Effect of Polygonatum cyrtonema Hua polysaccharides on gluten structure, in vitro digestion and shelf-life of fresh wet noodle

Siqing Xiong,Pengcheng Tao,Yuanguo Yu,Wenbing Wu,Yongxin Li,Gang Chen,Jinping Si,Huqing Yang

This study aimed to investigate the effects of raw Polygonatum cyrtonema Hua polysaccharides (RPCPs) and “zhi” P. cyrtonema Hua polysaccharides (ZPCPs) on the gluten structure, in vitro digestion, and shelf life of fresh wet noodles (FWN). The results demonstrated that incorporating PCPs improved the cooking and sensory qualities of FWN. Moreover, the shelf life of FWN was extended by 6 days with 1.5 % RPCPs (w/w) compared with the control FWN. Furthermore, incorporating 1.5 % ZPCPs led to a 1.2- and 0.2-fold increase in the disulfide bond and α-helix content, respectively, compared with the control FWN. This resulted in enhanced gluten structure, improved springiness and viscidity, and reduced cooking loss by 14.47 %–52.19 %. The scanning electron microscopy analysis revealed that the starch particles were entrapped by PCPs, leading to higher gelatinization temperature and lower setback value of FWN, thereby reducing the starch digestion ratio to 55.50 %. In summary, the findings suggested that FWN containing PCPs can extend shelf life, improve taste, and slow starch digestion staple.

Rapid on-site multi-indicator detection of Polygonatum cyrtonema Hua by handheld near-infrared spectroscopy with wavelength selection

Shuanglin Deng,Fang Wang,Meng Xiong,Bangxing Han,Hui Yan

The rapid, accurate, and on-site quantitative evaluation of the 3 components (total polysaccharide, total flavonoid, and total saponin) of Polygonatum cyrtonema Hua (PCH) is of great significance to ensure the value of its medicine and food, for this purpose, a handheld near-infrared (NIR) spectrometer was used. The raw NIR spectra were preprocessed by seven methods and their combinations, then three kinds of wavelength selection algorithms, the competitive adaptive reweighted sampling (CARS), random frog (RF) sampling, and Monte Carlo uninformative variable elimination (MCUVE) were applied, and the characteristic wavelengths for total polysaccharide, total saponin, and total flavonoid in PCH were obtained, respectively. Subsequently, the partial least squares (PLS) regression models were developed. The results showed that the combination of first derivative and standard normal variate was the optimal pretreatment method, and CARS had the highest performance of the wavelength selection for total polysaccharide and total flavonoid, while RF was the highest for total saponin. With these selected wavelengths, 3 PLS prediction models were built, and their performances were validated with unknown samples, the results showed that the determination coefficient of the prediction set (R 2 P ) of total polysaccharide, total saponin, and total flavonoid were 0.980, 0.948, and 0.942, respectively, and the corresponding root mean square error of prediction set (RMSEP) were 0.162, 0.142, and 0.052. Furthermore, the residual prediction deviations (RPDs) of the three components were all larger than 3, demonstrating the prediction performance of the models was high. Therefore, the three active functional components in PCH could be rapidly determined by a handheld NIR spectrometer, which would be beneficial for ensuring the quality of PCH.

Liposomes Enhance the Immunological Activity of Polygonatum Cyrtonema Hua Polysaccharides

Dong Liu,Tingting Hou,Chunye Geng,Lu Song,Xuefeng Hou,Yanjun Chen,Fang Wang,Wei Wang,Bangxing Han,Leilei Gao

Poor stability and difficult uptake of natural polysaccharides have been the main problems in their application. The purpose of this study was to optimize the preparation conditions of Polygonatum cyrtonema Hua polysaccharides liposomes (PCPL) and to investigate the immune enhancement activity of PCPL in vitro and in vivo, with a view to discovering new ways of natural polysaccharide application. The optimal preparation conditions of PCPL were as follows: the adding amount of Tween 80 of 0.5 %, the ultrasound time of 2 min and the ultrasound times of once. Under these conditions, the entrapment efficiency, drug loading rate and particle size of PCPL were 38.033 %±0.050, 2.172 %±0.003 and 146 nm, which indicated that PCPL with small particle size could be prepared by the reverse-phase evaporation method. Furthermore, PCPL promoted proliferation, phagocytosis, and secretion of nitric oxide and related cytokines in RAW264.7 cells. Moreover, PCPL improved spleen and thymus indices, increased the number or proportion of red blood cells, platelets, and lymphocytes in the blood, and ameliorated spleen and thymus atrophy in immunosuppressed mice. This study provides a new idea for applying Polygonatum cyrtonema Hua polysaccharides (PCP) and references for studying other polysaccharides.

Polygonatum cyrtonema Hua in Jiuhua Mountain: Structural Characterization and Biological Activity

Xueqing Geng,Xingchi Zhang,Yun Wang,Ting Yao,Zhuoting Gan

Polygonatum cyrtonemaHua in Jiuhua Mountain (JHPCH) is a nationally recognized geographically indicated medicinal plant known for its bioactive polysaccharides with diverse pharmacological activities. This study isolated a homogeneous polysaccharide JPCP-70-0 from JHPCH via hot water extraction, ethanol precipitation, and chromatographic purification. Structural characterization by GC-MS, FT-IR, and NMR identified it as a neutral fructan (2.88 × 104Da) with Glc/Fru (6.65%/93.35%), a backbone of →1)-β-D-Fruf-(2→, and side chains of →6)-β-D-Fruf-(2→). Using Caenorhabditis elegans as a model, we evaluated JPCP-70-0’s effects on nematode growth. Results showed concentration-dependent regulation: low concentrations promoted growth, locomotion, feeding, and reproduction, while high concentrations induced developmental inhibition and reduced activity. This work provides a theoretical basis for the rational development of JHPCH resources.

Antioxidant and antimicrobial potential of polysaccharides sequentially extracted from Polygonatum cyrtonema Hua

Ling Li,Kiran Thakur,Bu-Yan Liao,Jian-Guo Zhang,Zhao-Jun Wei

We examined the antioxidant and antimicrobial attributes of Polygonatum cyrtonema Hua polysaccharides (HBSS, CHSS, DASS, CASS). The UV spectra and FT-IR results showed the purity and acidic nature of polysaccharides with β -configurations. DASS (78.585% hydroxyl radical scavenging, 92.794% ABTS radical scavenging, 75.648% DPPH radical scavenging activity, 0.582 reducing power and 98.721% Fe 2+ chelation at 5   mg/mL) possessed the best antioxidant potential; while, HBSS (21.113% hydroxyl radical scavenging, 14.412% ABTS radical scavenging, 28.005% DPPH radical scavenging activity, 0.285 reducing power and 48.969% Fe 2+ chelation at 5   mg/mL) was observed with the lowest antioxidant action. The bacterial inhibition effect of four PCHPs on Gram-positive organisms ( B. subtilis and S. aureus with the maximum inhibitory diameters of 18.9 and 15.8   mm, respectively) was better than that of Gram-negative bacteria ( E. coli and S. typhimurium with the maximum inhibitory diameters of 10.9 and 8.4   mm, respectively). The bacterial sensitivity was observed in the following order: B. subtilis   >   S. aureus   >   E. coli   >   S. typhimurium . The trend of antibacterial action of PCHPs was in following order: DASS   >   CHSS   >   CASS   >   HBSS. DASS can be considered as natural anti-oxidant as well as spoilage inhibitors to enhance the nutritive and functional values of various foods.

Preparation, separation, and identification of antioxidant peptides from protein hydrolysate of Polygonatum cyrtonema Hua

Yanyan Zhang,Shengjun Ma,Haixia Li,Peizi Liu,Keyi Chen,Wei Cai

Research on Polygonatum cyrtonema Hua has focused extensively on its main active components, yet antioxidant peptides remained less explored. This study optimized the preparation process of antioxidant peptides from P. cyrtonema via protein hydrolysis using response surface methodology. The peptide mixture was separated into four fractions using ultrafiltration and G-25 gel chromatography, based on antioxidant capacity. Notably, the F2 exhibited significantly higher Fe 3 ⁺ reducing power (17.17%) and ABTS· radical scavenging rate (30.1%) compared to other fractions at the same concentration. Characterization by Nano-LC-Q Exactive Plus quadrupole Orbitrap MS revealed that F2 contained 11,704 peptides, markedly more than the endogenous (2,454) and crude protein peptides (2,334). Bioinformatics and molecular docking identified three peptides—QFLPF, FVPF, and FPNLF—as having the highest antioxidant potential. These peptides increased cell viability in oxidative damage-induced HepG2 cells by 6.39%, 14.51%, and 9.98%, respectively, compared to the model group. This study lays the groundwork for future mechanistic investigations into the antioxidant peptides of P. cyrtonema.

Structural elucidation and anti-nonalcoholic fatty liver disease activity of Polygonatum cyrtonema Hua polysaccharide

Wei Liu,Taili Shao,Lei Tian,Zhengrui Ren,Lan Gao,Zhiyan Tang,Zheng Fang,Pingchuan Yuan,Chunyan Liu,Jikun Li,Guodong Wang,Jun Han

The chemical structure and pharmacological activity of Polygonatum cyrtonema Hua polysaccharides have garnered significant attention in recent years. In this study, a homogeneous polysaccharide, PCP1, was extracted from P. cyrtonema Hua rhizomes and purified. Monosaccharide composition analysis showed that PCP1 is primarily composed of fructose, glucose, and mannose. Chemical structure analysis showed that the main chain of PCP1 is composed mainly of →1)-β-D-Fruf-(2→ and →1,6)-β-D-Fruf-(2→, with small amounts of →6)-α-D-Glcp-(1→, →4)-β-D-Manp-(1→, and β-D-Glcp-(1→. The side chain is β-D-Fruf-(2→ linked at C-6 of →1,6)-β-D-Fruf-(2→. In vivo experiments showed that PCP1 mitigates liver pathological damage, improves abnormal lipid metabolism and oxidative stress, promotes the production of short-chain fatty acids, and balances the composition of the intestinal microbiota in non-alcoholic fatty liver disease (NAFLD) mice. Thus, PCP1 can be used as a natural ingredient in functional foods for the treatment of NAFLD.

Zhao Yuanyuan,Wang Zhen,Fu Ranze,Xie Ruonan,Wang Bin,Li Qinglin

Introduction: Polygonatum cyrtonema Hua. (PC) is a traditional Chinese herb with a history of use in both food and medicine. For clinical use, processed PC pieces are most commonly used, while present research has focused on crude PC polysaccharides (PCPs).Methods: In this study, a new polysaccharide, PCP-F1, with a molecular weight of 37.46 kDa, was separated from four-time processed PCPs by column chromatography and evaluated by antioxidant activity. It was composed of glucose, mannose, galactose, rhamnose, and galacturonic acid with a molar ratio of 3.5: 2.5: 1.3: 1.8: 0.8.Results and Discussion: The methylation analysis and two-dimensional NMR measurement revealed that the configuration of PCP-F1 contained nine residues in the primary structural unit by the chain of →3)-α-D-Glcp, →2)-α-D-Glcp (6→, →1)-ꞵ-D-Glcp (2→, →2)-α-D-GalAp (3,4→, →1) -ꞵ-D-Manp (3→, →2)-α-D-Glcp (3→, branched for →3)-α-D-Glcp, →2)-ꞵ-D-Galp (4→, →1)-ꞵ-D-Glcp (2→, →2,4)-α-D-Manp (6→, →3)-α-L-Rhap (4→. Radical scavenging assays indicated that PCP-F1 could scavenge radicals with a high scavenging rate, suggesting PCP-F1 possesses good antioxidant activity. The study confirms the importance of processed PC and offers the potential for exploiting it as a functional food.

The Functional Components of By-Product Resources from the Aerial Parts of Polygonatum cyrtonema Hua

Jiabei Xia,Xingyu Mei,Huan Cheng,Shiguo Chen,Xingqian Ye,Jianle Chen

The rhizome ofPolygonatum, which has been consumed in China for nearly 2000 years, is rich in oligosaccharides, polysaccharides, saponins, and alkaloids and has various effects such as lowering blood sugar and anti-aging. Its aerial parts are often discarded as by-products and have been rarely studied; moreover, the nutritional composition and carbohydrate composition of these parts are not clear. In this study, the nutritional composition, amino acid evaluation, and the total phenolics content of the leaves, stems, fruits, and flowers ofPolygonatum cyrtonemaHua were analyzed, and the composition and content of carbohydrates were determined by high-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC–PAD). The results suggested that the leaves and flowers ofPolygonatum cyrtonemaHua (PC) contained 1.12–1.37% phenolic substances. The proteins of the aerial parts had high nutritional values, and the total amino acid content of the leaves and flowers, with the total leaf amino acids amounting to 11.77%, were followed by the flowers at 11.65%. The sugars in the flowers were mainly free monosaccharides at 10.38%, and the fruits were dominated by 9.44% fructo-oligosaccharides. This study provided new evidence for the utilization of the aerial parts ofPolygonatum. Keywords:Polygonatum cyrtonemaHua;nutrient composition;amino acid evaluation;composition of carbohydrate

Polygonatum cyrtonema Hua polysaccharide alleviates ulcerative colitis via gut microbiota-independent modulation of inflammatory immune response

Jin-Yang Shi,Yong-Jian Wang,Qian-Wen Bao,Ya-Min Qin,Pei-Pei Li,Qiao-Qiao Wu,Cheng-Kai Xia,De-Ling Wu,Song-Zi Xie

Polygonatum cyrtonema polysaccharides (PCP) exhibit ameliorative effects on colitis. However, whether the protective effects of PCP depend on the gut microbiota and how PCP regulates intestinal immune responses to alleviate colitis remain unclear. Therefore, this study investigated the effect of PCP against colitis focusing on the regulation of intestinal immune response. The PCP structure was reclassified as fructan. PCP treatment significantly reduced the symptoms of colitis. PCP restored IgA, ZO-1, Occludin, and MUC2 expression to enhance intestinal barrier function. Oral PCP administration markedly inhibited excessive inflammation-mediated immune response by modulating inflammatory cytokines secretion and Th17/Tregs cell balance and restored gut microbial composition. Interestingly, PCP still had a significant ameliorating effect on intestinal inflammation in colitis mice with gut microbial depletion by antibiotics. In the Caco-2/RAW264.7 co-culture inflammation model, PCP treatment improved the intestinal epithelial barrier function by regulating the inflammatory immune response through signal transduction pathways. Overall, these findings suggested that the alleviating effects of PCP on colitis are independent of gut microbiota, and that PCP can directly modulate the inflammatory immune response and intestinal barrier function, which in turn regulates gut microbiota. These findings will provide new insights into the action mechanism of natural polysaccharides in relieving colitis.

Inhibitory effect and mechanism of purified saponin from Polygonatum cyrtonema Hua on human ovarian cancer cell A2780

Qile Xia,Beiqiong Hao,Chenxing Liu,Yan Cao

Polygonatum cyrtonema Hua (PcH) is a traditional medicinal and edible homologous plant. Here, the anticancer activity of different purified PcH saponins (PcHs) on the ovarian cancer (OC) cell A2780 were investigated. The PcHs were purified by AB-8. Their effects on the apoptosis, migration and invasion of A2780 were explored by CCK8, wound healing assay, Hoechst 33258 staining, Annexin V-FITC/PI and intracellular reactive oxygen species (ROS) measurement. Moreover, western blot and realtime-PCR confirmation were performed for the associated protein and mRNA targets. The results showed that the content of PcHs increased from 0.31 ± 0.02 mg/mL to 0.79 ± 0.02 mg/mL after purification. The inhibition rate of purified PcHs on A2780 could reach 66.76 ± 0.56%. Specifically, it promoted cell apoptosis, disturbed the cell cycle, increased the ROS levels, regulated the expression levels of pro-apoptotic and anti-apoptotic proteins. PcHs can also regulate the expression level of proteins related to the invasion and migration characteristics of A2780, the mRNA expression level related to the P13K-Akt signal transduction process, and the extracellular matrix-receptor interaction pathway, as well as drug resistance. Overall, this study provides a new idea for the treatment of OC.

Zein/chitosan nanocomplexes loaded with polyphenols from Polygonatum cyrtonema Hua improve Pickering emulsion stability and β-carotene protection

Wang-Wei Zhang,Rui-Xi Zhang,Xiu-Xiu Zhang,Zhi-Jing Ni,Kiran Thakur,Jian-Guo Zhang,Zhao-Jun Wei

This study successfully developed a ternary ZCP-PCPP nanocomposite using zein (ZN), chitosan (CS), and the polyphenols extracted from Polygonatum cyrtonema Hua (PCPP) to stabilize Pickering emulsions. The optimal ZN-CS was 2:1, which resulted in the ZCP-PCPP nanocomposite with the strongest encapsulation ability for PCPP, uniform particle size, and strong structural stability. Infrared spectroscopy, fluorescence spectroscopy, and X-ray diffraction confirmed that ZN, CS, and PCPP interacted mainly through hydrogen bonding and hydrophobic forces. Stability analyses demonstrated that ZCP-PCPP markedly enhanced the physical stability and oxidative resistance of the emulsions. Furthermore, ZCP-PCPP nanoparticles at 3–5% (w/v) effectively protected β-carotene against thermal and ultraviolet degradation. These findings highlight that combining natural polymers with bioactive polyphenols provides an efficient strategy for producing stable Pickering emulsions with strong antioxidant protection potential.
