In this paper, using Styphnolobium japonicum, Cryptomeria fortunei and Pinus yunnanensis as raw materials, cellulose (CE) was first extracted by chemical method (CM1) and biological enzymatic method 2 (BM2), and then further prepared into nanocellulose fiber (CNF) by biological enzymatic method 1(CPM (CPM stands for cellulase and pectinase)). The CE and CNF were characterized by FTIR, XRD, SEM, DSC and TG. The results showed that the CNF prepared by CM1-CPM was cellulose type II, CNF prepared by BM2-CPM was cellulose type I. The best crystallinity of the CNF prepared by CM1-CPM reached 74.1% (C. fortunei CNF), the best yield was 24.0% (S. japonicum CNF) and the minimum average diameter was 62.6 nm (C. fortunei CNF). However, the best crystallinity of CNF prepared by BM2-CPM reached 80.1% (C. fortunei CNF), the best yield was 27.4% (C. fortunei CNF), and the minimum average diameter was 31.6 nm (C. fortunei CNF). And also, the thermal degradation analysis of the CNF prepared from the three raw materials revealed that the CNF prepared by BM2-CPM had better thermal degradability. Finally, the thermal stability analysis of the C. fortunei CNF was carried out, and it was found that the residual mass of CNF prepared by BM2-CPM was 48%, while the residual mass of C. fortunei CNF prepared by BM1-CPM was 22%, which showed the C. fortunei CNF prepared by BM2-CPM had better thermal stability. Therefore, the properties of nanocellulose prepared by BM2-CPM are optimal.

In this study, the pine nut ( Pinus yunnanensis Franch.) protein was hydrolyzed by alkaline protease and trypsin to prepare pine nut protein hydrolysate (PNPH). The chemical, intracellular and in vivo antioxidant capacity of PNPH were evaluated. PNPH owned the ability of scavenging free radicals, and it could protect the HepG2 cells from oxidative damage by preserving cell viability. Moreover, PNPH could reduce the malondialdehyde (MDA) content and improved the superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in serum, heart and liver of aging mice induced by D-galactose. Further, the PNPH was stepwise purified and identified, and 15 peptides were identified from purified fraction in PNPH. The three-dimension structures of identified peptides were predicted. Among all identified peptides, peptide 3, 7, 8 and 11 were presumed to possess good antioxidant activity. Overall, PNPH and purified peptides isolated from PNPH have potential application prospects in the field of natural antioxidants and anti-aging functional foods.

Pinus yunnanensis cones are a rich source of bioactive compounds; yet, the systemic exposure and mechanistic basis of their anti-breast cancer activity remain unclear. Herein, we systematically identify the systemically absorbed components of P. yunnanensis cone extract and elucidate its anti-breast cancer mechanisms using an integrated analytical and computational approach. An optimized ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method was developed to overcome severe matrix interference and insufficient sensitivity, facilitating the rapid identification of active constituents in the crude extract. UHPLC–MS/MS-based analysis of rat plasma and feces revealed 16 (14 prototypes and 2 metabolites) and 12 (10 prototypes and 2 metabolites) systemically absorbed components, respectively, derived from 28 identified compounds. Subsequent network pharmacology combined with molecular docking validation was used to screen the anti-breast cancer active components, core targets, and key pathways. Integrated network pharmacology predicted coniferaldehyde, phyllanflexoid A, 3′,4′,5,5′,6,7-hexamethoxyflavone, dihydrocaffeic acid, usnic acid, and 7-oxodehydroabietic acid, as core components, targeting TP53, STAT3, PIK3R1, SRC, and PIK3CA and involving key pathways such as VEGF signaling pathway, aldosterone-regulated sodium reabsorption, prolactin signaling pathway, PD-L1/PD-1 checkpoint pathway in cancer, and ErbB signaling pathway. This study establishes a robust UHPLC–MS/MS platform for the quality control of P. yunnanensis cones and provides critical insights into their translational potential for anti-breast cancer drug development and advancing separation science applications in natural product research.

Three sympatric Tomicus bark beetles, Tomicus yunnanensis , Tomicus brevipilosus and Tomicus minor that coexist in the trunks of a pine but occupy different spatiotemporal ecological niches, are the best-studied examples for addressing functional differentiation of chemosensory protein (CSP) orthologs. Here, totally 36 CSP-coding genes belonging to 12 groups of orthologs were identified from three Tomicus beetles. Tomicus CSP orthologs shared the high conservation and slight differences in sequence characteristics, phylogenetic relationships and tissue expression profiles. Binding assays revealed that 12 antenna-enriched Tomicus CSPs could respond strongly to non-host volatiles, pheromones and insecticides where each group of CSP orthologs presented diverse ligand-binding properties. CSP1 and CSP2 could interact strongly with benzaldehyde and salicylaldehyde (dissociation constant, K i < 14 μM) whereas CSP10 and CSP11 preferred to bind 1-hexanol and acetophenone (K i < 13 μM). All the proteins were tuned to chlorpyrifos with particularly high affinities (K i < 10 μM). Two conserved residues (glutamine, Q64 and leucine, L72) of CSP11 orthologs contributed to the specific binding to optimal ligands, but exhibited opposite binding properties between chlorpyrifos and other four compounds. Our study sheds light on the functional conservation and divergence of Tomicus CSP orthologs in olfactory reception and insecticide resistance, implying the correlation between ecological niche differentiation and olfactory specialization in three Tomicus bar beetles.

Ectomycorrhizal (EM) fungi play important roles in nutrient cycling and plant community establishment in forest ecosystems. Effects of EM formation on global alterations of the transcriptome and metabolome during plant-fungal interaction and the key metabolites involved in EM development are largely unknown. Here, dual RNA-Seq and untargeted metabolomic analyses were used to reveal stage-specific and core responses of Pinus yunnanensis and Lactarius deliciosus during mycorrhizal colonization. We found that L. deliciosus colonization in P. yunnanensis roots induced different transcriptional changes across three interaction stages, with a small core of genes consistently regulated at all stages. Concentrations of retinol (vitamin A) and retinoic acid increased while that of B group vitamins decreased during EM formation, which was coordinately regulated by these two plant-fungus partners, with L. deliciosus possibly playing a dominant role. Exogenous retinol altered the diameter and mantle thickness of P. yunnanensis - L. deliciosus EM tips and affected host plant growth and phosphorus acquisition . In the absence of L. deliciosus, exogenous retinol increased the root diameter and the number of root tips of P. yunnanensis . Furthermore, the concentration of auxin increased, but that of abscisic acid decreased during EM formation, and the genes involved in plant hormone signal transduction were gradually activated, and auxin and cytokinin signal transduction potentially played a positive role in this EM symbiosis. In conclusion, we propose that the interaction of P. yunnanensis and L. deliciosus alters vitamin metabolism, which may further affect plant hormone biosynthesis and signal transduction, modulating root morphology and EM traits.
