BACKGROUND Insect glutathione S-transferases (GSTs) play a crucial role in insecticide detoxification. However, there remains a distinct lack of information regarding the role of GSTs in the detoxification of Tolfenpyrad (TFP) in insects. RESULTS Real-time quantitative PCR showed significant upregulation of PxGSTs after exposure to TFP for 6 h. An in vitro inhibition assay showed that TFP could inhibit PxGSTδ, PxGSTε and PxGSTσ, and the most pronounced inhibitory effect was on PxGSTσ. Metabolism assays displayed that PxGSTσ was superior to other test PxGSTs in metabolizing TFP. The molecular docking of TFP and PxGSTσ revealed that the H-bond provided by the sidechains of Tyr107 and Tyr162 were key to the detoxification of TFP by PxGSTσ. Further tests using mutant PxGSTσ proteins at the sites of Tyr107 (PxGSTσY107A) and Tyr162 (PxGSTσY162A) corroborated that the individual replacement of Tyr107 and Tyr162 could greatly weaken the binding and metabolic abilities to TFP. CONCLUSION Metabolic interactions between the Plutella xylostella (L.) GSTs (PxGSTs) and TFP were deciphered. This study illustrates the molecular metabolism mechanism of PxGSTσ towards TFP and provides theoretical underpinnings for the design and optimization of novel TFP-like insecticides. © 2020 Society of Chemical Industry

BACKGROUND Insecticide resistance continuously poses a threat to agricultural production. Chemosensory protein-mediated resistance is a new mechanism of insecticide resistance discovered in recent years. In-depth research on resistance mediated by chemosensory proteins (CSPs) provides new insight into aid insecticide resistance management. RESULTS Chemosensory protein 1 in Plutella xylostella ( PxCSP1 ) was overexpressed in the two indoxacarb-resistant field populations and PxCSP1 has a high affinity with indoxacarb. PxCSP1 was upregulated when exposed to indoxacarb and the knockdown of this gene elevated sensitivity to indoxacarb, which demonstrate that PxCSP1 is involved in the indoxacarb resistance. Considering that CSPs may confer resistance in insects via binding or sequestering, we explored the binding mechanism of indoxacarb in PxCSP1 -mediated resistance. Using molecular dynamics simulations and site-directed mutation, we found that indoxacarb forms a solid complex with PxCSP1 mainly through van der Waals interactions and electrostatic interactions. Between these, the electrostatic interaction provided by the Lys100 side chain in PxCSP1 , and especially the hydrogen bonding between the NZ atom and the O of the carbamoyl carbonyl group of indoxacarb, are the key factors for the high affinity of PxCSP1 to indoxacarb. CONCLUSIONS The overexpression of PxCPS1 and its high affinity to indoxacarb is partially responsible for indoxacarb resistance in P. xylostella . Modification of indoxacarb's carbamoyl group has the potential to alleviate indoxacarb resistance in P. xylostella . These findings will contribute to solving chemosensory protein-mediated indoxacarb resistance and provide a better understanding of the insecticide resistance mechanism. © 2023 Society of Chemical Industry.

Oviposition is an important reproductive behavior that is triggered by mating in insects, and biogenic amines might be involved in its regulation. The effects of biogenic amines on oviposition have only been studied in a few insect species, and the findings to date have not been conclusive. In addition, there are few studies on the effects of biogenic amines on oviposition of the diamondback moth, Plutella xylostella L. Here, we tested how mating and biogenic amines regulate oviposition of P. xylostella by injecting amines and amine receptor antagonists into virgin and mated females and counting the number of eggs laid afterward. Biogenic amines of octopamine and tyramine could induce virgin adults of P. xylostella to lay eggs, while dopamine and serotonin had no such effect on oviposition. Furthermore, the octopamine antagonists mianserin, epinastine, and phentolamine inhibited oviposition by mated females. The tyramine antagonist yohimbine, dopamine antagonist SCH23390, and serotonin antagonist ketanserin did not block oviposition by mated females, and octopamine and tyramine-inducing oviposition by virgin females could be inhibited by the octopamine antagonists mianserin and epinastine instead of the tyramine antagonist yohimbine. We conclude that octopamine and its receptors are involved in mating-triggered oviposition in P. xylostella, while tyramine acts as a subsidiary. Further, the inducing effect of tyramine on oviposition is achieved via octopamine receptors instead of tyramine receptors. This experiment is helpful to further understand the role of biogenic amines in mating regulation and to provide a new strategy for controlling P. xylostella.

BACKGROUND The diamondback moth, Plutella xylostella , has developed resistance to almost all insecticides used for its control. The ‘push–pull’ method has been shown as an effective control strategy to address this resistance challenge of P. xylostella . The key focus of the strategy is the identification of attractive or repellent volatile components. The aim of this study was to identify attractive volatile compounds released from host plants. Identified compounds were applied in the biological control of this pest. RESULTS Nine active compounds released into the headspace of seven cruciferous plant species were identified using gas chromatography–electroantennographic detection and gas chromatography–mass spectrometry. Electroantennographic detection-active compounds included five green leaf volatiles (hexanal, trans -2-hexen-1-ol, cis -3-hexen-1-ol, cis -3-hexenyl acetate, and 1-penten-3-ol), three isothiocyanates (isopropyl isothiocyanate, allyl isothiocyanate, and butyl isothiocyanate), and nonanal. Except for nonanal, all the identified green leaf volatiles and isothiocyanates elicited strong electrophysiological and behavioral responses in P. xylostella . The strongest attractive compounds, trans -2-hexen-1-ol and isopropyl isothiocyanate, were further evaluated in oviposition and field-trapping assays. Results showed that they both lured female moths to lay eggs, and were highly attractive to P. xylostella adults in field, especially when used in combination with yellow and green sticky boards. However, a blend of the two compounds showed no synergistic effect, but rather an antagonistic effect. CONCLUSIONS Green leaf volatiles and isothiocyanates were identified as key olfactory cues for host selection of P. xylostella . Trans -2- hexen-1-ol and isopropyl isothiocyanate were identified as candidate attractive compounds to serve in a ‘push–pull’ strategy for P. xylostella control. © 2023 Society of Chemical Industry.

The long-term and excessive use of insecticides has led to severe environmental problems and the evolution of insecticide resistance in insects. Carboxylesterases (CarEs) are important detoxification enzymes conferring insecticide resistance on insects. Herein, the detoxification process of Plutella xylostella (L.) carboxylesterase 6 (PxEst-6), one representative P. xylostella carboxylesterase, is investigated with cypermethrin, bifenthrin , cyfluthrin and λ-cyhalothrin. RT-qPCR shows that PxEst-6 is highly expressed in the midgut and cuticles of the third instar larvae. Exposure to pyrethroid insecticides resulted in PxEst-6 up-regulation in a short time. Metabolic assays indicate that PxEst-6 has the capacity to metabolize these pyrethroid insecticides. The combination of molecular docking, binding mode analyses and alanine mutations demonstrated that His451, Lys458 and Gln431 were key residues of PxEst-6 for metabolizing pyrethroids and the acetate groups derived from pyrethroids were key sites for being metabolized by PxEst-6. H451- and K458-derived hydrogen bond (H-bond) interactions with the pyrethroid acetate groups and the polar interactions with the pyrethroid acetate group provided by the Q431 sidechain were crucial to the pyrethroids’ metabolism by PxEst-6. Our study contributes to revealing the reasons for pyrethroid resistance in P. xylostella , and provides a fundamental basis for the development of novel pyrethroid insecticides.

The diamondback moth (DBM), Plutella xylostella (L.), is a globally destructive pest of cruciferous vegetables. Excessive use of synthetic pesticides to control this species results in negative effects on the environment, human health, and nontargeted organisms. The essential oils (EOs) derived from plants may be developed as effective alternatives to conventional pesticides. In this study, thirteen EOs were extracted by hydrodistillation, respectively. Their chemical compositions were identified by gas chromatography-mass spectrometry (GC–MS). Furthermore, the biological activities of EOs such as toxicity, antifeedant activity, and growth inhibition effect, toward DBM larvae were investigated. Against DBM second-instar larvae, the most toxic EO was Pelargonium graveolens (LC 50 = 0.36 μg/μl) after 72 hr of exposure, followed by Polygonum hydropiper (LC 50 = 0.53 μg/μl). The Ocimum basilicum EO exhibited the highest antifeedant effect to third-instar larvae at at all set concentrations. At 15 μg/μl, the EOs of Acorus calamus , O. basilicum , and P . graveolens completely inhibited the feeding activities of larvae (100%). The Ruta graveolens EO showed the lowest relative growth rate and the highest growth inhibition rate towards third-instar larvae at diverse concentrations. And the EOs of P. hydropiper , A. calamus , and O. basilicum showed promising growth inhibition activities. Overall, the five EOs ( P . graveolens , O. basilicum , R. graveolens , P. hydropiper , and A. calamus ) showed moderate to high bioactivity, whereas eight EOs were found to be less active against DBM larvae. These results indicate that the five tested EOs are promising to be developed as novel botanical insecticides to control DBM population.

The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan obligate pest of cruciferous crops. Currently, DBMs management is largely dependent on broad-spectrum synthetic pesticides, which has led to the adverse effects on the environment, human health, and nontargeted organisms. Thus, alternative control methods are needed. The essential oils (EOs) derived from bioactive species may be developed as effective alternatives for the management of DBMs that is safe and eco-sustainable than conventional pesticides. In this study, we investigated electrophysiological (EAG) and behavioral responses of DBM adults to the thirteen non-host EOs of Acorus calamus , Ageratum conyzoides , Artemisia argyi , Chenopodium ambrosioides , Cymbopogon citratus , Foeniculum vulgare , Ocimum basilicum , Pelargonium hortorum , Perilla frutescens , Pogostemon cablin , Polygonum hydropiper , Rosmarinus officinalis , and Ruta graveolens in order to contribute to the development of low environmental impact management alternatives. EAG recordings revealed all the tested EOs significantly elicited EAG responses of DBM females, and the males were sensitive to the eleven EOs except A. conyzoides and O. basilicum . Meanwhile, EAG recordings of DBMs exhibited dose-dependent responses to these tested EOs in most cases. In Y-tube olfactometer bioassays, the EOs of A. calamus , C. citratus , P. hortorum , P. hydropiper , and R. graveolen demonstrated prominent repellent effects on DBM females at concentrations of 10 μg/μL or higher. The results of oviposition bioassays found the number of eggs on the two plants with the five tested EOs were less than that on the two plants with paraffin oil. And the oviposition indexes of the plants that fixed with the EOs of P. hortorum , R. graveolen , A. calamus , P. hydropiper , C. citratus , and paraffin oil were –71.81%, –59.43%, –57.46%, –51.11%, –35.41%, and 0.09%, respectively. These findings indicate that the five EOs, especially P. hydropiper EO, provide active and safe tools for natural repellents to prevent and control DBMs infestations of cruciferous vegetables.

Simple SummaryPlutella xylostella(L.) (Lepidoptera: Plutellidae) is the most destructive pest of cruciferous crops around the world. The inappropriate use of insecticides to manage it has brought the development of resistance to several pesticide active ingredients. Keeping in view the resistance problem and long-term application, essential oils of plants may provide an environment-friendly solution to manage pests. Although the essential oil ofArtemisia lavandulaefoliaDC. was proven to exhibit promising insecticidal activities againstP. xylostella, which components have insecticidal activities are unknown. In the present study, four bioactive components were identified fromA. lavandulaefoliaessential oil by GC/MS, and their insecticidal activities againstP. xylostellawere also evaluated. Among them, (-)-4-terpineol was proven to be mostly responsible for its insecticidal activity againstP. xylostella. This effect was associated with decreased activity of GST, CAT, AChE and Na+/K+-ATPase. Overall, our findings indicated that (-)-4-terpineol may offer an alternative source for the control ofP. xylostella.AbstractPlutella xylostella(L.) is one of the most serious pests of cruciferous vegetables. Our previous work demonstrated that the essential oil ofArtemisia lavandulaefoliaDC. exhibits promising insecticidal activities againstP. xylostella. This study further characterizes the key components that are responsible for the insecticidal effect. In total, 47 compounds (96.52% of the total compounds) were identified from the total oil using GC-MS, and the major compounds were eucalyptol (21.57%), D(+)-camphor (17.33%), (-)-4-terpineol (9.96%) and caryophyllene oxide (10.96%). Among them, (-)-4-terpineol showed significantly larvicidal and fumigant activities againstP. xylostella. The LD50of (-)-4-terpineol was 43.15 mg/mL at 12 h and 31.22 mg/mL at 24 h for 3rd instar larvae, and the LC50for adults was 8.34 mg/mL at 12 h and 7.35 mg/mL at 24 h. In addition, the adults treated with (-)-4-terpineol showed varying degrees of inhibitory activity toward glutathione S-transferase, catalase, acetylcholinesterase and Na+/K+-ATPase at different post-treatment intervals and concentrations. The results indicate that (-)-4-terpineol has promising insecticidal activities againstP. xylostella, and it has good inhibitory effects on the four enzymes ofP. xylostellaadults. Keywords:Plutella xylostella;bioactive compounds;(-)-4-terpineol;insecticidal activities

BACKGROUND The emergence and increasing intensification of insect pest resistance greatly shortens the service life of commercial insecticides. Hence, the continuous development of novel insecticides is extremely necessary. Discovering new insecticide molecules based on natural products with insecticidal activity has been a popular technique and has made outstanding contributions in this field. RESULTS A series of betulin-cinnamic acid-related hybrid compounds was prepared by molecular hybridization through the bridge of acyloxy groups and their chemical structures were unambiguously confirmed. Against A. pisum , compounds 2e , 3a-3e , 3g , 3i , 3j , and 3m showed complete antifeedant activity at 2 mg/mL; compound 5b exhibited 2.7 times the aphicidal activity of the precursor betulin. Against P. xylostella , compound 2l displayed 3.9 times the insecticidal activity of betulin. Meanwhile, some interesting structure–activity relationships were obtained. Furthermore, compound 2l could destroy the ultrastructure of midgut cells and significantly inhibit the activity of α-amylase in P. xylostella larvae. Molecular docking indicated that the lupane skeleton and p -nitrocinnamoyloxy groups played a key role in the binding of compound 2l to α-amylase. CONCLUSION Compounds 5b and 2l could be further optimized structurally as the secondary lead compounds for the discovery of novel insecticide molecules. © 2025 Society of Chemical Industry.

The diamondback moth (DBM), Plutella xylostella (L.), is a major pest affecting cruciferous vegetables, and seriously affects the quality and yield of these vegetables. Diafenthiuron is a traditional thiourea-based insecticide , but it is rarely used to control pests on cruciferous vegetables due to its phytotoxicity on these vegetables under high temperature and light conditions. Thus, there is an ongoing need for more effective pesticides that can be used on cruciferous vegetables, possibly including new formulations of diafenthiuron. A new thiourea insecticide, methylthio-diafenthiuron, is intended to optimize the structure of diafenthiuron not only to preserve its insecticidal bioactivity but also to overcome its phytotoxicity to cruciferous vegetables, aiming to control insect pests on cruciferous vegetables. In this study, we compared the toxicity of methylthio-diafenthiuron to some frequently used insecticides on the third-instar larvae of DBM. The parental pupal duration was significantly longer under the treatment than in the control, but the pupal weight, fecundity, and hatching rate significantly decreased. By studying the changes in three detoxifying enzymes within 72 h after treatment with a sublethal concentration, the activity of CarE and ODM in the treatment group significantly increased at first and then decreased. In addition, methylthio-diafenthiuron clearly inhibited three kinds of ATPases in the DBM and significantly reduced the eclosion rate of the pupae . This research provides valuable information for the assessment and rational application of methylthio-diafenthiuron for the control of pests on cruciferous vegetables.

Insect antennae play a fundamental role in perceiving and recognizing a broad spectrum of conventional semiochemicals and host plant-derived odors. As such, genes that are tightly associated with the antennae are thought to have olfactory-related roles related to signal transduction mechanisms. Several mechanisms suggest that enzymatic inactivation could contribute to the signal termination process, such as odorant-degrading enzymes (ODEs). To date, a few ODEs have been identified and characterized in detail in insect herbivores, but little is known about aldehyde oxidases (AOXs); moreover, direct in vivo experimental evidence is needed. AOXs are a major family of metabolic enzymes that oxidize a variety of aromatic aldehydes, and they may also play a significant role in detoxification and degradation of environmental chemical cues. Here, we report on the identification and characterization of a novel cDNA encoding the putative odorant-degrading enzyme, PxylAOX3, from the antennae of the diamondback moth , (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae). The purified recombinant protein showed a wide-range of substrate zymography oxidizing both sex pheromone compounds as well as plant-derived aldehydes with distinct activities. Our data suggest PxylAOX3 might be involved in the degradation of many structurally diverse aldehyde odorants. Furthermore, PxylAOX3 could participate in olfactory neuron protection by inactivation of redundant odorants and xenobiotic detoxification, making it a potential target for pesticide development as well.
