Biodegradation of decabromodiphenyl ethane (DBDPE) by white-rot fungus Pleurotus ostreatus: Characteristics, mechanisms, and toxicological response

Shutao Wang,Wanlun Li,Lu Liu,Hong Qi,Hong You

Decabromodiphenyl ethane (DBDPE) can pose a potential toxic threat to human beings and the environment. P. ostreatus , as one of the typical white-rot fungi, can effectively degrade various refractory pollutants. The biodegradable characteristics of DBDPE by P. ostreatus , as well as the mechanisms, and toxicological response were investigated in this study. The removal rate reached 47.73% and 43.20%, respectively, for 5 and 20 mg/L DBDPE after 120-h degradation by P. ostreatus . As a coexisting substance, Pb could inhibit the biodegradation. It is found that both the intracellular enzyme (P450) and extracellular enzymes (manganese peroxidase (MnP), lignin peroxidase (LiP), and laccase (Lac)) played a very important role in the biodegradation of DBDPE, of which Lac dominated the degradation. The toxic response was monitored during the degradation. The activities of SOD and CAT were enhanced to eliminate excess ROS in P. ostreatus triggered by DBDPE. In addition, debromination, hydroxylation, and oxidation were inferred as the main degradation pathways preliminarily. The findings provide a theoretical basis for the application of microbial degradation of DBDPE contamination.

Structure characterization, simulated digestion, and microbial modulation of Pleurotus ostreatus polysaccharides

Kai Ye,Chujing Fu,Hengjun Du,Shiguo Chen,Donghong Liu,Gaoxing Ma,Hang Xiao

Edible mushrooms polysaccharides are widely recognized for their biological activities, yet the prebiotic potential of Pleurotus ostreatus polysaccharides (POP) remains underexplored. In this study, POP was isolated, structurally characterized, and evaluated for digestibility and impact on gut microbiota. Structural analysis revealed that POP comprises →4)-α-Glcp-(1→, →3)-β-Glcp-(1→, →4,6)-α-Glcp-(1→ and →3,4)-α-Glcp-(1→ linkages as the backbone, with side chains of 1,6-α-Galp and α-Manp residues. POP was resistant to simulated gastrointestinal digestion. During fecal fermentation, POP significantly increased microbial diversity, with Chao1 and Shannon indices rising by 26.95 % and 32.67 %. Furthermore, POP promoted the relative abundance of beneficial genera like Prevotella (8.1-fold increase), Bacteroides (3.8-fold), and Faecalibacterium (4.4-fold), while reducing opportunistic pathogens including Fusobacterium and Escherichia . Total short-chain fatty acid (SCFA) production reached 54.7 mM at 24 h, particularly acetate, propionate, and butyrate. These results highlight the prebiotic potential of POP and support its application as a functional food ingredient for improving gut health.

Bioactive metabolites in Hypsizygus marmoreus spent substrate enhanced Pleurotus ostreatus growth

Yuan Guo,Yajie Zhang,Dianpeng Zhang,Yu Liu,Qiong Yu,Shuang Song,Caige Lu,Jianping Xu,Shouxian Wang

The rapid accumulation of spent mushroom substrate (SMS) presents a critical environmental challenge globally, but efficient and cost-effective recycling approaches remain limited. Here, to advance valorization of SMS, a proof-of-concept study was conducted in which Hypsizygus marmoreus SMS ( Hm SMS) was recycled in the cultivation of Pleurotus ostreatus . Impacts of the Hm SMS-derived bioactive metabolites were further examined systematically. Results showed that integration of 10 %–30 % Hm SMS shortened the harvest time by 2–6 days, significantly improved fruiting body differentiation and productivity of P. ostreatus . The Hm SMS-derived water-soluble and volatile metabolites significantly enhanced P. ostreatus growth, albeit through different metabolic pathways: the former act by reprogramming of the amino acid-related metabolisms, whereas the latter act by upregulating purine metabolism, nucleotide metabolism, and the biosynthesis of phenylalanine, tyrosine and tryptophan. Chemical profiles of SMS were comprehensively investigated using a combined multi-omics approach. The Water-soluble metabolites within Hm SMS comprised primarily lipids and lipid-like molecules (26.8 %; LC–MS) and organic acids and derivatives (31.9 %; GC–MS). Volatile metabolites in Hm SMS were primarily lipids and lipid-like molecules (33.6 %; GC–MS). Five of the examined water-soluble metabolites (fulvic acid, dl-arabinose, erythritol, l-valine and l-isoleucine) significantly promoted P. ostreatus productivity. These findings highlight the untapped potential of SMS-derived bioactive metabolites and imply their potential high-value valorization in forestry and agricultural practices.

A comparative study of the extraction of polysaccharides from Pleurotus ostreatus residues using different techniques

Liang Fengxiang,Liu Wenqi,Liu Jia,Xue Yanting,Li Yuxian,Zhang Haipeng,Guan Lili

The edible mushroom industry generates substantial agricultural by-products (e.g., Pleurotus ostreatus residues, POR) rich in polysaccharides. This study innovatively applied multiple extraction methods to obtain Pleurotus ostreatus residues polysaccharides (PORPs) for the first time, systematically investigating their physicochemical, structural, functional, and bioactive properties, thereby filling the existing knowledge gap. Among the six PORPs extracted, hot water-extracted PORPs (PORP-H) and hot alkali-extracted PORPs (PORP-B) showed higher yields; ultrasonic-assisted extracted PORPs (PORP-U) and hot water-hot alkali sequentially extracted PORPs (PORP-HB) exhibited distinct functional advantages, thus highlighting the specificity of each method in tailoring PORP characteristics. All six PORPs consisted of 9 monosaccharides, shared analogous functional groups, displayed good thermostability, contained plentiful branched chains, and lacked a triple-helix structure, with molecular weights ranging from 432.50 kDa (PORP-U) to 590.10 kDa (PORP-H). PORP-B showed the strongest in vitro antioxidant activity; acid-extracted PORPs (PORP-A) effectively promoted the proliferation of lactic acid bacteria (LAB), indicating potential prebiotic effects. PORP-H and PORP-HB exhibited superior foaming capacity (FC) (135.25 ± 1.06% and 132.67 ± 2.75%) and foaming stability (FS) (120.25 ± 0.35% and 119.67 ± 0.76%). PORP-U had better water-holding capacity (WHC) (1.42 ± 0.21 g water/g) and emulsifying activity index (EAI) (12.85 ± 0.33 m 2 /g), while PORP-H demonstrated the best emulsifying stability index (ESI) (73.99 ± 2.25 min). These findings demonstrate that PORPs extracted by appropriate methods possess high bioactivity and functional properties, supporting their promising application in functional foods. Graphical

Total antioxidant activity assessment of Pleurotus ostreatus using a 3D-printed hydrogel nanozyme with peroxidase-like activity

Lu Gao,Tao Zhang,Xinyu Huang,Mengyuan Tan,Tianzhu Guan,Zhirong Wang,Yi Li,Xuechao Xu

A facile, cost-effective, and portable total antioxidant capacity (TAC) evaluation system was developed for Pleurotus ostreatus using a 3D-printed ferric coordination polymer (FCP) hydrogel nanozyme. Initially, the prepared peroxidase-like FCP nanozyme can catalyze the generation of hydroxyl radicals to oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of H 2 O 2 . Then, hydroxyl radicals can be neutralized by antioxidants, thereby inhibiting the TMB chromogenic reaction. Vitamin C (Vit C), vitamin E (Vit E), cysteine (Cys), and glutathione (GSH) were successfully used as reference antioxidant substrates. Moreover, a 3D-printed FCP hydrogel nanozyme was developed and incorporated into a smartphone-assisted point-of-care testing (POCT) system. TAC was evaluated by this POCT system, offering a limit of detection (LOD) as 0.34 mmol/L. The system was successfully applied to assess TAC in extracts from wild and commercially cultivated P. ostreatus . The results revealed TAC differences among these samples. Hence, this study provides a promising strategy for rapid, visual, and quantitative TAC detection in food.

Recycling and reutilizing of Pleurotus ostreatus absorbed with nano-polystyrene and vanadium ions for aqueous supercapacitors and batteries

Qianghong Wu,Youzhi Wu,Fen Ran

The harm of vanadium and nano-plastic pollution to organisms and the destruction of ecological environment are increasingly concerned by the world. However, due to the multiple valence states of vanadium and the high temperature cracking of nano-plastics, which can provide additional capacity and effectively improve the structural morphology and graphitization degree of materials, so it has great development potential in energy storage field. In this work, Pleurotus ostreatus is used to absorb both vanadium and nano-plastics, including nano-PS, nano-PS-NH 2 , and nano-PS-COOH, which are used to regulate the absorption of vanadium ions by Pleurotus ostreatus. Vanadium ions, nano-plastics, and their complexes can coordinate and decompose with various polymers of Pleurotus ostreatus. The biochar-vanadium composites prepared by this method have high specific surface area, high graphitization degree, rich hierarchical porous structure, and N, O, S, and other heteroatoms, showing good spatial confinement effect and heteroatom anchoring effect. The series of optimization processes make the electrode materials exhibit excellent electrochemical performance in supercapacitors. When the current density is 0.5 A·g −1 , the specific capacitance reaches 582.6 F·g −1 ; at 50 A·g −1 , the specific capacitance is still as high as 325 F·g −1 . After 30, 000 cycles, the specific capacitance can still reach 361.6 F·g −1 at 2 A·g −1 . The Coulomb efficiency is about 99 %. In addition, the electrode material can also be applied in zinc-ion batteries.

Coating starch improves the quality of “Pepper-salt Pleurotus ostreatus”: Insights from rheologicalproperties, microstructure and water–oil distribution

Han Xie,Wenfeng Li,Zhenan Rao,Jichun Zhao,Fuhua Li,Wenjun Wang,Jian Ming

This research investigated how native starches in batters affect texture and oil absorption of “Pepper-salt Pleurotus ostreatus ”. Corn starch (CS), potato starch (PS), glutinous rice starch (GRS), pea starch (PES), sweet potato starch (SPS) and cassava starch (CAS) were evaluated. GRS and CS coated products showed better sensory quality; PES coated samples were greasy with poor texture. GRS exhibited the highest pick-up and apparent viscosity, forming a stable coating that converted into a porous, crispy crust. CS generated a compact and stable crust with the lowest oil content (13%) due to its dense crystalline structure and limited swelling, while high b* produced bright, attractive golden-yellow color. In contrast, PES formed a coarse-pored crust with the highest oil content (30%), resulting in less durable crispness. SEM, FTIR, SAXS and LF-NMR confirmed these findings. A blend of CS and GRS is recommended to optimize crispness while minimizing oil content in pre-fried foods

Effective degradation of zearalenone by dye-decolorizing peroxidases from Pleurotus ostreatus and its metabolic pathway and toxicity analysis

Shuai Ding,Chen Lin,Qiuyun Xiao,Fa Feng,Junfeng Wang,Xing Zhang,Shengjing Yang,Lingling Li,Fei Li

The widespread detection of zearalenone (ZEN) in cereal crops and feeds poses a significant threat to both humans and animals. Consequently, the urgency for the international community to address this issue is evident in the demand for safe and effective measures to mitigate zearalenone contamination and explore detoxification methods. In this study, a dye-decolorizing peroxidase ( Po DyP4) from Pleurotus ostreatus is characterized for its impressive ZEN degradation effectiveness. Po DyP4 was demonstrated that the ability to almost completely degrade ZEN at pH 6.0 and 40 °C for 2 h, even at high concentrations of 1 mM. The promotion of enzymatic degradation of ZEN was most pronounced in the presence of Mg 2+ , while Cu 2+ and Fe 2+ exhibited a notable inhibitory effect. The degradation mechanism elucidated the detoxification of ZEN by Po DyP4 through hydroxylation and polymerization reactions. The resulting metabolic products displayed significantly reduced toxicity and minimal impact on the viability and apoptosis of mouse spermatocytes GC-2 cells, in comparison to the original ZEN. Hydrophobic contacts and hydrogen bonds were found to be crucial for ZEN- Po DyP4 stability via molecular docking. This finding suggests that Po DyP4 may have a promising application in the field of food and feed for zearalenone detoxification.

Antioxidant capacities and non-volatile metabolites changes after solid-state fermentation of soybean using oyster mushroom (Pleurotus ostreatus) mycelium

Mengxin He,Qing Peng,Xiaoqing Xu,Bo Shi,Yu Qiao

Given the abundance of beneficial properties and enzymes secreted by edible oyster mushrooms, their mycelium could serve as a starter for fermented foods to enhance their nutritional and bioactive quality. This study aimed to investigate the effects on the nutritional ingredients, antioxidant activity, and non-volatile metabolites during solid-state fermentation (SSF) of soybeans by Pleurotus ostreatus mycelium. The results indicated that the contents of dietary fiber and starch in fermented soybeans decreased, while the amounts of protein and lipid increased after SSF (P < 0.05). Analysis of the total phenolic content (TPC) and antioxidant activities of the fermented soybeans revealed that the methanolic extracts significantly increased TPC and antioxidant activities against intracellular reactive oxygen species (ROS) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages, as well as against DPPH and ABTS radicals in vitro. A total 154 differential metabolites were identified after SSF, and a Spearman correlation study revealed a direct relationship between antioxidant activities and certain metabolites including phenolic compounds, oligopeptides, and free fatty acids etc. Among these metabolites, phenolic compounds produced by the shikimic acid pathway were diverse in variety and had the greatest multiple differences. The study discovered that a potential mechanism involving SSF with P. ostreatus mycelium increased the antioxidant activity of soybeans.

Energy-efficient and formaldehyde-free biomass-based boards derived from Pleurotus Ostreatus substrate waste: A novel sustainable utilization strategy

Yalan Yan,Yan Li,Bo Wang,Xin Zhang,Xu Zeng,Shuang Ding,Bo Ren,Xiaodong Yang

The preparation of formaldehyde-free biomass-based boards from waste mushroom substrate simultaneously tackles environmental pollution from landfill/incineration of mushroom waste and wood resource shortage, offering novel insights for ecological sustainable development. However, current formaldehyde-free biomass-based board production from laboratories suffers from extremely high hot-pressing energy consumption (hot-pressing time ≥ 2 h), conflicting with the energy-saving demands under the “Double Carbon” initiative. This study proposes a novel strategy using citric acid (CA)-epoxidized soybean oil (ESO) for synergistic pretreatment of Pleurotus ostreatus ( P. ostreatus ) substrate powder to prepare mushroom substrate-based boards (MSB) using hot-pressing technique. Both the mechanical properties and water resistance of the prepared MSBs are dramatically improved, and meet the standards for dry-condition furniture-grade and load-bearing particle-boards specified in GB/T 4897–2015, while reducing hot-pressing energy consumption by over 63 % compared with the P. ostreatus substrate-based boards without CA and ESO pretreatment. Further FT-IR spectroscopy measurement reveals that pretreatment agents enhance hot-pressing efficiency via esterification and amidation reactions with lignocellulose/protein in P. ostreatus substrates. The proposed strategy provides a technically and economically viable energy-saving solution for business application of MSBs.

The Interference of Mnsod3 Enhances the Tolerance of Pleurotus ostreatus Mycelia to Abiotic Stress by Reshaping the Cell Wall

Ludan Hou,Tonglou Li,Baosheng Zhang,Zehua Zhang,Bing Deng,Lijing Xu,Xueran Geng,Yanfen Cheng,Mingchang Chang,Junlong Meng

In recent years, the response mechanism ofPleurotus ostreatusto abiotic stress has received widespread attention. MnSOD is an important antioxidant enzyme that has been widely studied in animals and plants because of its functions. However, there is little research on the function and regulatory mechanism of MnSOD in the growth and development of edible fungi. This study investigated the role ofMnsod3in the growth and development ofP. ostreatus. The results showed that during the nutritional growth stage, heat stress can cause the cell wall of mycelia to shrink and the cells to exhibit cytoplasmic wall separation. RNA-seq revealed thatMnsod3interference is strongly correlated with increased transcript levels of cell wall synthase genes and with increased tolerance to cell wall disruptors. During the primordium formation stage, the mycelial cell wall also significantly wrinkled under cold and light stresses. RNAi ofMnsod3alleviated the cell wall wrinkling caused by cold and light stress, restored the smoothness of the cell walls, and increased mycelial tolerance to abiotic stress. This may be related to the slower formation rate of primordia, but the specific molecular mechanism still needs further research. and slowed the rate of primordium formation. In summary,Mnsod3plays an important role in the growth and development ofP. ostreatusunder abiotic stress and plays a critical regulatory role in cell wall remodeling under abiotic stress.

Oat protein isolate-Pleurotus ostreatus β-glucan conjugate nanoparticles bound to β-carotene effectively alleviate immunosuppression by regulating gut microbiota

Lei Zhong,Qiuhui Hu,Qiping Zhan,Mingwen Zhao,Liyan Zhao

Individuals with immune disorders cannot establish an adequate defense to pathogens, leading to gut microbiota dysbiosis. β-Carotene can regulate immune response, but its bioavailability in vivo is very low. Herein, we developed a glycosylated oat protein-based nanoparticle to improve the application of β-carotene for mitigating cyclophosphamide-induced immunosuppression and gut microbiota imbalance in mice. The results showed that the nanoparticles facilitated a conversion of β-carotene to retinol or retinyl palmitate into the systemic circulation, leading to an increased bioavailability of β-carotene. The encapsulated β-carotene bolstered humoral immunity by elevating immunoglobulin levels, augmenting splenic T lymphocyte subpopulations, and increasing splenic cytokine concentrations in immunosuppressed mice. This effect was accompanied by the alleviation of pathological features observed in the spleen. In addition, the encapsulated β-carotene restored the abnormal gut microbiota associated with immunosuppression, including Erysipelotrichaceae, Akkermansia, Bifidobacterium and Roseburia. This study suggested that nanoparticles loaded with β-carotene have great potential for therapeutic intervention in human immune disorders by specifically targeting the gut microbiota.
