Plasmalogens are a special class of glycerophospholipids characterized by a vinyl ether bond (-C = C-O-) at the sn -1 position of the glycerol backbone. Altered plasmalogen profiles have been observed in neurodegenerative diseases and cancers. Profiling of plasmalogens requires specifying the vinyl ether bond and differentiating them from various types of isobars and isomers. Herein, by coupling C = C derivatization via offline Paternò–Büchi reaction with liquid chromatography-tandem mass spectrometry, we have developed a sensitive workflow for analysis of plasmalogens from biological samples. Using bovine heart lipid extract as a model system, we profiled more than 100 distinct structures of plasmenylethanolamines (PE-Ps) and plasmenylcholines (PC-Ps) at the C = C location level, far exceeding previous reports. Analysis of human glioma and normal brain tissue samples revealed elevated n-10 C = C isomers of PE-Ps in the glioma tissue samples. These findings suggest that the developed workflow holds potential in aiding the study of altered metabolism of plasmalogens in clinical samples. Graphical Abstract

Background: In acute liver injury (ALI), cell membrane damage could induce an inflammatory response and oxidative stress. As a membrane glycerophospholipid, plasmalogens (PLS) are crucial in regulating the cell membrane properties and exhibit beneficial effects in various liver diseases. However, the specific regulatory effects of PLS in the ALI remain unknown. Methods: We utilized CCl4 to induce ALI in AML12 hepatocytes and C57BL/6J mice and examined oxidative stress indicators and inflammatory cytokine levels. Our study further validated the effect of PLS on cell membrane integrity by Lactate Dehydrogenase (LDH) release assay and Dil/Calcein assay, and molecular dynamics (MD) simulations were employed to elucidate the molecular mechanisms by which PLS affected cell membranes. Results: PLS attenuated hepatocyte damage both in vivo and in vitro. Moreover, PLS increased levels of SOD, GSH, and CAT and inhibited the production of malondialdehyde. PLS succeeded in decreasing proinflammatory cytokines (TNF-α, IL-1β, and IL-6) while increasing anti-inflammatory cytokines (IL-10). Furthermore, PLS effectively maintained the cell membrane integrity. The MD simulations well explained the molecular mechanisms: a high level of PLS modulated the cell membrane properties, enabling them to be more flexible, elastic, and less prone to rupture. Conclusions: Our study illustrated the effect and molecular mechanisms of PLS against ALI, potentially broadening its application in liver diseases.

Seafood provides important nutrients to our diets, particularly omega-3 polyunsaturated fatty acids (PUFAs). Compared with other common seafood, sea snails represent an underutilized resource that may be exploited to help meet global PUFA demands. However, sea snails exhibit complex fatty acid (FA) profiles, making an accurate and detailed characterization a considerable analytical challenge. By employing an advanced mass spectrometry (MS) technique, i.e., covalent adduct chemical ionization (CACI), we characterized the FA profiles of five commonly consumed sea snail species. A straightforward method was established to distinguish monounsaturated fatty acid methyl esters (FAMEs) from isobaric dimethyl acetals (DMA), verifiable by retention-time models. FAME double-bond positional isomers were further identified by CACI-MS/MS diagnostic ions. Sea snails are abundant in plasmalogens at 82–333 mg/100 g food and long-chain omega-3 PUFA up to 174 mg/100 g food. Therefore, sea snails, particularly saltwater varieties, show promise as mainstream seafood because of their high nutritional values.

Plasmalogens (Pls) are bioactive substances enriched in the brain with a regulatory effect on Alzheimer’s disease (AD), while their metabolomic influence accompanying AD and the underlying mechanisms remain unclear. Here, we extracted and purified Pls (purity of ≥90%) from mussels and applied unbiased metabolomics using ultraperformance liquid chromatography Q-Exactive Orbitrap mass spectrometry to analyze the variation of metabolites in the major metabolic pathways of AD and revealed the cognitive improvement effect of Pls using an experimental AD zebrafish model. The results showed that 37 differential endogenous metabolites were identified, among which glycerophosphocholine, choline, S-adenosylmethionine (SAM), l-glutamine, linoleic acid, 9(S)-HPODE, methionine, and creatine were the major abnormally regulated metabolites, and the first four metabolites were viewed as potential endogenous markers. This study suggested that systemic metabolic profiling could reveal the potential metabolic networks of AD and illuminated the protective effect of Pls on AD through biochemistry mechanisms and metabolic pathways.
