CuCl2 and CuO-rGO treatments. These findings suggest that CuO-rGO has great potential as a practical agricultural agent against oomycete pathogens. Further investigations are essential to elucidate the mechanisms underlying the oxidative stress and antioxidant systems induced by different copper forms in the future.

Glyceollins are structurally complex potent antimicrobial isoflavonoid phytoalexins produced by the crop soybean ( Glycine max ), yet their biosynthesis remains elusive, making it impossible to carry out synthetic biology-based production and engineering for further development. Here, via assembling synergistic engineering strategies, we successfully rewired the metabolic fluxes in Nicotiana benthamiana leaves for high-yield production of isoflavonoid precursor daidzein (7.04 g kg −1 dry weight (dw)), allowing for efficient screening and identification of six cytochrome P450 monooxygenases, namely glyceollin synthases, that furnish the pyrano/furano E ring and complete the 15-step biosynthetic pathways of diverse glyceollins. We establish that purified glyceollins are important for plant defense as they can effectively suppress the growth of Phytophthora sojae in vitro. Our engineered plant chassis can provide facile access to bioactive isoflavonoids, as manifested by the de novo total biosynthesis of glyceollins (for example, I, II, III and VII at up to 5.9 g kg −1 , dw) and medicarpin (0.72 g kg −1 , dw) for enhanced pathogen resistance and medicinal value.

Borrelidin has high and specific antifungal activity against Phytophthora sojae. To explore the antifungal mechanism of borrelidin against P. sojae, the relationship between the antifungal activity of borrelidin and the concentration of threonine was evaluated. The results demonstrated that the growth-inhibitory effect of borrelidin on the growth of P. sojae was antagonized by threonine in a dose-dependent manner, suggesting that threonyl-tRNA synthetase (ThrRS) may be the potential target of borrelidin. Subsequently, the inhibition of the enzymatic activity of ThrRS by borrelidin in vitro was confirmed. Furthermore, the detailed interaction between ThrRS and borrelidin was investigated using fluorescence spectroscopy and circular dichroism (CD), implying a tight binding of borrelidin to ThrRS. Taken together, these results suggest that the antifungal activity of borrelidin against P. sojae was mediated by inhibition of ThrRS via the formation of the ThrRS–borrelidin complex.

Interspecies interactions play a key role in soil-borne disease suppression in intercropping systems. However, there are limited data on the underlying mechanisms of soil-borne Phytophthora disease suppression. Here, a field experiment confirmed the effects of maize and soybean intercropping on Phytophthora blight of soybean caused by Phytophthora sojae. Experimentally, the roots and root exudates of maize were found to attract P. sojae zoospores and inhibit their motility and the germination of cystospores. Furthermore, five phenolic acids (p-coumaric acid, cinnamic acid, p-hydroxybenzoic acid, vanillic acid, and ferulic acid) that were consistently identified in the root exudates and rhizosphere soil of maize were found to interfere with the infection behavior of P. sojae. Among them, cinnamic acid was associated with significant chemotaxis in zoospores, and p-coumaric acid and cinnamic acid showed strong antimicrobial activity against P. sojae. However, in the rhizosphere soil of soybean, only p-hydroxybenzoic acid, low concentrations of vanillic acid, and ferulic acid were identified. Importantly, the coexistence of five phenolic acids in the maize rhizosphere compared with three phenolic acids in the soybean rhizosphere showed strong synergistic antimicrobial activity against the infection behavior of P. sojae. In summary, the types and concentrations of phenolic acids in maize and soybean rhizosphere soils were found to be crucial factors for Phytophthora disease suppression in this intercropping system.
