Antibacterial Activity and Mechanism of Pinus pumila (Pall.) Pinecone Essential Oil Against Escherichia coli

Zhou Caixue,Dai Shuxia,Yin Li,Zhuo Yu,Zhao Nannan,Li Dehai,Sun Changyan

Purpose In this paper, we use Pinus pumila (Pall.) Pinecone to extract essential oil and improve its utilization. Methods The study used hydrodistillation to extract essential oil from Pinus pumila (Pall.) pinecone. Inhibition analysis of E. coli using extracted essential oils was evaluated to reveal the inhibitory activity by assessing growth curves, conductivity, nucleic acids, proteins, enzyme activities, and protein synthesis. Results The primary constituents, as shown by GC-MS chemical composition study, were α-pinitol, β-pinene and limonene. The results of the bacteriostatic activity indicated that the inhibitory zone (DIZ) had a diameter of 14.93 ± 0.14 mm, the minimum bactericidal concentration (MBC) was 16.0 µL/mL, and the minimum inhibitory concentration (MIC) was 8.0 µL/mL. Sweeping electron microscopy and laser confocal also demonstrated the disruption of cell wall and cellular enzymes, analyzed by CATase and ATPase activities, which showed a decrease in enzyme activity, and SDS-PAGE analysis showed its ability to inhibit protein synthesis, which leads to cellular damage. Conclusion The present study emphasizes the sustainable use of Pinus pumila (Pall.) Pinecone for economic opening of food preservation in addition to inhibition of spoilage bacteria.

Modification-based Pinus pumila polysaccharides and their effects on osteoblast MC3T3-E1

Hua Zhang,Zhao-Yuan Feng,Zhen-Zhou Li,Yi-Sen Wang,Zhe-Xuan Mu,Xiao-Hong Lv,Zhen-Yu Wang,Zi-Luan Fan

In this study, a polysaccharide component was extracted from Pinus pumila using biomimetic-microwave assisted alkali tandem extraction and then purified by DEAE-52 and Sephadex G-200. A new purified polysaccharide (PSP c-a) was determined through in vitro tracking. The structural features were characterized using a HPGP, FTIR, IC and NMR spectroscopy. The analysis showed that there may be →3, 6)-α-Galp (1 → 3, 4)-α-Glcp (1 → 5)-α-Manp (1→ in the backbone of PSP c-a, →3, 4)-α-Glcp (1→ and →3, 4)-α-Glcp (1 → α-Arap (1→ in the branch of PSP c-a. To improve the effects of PSP c-a on osteoblast MC3T3-E1, PSP c-a was biologically and chemically modified by α-glucosidase together with Sr. Finally, a new thermostable complex Sr-PSP c-a-1 was synthesized. The effects of PSP c-a and Sr-PSP c-a-1 on osteoblasts MC3T3-E1 were compared. It was found that both were found to have a positive effect on MC3T3-E1, and Sr-PSP c-a-1 was more effective than PSP c-a. Therefore, Sr-PSP c-a-1 should be a potential candidate for promoting skeletal tissue regeneration.

An approach for extraction of kernel oil from Pinus pumila using homogenate-circulating ultrasound in combination with an aqueous enzymatic process and evaluation of its antioxidant activity

Fengli Chen,Qiang Zhang,Huiyan Gu,Lei Yang

In this study, a novel approach involving homogenate-circulating ultrasound in combination with aqueous enzymatic extraction (H-CUAEE) was developed for extraction of kernel oil from Pinus pumila . Following comparison of enzyme types and concentrations, an enzyme mixture consisting of cellulase, pectinase and hemicellulase (1:1:1, w/w/w) at a concentration of 2.5% was selected and applied for effective oil extraction and release. Several variables potentially influencing extraction yields, namely, homogenization time, incubation temperature, incubation time, mark-space ratio of ultrasound irradiation, ultrasound irradiation power, liquid-solid ratio, pH and stirring rate, were optimized by Plackett-Burman design. Among the eight variables, incubation temperature, incubation time and liquid-solid ratio were statistically significant and were further optimized by Box-Behnken design to predict optimum extraction conditions and ascertain operability ranges for maximum extraction yield. Under optimum operating conditions, extraction yields of P. pumila kernel oil were 31.89   ±   1.12% with a Δ5-unsaturated polymethylene interrupted fatty acid content of 20.07% and an unsaturated fatty acid content of 93.47%. Our study results indicate that the proposed H-CUAEE process has enormous potential for efficient and environmentally friendly extraction of edible oils.

Essential oil extraction from fresh needles of Pinus pumila (Pall.) Regel using a solvent-free microwave-assisted methodology and an evaluation of acetylcholinesterase inhibition activity in vitro compared to that of its main components

Xiaojin Peng,Xinyu Yang,Huiyan Gu,Lei Yang,Hongfei Gao

A solvent-free microwave-assisted method and the screw extrusion of raw materials are combined and successfully applied to obtain the essential oil of fresh needles of Pinus pumila (Pall.) Regel. Single-factor screening and the Box-Behnken design method were applied to optimize the extraction of essential oils, and the yield of essential oil under the optimal conditions was 12.00 ± 0.60 mL/kg, which was highly consistent with the results predicted by the Box-Behnken design model. The process developed offers advantages in terms of the yield of essential oil and a reduced extraction time (31 min in comparison to 4 h for conventional hydrodistillation). The microstructures, chemical structures and thermal stability of needle samples before and after extraction were also investigated. The main components found in the essential oil extracted from the fresh needles of P . pumila were (+)-3-carene (22.75 %), δ-cadinene (12.12 %), terpinolene (12.11 %) and β-bisabolene (9.12 %). Furthermore, the semi-inhibition concentrations (IC 50 ) of acetylcholinesterase inhibition activity of the essential oil, terpinolene, (−)- trans -caryophyllene and (+)-3-carene were determined to be 0.76 ± 0.04, 0.55 ± 0.03, 0.45 ± 0.02 and 0.64 ± 0.03 mg/mL, respectively. In general, P . pumila needle essential oil showed notable inhibitory ability to acetylcholinesterase, and the components terpinolene, (−)- trans -caryophyllene, and (+)-3-carene play major roles in the process of essential oils inhibiting acetylcholinesterase. Additionally, the acetylcholinesterase inhibition activities of all the tested samples were mostly related to the corresponding concentrations. The acetylcholinesterase inhibition activity of kojic acid was also determined in parallel experiments as a positive control.
