Optimization of arsenic extraction in rice samples by Plackett–Burman design and response surface methodology

Li Ma,Lin Wang,Jie Tang,Zhaoguang Yang

Statistical experimental designs were employed to optimize the extraction condition of arsenic species (As(III), As(V), monomethylarsonic acid (MMA) and dimethylarsonic acid (DMA)) in paddy rice by a simple solvent extraction using water as an extraction reagent. The effect of variables were estimated by a two-level Plackett–Burman factorial design. A five-level central composite design was subsequently employed to optimize the significant factors. The desirability parameters of the significant factors were confirmed to 60   min of shaking time and 85   °C of extraction temperature by compromising the experimental period and extraction efficiency. The analytical performances, such as linearity, method detection limits, relative standard deviation and recovery were examined, and these data exhibited broad linear range, high sensitivity and good precision. The proposed method was applied for real rice samples. The species of As(III), As(V) and DMA were detected in all the rice samples mostly in the order As(III)   >   As(V)   >   DMA.

Optimization of ultrasonic circulating extraction of samara oil from Acer saccharum using combination of Plackett–Burman design and Box–Behnken design

Fengli Chen,Qiang Zhang,Shimin Fei,Huiyan Gu,Lei Yang

In this study, ultrasonic circulating extraction (UCE) technique was firstly and successfully applied for extraction of samara oil from Acer saccharum . The extraction kinetics were fitted and described, and the extraction mechanism was discussed. Through comparison, n -hexane was selected as the extraction solvent, the influence of solvent type on the responses was detailedly interpreted based on the influence of their properties on the occurrence and intensity of cavitation. Seven parameters potentially influencing the extraction yield of samara oil and content of nervonic acid, including ultrasound irradiation time, ultrasound irradiation power, ultrasound temperature, liquid-solid ratio, soaking time, particle size and stirring rate, were screened through Plackett–Burman design to determine the significant variables. Then, three parameters performed statistically significant, including liquid-solid ratio, ultrasound irradiation time and ultrasound irradiation power, were further optimized using Box–Behnken design to predict optimum extraction conditions. Satisfactory yield of samara oil (11.72   ±   0.38%) and content of nervonic acid (5.28   ±   0.18%) were achieved using the optimal conditions. 1% proportion of ethanol in extraction solvent, 120   °C of drying temperature and 6.4% moisture were selected and applied for effective extraction. There were no distinct differences in the physicochemical properties of samara oil obtained by UCE and Soxhlet extraction, and the samara oil obtained by UCE exhibited better antioxidant activities. Therefore, UCE method has enormous potential for efficient extraction of edible oil with high quality from plant materials.

Assessment of the effects of nutrients on biomass and lipid accumulation in Dunaliella tertiolecta using a response surface methodology

Xiao-Ying Qv,Yan-Yun Guo,Jian-Guo Jiang

Bio-energy is arousing the common interest of worldwide researchers to investigate it as a substitute for fossil resources. Compared to other bio-energy sources, algae energy has many advantages. This research was aimed to increase the lipid productivity of Dunaliella tertiolecta, an oleaginous microalgae, by optimizing algal culture medium. Plackett–Burman Design (PBD) was adopted to screen out the critical factors from seven nutritional elements. Box–Behnken design (BBD) was used to seek the optimal nutritional conditions for achieving maximum lipid productivity. Results showed that four nutrients, i.e. NaNO3, NaCl, NaHCO3 and NaH2PO4 2H2O, played a critical role in the accumulation of lipid. GC analysis verified that the four critical nutrients mentioned above had different influences on fatty acid methyl esters, moreover, NaCl had the most prominent effects on fatty acid composition of D. tertiolecta. NaCl 87.69 g L−1, NaNO3 1.77 g L−1, NaH2PO4·2H2O 1.92 g L−1, NaHCO3 5.57 g L−1 were proven to be the optimal culture conditions, under which the lipid productivity in the experiment was 12.576 ± 0.12 mg per L per day, which is approaching the theoretically predicted lipid productivity of 12.925 mg per L per day. The lipid productivity was increased by 31.4% under the optimal culture conditions compared to that under the initial medium composition, indicating that optimization of nutrients could effectively improve the lipid production of D. tertiolecta.

The fermentation optimization for alkaline protease production by Bacillus subtilis BS-QR-052

Sun Biao,Zou Kai,Zhao Yingqing,Tang Yinfang,Zhang Fuming,Chen Weijing,Tang Xiaoting,Chang Chenran,Zheng Yan

Introduction: Proteases exhibit a wide range of applications, and among them, alkaline proteases have become a prominent area of research due to their stability in highly alkaline environments. To optimize the production yield and activity of alkaline proteases, researchers are continuously exploring different fermentation conditions and culture medium components. Methods: In this paper, the fermentation conditions of the alkaline protease (EC 3.4.21.14) production by Bacillus subtilis BS-QR-052 were optimized, and the effect of different nutrition and fermentation conditions was investigated. Based on the single-variable experiments, the Plackett–Burman design was used to explore the significant factors, and then the optimized fermentation conditions, as well as the interaction between these factors, were evaluated by response surface methodology through the Box–Behnken design. Results and discussion: The results showed that 1.03% corn syrup powder, 0.05% MgSO4, 8.02% inoculation volume, 1:1.22 vvm airflow rate, as well as 0.5% corn starch, 0.05% MnSO4, 180 rpm agitation speed, 36°C fermentation temperature, 8.0 initial pH and 96 h incubation time were predicted to be the optimal fermentation conditions. The alkaline protease enzyme activity was estimated to be approximately 1787.91 U/mL, whereas subsequent experimental validation confirmed it reached 1780.03 U/mL, while that of 500 L scale-up fermentation reached 1798.33 U/mL. This study optimized the fermentation conditions for alkaline protease production by B. subtilis through systematic experimental design and data analysis, and the activity of the alkaline protease increased to 300.72% of its original level. The established model for predicting alkaline protease activity was validated, achieving significantly higher levels of enzymatic activity. The findings provide valuable references for further enhancing the yield and activity of alkaline protease, thereby holding substantial practical significance and economic benefits for industrial applications.

Efficient Ultrasonic-assisted Aqueous Enzymatic Method for Pecan Nut Kernel Oil Extraction with Quality Analysis

Anning Liu,Huijuan Jing,Xiaojing Du,Chaoyang Ma,Hongxin Wang

In this study, the recovery of pecan nut kernel oil (PNKO) obtained by mechanical pressing (MP) was compared with that obtained by ultrasonic-assisted aqueous enzymatic extraction (UAAEE). At the same time, contents of substances with proven bioactivity, fatty acid compositions, physicochemical properties and antioxidant activities of PNKO were also assessed. Obviously, the oil yield obtained by UAAEE (71.32%) was higher than that obtained by MP (56.81%). Therefore, Plackett-Burman design (PBD), as well as Box-Behnken design (BBD), further optimized the UAAEE process, and the highest oil yield of 78.83% was acquired under the following conditions: incubation time of 2 h, the mixed enzyme (cell ulase+hemicellulase+pectinase+neutrase, w/w/w/w=1/1/1/1) concentration of 2.9%, liquid-solid ratio of 4 mL/g, pH of 4, particle size of 300 μm, ultrasonic time of 20 min, ultrasonic power of 432 W and reaction temperature of 53°C. The PNKO obtained by the two methods possessed similar fatty acid composition, but that obtained by UAAEE had comparatively low acid value and peroxide value. Moreover, rich content of total phenolics, squalene and phytosterols, as well as strong scavenging activities against DPPH and ABTS+ free radicals, were also contained in the PNKO obtained by UAAEE. These results demonstrated that UAAEE was an efficient method to acquire pecan nut kernel oil with excellent quality and high recovery.

pH-responsive functionalized surface active ionic liquid as an enhanced medium for efficient extraction and in-situ separation of flavonoids in Vitex negundo L. Leaves

Jiabo Cheng,Mengfei Tian,Zarmina Gul,Chunyu Liang,Bin Qiao,Yuan Gao,Chunjian Zhao,Chunying Li

In this study, a functionalized surface active ionic liquid (SAIL) was designed and prepared, which exhibits pH-responsive behavior and high yield for 6 target flavonoids from the leaves of Vitex negundo L.. Firstly, a series of functionalized SAIL [N-C n , N'–CO 2 H-Im]Br (n = 10, 12, 14, 16) were prepared and used for extraction of the target components, [N-C 14 , N'–CO 2 H-Im]Br showed the highest yield. Secondly, optimized extraction conditions obtained by Plackett-Burman design (PBD) and response surface methodology (RSM) were concentration of 45.5 mM, liquid–solid ratio of 11.6 (mL/g), extraction time of 38.5 min, extraction temperature of 40 °C, and ultrasonic power of 400 W. The yield obtained by [N-C 14 , N'–CO 2 H-Im]Br of vitexin, isovitexin, hyperoside, luteolin, quercetin, apigenin is 2.04, 1.81, 1.24, 1.51, 2.47 and 1.21 mg/g, respectively, which is 1.11–1.56 folds higher than non-functionalized SAIL ([C 14 min]Br) and 1.50–5.36 folds higher than traditional solvents. More importantly, the solvent system can realize the in-situ separation of the target compound by adjusting pH. When pH is changed from 6.1 to 4.9, the system is divided into two phases, and the target compounds were separated to the upper phase with a high recovery rate. Finally, molecular simulation was used to reveal the extraction mechanism of functionalized SAIL on target compounds. The results showed that the target flavonoids were compatibilized to the palisade region of micelles, and the weak interaction between the target flavonoids and functionalized SAIL was mainly van der Waals force.

Composition, characteristics and antioxidant activities of fruit oils from Idesia polycarpa using homogenate-circulating ultrasound-assisted aqueous enzymatic extraction

Kexin Hou,Xuebing Yang,Meili Bao,Fengli Chen,Hao Tian,Lei Yang

An efficient approach to isolate fruit oil from Idesia polycarpa using homogenate-circulating ultrasound-assisted aqueous enzymatic extraction (H-CUAEE) was developed. An enzyme cocktail comprising cellulase, hemicellulase, and pectinase proved to be the most effective in extracting oils. Nine potential variables that possibly affected the extraction yields – the liquid-solid ratio, incubation temperature, incubation time, amount of enzyme, homogenate time, ultrasound irradiation power, mark-space ratio of ultrasound irradiation, rate of agitation, and pH – were investigated using a Plackett–Burman design in order to screen out the significant variables. The liquid-solid ratio, incubation temperature, and incubation time were significant on a statistical basis and these three parameters were studied through a Box–Behnken design for the prediction of the best experimental conditions that maximize extraction yield of the fruit oil. A maximum fruit oil recovery of 79.36% with an error of 0.17% was achieved under the optimal extracting conditions: liquid-solid ratio of 7.13, the incubation time of 2.94 h, and the incubation temperature of 48.74 °C. The H-CUAEE process has expansive prospects for the preparation of oils from oil plants.

Carboxyl-functionalized magnetic amide-linked covalent organic framework for efficient extraction of multi-class mycotoxins from soybeans prior to HPLC-MS/MS analysis

Dan Wei,Qiao Deng,Jianliang Li,Jingjing Xu,Ming Guo,Huizhen Wu,Ju Zhang,Jianchang Jin,Xu Wang

Mycotoxins co-contamination in soybean poses great risks to human health, however, due to their diverse structures, it is difficult to simultaneously extract and detect these multi-mycotoxins. By incorporating modification of hydrophilic carboxyl groups with the hydrophobic properties of covalent organic frameworks, a stable cubic carboxyl-functionalized magnetic amide-linked covalent organic framework were fabricated, characterized and utilized as adsorbents for magnetic solid phase extraction (MSPE) of eleven frequently detected mycotoxins from soybeans followed by HPLC-MS/MS analysis. The key MSPE parameters were screened using the Plackett-Burman design and optimized with the Box-Behnken design for response surface methodologies. Under the optimal experimental conditions, the linear range was 0.2–50 μg/kg, the limits of detection were 0.1–1.4 μg/kg. The established method was applied for the soybean analysis with satisfactory recoveries (70.8–108.3%) and precision (intra- and inter-relative standard deviations of 2.1–9.2% and 1.6–11.1%, respectively). With proper modification, this method may have great application potential for simultaneously analyzing and monitoring the multi-mycotoxin contamination in foods to ensure consumer safety and public health.

Kexin Hou,Meili Bao,Lu Wang,Hua Zhang,Lei Yang,Haitian Zhao,Zhenyu Wang

In this study, in order to simultaneously extract procyanidins and essential oil from pinecones of Pinus koraiensis , aqueous enzymatic pretreatment ionic liquid–lithium salt based microwave–assisted extraction (AEILLME) with a modified hydrodistillation concatenated Soxhlet extraction apparatus was proposed. 1-Butyl-3-methylimidazolium bromide solution was selected as extraction solvent , while LiCl and mixture of cellulase and pectinase (1:1, w/w) as additives by single factor experiment. Other parameters that affect the yield of target components were investigated and optimized by Plackett–Burman design and Box–Behnken design. The obtained optimal conditions were: microwave irradiation power 581.49 W, microwave irradiation time 15.95 min and liquid–solid ratio 11.91 mL/g. Under the optimized conditions, the predicted yields of oligomeric procyanidins, polymeric procyanidins and essential oil were 13.02, 32.19 and 7.76 mg/g, respectively. Analysis of chemical constituents in the essential oil was performed on gas chromatography–mass spectrometry and there was no significant difference on the components of essential oil obtained by proposed method and traditional methods. Ionic liquids were successfully recycled and reused, which could lower the process cost and being beneficial to industrial production. This study provides an approach with good prospects for the application in extracting active components from natural plant material.

Yi Xiaotong,Liu Chang,Liu Xueke,Wang Peng,Zhou Zhiqiang,Liu Donghui

Magnetic partially carbonized cellulose nanocrystals (MPC-CNC) were obtained by sulfuric acid treatment of microcrystalline cellulose (MCC) and then loaded with magnetic Fe 3 O 4 nanoparticles. The material is shown to be a viable material for magnetic solid phase extraction of triazine and triazole pesticides from water. The pesticides (specifically: simazine, ametryn, prometryn, terbutryn, atrazine, triadimenol, epoxiconazole, myclobutanil, triadimefon and tebuconazole) were quantified by ultra HPLC in tandem with a triple quadrupole mass spectrometry (UHPLC-MS/MS). The effects of NaCl concentration, amount of adsorbent, vortex time, sample volume and pH value on extraction efficiency were optimized by Plackett-Burman design and Box-Behnken design methods. Under the optimal conditions, the method shows the following figures of merit: (a) Linear responses in the range from 0.02–10 μg L −1 ; (b) detection limits between 2.2 to 6.1 ng L −1 (for S/ N  = 3); (c) recoveries from spiked samples of 73.7–117.1% with relative standard deviations (RSDs) of 2.0–15.7%; and (d) an enrichment factor of 75. The method was successfully applied to the determination of the pesticides in five environmental water samples. Schematic representation of the process of magnetic solid phase extracting pesticides in water using MPC-CNC. Ultrasound-assisted extraction of polyphenols from Phyllanthi Fructus: Comprehensive insights from extraction optimization and antioxidant activity

Haixia Che,Rui Zhang,Xianyao Wang,Hong Yu,Xuepeng Shi,Jie Yi,Jiawen Li,Qi Qi,Ruizhi Dong,Qian Li

Phyllanthi Fructus (PF) is a valuable botanical resource with a long history of traditional use, known for potent antioxidant and anti-inflammatory effects attributed to its rich contents of bioactive compounds, particularly polyphenols. However, current extraction techniques limit the utilization of polyphenols from PF. This study aimed to achieve the maximum polyphenol yield and improve the antioxidant activity of PF extracts to promise PF’s prospects for modern healthcare. Firstly, ultrasonic-assisted extraction (UAE) was employed to extract the polyphenols in PF and a combination of Plackett-Burman designs (PBD) and response surface methodology (RSM) was applied to optimize UAE’s conditions. Next, cellular superoxide dismutase (SOD) and malondialdehyde (MDA) were used to assess the antioxidant activity of extracted polyphenols. Ultra-Performance Liquid Chromatography coupled with Quadrupole Time-of-Flight Mass Spectrometry (UPLC-Q-TOF MS) was utilized to characterize polyphenol components in the PF extracts. Finally, network pharmacology and molecular docking analysis were performed to screen the potential target proteins of polyphenols from PF. As a result, the optimized polyphenol yield was 213.49 mg/g, and the antioxidant activities, measured by ability of DPPH scavenging, ABTS+ scavenging, and FRAP were 76.95 %, 2.24 mmol/g, 2.34 mmol/g, respectively. PF extracts also showed good antioxidant capabilities at cellular level. 26 polyphenol components were identified in the PF extracts. Among these, ellagic acid, myricetin, and eriodictyol may exert antioxidant effects by interacting with AKT serine/threonine kinase 1 (AKT1). In conclusion, our study provides valuable insights into the optimizing PF extraction and underscores its potential applications in enhancing natural polyphenols extraction using UAE with a combination of PBD and RSM. These findings offer a promising avenue for the development and utilization of PF, and could serve as a reference for similar extraction processes in the future.

Hybridising plasma functionalized water and ultrasound pretreatment for enzymatic protein hydrolysis of Larimichthys polyactis: Parametric screening and optimization

Okon Johnson Esua,Da-Wen Sun,Jun-Hu Cheng,Huifen Wang,Chen Chen

Hybridising plasma functionalized water and ultrasound pretreatment for the enzymatic hydrolysis (HPUEH) of Larimichthys polyactis was evaluated by adopting Plackett-Burman design for parametric screening of six key variables, and Box-Behnken design for optimizing three most significant variables including plasma generating voltage (P V ), ultrasound treatment time (U T ), and enzyme concentration (E C ). The models developed for predicting the degree of hydrolysis (DoH), protein recovery (PVY), and soluble protein content (SPC) were sufficiently fitted to the experimental data (R 2  ≥ 0.966) with non-significant lack of fit and used for determining the optimum conditions as P V of 70 V, U T of 15 min, and E C of 1.787%, with predictive values of 27.74%, 85.62%, and 3.28 mg/mL for DoH, PVY, and SPC, respectively. HPUEH presented hydrolysates with smaller peptide sizes and molecular weights, enhanced DoH, PVY, SPC, amino acids and antioxidant activity, but reduced emulsifying and foaming properties when compared with conventional enzymatic hydrolysis.
